Arabidopsis thaliana as Bioindicator of Fungal VOCs in Indoor Air

  • In this paper, we demonstrate the ability of Arabidopsis thaliana to detect different mixtures of volatile organic compounds (VOCs) emitted by the common indoor fungus, Aspergillus versicolor, and demonstrate the potential usage of the plant as a bioindicator to monitor fungal VOCs in indoor air. We evaluated the volatile production of Aspergillus versicolor strains SRRC 108 (NRRL 3449) and SRRC 2559 (ATCC 32662) grown on nutrient rich fungal medium, and grown under conditions to mimic the substrate encountered in the built environment where fungi would typically grow indoors (moist wallboard and ceiling tiles). Using headspace solid phase microextraction/gas chromatography-mass spectrometry, we analyzed VOC profiles of the two strains. The most abundant compound produced by both strains on all three media was 1-octen-3-ol. Strain SRRC 2559 made several terpenes not detected from strain SRRC 108. Using a split-plate bioassay, we grew Arabidopsis thaliana in a shared atmosphere with VOCs from the two strains of Aspergillus versicolor grown on yeast extract sucrose medium. The VOCs emitted by SRRC 2559 had an adverse impact on seed germination and plant growth. Chemical standards of individual VOCs from the Aspergillus versicolor mixture (2-methyl-1-butanol, 3-methyl-1-butanol, 1-octen-3-ol, limonene, and ${\beta}-farnesene$), and ${\beta}-caryophyllene$ were tested one by one in seed germination and vegetative plant growth assays. The most inhibitory compound to both seed germination and plant growth was 1-octen-3-ol. Our data suggest that Arabidopsis is a useful model for monitoring indoor air quality as it is sensitive to naturally emitted fungal volatile mixtures as well as to chemical standards of individual compounds, and it exhibits relatively quick concentration- and duration-dependent responses.

Changes in terpenes of three kinds of pine needles during litter decomposition

  • This study was designed to evaluate changes in the terpene composition of 3 types of pines (Pinus densiflora, Pinus thunbergii and Pinus rigida), while decomposing their leaf litter. Needle litters were placed at two different organic layer depths, one on the surface and the other beneath the litter layer. Changes in the terpene composition of this litter were detected using a gas chromatograph-mass spectrometer. Among the monoterpenes acquired from the fresh needles of P. densiflora and P. rigida, $\alpha$-pinene (12.05% and 19.87%, respectively) was the major one, followed by $\beta$-pinene (2.90% and 14.07%). However, from the needles of P. thunbergii, $\beta$-pinene (20.77%) was the major one, followed by $\alpha$-pinene (10.79%). Among the sesquiterpenes detected in P. densiflora, trans-caryophyllene (3.12%) was the highest composition compound, whereas germacrene-D (6.09%) for P. thunbergii and 1,6-cyclodecadiene (7.41%) and endo-1-bourbonanol (7.41%) for P. rigida were the highest content compounds. However, the total amounts of terpenes decreased sharply by 40-85.4% in all three types of pine needle after 90-120 days of the experiment. The concentration of each terpene differed during decomposition, and the majority of compounds disappeared from beneath the litter layer. It was determined that three types of reducing patterns of each compound appeared on the rate of loss of concentration during decomposition; one pattern decreasing sharply during the initial period, another pattern steadily or slowly decreasing, and a newly detected pattern at low concentration occurring during decomposition.

Components of Pine Needles Extract and Functionality of the Dyed Fabrics (솔잎 추출물의 성분 분석 및 염색물의 건강안전 기능성 평가)

  • The pine needles can be used for four seasons in normal living and it can be taken friendly everywhere as it is distributed over 50% in Korea. The pine needles consist of vitamins, protein, minerals, essential oil and enzyme related to antimicrobial activity. It has effect like high blood pressure, neuralgia and hanged over by terpene, glucokinin, rutin, apigenic acid and tannin. Also the extract of them can be used for dyeing of fabrics. However, the extract components and effects of them are not well known yet. Therefore, the purpose of this study was to investigate the volatile components of the pine needles extract and functionality. The pine needles extract was dyed into various fabrics(nylon, silk, wool and soybean) and mordanted with Al, Cu, Cr, Fe and Sn. The extracted aroma compounds were compared by gas chromatography-mass spectrometry. The major volatile compounds of pine needles verified by using SPME were alpha-pinene, beta-pinene, beta-phellandrene, caryophyllene, ethanon, benzen. A total of 15 compounds were identified by using the SPME fibers. In the UV-visible spectra, the maximum absorption of wavelength of the pine needles ethanol extract appeared at 460, 630nm for chlorophyll component and at 237, 281nm for tannin component with the pine needles distilled water extract. Most of sample showed high antibacterial effect in none mordant but wool fabric showed high antibacterial effect in mordants. The result of UV block test showed a superior ability of blocking ultraviolet ray infiltration in all sample.

