Continuous Water Toxicity Monitoring Using Immobilized Photobacterium phosphoreum

  • Water toxicity monitoring based on the continuous cultivation of Photobacterium phorphoreum is presented. Normally, after 10 days of operation, a dark variant, which emits no light, appears and dominates the population, resulting in a rapid decrease in bioluminescence. Therefore, to overcome this problem, a fluidized-bed reactor is used in which alginate-immobilized cells are grown and leaking cells are continuously released into the effluent Experimental results revealed that the dominance of dark variants was suppressed inside the immobilized beads, thereby mitigating the rapid loss of bioluminescence. Plus, a high dilution rate (1.2 h$\^$-1/) prevented the occurrence of other microbial contamination in the reactor The concentration and bioluminescence of the released cells were sufficient to measure the water toxicity for more than 4 weeks.

An Economic Analysis of Oak Mushroom Cultivation in Korea (표고버섯 재배에 대한 경제성 분석)

  • This study analyzed an economic efficiency of oak mushroom cultivation in order to provide data needed for formulating related policies. 88 farmhouses in major producing areas were interviewed with. The average production cost per 100 bed-logs was 151,699 won. Among production costs, material costs accounted for 56.0% and labor costs 41.8%. Oak mushroom cultivation was labor-intensive and labor input was a significant factor determining the production costs. There was 3 7.2% gap in production costs between Chungbuk, the highest cost area and Gyeonggi, the lowest cost area of oak mushroom due to difference in labor input. Average revenue per 100 bed-logs was 178,627 won. Average income per 100 bed-logs, which is variable production costs of 116,613 won subtracted from the revenue, was 62,014 won and thus income rate was 34.7%. Income rate in Jeonbuk, the highest area, was higher than that in Gyeonggi, the lowest area, by 15.8% points due to difference in quantities produced, grade, and variable production costs, in particular, labor costs. The result of the economic analysis indicates that reduction in establishment and labor costs, accounting for the highest portion in production costs is priority issue for the enhancement of the competitiveness of the oak mushroom industry.

Conservation Measure of Sajapyeong Alpine Wetland (사자평 고산습지의 보전대책)

  • The formation of Sajapyeong Alpine Wetland was influenced by factors of drainage basin and its geology, and fire-shifting cultivation. Sajapycong drainage basin had a narrow outlet, Sijeon-cheon in it flowed relatively slowly. Bedrock in basin was weak to mechanical weathering, many rock detritus were, produced. Deforestation for reclamation using fire accelerated topsoil loss. Thus much sediments was supplied to Sijeon-cheon and deposited in the channel bed, and wetland was formed on channel marginal footslope. In Sajapyeong moor were Gullies formed along road. Because they blocked sediments and throughflow transferring into moor, moor became dry land. In order to prevent this drying, we have, to raise water level of a drain ditch to level of weathered bedrock to transfer throughflow into moor, modify the shape of ditch to be naturally irregular, and construct large boulders step on the Sijeon-cheon bed to prevent from lowering of its bed.

The Effects of Administration of Physiologically Activating Substance on the Log Cultivation of Lentinus edodes (생리활성화물질(生理活性化物質) 투여(投與)가 표고버섯 원목재배(原木栽培)에 미치는 영향(影響))

  • This study was aimed to reduce the period of bed log through the protection of harmful fungi and to investigate the effects of physiologically activating substance on the yield increase of Lentinus edodes. Extracts of Allium fistulosm, Hordeum vulagare var. hexastichon, Lentinus edodes. Daucus carota var. sativa, and Citrus junos were used as a physiologically activating substances. The degree of mycelial growth tested by color change after Benzene - azo - ${\alpha}$ - naphthylamine. One percent extract of Album fistulosm was most highly effective as a physiologically activating substance on mycelial biomass growth of Lentinus edodes. Best effects of application of physiologically activating substance was observed in Quercus variabilis bed logs. These results suggested that supplement with physiologically activating substance to the bed log would be beneficial for the production of Lentinus edodes.

