수확 후 자외선 조사에 의한 '부유' 단감의 과피 착색 증진 (Enhancement of Skin Color by Postharvest UV Irradiation in 'Fuyu' Persimmon Fruits)

  • 에틸렌 작용억제제인 1-MCP의 처리와 함께 수확 후 '부유' 단감 과실에 호르메시스유발원으로 자외선을 조사하였을 때 carotenoid 색소 함량 변화와 함께 에틸렌 생성, 호흡 및 과육 경도의 변화를 조사하였다. 수확 당시 '부유' 단감 과실의 주요 carotenoid 색소는 ${\beta}$-carotene, lycopene, ${\beta}$-cryptoxanthin이었으며, 이 중 lycopene 색소 함량은 수확 이후 실온 저장 기간 중에 크게 증가하는 양상을 보였다. 자외선 조사는 단감에서 ${\beta}$-carotene과 lycopene 색소의 함량을 증가시켜 과피의 착색을 증진하였으나 동시에 자외선 스트레스에 따른 과육의 경도 저하를 촉진하였다. 그러나 자외선 조사에 따른 carotenoid 색소 함량의 증가는 1-MCP 처리의 영향을 받지 않은 반면 과육의 연화는 1-MCP 처리에 의해 크게 지연되었다. 따라서 수확 후 '부유' 단감 과실에 1-MCP 처리와 자외선 조사를 병행할 경우 연화 억제와 과피의 착색 중진을 통한 품질의 향상이 가능할 것으로 생각된다.

흡연대학새의 비타민 A와 $\beta$-Carotene 섭취량 및 혈청 총 Carotenoids 수준과이 관계 (Relation between Vitamin A and $\beta$-Carotene Intakes and Serum Total Carotenoids Levels in Smoking College Students)

  • The aim of this study was to assess the association between total vitamin A, retinol and carotene intakes and serum concentration of total carotenoids in Korean male smokers and nonsmokers. Food frequency and nutrient intakes of 64 healthy college students were estimated by the 24 hour recall method and food frequency questionnaire and analyzed by smoking status. Serum total cartenoids and lipid profiles were measured in 28 smokers and 36 nonsmokers. The mean total vitamin A, retinol and carotene intakes of smokers were not significantly different from nonsmokers. However, carotene intakes of the smokers who have smoked more than 5 packyears were significantly lower than those of nonsmokers. Smokers consumed green yellow leafy vegetables more often(P=0.005) and fresh fruits less often(P=0.017) than nonsmokers. The serum total carotenoids of smokers consuming the same amount of retinol and carotene as nonsmokers, 1.79 g/ml, were 17% lower than nonsmokers, 2.15 g/ml(P<0.05). The lipid profiles including total cholesterol, LDL cholesterol, VLDL cholesterol and triglycerides of smokers were not significantly different from nonsmokers, however, the concentration of HDL cholesterol of smokers were significantly higher than that of nonsmokers. The serum carotenoids concentration was positively associated with fruit consumption frequency in smokers, and also was positively associated with green yellow leafy vegetable consumption frequency in non smokers. There were no correlations between dietary and serum carotenoids for smokers as well as nonsmokers. It was concluded that smoking could be inversely related to the serum concentration of carotenoids at constant carotene intakes and smokers might require more carotene to reach the same concentration comparable to nonsmokers.

국내에서 상용되는 농산물의 카로티노이드 함량 분석 (Analysis of Carotenoids in Commonly Consumed Agricultural Products in Korea)

  • A total of 51 vegetables and fruits, commonly consumed agriculture products in Korea, were analyzed for their α-carotene, β-carotene, and β-cryptoxanthin contents as provitamin A. The beta-carotene content (㎍/100 g) was high in a few green leaf vegetables such as coriander (5,924.07), gegeol radish leaf (5.855.72), and curried mallow (5,138.01), while α-carotene and β-cryptoxanthin contents were not detected. The β-carotene in 8 kinds of 20 general vegetables was detected in the range of 214.06~1,437.67 ㎍/100 g, while α-carotene was detected at 460.17 ㎍/100 g in only old pumpkin. The β-cryptoxanthin was detected in the range of 106.55~315.49 ㎍/100 g in Japanese elm, watermelon, white cucumber, and lettuce. However, carotenoids were not detected in 10 kinds of agricultural products including oriental melon, potato, etc. In fruits, the beta-carotene contents ranged from 165.72~3,997.39 ㎍/100 g, showing maximum value in apple mango and minimum value in persimmon. The β-cryptoxanthin was detected at 232.22 ㎍/100 g in only passion fruit, while the α-carotene was detected at 77.25 ㎍/100 g in only darae. Thus, based on the analyzed results of carotenoids of agriculture products consumed or cultivated in Korea, and it was found that green leaf vegetables comprise high beta-carotene overall.