한우 수정란 생산에 있어서 체외성숙의 생물학적 요인 III. 소 난포란의 체외성숙에 있어서 단백질 변화

  • 체외에서 한우 난포란의 감수분열과 배발달 능력의 획득에는 단백질 합성이 수반되어야 한다. 그러나 이러한 변화와 관련된 연구 보고는 거의 전무한 실정이다. 본 실험은 난자의 핵성숙과 관련된 세포질내 단백질 변화를 파악하기 위하여, 체외성숙 시간에 다른 배발달율과 세포질내 단백질을 비교하여 배발달능력 획득과 관련 있는 단백질을 규명하고자 실시하였다. 한우 난소에서 2-8mm의 가시난포로부터 난포란을 회수하였다. 회수된 난포란은 10% FBS와 호르몬이 첨가된 TCM199 용액에서 18시간 또는 24시간 체외성숙을 실시하였다. 난자 세포질내 단백질 변화는 2D gel electrophoresis를 이용하였고, 유의적인 변화를 나타낸 spot은 peptide mass fingerprinting을 통하여 단백질 동정을 실시하였다. 체외수정은 fer-TALP 용액을, 체외배양은 CR1aa 용액을 배양 3일째까지는 0.3% BSA, 그 이후에는 10% FBS와 난관상피세포를 첨가하여 사용하였다. 통계분석은 t-test를 이용하였다. 난자의 세포질에 대한 이차원전기영동 결과 29개의 단백질 spot들을 확인하였다. 한편 체외성숙 18시간째에 PB가 출현된 난자는 PB가 출현되지 않은 난자에 비하여 15개의 spot에서 유의적인 변화를 나타냈다. 이들 중 4개의 단백질 spot은 낮았고, 11개 spot의 수준은 높은 경향이었다. 체외성숙 18시간째와 24시간째의 배발달율을 조사한 결과 18시간째에서 유의적으로 높은 배반포 발달율을 나타냈다. 그리고 체외성숙 18시간과 24시간째 난자의 세포질내 단백질 spot들의 변화를 비교한 결과 PB가 출현된 난자 세포질에서 단백질의 변화와 유사한 경향이었다. 그러나 2개의 단백질 spot은 상반된 경향을 나타냈다. 따라서 본 실험에서 난자의 핵성숙과 관련 있는 15개의 spot을 확인하였고, 이들 단백질 spot중에서 2개가 배발달 능력과 관련이 있을 것으로 사료된다.보다는 육질등급에 많은 영향을 받는 것으로 판단된다. 한편 육질 1등급에서 배발달율이 낮은 이유는 육질 향상을 목적으로 암소를 비육 하는 경우 발생하는 번식장애와 밀접한 관계가 있는 것으로 사료된다.각각 가장 높았다. 배양 8일째 배반포의 세포수에 있어서 총세포수와 TE 세포수는 차이가 없었으나, ICM 세포수가 l0mg 첨가군에서 가장 높았다. 본 실험 결과에서 체외성숙 배지에 NEAA와 EAA 첨가가 배발달율에는 효과가 없었지만, 첨가농도의 증가에 따라 ICM 세포수가 증가하였다. 한편 체외성숙 배지에 LAH 첨가는 첨가 농도가 높을수록 배발달율은 낮았지만 ICM 세포수는 증가하였다.에 Csk가 관여하고 있음을 알 수 있다. 결론적으로 성적 성숙에 따른 생쥐 정소 내 Src-Csk loop의 발현과 Src kinase 활성의 변동은 정소 내 간충조직, 세정관 상피의 증식 및 기능적 분화 과정을 매개하는 생리적 활성분자 수용체 하위의 신호전달 과정에 Src-Csk loop에 의한 조절가능성을 확인할 수 있었다.rugrene의 향기성분이 주요 성분군으로 확인되었다. 2. 생강나무에서 생강의 향기를 발산하는 성분으로는 $\beta$-myrcene, o-terpinolene, phellandrone, ι-limonene, $\beta$-eudesmol, $\delta$-cadinone, elemol, trans-caryophyllene으로 동정되었으며 그 중에서도 phellandrene, $\beta$-eudesmol이 주된 역할을 하는 성분으로 확인하였다. 유의적인 관련성이 나타났고, 복부 비만의 지표인 허리엉덩이둘레비는 GPT, alkaline phosphatase, 공복시 혈당 및 MCV 등 다양한 건강지표와 관련성을 나타내어 향후 비만에 있어 다양한 혈액 성상의 변화 및 역할규명에 대한 연구가 이루어져야 할 것으로 본다.hat

