Root Bark of Morus alba L. and Its Bioactive Ingredient, Ursolic Acid, Suppress the Proliferation of Multiple Myeloma Cells by Inhibiting Wnt/β-Catenin Pathway

  • The root bark of Morus alba L. has cytotoxic activity against several types of cancer cells. However, little is known about its chemopreventive mechanisms and bioactive metabolites. In this study, we showed that M. alba L. root bark extracts (MRBE) suppressed β-catenin response transcription (CRT), which is aberrantly activated in various cancers, by promoting the degradation of β-catenin. In addition, MRBE repressed the expression of the β-catenin/T-cell factor (TCF)-dependent genes, c-myc and cyclin D1, thus inhibiting the proliferation of RPMI-8226 multiple myeloma (MM) cells. MRBE induced apoptosis in MM cells, as evidenced by the increase in the population of annexin VFITC-positive cells and caspase-3/7 activity. We identified ursolic acid in MRBE through LC/mass spectrum (MS) and observed that it also decreased intracellular β-catenin, c-myc, and cyclin D1 levels. Furthermore, it suppressed the proliferation of RPMI-8226 cells by stimulating cell cycle arrest and apoptosis. These findings suggest that MRBE and its active ingredient, ursolic acid, exert antiproliferative activity by promoting the degradation of β-catenin and may have significant chemopreventive potential against MM.

  • In the study, we endeavored to investigate the effect of phenylephrine, isoprenaline and prazosin on the tissue-specific vascular contractility and to determine the mechanism involved. There were few reports addressing the question whether thin or thick filament modulation is included in phenylephrine, isoprenaline and prazosin-induced regulation. We hypothesized that isoprenaline and prazosin play a role in tissue-dependent regulation of vascular contractility. Denuded arterial muscles of Sprague-Dawley male rats were suspended in organ baths and isometric tensions were transduced and recorded using isometric transducers and an automatic data acquisition system. Interestingly, sustained continuous contraction of thoracic and abdominal aorta. Furthermore, isoprenaline and prazosin together with phenylephrine inhibited transiently and persistently vasoconstriction of thoracic and abdominal aorta suggesting that additional mechanisms (e.g. decreased receptor density, chemical interaction, postreceptor signaling or distribution of agonists) might be included in the modulation of vascular contractility.

Anti-atopic Effects of Castanea crenata Inner Shell Extracts Fermented by Lactobacillus bifermentans (Lactobacillus bifermentans로 발효한 율피의 항아토피 효과)

  • Atopic dermatitis (AD) is a common skin disease characterized by chronic and relapsing inflammatory dermatitis with immunological disturbances. In spite of the continuous increase in the incidence of AD, it is regrettable that till date there is no effective treatment to treat the same. Therefore, the present study was designed to examine the possible anti-atopic effects of Castanea crenata inner shell extracts fermented by Lactobacillus bifermentans (FCS) in 2,4-dinitrochlorobenzene (DNCB) induced AD in NC/Nga mice. Based on the results of HPLC analysis, we found that FCS contains anti-inflammatory factors such as gallic acid (10.18 mg/g) and ellagic acid (2.14 mg/g). The groups that we have used in this study included 0.1%, 1%, 5% fermented Castanea crenata inner shell extracts (FCS 0.1, FCS 1, FCS 5), 1,3-butylene glycol treated control (AD), and normal mice. After topical FCS treatment, we observed that the clinical severity score for AD was lower in both the FCS 1 and FCS 5 groups than the AD group. We also proved beyond doubt that there was improvement of melanin, erythema and skin moisture indices in the FCS 5 group. Spleen index and gene expression levels of pro-inflammatory cytokines such as IL-$1{\beta}$ and TNF-${\alpha}$ were significantly decreased in the FCS 5 group compared to the AD group (P<0.05). Further, we also found that the level of serum immunoglobulin E (IgE) in the FCS-treated group was decreased in a concentration-dependent manner. The results of our study suggest that FCS can be effectively used as a cosmeceutical ingredient for both the prevention and improvement of AD.

