  • There have been many studies on monitoring of biodiversity changes and preservation of Gwangneung Forest Biosphere Reserve (GFBR) in South Korea in recognition of the rare ecosystem that has been preserved for a long period. However, there are few studies on diversity and community characteristics of benthic macroinvertebrates as an indicator of stream health of GFBR. The purpose of this study was to assess the water quality of Bongseonsa Stream that penetrated through Gwangneung Forest and the nearby torrents by analyzing the benthic macroinvertebrates community during April to September 2016. The investigation collected a total of 114 species of benthic macroinvertebrates belonging to 56 families, 17 orders, 8 classes, and 5 phyla from the Bongseonsa Stream and Kwangneung Stream. Ephemeroptera and Trichoptera were the largest groups in species diversity with 30 species (32.3%) and 16 species (17.2%), respectively, and Tubificidae sp., Baetis fuscatus, Antocha KUa, and Cheumatopsyche brevilineata, which usually habit in contaminated streams, appeared frequently. Among the feeding function groups, the gatherers and hunters appeared relatively frequently, and the shredders and scrapers appeared frequently in the torrents. Among the habitat oriented groups, the clingers and burrower appeared more frequently and represented the microhabitats in the shallow areas. The result of the analysis of benthic macroinvertebrates community showed that the dominant index was $0.48{\pm}0.10$ in average while it was lowest with 0.33 in GS 8 of the Gwangneung Forest torrent and highest in BS 1 of Bongseonsa Stream. The diversity and richness indices were inversely proportional to the dominant index and were 2.53 and 4.22, respectively, in GS 8 where the dominant index was low. The result of the analysis of community stability showed that area I, which had high resistance and restoration, was high in Bongseonsa Stream while the area III, which had low resistance and restoration, was high in Gwangneung Forest, indicating that the water system in Gwangneung Forest had a wider distribution of specifies sensitive to agitation. The biological water quality assessment showed ESB of $50.88{\pm}17.69$, KSI of $1.11{\pm}0.57$, and BMI of $78.55{\pm}11.05$. GS 8 of Gwangneung Forest torrent was judged to be the highest priority protective water area with the best water environment and I class water quality with ESB of 63, KSI of 0.55, and BMI of 89.9. On the contrary, BS 1 of Bongseonsa Stream was judged to be the high priority improvement area that had the lowest water quality rating of III with ESB of 25, KSI of 2.13, and BMI of 62.7. Although the diversity of water beetle was higher in the water system of nearby Bongseonsa Stream than the water system inside the Gwangneung Forest, the annual community structure appeared to have distinct differences.

  • In a previous report, 50 potato cultivars were screened for infestation levels by the coppery click beetle (Selatosomus puncticollis Motschulsky) in the field. Subsequently, we selected 10 potato cultivars (Anco, Atlantic, Bintje, Dejima, Denali, Jopung, Irish Cobbler, Namsuh, Shepody, and Superior) to evaluate feeding preferences by wireworms, and to analyze some factors associated with resistance. The injury rates and number of holes in potato tubers damaged by larvae of S. puncticollis were checked in the field and laboratory. Additionally, some of their chemical characters (contents of glycoalkaloids, total-nitrogen, Ca, K, Mg, sugars, and starch) were quantified. And finally correlation analysis was conducted to see whether there is a possible relationship between these characteristics and the damage level. The tuber injury rates by S. puncticollis larvae were generally high showing 19% to 73% of damage level. The highest number of tuber hole damaged by S. puncticollis larvae was found on cv. Namsuh, but generally fewer on cvs. Anco, Atlantic, Bintje, Denali and Superior. No activities for $\alpha$-chaconine, $\alpha$-solanine and $\alpha$-tomatine at a concentration of 2,500 ppm were found to S. puncticollis larvae. The contents of glycoalkaloids in tuber were different depending on cultivars. In tubers, cv. Superior contained the highest level of 18.8 mg%, but cv. Irish Cobbler had the lowest level of 6.39 mg%. Concentrations of reducing sugars and total free sugars in tubers of cv. Namsuh were 0.71 % and 2.95%, but 0.26% and 1.77% in those of cv. Dejima, respectively. For the content of potato starch, cvs. Bintje, Dejima and Irish Cobbler showed higher level, but cvs. Jopung and Shepody lower. The highest contents of total nitrogen, Ca, K and Mg in tuber were found on cvs. Anco, Atlantic, Anco and Jopung, respectively. From correlation analyses, injury rate by S. puncticollis larvae was correlated with total nitrogen content (r = -0.71435*) and total sugar content in tuber (r = 0.78018*). Such information will become essential in developing integrated pest management programs and also in breeding new potato cultivars resistant to the wireworms.