Anti-Porcine Epidemic Diarrhea Virus (PEDV) Activity and Antimicrobial Activities of Artemisia dubia Essential Oil (참쑥(Artemisia dubia) 오일의 돼지 유행성 설사 바이러스(Porcine Epidemic Diarrhea Virus)에 대한 항바이러스 항균활성)

  • The chemical composition, anti-porcine epidemic diarrhea virus (PEDV) activity and antimicrobial activity of Artemisia dubia essential oil were evaluated in this study. Fifty eight compounds from A. dubia essential oil were identified through analysis by gas chromatography-mass spectrometry (GC-MS). The major constituents of the oil were camphor (17.18 %), germacrene-D (15.70%), trans (${\beta}-$) racaryophyllene (6.79%), ene thujones (6.57%), 1, 8-cineole (5.94%) and camphene (5.08%). The essential oil was evaluated for antiviral activity against PEDV in Vero cells using a cytopathic effect (CPE) reduction method. The oils actively inhibited PEDV replication with a 50% inhibitory concentration ($IC_{50}$) of 43.7 ${\mu}^3/mL$. The 50% cytotoxicity concentration ($CC_{50}$) of the oils was over 100 ${\mu}/mL$ and the derived therapeutic index was >2.3. Similar analysis of the ribavirin revealed that they have a relatively weaker efficacy when compared to the oils. The antimicrobial activity of the essential oil against 5 microorganisms was evaluated by the disc diffusion method. The essential oil exhibited antimicrobial activity against 5 tested microorganisms with a clear zone of 8-22 mm. Among the tested microorganisms, Streptococcus pyogenes was the most sensitive and Candida albicans the least. Therefore, in can be concluded that essential oils of A. dubia may have interesting applications for microbial control or the control of PEDV-derived diseases.

Modulation of the Bacterial Mutagenicity for food-borne Mutagens by Hexane Fraction from Saururus chinesis (Lour.) Bail (삼백초 Hexane 분획물의 Heterocyclic Amine 돌연변이성 조정효과)

  • Antimutagenic activity of Saururus chinesis (Lour.) Bail was investigated for food-borne mutagens using S. typhimurium TA98. Methanol extract from Saururus Chinesis (Lour.) Bail was fractionated into hexane, chloroform, ethylacetate and butanol fractions, followed by determination of antimutagenic activity for food-borne mutagenic heterogenic amines (HCA). The hexane fraction exhibited a strong antimutagenic activity for 2-amino-3-methylimidazo[4,5-f]quinoline (IQ), 2-amino-3,8-dimethylimidazo[4,5-f] quinoxaline (MeIQ), 2-amino-3,4-dimethyl-3H-imidazo[4,5-f]quinoline (MeIQx), 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP), 3-amino-1-methyl-5H-pyroid[4,3-b]indole acetate (Trp-2-A); however its fraction rather enhanced the bacterial mutagenicity of 2-amino-3,4,8-trimethyl-3H-imidazo[4,5-f]quinozaline (4,8-diMeIQx) and 2-amino-3,7,8-trimethyl-3H-imidazo[4,5-f]quinoxline (7,8-diMeIQx). Active principle in the fraction was found to be two major compounds (${\gamma}$-crene B and epi-bicyclosesquiphellandrane) and 6 minor compounds (${\delta}$-caryophyllene, ${\gamma}$-elemene, ${\beta}$-cabebene, ${\delta}$-cadinene, ${\delta}$-selinene, and patchoulene). Modulation effect for the mutagenic activity of the food-borne mutagenic HCA by the fraction might be derived from a cumulative effect of each individual compounds. Hence, this hexane fraction might be use to reduce the production of mutagenic HCA during cooking process of protein-rich foods.

Analysis of Volatile Compounds in Perilla frutescens var. acuta by Solid Phase Microextraction (SPME에 의한 소엽의 향기성분 분석)

  • This study was conducted to find the appropriate fiber for extraction of volatile compounds from Perilla frutescens var. acuta. by solid phase microextraction (SPME). Two SPME fiber, carboxen/polydimethylsiloxane (CAR/PDMS) and polydimethylsiloxane (PDMS) were used to determine the selectivity of the fibers to the different flavor compounds present in the Perilla frutescens var. acuta. Thirty-nine compounds were identified in the volatile compounds extracted by CAR/PDMS fiber, including 6 aldehyde, 1 alcohol, 10 hydrocarbons, 17 terpene hydrocarbons, 2 ketones and 3 benzenes. In PDMS fiber, 3 aldehydes, 2 alcohols, 13 terpene hydrocarbons and 2 miscellaneouses were identified. Perillaldehyde was found to be major volatile flavor component of fresh Perilla frutescens var. acuta. Perillaldehyde and terpene hydrocarbons were more identified in PDMS fiber. These results suggested that the selectivity of PDMS fiber was better than that of CAR/PDMS fiber in Perilla frutescens var. acuta..