The Influence of a General Hospital Nurse's Emotional Labor, Emotional Intelligence on Job Stress (일개 종합병원 간호사의 감정노동과 감성지능이 직무스트레스에 미치는 영향)

  • The purpose of this study is to investigate how the emotional labor and emotional intelligence of nurses working at a general hospital affect their job stress, and how the integration factor of emotional labor and emotional intelligence affects their job stress. The subjects of research were the nurses working at general hospital in Seoul city from March 11-29, 2013. The collected data was analyzed after computerized statistical processing using SPSS 18.0 and AMOS 18.0. It was found that the frequency of emotional expressions, one of emotional labor variables, significantly negatively influenced job stress(${\beta}=-.301$, p<.01), and that the attention required for the norms of emotional expressions significantly positively affected job stress(${\beta}=.277$, p<.01). Among emotional intelligence variables, understanding of self-emotion and control of emotion were found to significantly negatively affected job stress. Given the study result, in order to alleviate nurses' job stress, it is necessary to have positive emotional expressions with patients, come up with a plan to show nurses' emotions which they fail to express because of the norms of emotional expressions in hospital, and make their effort to improve understanding of their own emotions and the capability of controlling emotions.

The Effect of EEG And Physiological Changes To Participation in Progressive Relaxation Technique Of The University Archery players (호흡을 통한 신체적 이완이 대학 양궁선수의 뇌파 및 생리적 변인에 미치는 영향)

  • The purpose of this study is to monitor the physiological changes by measuring EEG, blood pressure, heart rate, etc after applying progressive relaxation technique to university archery players in order to verify the effect of progressive relaxation through respiration, and, ultimately, improve an athlete's archery performance. This study chose 13 female archery athletes. The EEG, blood pressure, and heart rate of the athletes were measured before applying Jacobson's progressive relaxation technique for 12 weeks, during which the EEG, blood pressure, and heart rate were measured 3 times a week after 15 to 20 minutes of training before every measurement. We used the SPSS18.0 program for technical analysis to check the general features of the athletes and carried out the paired t-test to examine the changes made to the EEG, blood pressure, and heart rate of a single test group before and after the experiment. All statistical level of significance was maintained at ${\alpha}=0.5$. The results are as follows. First, after 12 weeks of appliance, between the targets' relative ${\alpha}$ wave and ${\beta}$ wave, the relative ${\alpha}$ wave showed a statistically significant increase while the relative ${\beta}$ wave showed no changes. Second, after 12 weeks of appliance, in terms of blood pressure, the systolic pressure showed no changes while the diastolic pressure showed a statistically significant change. Third, there was no statistically significant change to the heart rate. In conclusion, progressive relaxation through breathing has a positive effect on the archery athletes.

Investigation of the IL-1β, TNF-α and iNOS gene differential expression in Raw 264.7 cells by the water extract of Angelicae Radix from Korea, China and Japan (참당귀, 중국당귀, 일당귀 열수 추출물의 RAW 264.7 대식세포에서 IL-1β, TNF-α, iNOS 유전자 차등 발현 연구)

  • We tried to analyze the inflammation reactions by treatment of AG, AS and AA in murine RAW 264.7 cells. To investigate the effect of AG, AS and AA on cell viability of RAW 264.7 cells, AG, AS and AA were treated for 24 h and MTS assay was performed. Cell viabilities were increased in $1,600{\mu}g/ml$ concentration by AS, AA and AG treatments, respectively. The mRNA expression levels of $IL-1{\beta}$, $TNF-{\alpha}$ and iNOS were increased by AG and AA treatment at a concentration of $200{\mu}g/ml$ in RAW 264.7 cells without Lipopolysaccharide (LPS) treatment. The mRNA expression levels of $IL-1{\beta}$, $TNF-{\alpha}$ and iNOS were increased by AG and AA 6 h treatment at a concentration of $200{\mu}g/ml$ with LPS treatment. In this study, we observed that AG, AS and AA show various activities on inflammation reaction depend on their treatment time. In the future, studies should be conducted to investigate the effects of AG, AS and AA on the various inflammatory responses of macrophages.