Effects of various bed soil on the growth and yield of organically grown 2-year-old ginseng in the shaded plastic houses (하우스 인삼재배에서 상토의 조성이 유기농 2년근 인삼의 생육 및 수량에 미치는 영향)

  • This research was conducted to investigate the influence of organic bed soil substrates on growth and yield of organically grown ginseng transplantation in a shaded plastic houses. The pH and EC of the substrates used for this study were 5.93-6.78 and 0.03-0.15 dS/m, respectively. The concentrations of NH4-N and $NO_3$-N were 14.01-68.63 mg/L, 5.60-58.83 mg/L respectively. and the average quantum in the shaded plastic houses was 11-15% of natural light. The maximum temperature in the shaded plastic houses is higher ($3-7^{\circ}C$) than that of outside open field from the last part of April to early in August. Emergence date of ginseng was on March 21 in the mongolian type shaded plastic house, and was on March 29 in normal type shaded plastic house. Both roots and shoot growth of ginseng were excellent in the bed soils with PPV-2, compared with other compounds used. We concluded that the PPV-2 could be promising a good bed soil substrate for organic ginseng cultivation in shaded plastic house.

Spot Cooling System Development for Ever-bearing Strawberry by Using Low Density Polyethylene Pipe (연질 PE관을 이용한 여름딸기 부분냉방기술 개발)

  • The effects of spot cooling on growing ever-bearing strawberry in hydroponic cultivation during summer by spot cooling system was estimated in plastic greenhouse located in Pyeongchang. The temperature of cooling water was controlled by heat pump and maintained at the range of $15{\sim}20^{\circ}C$. Cooling pipes were installed in root zone and very close to crown. Spot cooling effect was estimated by applying system in three cases which were cooling root zone, crown plus root zone, and crown only. White low density polyethylene pipe in nominal diameter of 16 mm was installed on crown spot, and Stainless steel flexible pipe in nominal diameter of 15A was installed in root zone. Crown and root zone cooling water circulation was continuously performed at flowrates of 300 ~ 600 L/hr all day long. Strawberry yields by test beds were surveyed from Aug. 1 to Sep. 30. The accumulated yield growth rate compared with a control bed of crown cooling bed was 25 % and that of crown plus root zone cooling bed was 25 % and that of root zone cooling bed was 20 %. The temperatures of root spot in root zone cooling was maintained at $18{\sim}23.0^{\circ}C$ and that of crown spot in crown cooling was maintained at $19{\sim}24^{\circ}C$. Also, the temperatures of root spot in crown plus root zone cooling bed was maintained at $17.0{\sim}22.0^{\circ}C$ and that of crown spot was maintained at $19{\sim}25^{\circ}C$.

Effects of Straw Mulching and Nitrogen Fertilization on the Growth of Direct Seeded Rice in No-tillage Rice / Vetch Cropping System

  • No-till direct seeding cultivation of rice has major advantages such as saving of labor and cost by eliminating tillage, preparation of seed bed and trans-planting procedure compared to the conventional transplanting cultivation. A field experiment was conducted to evaluate the effects of straw treatment and nitrogen levels on the rice growth in no-till direct-seeding cultivation. Rice straw, vetch straw, and the mixture of both of the straws were mulched on the surface of soil before seeding while 4 levels of nitrogen fertilizer, 0, 7, 9, and 11 kkg/10a respectively, were applied at 3 split times, 3-weeks after sowing, 5-weeks after sowing and the panicle initiation stage. Mulching of vetch straw significantly reduced seedling establishment of rice which may be attributed to low oxidation-reduction potential of soil by vetch mulching treatment. Vetch straw increased the concentration of soil ammonium leading to an extension of the greenish leaf to panicle initiation stage. Agronomic nitrogen use efficiency (AD $E_{N}$) in heavy-mixed straw mulching plots was lower than other treatments. Grain yield and AU $E_{N}$ in the vetch treatment were less affected by fertilized N levels. Conclusively, it is suggested that heavy straw mulching was not efficient for rice seedling establishment and nitrogen usage.e.