  • The beetle Popillia flavosellata has been no reported its functional effects. In this study, we investigated the anti-inflammatory effect of P. flavosellata ethanol extract (PFE) on RAW 264.7 mouse macrophage cells treated with lipopolysaccharide (LPS) for the induction of inflammation. First, we examined the cytotoxicity of PFE in the RAW 264.7 cells at a concentration of 2,000 μg/ml or less. To evaluate the anti-inflammatory effects of PFE, we investigated the expression levels of proinflammatory cytokines, such as tumor necrosis factor (TNF)-α and interleukin (IL)-6, and proinflammatory enzymes, such as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in LPS-induced RAW 264.7 cells. In addition, we examined whether PFE inhibited the translocation of nuclear factor kappa B (NF-κB) p65 into the nucleus in the LPS-induced RAW 264.7 cells. We found that the protein levels of TNF-α and IL-6 were decreased in the LPS-induced RAW 264.7 cells after the treatment with PFE in a dose-dependent manner. In addition, we confirmed that PFE inhibited the translocation of NF-κB p65 into the nucleus, as well as the protein expression levels of iNOS and COX-2. Accordingly, we propose that PFE exerts an anti-inflammatory effect through the down-regulation of NF-κB p65, TNF-α, IL-6, iNOS, and COX-2 via the toll like receptor (TLR)-4 inflammatory signaling pathway.

  • Enterobacteria were isolated in the gut of the predacious multicolored Asian ladybird beetle, Harmonia axyridis, and their effects to the development of H. axyridis were examined. Populations of H. axyridis in this experiment were collected from Kimjae at Cheonbuk province (JK population), Geumsan at Chungnam province (CK population) and laboratory population at Laboratory of Insect Physiology in Chungnam National University, Daejeon. Thirty-four enterobacteria isolates were purified and isolated from the digestive tract of H. axyridis, and a total of 4 strains were classified into group by analysis of 16S rRNA gene sequences. About 70% of total isolates were phylogenetic groups of Bacillus genus and Staphylococcus genus, and they were commonly separated from the digestive tract of H. axyridis. After investigating their susceptibility against antibiotics with 18 representative enterobacteria isolates, ofloxacin and penicillin were selected for examination in this study of their ability to inhibit the growth of all of isolates. In order to remove the enterobacteria from the aphids, ofloxacin and penicillin were given to the green peach aphid, Myzus persicae, and the turnip aphid, Lipaphis erysimi. These aphids were provided to H. axyridis as prey. The weight of pupa, developmental periods of each larval instar, the number of eggs and their hatching ratio of H. axyridis with treatment aphids were lower compared with non-treatment aphids. Staphylococcus saprophyticus is a representative enterobacteria and commonly isolated from the digestive tract of H. axyridis. In the absence of S. saprophyticus, the developmental periods of each larval instar increased; however, the weights of pupa, the number of eggs, and their hatching ratio decreased.

  • This experiment was carried out to investigate the penetration velocity and diffusion ability of methyl bromide fumigants under the condition of natural gravity, and the effect of in control of yellow corn in the vortical silo. The results were as fallows 1. The methyl bromide remainded concentration on the surface of the vertical silo was rapidly reduced from over 100mg/l to under 20mg/l, within 2 hours after supplying methyl bromide fumigant 2. Even 3m depth place from the surface of the vertical silo filled with yellolw corn was well penetrated within 30 minutes, but its remained methyl bromide gas was reduced gradually from over 100mg/l to 30mg/l after 12 hours duration. 5. In case of 13m place tile penetration ability of methyl bromide was over 100mg/l within one hour and kept the same concentration of it for 12 hours, its remainded methyl bromide preserved over 40mg/l until 48 duration time. 4. Less methyl bromide gas was detected at the bottom place than 3m and 13m depth of the vertical silo. It showed bellow 20mg/l of methyl bromide gas only through the whole duration time. 5. Comparing to the vertical penetration velocity of mehtyl bromide gas, the horizontal diffusion ability was not so good. Therefore, remained methyl bromide gas of 3m depth at the wall side of silo was about half than that of center circle where the nearer place from the methyl bromide supplying point, and the methyl bromide gas of 13m depth placed center circle was detected about 15 to 20 times than that of wall side. 6. For the purpose of examining the mortality of methyl bromide, the testing insects (Lesser Rice weevil and confused flour beetle) placed on the surface and bottom side of the silo were killed completely after 24 hrs fumigation, and the same results showed also at the 3m, 7m and 13m depth after 48 hrs fumigation.

