In order to explain the connection between ginseng rust spot and soil ecological conditions, the bed soils and ginseng roots were sampled at different microrelief units and the reducing substances of the bed soils and iron forms of the ginseng root epi dermises were determined. The results showed that the occurrence of the rust spot was connected with the ecological conditions of the soils and the metabolism of the plant which was caused by the excessive $Fe^{2+}$ in the soil solution. Ginseng rust spot was the enrichment of iron which was mainly composed of organic complex irons. Including active ferrous active ferric and non active ferric forms and they were transformed into each other following the change of soil moisture and temperature regimes. According to the regularity of growth and decline of reducing substances in soil and rust index of ginseng roots as well as the difference of adaptability to excessive $Fe^{2+}$ in soil among different year-old seeding, a new comprehensive measure based on the connection of ameliorating soil and improving cultivation system was recommended to prevent the occurrence of ginseng rust spot.
${\gamma}$-Aminobutyric acid(GABA) is a four carbon non-protein amino acid that has several well-known physiological functions, such as a postsynaptic inhibitory neurotransmitter in the brain and induction of hypotensive and tranquilizer effects. A lactic acid bacterium was isolated from button mushroom bed, which is showing high GABA productivity by TLC or HPLC analysis. The strain was identified as Lactobacillus hilgardii by analysis of 16S rDNA gene sequence. When the maximum production of GABA by L. hilgardii was investigated with various concentration of monosodium glutamate, the yield of GABA reached to be 53.65 mM at 1% mono sodium glutamate (MSG) in flask cultivation. A Glutamate decarboxylase (GAD) enzyme, which was known to convert MSG to GABA, was purified from a cell-free extract of L. hilgardii and the molecular weights of purified GAD was estimated to 60,000 by SDS-PAGE. The optimum pH and temperature of GAD were at pH4.6 and at $37^{\circ}C$, respectively. The GAD activity was increased by the addition of sulfate ions such as ammonium sulfate, sodium sulfate and magnesium sulfate, indicating that the increase of hydrophobic interaction causes the increase of GAD activity.
Sodium hypochlorite alkaline was tested against Pseudomonas tolaasii causing bacterial blotch on cultivated oyster mushroom (Pleurotus ostreatus). The minimum inhibitory concentration of sodium hypochlorite against P. tolaasii contained active chlorine (AC) at 1.4 mg/l on plate assay. The highest cultivation yield was obtained from the treatment of AC 5.7 mg/l. Treatment of sodium hypochlorite at the rate of higher than AC 11.4 mg/l resulted in reduced yields at the harvest. However, the population of total bacteria on the bed surface treated with AC 5.7 mg/l of sodium hypochlorite was maintained to some extent. Inhibitory concentration against total bacteria on the bed surface was over AC 22.8 mg/l. Mushroom mycelium was damaged and its growth strongly inhibited at the concentration of AC 200 mg/l. Mushroom caps showed yellowish symptom by chemical injury by treatments of AC 74.1 mg/l or higher. Sporocarps infected by P. tolaasii were irrevocable at any concentration of sodium hypochlorite. Routine watering with AC 5.7 mg/l from mushroom initiation to the end of picking resulted in reduced bacterial blotch incidence of 40% and 86% at two mushroom farms. The treatment resulted in higher quality mushroom production compared to that conventionally watered with tap water alone.
