Structures of Dotorioside I and II Obtained from the Fruits of Quercus acutissima $C_{ARRUTHERS}$ (도토리에서 분리한 Dotorioside I, II의 구조)

  • From the methanolic extractive of the fruits of Quercus acutissima $C_{ARRUTHERS}$(Fagaceae) a mixture(QC-A saponin) of two ester glycosides, which were named as dotorioside I(3) and ll(4), was separated by silica gel column chromatography and HPLC. The structures of these two glycosides including their genuine aglycones(1,2) were elucidated as 1: $2{\alpha}$, $3{\beta}$, $19{\alpha}$, 23-tetrahydroxyolean-12-en-28-oic acid, 2: $2{\alpha}$, $3{\beta}$, $19{\alpha}$, 23-tetrahydroxyurs-12-en-28-oci acid, 3: 28-O-${\beta}$-D-glucopyranosyl ester of 1, 4: 28-O-${\beta}$-D-glucopyranosyl ester of 2, respectively, on the basis of chemical and spectral evidence.

  • The catalytic oxidation of volatile organic compounds (methanol. acetaldehyde) has been characterized using the copper phthalocyanine catalyst in a fixed bed flow reactor under atmospheric pressure. The catalytic activity for pretreatment conditions was examined by this reaction system. The catalytic activity was ordered as follows: metal free-PC<Cu ($\alpha$)-PC<Cu ($\beta$)-PC The formaldehyde, carbon monoxide as a partial oxidation product of methanol and acetaldehyde over Cu ($\alpha$)-PC catalyst were detected and the conversions of methanol and acetaldehyde were accomplished above 95% over Cu ($\alpha$) -PC, Cu ($\beta$) - PC catalyst at 35$0^{\circ}C$. The pretreated metal free -PC, Cu($\alpha$)-PC, Cu($\beta$)-PC catalysts have been characterised by TGA, EA and XRD analysis. The catalytic activity pretreated with air and $CH_3$OH mixture (P-4) or air only (P-5) was very excellent. XRD and EA results showed that Cu($\alpha$)-PC, Cu($\beta$)-PC were destroyed an(1 new metal oxide such as CuO were formed.

Mechanical Properties of $\beta$-Sialon Prepared from Wando Pyrophyllite (완도납석으로부터 제조된$\beta$-Sialon의 기계적 성질)

  • $\beta$-Sialon powders were prepared by the reduction-nitridation reaction from the mixture of Wando-pyrophyllite and graphite at 135$0^{\circ}C$ in $N_2-H_2$ atmosphere. The $\beta$-Sialon powders were sintered at 175$0^{\circ}C$ by the pressureless sintering for 90min and the hot-pressing for 60 min respectively. All the sintered bodies showed their relative densities higher than 94% The values of M.O.R fracture toughness(KIC) and hardness showed 32.9kpsi 2.9MN/$m^{1.5}$ , 12.1GN/$m^2$ for the pressure-less sitnered bodies and 48.6kpsi, 4.6 MN/4m^{1.5}$ 15.3GN/$m^2$ for the hot-pressed bodies respectively.

  • Further investigation of Lycium chinense fruits gave a mixture of (6'-O-palmitoyl)- and (6'-O­stearoyl)-$\beta$-sitosterol-3-O-$\beta$-D-glucopyranoside (1) and two glycolipids, 1-O-(9Z, 12Z, 15Z-octa­decatrienoyl)-2-O-(9Z, 12Z, 15Z-octadecatrienoyl)-3-O-$\beta$-D-galactopyranosyl glycerol (2) and 1-O-(9Z, 12Z-octadecadienoyl)-2-O-(9Z, 12Z, 15Z-octadecatrienoyl)-3-O-$\beta$-D-galactopyranosyl glycerol (3). These compounds were newly isolated as constituents of L. chinense.