Unrecorded Fungal Species Isolated from Indoor Air in the Log Bed- and Sawdust Media-based Mushroom Cultivation Houses (원목 및 톱밥배지 버섯 재배사 내 실내 공기서 분리한 미기록 진균 보고)

  • Oak mushroom is cultivated using logs and sawdust media as substrates. In this study, fungi were isolated during a monitoring of indoor air in the oak mushroom cultivation houses located in Cheongyang-gun of Chungnam, Geoje-gun of Gyeongnam, Gumi-si of Gyeongbuk, Jangheung-gun of Jeonnam and Yeoju-si of Gyeongggi-do. Identification of the fungi based on morphology and molecular analysis of the internal transcribed spacer region and 28S rDNA, translation elongation factor translation elongation factor 1 a gene, and ${\beta}-tubulin$ gene revealed that six fungi, Cenangium acuum, Neopestalotiopsis surinamensis, Metarhizium marquandii, Periconia macrospinosa, Trichoderma petersenii, and Trichoderma paratroviride that have not been recorded previously in Korea.

Optimization of the Bacteriophage Cocktail for the Prevention of Brown Blotch Disease Caused by Pseudomonas tolaasii

  • Brown blotch disease, caused by Pseudomonas tolaasii, is one of the most serious diseases in mushroom cultivation, and its control remains an important issue. This study isolated and evaluated pathogen-specific bacteriophages for the biological control of the disease. In previous studies, 23 varieties of P. tolaasii were isolated from infected mushrooms with disease symptoms and classified into three subtypes, Ptα, Ptβ, and Ptγ, based on their 16S rRNA gene sequences analysis and pathogenic characters. In this study, 42 virulent bacteriophages were isolated against these pathogens and tested for their host range. Some phages could lyse more than two pathogens only within the corresponding subtype, and no phage exhibited a wide host range across different pathogen subtypes. To eliminate all pathogens of the Ptα, Ptβ, and Ptγ subtype, corresponding phages of one, six, and one strains were required, respectively. These phages were able to suppress the disease completely, as confirmed by the field-scale on-farm cultivation experiments. These results suggested that a cocktail of these eight phages is sufficient to control the disease induced by all 23 P. tolaasii pathogens. Additionally, the antibacterial effect of this phage cocktail persisted in the second cycle of mushroom growth on the cultivation bed.

Evaluation of Growth and Yield When Harvesting Italian Ryegrass Transplanted After Cultivation of Paddy Rice

  • Seed production of italian ryegrass has a problem of lodging during ripening and a decrease in quality due to difficulty in drying seeds during harvest. Therefore, in order to produce high-quality Italian ryegrass in paddy fields, it was carried out to reduce the density and solve the lodging problem through transplanting. In this experiment, Lolium multiflorum cv. Kowinearly was transplanted in autumn from a paddy field in Sindong-ri, Gwansan-eup, Jangheung-gun, Jeollanam-do. var. Kowinearly was made into a bed at 90 g/box and stacked in boxes. It was transplanted on October 27th after 2 days of germination at 30℃ and 15 days of seedling and greening. When transplanting, they were transplanted at intervals of 30×14cm. The existing cultivation method, drill seedling, was sowed at a level of 50 kg/ha, and both transplanting and drilling were carried out at a nitrogen fertilization rate of 45 kg/ha. The number of ears during transplant cultivation was 1,016/m2 and the drilling tended to be higher at 2,278/m2, but this was probably due to the difference in seeding amount. The seed number of an ear tended to be 56% higher in transplantation, which had a significant impact on yield. The seed yield was 2,096 ka/ha in transplantation, which was 21% higher than that of drilling. When looking at the relationship with weed occurrence, there were areas where all the weeds, such as amul foxtail, occurred due to the low density. Even in the same transplanting area, the seed yield was about 1,000kg/ha less in the area where the weeds were abundant. It seems that weed management is important in paddy cultivation. Therefore, it seems necessary to develop an exclusive herbicide for Italian ryegrass cultivation.