베타-카로텐 탑재 하이드로 젤 농도와 저작에 따른 지방소화율과 생체접근율의 변화 (Influence of Starch Concentration and Mastication on the Lipid Digestion and Bioaccessibility of β-carotene loaded in Filled Hydrogels)

  • Purpose: This study was conducted to examine the effects of the starch concentration of filled hydrogel and the addition of ${\alpha}-amylase$ and simulated mastication processing in an oral phase on lipid digestion and ${\beta}-carotene$ bioaccessibility of filled hydrogels. Methods: Lipid digestion and ${\beta}-carotene$ bioaccessibility of the filled hydrogels were measured after the samples were passed through an in vitro gastrointestinal tract model consisting of oral, gastric, and small intestinal phases. Results: The initial rate and final extent of lipid digestion were higher in the filled hydrogels than in the emulsion when the filled hydrogels were treated in an oral phase without simulated mastication processing and addition of ${\alpha}-amylase$, regardless of starch concentration. However, when the filled hydrogels were minced using mortar and pestle for 2 min and were exposed to ${\alpha}-amylase$, the filled hydrogel fabricated with 5% starch showed the lowest lipid digestion rate and extent compared to the emulsion and other filled hydrogels. Bioaccessibility of ${\beta}-carotene$ was higher in the filled hydrogels than in the emulsion, regardless of the digestion method performed in an oral phase and starch concentration. However, there were appreciable differences in bioaccessibility of the filled hydrogels depending on whether or not simulated mastication and addition of ${\alpha}-amylase$ were employed. Conclusion: These results suggested that the rheological properties of initial filled hydrogels and simulated mastication processing in an oral phase plays an important role in determining the lipid digestion and ${\beta}-carotene$ bioacccessibility entrapped within filled hydrogels.

감자에서 추출한 phenolics의 갈변화에 미치는 $\beta$carotene의 영향과 polyphenol oxidase 특성 (Effects of $\beta$Carotene on the Browning of Phenolics Extracted from Potato and Polyphenol Oxidase Characteristics)

  • 한국의 민간요법에서 감자와 당근의 주스 혼합액이 위궤양의 치료에 사용되어져 왔다는 사실을 근거로 전보에서 당근이 감자의 갈변에 어떤 영향을 미치는지를 알아보았다. 본 논문에서는 감자의 갈변을 저해하는 감자속의 어떤 물질의 효과를 배제시키기 위하여 감자에서 polyphenolics만을 추출하여 여기에 효소를 가한 뒤 당근을 첨가하여 갈변화를 측정하였고 감자의 PPO활성에 베타-카로틴이 어떤 영향을 미치는지를 살펴 보았다. 그 결과 시료로 사용한 총 polyphenolics의 함량은 1. 854mg/g D.W.였고 PPO활성도는 100unit이였다. 감자에서 추출한 polyphenolics의 delta 'L'값은 급격히 감소하였으나 당근주스의 혼합액은 조금씩 감소하였다. Polyphenolics와 합성 베타-카로틴을 섞은 용액은 반응 50분 후에 약간의 감소를 보였다. 동시에 감자에서 추출한 polyphenolics를 당관에서 추출한 카로틴 혹은 베타-카로틴을 가한 후 갈변차를 알아본 결과 당근에서 추출한 카로틴 용액을 섞은 것은 delta 'L'값이 증가하였고 1 시간 후 합성 베타-카로틴과 비슷하게 감소하였다. 한편 감자에서 추출한 PPO의 갈변화에 기질과 효소의 농도가 어떤 영향을 미치는지를 알아본 결과 기질농도 10%일때 최적효소의 농도는 10%였으며(대조군의 농도비는 완충액 2.6 : 기질 0.3 : 효소액 0.1이었음). 효소농도 3.3%일때 최적 기질의 농도는 13.3%이었고 베타-카로틴의 농도는 PPO활성도에 거의 영향을 미치지 않는 것으로 나타났다.

비타민 A 보충 식이 및 에탄올의 만성적 급여가 흰쥐의 체내 산화적 손상과 항산화체계에 미치는 영향 (Effect of Dietary Supplementation of Vitamin A and Chronic Consumption of Ethanol on Oxidative Damage and Antioxidant System in Rats)