Comparison of Saccharina japonica-Undaria pinnatifida Mixture and Minoxidil on Hair Growth Promoting Effect in Mice

  • Background Algae have traditionally been used for promotion of hair growth. Use of hair regrowth drugs, such as minoxidil, is limited due to side effects. The aim of this study was to examine a mixture of Saccharina japonica and Undaria pinnatifida (L-U mixture) on hair growth and to compare the promoting effect of hair growth by a 3% minoxidil and a L-U mixture. Methods To evaluate the hair growth-promoting activity, saline, 50% ethanol, 3% minoxidil, and the L-U mixture were applied 2 times a day for a total of 14 days on the dorsal skin of C57BL/6 mice after depilation. Analysis was determined by using a high-resolution hair analysis system, real-time polymerase chain reaction, and H&E staining. Results On day 14, the hair growth effect of the L-U mixture was the same as that of the 3% minoxidil treatment. The L-U mixture significantly (P<0.05) stimulated hair growth-promoting genes, as vascular endothelial growth factor (VEGF) and insulin-like growth factor -1. Increase of VEGF was observed in the L-U mixture group compared with minoxidil and the negative control. In contrast, the L-U mixture suppressed the expression of transforming growth factor-${\beta}1$, which is the hair loss-related gene. In histological examination in the L-U mixture and minoxidil groups, the induction of an anagen stage of hair follicles was faster than that of control groups. Conclusions This study provides evidence that the L-U mixture can promote hair growth in mice, similar to the effect from minoxidil, and suggests that there is potential application for hair loss treatments.

The High Production of Cellulolytic Enzymes using Cellulosic Wastes by a Fungus, strain FJ1. (섬유소폐기물을 이용한 사상균 FJ1의 섬유소 분해효소의 고생산)

  • A filamentous microorganism, strain FJ1, was isolated from completely rotten wood for the production of cellulolytic enzymes. For the production of the enzymes, cellulolsic wastes were used as carbon sources of strain FJ1 and rice straw showed higher enzyme activities than sawdust and pulp. The activities of CMCase, xylanase, $\beta$-glucosidase, and avicelase were 2.95, 5.89, 0.45, and 0.12 unit/ml by use of rice straw, respectively. To enhance production of the enzymes, the mixture substrate of rice straw and cellulosic materials were investigated as carbon sources. The highest activities of CMCase, $\beta$-glucosidase, and avicelase were found in the mixture of rice straw (0.5%, w/v) and avicel (0.5%, w/v), and the highest xylanase was obtained at the mixture ratio of 0.71%(w/v) and 0.29%(w/v). Addition of 0.1%(w/v) peptone showed enhanced production of the cellulolytic enzymes in which the activities of CMCase, xylanase, $\beta$-glucosidase, and avicelase were 19.23, 27.18, 1.28, and 0.53 unit/ml, respectively. The production of the enzymes using rice straw was efficiently induced in the presence of avicel and pulp containing cellulose. In particular, a medium composed of rice straw (0.5%, w/v) and pulp (0.5%, w/v) yielded larger cellulolytic enzymes: CMCase 24.3 unit/ml, xylanase 38.7 unit/ml, $\beta$-glucosidase 1.5 unit/ml, and avicelase 0.6 unit/ml. The filamentous microorganism, strain FJ1 utilized various cellulosic wastes as carbon sources and will be expected as a favorable candidate for biological saccharification of cellulosic wastes.