  • Korea has put in significant efforts to increase the number of wildlife crossings between fragmented habitats to prevent loss of biodiversity and to encourage the habitat connectivity in Korea. However, there is a lack of biological data on the effect of vegetation structure in these wildlife crossings and guidelines for design and management of wildlife crossing structures in Korea. Therefore, we selected ground beetle assemblages as model organisms to compare the effect of vegetation structure in wildlife crossings, i.e. bare ground- and shrub-type corridors, in agro-forested landscapes. For this study, 4,207 ground beetles belonging to 33 species were collected through pitfall trapping along the northern forest-corridor-southern forest transects from late April to early September in 2015. Dominant species, abundance, and species richness of ground beetles were significantly higher in the shrub-type corridors than the bare ground-type corridors. Also, the species composition of bare ground-type corridor was significantly different compared to the other habitats such as shrub-type corridor and forests. Similarly, environmental variables were also influenced by vegetation management regimes or trap locations. Collectively, our study clearly indicates that the movement of forest associated ground beetles between forest patches can increase as the vegetation in wildlife crossings becomes complex. Although further studies are needed to verify this, there are indications that the current wildlife crossings that comply with the guidelines may be unfriendly to the movement of ground dwelling arthropods as well as ground beetles. To enhance the ecological function of wildlife crossings, the guidelines need to be rectified as follows: 1) Shrubs or trees should be planted along the corridor verges to provide refuge or movement paths for small mammals and ground dwelling arthropods, and 2) Open spaces should be provided in the middle of the corridors to be used as a path for the movement of large mammals.

  • Beetle larvae have been used as a traditional medicine to treat various human liver diseases. To prove the liver protective function of Allomyrina dichotoma larvae (ADL), we induced liver damage by the intraperitoneal injection of a hepatotoxic reagent, diethylnitrosamine (DEN), to C3H/HeN male mice and orally administered freeze-dried ADL powder. ADL powder lessened DEN-induced hepatotoxicity considering the reduced signs of acute and chronic hepatotoxicities, such as the ALP level in the blood serum, TUNEL-positive hepatocytes, ductural reactions, steatotic hepatocytes, and collagen deposition of the Masson’s trichrome staining. In addition to hepatoprotection, the anti-cancer activity of ADL has been examined. The ADL powder was extracted with ethanol and then fractionated with hexane, ethyl acetate, and water by a solvent partition technique. The ethyl acetate fraction showed cytotoxicity to various cancer cells through induction of apoptosis and necrosis, as well as the perturbed metabolism of the cancer cell to trigger autophagy. Collectively, ADL contains bioactive substances that can protect hepatocytes from toxic chemicals and trigger cell death in cancer cells. Thus, further purification and analyses of ADL fractions could lead to the identification of novel bioactive compounds.

  • With the objective of developing a functional food source, we established optimal processing conditions for the larvae of Allomyrina dichotoma, which have been used in traditional medicine to treat hepatic disorders in Korea. Without suitable processing, the larvae are difficult to consume as a food because of their disgusting taste and smell; moreover, in this form they might be a potential microbial hazard. In this study, we investigated the effect of feeding material, sterilization, and powdering after freeze-drying on the food quality of the larvae of A. dichotoma and on cytotoxicity against Raw 264.7 cells. Three to five days feeding with the sawdust from discarded oak-trees is sufficient for the breeding process. The sawdust was sterilized by vapor for five minutes. Sterilization of the larvae at a high temperature ($115^{\circ}C$ for 5 min, 0.9 $kgf/cm^2$) is necessary to eliminate pathogenic bacteria and fungi. The results of the cytotoxicity assay showed no toxicity in the prepared extract from larvae of A. dichotoma. In addition, to prepare the larvae for human consumption, various feeds were used and the smell, color, and taste were evaluated. Our results suggested that larvae of A. dichotoma could be developed as food source when a suitable processing method is established.

  • This study was conducted to investigate the effect of methanol extract of Allomyrina dichotoma larva (MEAL) on carbon tetrachloride $(CCl_4)-induced$ hepatotoxicity in mice. ICR mice were divided into 5 groups [Vehicle control, $CCl_4\;(10{\mu}g/g)$ alone, $CCl_4$ plus a low dose $(50{\mu}g/g)$ of MEAL, $CCl_4$ plus a high dose $(100{\mu}g/g)$ of MEAL]. Silymarin $(2{\mu}g/g)$ was used as the reference in the experiment. Administration of MEAL tended to decrease the serum alanine transaminase (ALT) activity induced by $CCl_4$ treatment in mice. Hepatic concentration of thiobarbituric acid-reactive substances (TBARS) in a high-dose group of diet decreased to the level of silymarin-treated group. Hepatic activity of glutathione S-transferase in MEAL-treated group was lower than that of $CCl_4-treated$ group. Serum concentration of bilirubin was significantly increased by $CCl_4$ treatment, but MEAL or silymarin recovered the level. These results suggest that MEAL may exert the protective effect against $CCl_4-induced$ hepatotoxicity in mice. However, more intensive studies would be needed to elucidate the protective mechanism of the beetle on hepatotoxicity of mice.