This experimemt was conducted to determine the effect of ridge-up bed with different height (0, 20, 40, 60 cm) on the fruit quality of 'Miyagawa Wase' satsuma mandarin cultivated in a plastic film house. Soil moisture was measured at the soil depths of 10, 30, and 60 cm for each height of ridge-up bed. The time required to reach the condition, -1.4 Mpa at 30 cm deep in soils, which are supposed to be the ideal soil moisture potential and soil depth for high quality satsuma mandarin production, was 100, 60, and 30 days for 20, 40, and 60 cm ridge-up bed, respectively, and more than 100 days for 0 cm. Peel chromaticity by 'a' value was increased as the ridge-up height went up, so that plants grown at the bed with 60 cm height had 11 degree higher value than those of plants grown at the bed with 0 cm height. The 'a/b' values also had a similiar trend as value 'a'. The reducing sugar level of the fruit juice, which was composed of glucose and fructose, was increased as the height of ridge went up, showing significant difference between the ridge heights. The sucrose level had the same trend as the reducing sugar level although the difference between the ridge heights were not significant. Generally, the soluble solid level is considered to be representing the fruit quality. It had 11.4, 12.1, 12.5, $12.8^{\circ}Brix$ for 0, 20, 40, and 60 cm ridge-ups, respectively, showing $1.4^{\circ}Brix$ difference between 0 and 60 cm ridges. Acidity basically showed the same result as that of the soluble solid level depending on the height of the ridges.
Since 1990, destructive yellow rots on Ganoderma lucidum caused by a soilborne fungus have been occurred in major cultivation areas of Korea. Incidences of the disease were 61% in Chulwon and 94% in Kanghwa area where the mushroom has been cultivated for 10 years, whereas the disease has not been found yet in new cultivation areas such as Moonkyung and Hongsung. when severely infected, inner tissues of bed-logs showed severe yellow and fruiting bodies of the mushroom was not produced. Infected tissues of bed-logs were readily distinguished from those of healthy ones by a distinctive brown border line. When the disease progressed, mycelia of Ganoderma lucidum were totally destroyed, and abundant ascocarps of the pathogen were formed inside the tissues of bed-logs showing yellowish green. The fungus derived from a single ascospore strongly lysed mycelia of Ganoderma lucidum growing on bottle media, and non-volatile components secreted by the pathogen were also highly inhibitory to mycelial growth of the mushroom fungus. The pathogen was identified as Arthrographis cuboidea based on its distinctive cultural and morphological characters. The fungus produced arthroconidia and unbrached conidiophores. The width of fungal conidia was distinctively wide as compared with the length. Colonies of the fungi were pale yellow to yellowish green on agar media. As a causal pathogen of yellow rot of Ganoderma lucidum., this fungus has not been reported yet in Korea.
Yields of ginseng seedlings and cultivation methods were investigated in 29 Ban-Yang-Jik (semimodified soil) nurseries to obtain some information about the production of ginseng seedling. The average number of available seedlings (useful for transplanting in the main field) per Kan ($180{\times}90$ cm) was 362. The percentage of available seedlings to the total seedlings harvested (rate of available seedlings) was 45 %. Although there were severe variations in the numbers and rates of available seedlings among the nurseries surveyed, the number of available seedlings were reduced due to both the short weighted and poor shaped (obese shape with poor root development) seedlings. The number of poor shaped seedlings was negatively correlated with height of seed-bed and frequencies of irrigation. Quadratic relations were noticed between rate of available seedling and heights of front post and rear post of shade. On the other hand, negative correlation was recognized between rate of available seedling and breadth of shade, but positive correlation was noted between rate of available seedling and frequencies of irrigation.
This study attempted to estimate the degree of management of oak mushroom farms in Chungcheongnam-do and to provide information to establish oak mushroom cultivation-related policies. The oak mushroom management standard diagnostic table consists of three major categories, growing condition, inoculation management, cultivation management and management administration, along with 20 subcategories. Thus, 209 households of oak mushroom farms were surveyed from 2015 to 2018 in Gongju, Cheongyang, Buyeo and Seochun in Chungcheongnam-do. The average score for the 20 subcategories was 71.5 points (representing a significant level), indicating that these areas have excellent management conditions. The analysis of the management performance indicators revealed a high number of indicators with scores of five or above. The total score was higher, and the amount per bed log and the rate of top-grade products in the total output were also higher, indicating a significant correlation. These findings will provide consulting services to oak mushroom growers as they highlight the correlation between the higher scores of indicators in the oak mushroom management standard diagnostic table and the management performance of farmers. We found that the scores of the indicators for management administration, such as management record and analysis and fund plan were relatively lower than those of other indicators. It is assumed that the owners aging has led to negligence in recording the details on incomes, expenditures, and work and lowered the willingness to make substantial profits. Therefore, it is essential to overcome these problems for profitable oak mushroom farming.