  • Egasyn is accessory protein of ${\beta}$-glucuronidase(${\beta}$-G) in the liver microsomes. Liver microsomal ${\beta}$-G is stabilized within the luminal site of the microsomal vesicles by complexation with egasyn which is one of carboxylesterase isozymes. We investigated the effects of organophosphorus compounds(OPs) such as insecticides on the dissociation of egasyn-${\beta}$-glucuronidase(EG) complex. The EG complex was easily dissociated by administration of OPs, i.e., Fenitrothion, EPN, Phenthionate, and bis-p-nitrophenyl phosphate(BNPP), and resulting ${\beta}$-G dissociated was released into blood, leading to the rapid and transient increase of plasma ${\beta}$-G level with a concomitant decrease of liver microsomal ${\beta}$-G level. In a case of phenthionate treatment, less increase in plasma ${\beta}$-G level was observed, as compared with those of other OPs. This may be explained by a fact that phenthionate was easily hydrolyzed by carboxylesterase. Similarly, carbamate insecticides such as Carbaryl caused rapid increase of plasma ${\beta}$-G level. In contrast, no significant increase of plasma ${\beta}$-G level was observed when pyrethroid insecticides were administered to rats. This is due to a fact that pyrethroids such as Phenthrin and Allethrin were easily hydrolyzed by A-esterase as well as carboxylesterase. On the other hand, addition of OPs to the incubation mixture containing liver microsomes caused the release of ${\beta}$-G from microsomes to the medium. From these in vivo and in vitro data, it is concluded that increase of the plasma ${\beta}$-G level after OPs administration is much more sensitive biomarker than cholinesterase inhibition to acute intoxication of OPs and carbamates.

  • Three sesquiterpene lactone compounds, two novel(1.betha.,3.betha.-dihydroxy-6.betha.,11.betha.,4.alpha.,5.alpha.,7.alpha.H -eudesm-12, 6-olide-1-O-.betha.-D-glucopyranoside, 1.betha.,3.betha.-dihydroxy-6.betha.,11.betha.,4.alpha.,5.alpha.,7.alpha.H-eudes m-12,6-olide-1-O-.betha.-D-glucopyranoside) and 1.betha.,3.betha.-dihydroxy-6.betha.,11.betha.,4.alpha.,5.alpha., 7.alpha.H-eudesm-12,6-olide were isolated from the aqueous fraction of MeOH extract of the roots from Taraxacum hallaisanensis (Compositae) employing Amberlite XAD-2, ODS-gel, silica gel and Sephadex LH-20 column chromatographics. Another known compound, (-)-epicatechin, was isolated from the aqueous fraction of the MeOH extract. The total MeOH extract also contained phytosterol and a mixture of .betha.-amyrin acetate, .alpha.-amyrin acetate and lupeol acetate. Structures of isolated compounds were elucidated by spectroscopic parameters of IR, Mass, /sup 13/C-NMR, /sup 1/H-NMR, /sup 1/H-/sup 1/H COSY, /sup 13/C-/sup 1/H COSY and HMBC.

  • The medium additives such as fatty acid, lipid, mannose, folic acid, $CaCl_2$ were examined to enhance recombinant ${\beta}-galactosidase\;({\beta}-gal)$ production in T-flask and air-lift bioreactor. The addition of each component. such as cholesterol, tocopherol, tricaprylin, mannose and folic acid at AcNPV infection of Tn5B1-4 cells enhanced ${\beta}-gal$ production, whereas the addition of $CaCl_2$ did not increase ${\beta}-gal$ production. The recombinant ${\beta}-gal$ production using the infection medium supplemented with a mixture of 0.34 mM cholesterol, 2.2 mM mannose and 0.045 M folic acid was enhanced 2 fold in an air-lift bioreactor, compared to the basal medium.

  • An antifungal mixture of six members (component A to F), KRF-001 produced by Bacillzts subtilis subsp. krictiensis was isolated from the fermentation broth. Molecular weight of component A to F was determined by FAB-MS to be 1042, 1056, 1056, 1070, 1070 and 1084 respectively. Various instrumental analyses (amino acid analysis, GC-MS, $^1H-NMR, ^1HH$ COSY NMR) revealed that the mixture was a homologous cyclic peptide composed of each one mole of glutamine, proline, tyrosine, serine, unusual $\beta$-amino acid and three moles of asparagine. The structural differences of component A to F were found in carbon number and terminal structure of the unusual $\beta$-amino acid. After determination of the sequence and stereochemistry of those amino acids, the tentative structure of KRF-001 was determined.