  • 본 연구는 에탄올을 급여하는 동안 일어나는 지질과산화에 의한 간 손상과 이에 대하여 $\beta$-carotene, retinyl acetate, 13-cis-retinoic acid의 효과를 비교ㆍ검토하고자 실시되었다. 혈 장내 MDA 함량은 에탄올 급여군 중 비타민 A를 결핍시킨 FE군과 $\beta$-carotene을 공급한 DE군에서는 감소되지 않았으나, retinyl acetate와 13-cis-retinoic acid 공급시킨 RE군과 RAE군에서 감소되었다. 간 미토콘드리아내 MDA함량은 에탄을 급여군 중 FE군과 $\beta$E군에서 높은 함량을 보인데 반해 RE군과 RAE군에서는 낮은 수준을 보였다. 에탄올 비급여군인 pair-fed군중에서는 비타민 A를 결핍시킨 FP 군에 비해 $\beta$-carotene과 retinyl acetate를 각각 공급한 5P와 RP군의 MDA 함량은 감소하였는데 반해 retinoic acid를 공급한 RAP군의 MPA 수준은 차이를 보이지 않았다. 혈장내 AP 활성도는 에탄올과 함께 $\beta$-carotene과 retinoic acid를 공급시킨 군에서 각각의 pair-fed군에 비하여 높은 활성도를 보였으며 간 마이크로솜내 G6P 활성도는 에탄올 급여군 중 비타민 A 결핍군만 pair-fed군에 비해 유의적으로 감소되었으며 비타민 A를 보충시켰을 때에는 활성도가 증가되었다. 간 사이토졸내 SOD 활성도는 에탄을 급여와 함께 비타민 A를 결핍시킨 군에 비해 13-ciz-retinoic acid을 공급시킨 군이 가장 높았으며 Pair-fed 군 중에서는 retiny acetate 공급군에서 가장 낮은 높은 활성도를 보였다. 간 사이토졸내 GSH-Px 활성도는 에탄올 급여군과 pair-fed 군 사이에 일정한 경향은 없었으며 간 사이토졸네 GST 활성도는 에탄올 급여군 중 $\beta$-carotene과 retinyl acetate 공급군이 각각의 pair-fed군에 비해 증가되었다. 간조직내 $\beta$-carotene함량은 $\beta$-carotene, 공급군에서만 검출되었고 에탄을 급여군이 pair-fed군에 비해 6배정도 높은 함량을 보였으며 혈장과 간조직 내 retinol 함량은 에탄올 급여로 낮은 함량을 보였으나 비타민 A형태 중 retinyl acetate 공급군이 가장 높았다. 간조직 내 retinoic acid 함량은 에탄을 급여군 중 $\beta$-carotene과 retinyl acetate 공급군이 각각의 Pair-fed 군에 비해 유의적인 감소를 보였다. 상기 연구 결과를 보면 에탄올 급여로 지질과 산화물 함량이 증가되고 관련 항산화 효소계의 활성도 변화로 보아 간 손상에 지질과산화 반응이 관여된 것으로 보여 지며 에탄올의 영향에 대한 비타민 A의 효과에는 retiny1 acetate가 항산화제로 작용하여 간조직의 급격한 손상을 효율적으로 경감시킬 수 있는 것으로 나타났다.

Inhibition of Proliferation and Induction of Apoptosis by the Combination of β-carotene and 1,25-dihydroxyvitamin D3 in Human Esophageal Cancer EC9706 Cells

  • Esophageal cancer is a common malignant tumor occurring in human esophageal epithelial tissue. The primary purpose of this paper was to define the effects of ${\beta}$-carotene and 1,25-dihydroxyvitamin $D_3$, alone and in combination, on cell proliferation, cell cycle and apoptosis of human esophageal cancer EC9706 cells. Treatment with different concentrations of ${\beta}$-carotene and/or 1,25-dihydroxyvitamin $D_3$. MTT assay showed that ${\beta}$-carotene and 1,25-dihydroxyvitamin $D_3$ significantly inhibited proliferation of EC9706 cells in a dose- and time-dependent manner. Further studies also demonstrated that ${\beta}$-carotene alone or 1,25-dihydroxyvitamin $D_3$ alone caused a marked increase on the induction of apoptosis in EC9706 cells. The percentage of G0/G1-phase cells significantly increased on addition of 1,25-dihydroxyvitamin $D_3$ alone, but there were no significant changes with ${\beta}$-carotene alone. These two agents in combination synergistically inhibited cell growth and induced apoptosis. Therefore, our results indicate that ${\beta}$-carotene and 1,25-dihydroxyvitamin $D_3$ in combination may provide a novel strategy for preventing and treating esophageal cancer.

여대생의 Carotenoids 식품 보충 섭취에 따른 혈청 비타민 A 함량 변화 (Serum Vitamin A Response to SHort-term Carotenoids Foods Supplementation in Female University Students)

  • Korean intakes of Vitamin A have not reached RDA for a long time according to the National Nutrition Survey. The pupose of this study was to examine the serum response to short-term carotenoids-rich vegetable juices supplementation. A total of 31 female students were recruited for this study. Their average serum retinol content before supplementation began was 14.74$\mu\textrm{g}$/100ml, and 87% of them had lower than adequate levels. Serum $\beta$-carotene, $\alpha$-carotene, lycopene and lutein contents were 0.77, -, 0.74 and 8.29$\mu\textrm{g}$/100$m\ell$ respectively. The serum responses to 3 kinds of juice supplementation for 3 weeks are summarized as follows. A small but steady increase in serum retinol concentration was detected with the supplementation (p<.05), but the kinds of juice made no significant difference. Serum $\beta$-carotene contents also increased with the supplementation(p<.01), but the kinds of juice again made no significant difference. Serum lycopene increased with the lycopene-containing vegetable juice and tomato juice supplementation, but the increase wasn't significant. There was no change in serum lycopene levels with the lycopene free carrot juice supplementation. Finally, there were no signifnicant changes in serum lutein and $\alpha$-carotene levels with the 3 kinds of juice supplementation.