Effect of Priming, Temperature and Light Quality on Germination of Pokeweed(Phytolacca americana) Seed (Priming, 온도 및 광질이 미국자리공 종자의 발아에 미치는 영향)

  • Pokeweed, a polycarpic plant, has been used as herbage medicine, vegetable or dye. It, however, is known as an aggressive plant in the vicinity of the industrial area evolving air pollutants. The experiment was done to determine the effect of priming using nitrates, germination temperature and light quality on germination of its seed to get information on the optimum germination process as well as its establishment. The daily percent germination was measured to 12 days after sowing since its seeds were treated by two different nitrates [KNO$_3$, Ca(NO$_3$)$_2$]. their different concentrations (0, 50, 150, 300mM), then treatment duration (1, 3, 6 days), different germination temperature (day /night; 30/30, 30/20, 20/30, 20/2$0^{\circ}C$) and light quality (red, white, dark) before or during germination. The percent germination was greater in the KNO$_3$ treatment than in the Ca(NO$_3$)$_2$ but in the priming treatment with KNO$_3$ in comparison with no-priming. In the priming treatment with KNO$_3$, the percent germination was increased with its increased concentration to 150mM although decreased with delayed duration to 6 days. Regardless of light quality, the greater percent germination was shown in the order of 2$0^{\circ}C$ constant and 20/3$0^{\circ}C$ alternative, 3$0^{\circ}C$ constant, 30/2$0^{\circ}C$ alternative temperature. The germination was less in the dark during germination than in the illumination in which the red light had greater percent germination compared to white light. The seeds primed with KNO$_3$ were germinated under the alternative temperature even in the dark condition.

Effects of Treatments on the Distribution of Volatiles in Artemisia princeps Pampan (쑥의 처리조건에 따른 휘발성 성분 변화)

  • Volatiles in Artemisia princeps Pampan. cv. sajabal (sajabalssuk) and A. princeps Pampan. (ssuk) treated with different processing were analyzed using headspace-solid phase microextraction (HS-SPME)/gas chromatography- a mass selective detector (GC-MS). Sajabalssuk and ssuk were treated with steam distillation (SD) and freeze-dried/steam distillation (FD/SD) while controls were raw sajabalssuk and raw ssuk. Sajabalssuk had significantly more total volatiles than ssuk in control and FD/SD treated samples (p<0.05). Major volatiles in raw sajabalssuk were 2-hexenal, 1,8-cineol, trans-caryophyllene, and hexanal while those in raw ssuk were 1-hexanol, ${\beta}$-myrcene, limonene, and 2-hexenal, which implies that substantial lipid oxidation occurred in raw samples. Sajabalssuk with SD and FD/SD treatment had higher peak areas of 1,8-cineole, 4-terpineol, 1-octen-3-ol, and ${\alpha}$-terpineol while ssuk with SD and FD/SD treatment possessed 1,8-cineol, camphor, borneol, artemisia ketone, ${\alpha}$-thujone, and 1-octen-3-ol, which showed that steam distillation produced more volatiles from terpenoids than raw samples. Based on the results of HS-SPME/GC-MS, relative amounts of volatiles from lipid oxidation including 2-hexenal, hexanal, and 1-hexanol were reduced in sajabalssuk and ssuk with freeze-drying and/or steam distillation treatment.

  • The transitivity of Chemical constituents by Pleurotus ostreatus cultivated in different raw sawdusts, which are Juglans mandchurica, Cudrania tricuspidata and Lindera glauca, was investigated. The HPLC chromatography patterns on the chemical constituents of P. ostreatus showed the similar chromatography patterns in all different raw sawdusts and control sawdust. The unknown chemical constituents of P. ostreatus cultivated in the 10%, 20% mixed medium added 10 %, 20% different raw sawdusts, respectively, were increased. But the significance results in the mixed medium added 50% different raw sawdusts were not showed. The chromatography patterns of mycelia grown in media added the 80% MeOH extracts of three tree species showed the similar patterns in comparison with control mycelia. In the results, the secondary metabolites of functional media were not degrade and changed to other derivatives compounds by P. ostreatus.