Ground bed cultivation method for shiitake mushroom (Phogo; Lentinula edodes) has been advanced to use the oak-sawdust composted by microorganisms. This method has some advantages in saving the labors and improving productivity. The aim of this study is to analyze microbial community in oak sawdust and to provide the information for efficient cultivation of shiitake mushroom. In process of composting, the proportions of thermophiles increased from 10% to 80%. This results show the microbial community succession from mesophilic bacteria to thermophilic bacteria. T-RFLP analysis and nucleotide sequencing of 16S rRNA gene demonstrated that the change of dominant bacteria coupled with shifts in each stages. The family of Enterobacteriaceae as a mesophilic bacteria were dominant (100%) in oak sawdust sample, but Amycolatopsis (49.0%) and Saccharopolyspora (26.5%) as a thermophilic Actinomycetes were dominant in the last day of composting. In hyphal growth stage, Leuconostoc (psychrophilic bacteria) was dominant (75.0%). The composting process of sawdust is very important for growth of mushroom mycelium. Therefore, there is need for the further study for the Amycolatopsis as thermophilic Actinomycetes.

Plant factories are plant cultivation systems which produce farm products uniformly under the controlled environmental condition regardless of seasons and places. Thermal flow in the plant factory is an important parameter in cultivating plants. In this research, we study thermal flow characteristics for a hybrid plant factory with multi-layer cultivation shelves using computer simulation techniques. In order to obtain numerical solutions for thermal flow characteristics, a finite volume method was applied. We consider a low-Reynolds-number ${\kappa}-{\epsilon}$ turbulence model, incompressible viscous flows, and pressure boundary conditions for numerical simulation. Commercial software Solid Works Flow Simulation is then used to investigate characteristics of thermal flows in the plant factory applying several different inflow air velocities and arrangements of cultivation shelves. From numerical analysis results, we found that temperatures in cultivation shelves were uniformly distributed for Case 3 when the inflow air velocity was 1.6 m/s by using a blower in the plant factory. However in Case 1 lower temperature distributions were observed in test beds, TB2 and TB3, which indicated that additional temperature control efforts would be required. Average shelf temperature increased by $3^{\circ}C$ using artificial light source (DYLED47) with 50% blue and 50% red LED ratios. Korea Academia-Industrial cooperation Society.

This study investigated a suitable method that could be applied for Asian chain fern [Woodwardia japonica (L. f.) Sm.] to propagate gametophytes and promote sporophyte formation. The gametophytes used in all experiments were obtained from germinated spores in vitro and were subcultured at 8-week intervals. The most appropriate media for gametophyte propagation was identified by culturing 300 mg of gametophyte in Murashige and Skoog (MS) basal medium (1/8, 1/4, 1/2, 1, 2), and Knop medium for 8 weeks. As a result, fresh weight of the gametophyte was increased by 56.7-fold on MS medium. Moreover, antheridium formation as well as gametophyte growth was improved on MS medium, especially. To improve the sporophyte formation ex vitro, 1.0 g of gametophyte was ground with distilled water and spread on eight combinations onto four different culture mediums, such as bed soil, peat moss, perlite and decomposed granite. Then generation and growth of sporophytes were investigated after cultivation for 10 weeks. As a result of this experiment, peat moss had a promotive effect of sporophyte formation at single-use and mixed culture soils. In particular, a mixture of bed soil, peat moss and perlite in a 1:1:1 ratio (v/v/v) led to the accelerated formation (782.5 ea/pot) and the frond growth of sporophytes. This included increases in length and width of fronds. However, promotive effect of gametophyte growth and sporophyte formation was not found at single-use and treatment with high ratio of bed soil.