Factors Influencing Acculturative Stress of Migrant Workers (외국인 근로자의 문화적응 스트레스 영향요인)

  • The purpose of this study was to identify acculturative stress, social support and self esteem in migrant workers. The data of 181 subjects were collected from June 1st to September 27, 2016, and analyzed using the SPSS WIN 23.0 program. Results: There was a significant negative correlation among acculturative stress, social support(r=-.465, p<.001) and self esteem(r=-353, p<.001). Stepwise multiple regression analysis showed self esteem(${\beta}=-.21$, p<.001) and job(${\beta}=-.02$, p<.05) were significant factors affecting acculturative stress(F=27.672, p<.001). The results of the study indicate that the efforts, to improve self esteem, and to increase social support, are required to decrease acculturatives stress among migrant wokers. The results are expected to be utilized in development of supportive program for migrant workers adaptation. In the following study, the analysis of structural equation modeling about additional factors of acculturative stress among migrant workers will be needed.

Moderating Effects of Interpersonal Relation and Social Network on the Relationship between Depression and Health Behavior in Elderly (노인의 우울과 건강증진행위와의 관계에서 대인관계와 사회적 지지의 조절효과)

  • This study was conducted to identify moderating effects of interpersonal relation and social support on the relationship between depression and health promoting behaviors in the elderly. The participants were 286 elderly using two elderly welfare centers and one cultural center in D city. Data were collected from October 12th to 20th December, 2016, and were analyzed by t-test, ANOVA, multiple regression using SPSS 20.0. The result were as follows; The result were as follows; 1) Mean score of variables was Depression: 20.47, interpersonal relationship: 21.27 points, social support: 20.92 points, and health promotion behaviors: 111.69. 2) Depression had negative relationship with interpersonal relationship(r=-.283, p<.000), social network (r=-.391, p<.000), health promotion behaviors(r=-.611, p<.000), and interpersonal relationship had positive relationship with social support(r=.353, p<.000) and health promotion behaviors(r=.372, p<.000) significantly. 3) Social support had a moderating effect in the relationship between depression and health promoting behaviors(${\beta}=.448$, p=.011), while interpersonal relationship did not show moderation effect(${\beta}=.380$, p=.135). Based on these results, health program for elderly including social support is recommended to promote health behavior of the elderly in the community.

Influences of Nutritional Status and Depression on Satisfaction with Life in Middle-Aged Long-Term Care Patients (중년기 장기요양 환자의 영양 상태, 우울이 삶의 만족도에 미치는 영향)

  • This study aimed to identify influences which nutritional status and depression have on satisfaction with life on middle-aged long-term care patients. This study is a descriptive study conducted with 120 patients aged 40-65 from February 1st to March 31th, 2015 in hospital. Data were analyzed using t-test, ANOVA, Scheff's test, Pearson's correlation coefficients, multiple regression. The average of nutrition status, depression, satisfaction of life was $15.85{\pm}3.85$, $24.70{\pm}15.85$, $3.54{\pm}1.22$ each. The findings show nutritional status(r=.281, p<.005) have correlation(+) with satisfaction of life, otherwise, nutritional status(r=.439, p<.001) and satisfaction of life(r=-.-574, p<.001) have correlation(-) with depression both. Influencing factors on satisfaction of life were subjective health(${\beta}=.387$, p=.000) and depression(${\beta}=.251$, p=.010), explained 36.0% of the variance. Therefore, management program for improving satisfaction of life is needed on middle-aged long term care patients.

Prediction Model for Health Related to Quality of Life of Married Immigrant Women in Multicultural Families (다문화가정 결혼이주여성의 건강관련 삶의 질 예측모형)

  • This study is to determine health related to Quality of Life(QOL) of married immigrant women in multicultural families, factors of predictors, and influence of these factors to develop and verify the structural model for development of an effective nursing mediation strategy for improved QOL. The participants were 254 women who immigrated to Korea for marriage. The hypothetical model is based on the health improvement model by Pender. Immigrant women's perceived barriers, perceived benefits, self-esteem, and perceived health had an impact on their health promotion behavior(HPB). These variables explained 55.2% of the QOL regarding health, and perceived health conditions, self-efficacy, self-worth, and level of cultural adaptation explained 29.3% of acts towards improving health. The results of this study suggest that development of efficient policy considering factors affecting health related to QOL of married immigrant women in multicultural families.
