Anthracycline계 항암성 항생물질 DA-125의 Beagle dog에 대한 26주 반복정맥투여독성시험 (Toxicity Studies of DA-l25, an Anthracycline Antitumor Antibiotic : Intravenous Repeated Doses for 26 Weeks in Beagle Dogs)

  • This study was performed to investigate the toxicity of DA-125 in beagle dogs, an anthracycline antitumor antibiotic. The dogs were administered DA-125 i.v. at 0.0023, 0.0375, 0.15 and 0.6 mg/kg/day, 6 days/week for 26 weeks. At 0.6 mg/kg, all male and female dogs were either sacrificed moribundly or dead during the 26-week treatment. The dogs revealed inactivity, salivation, dark bloody discharge, swelling of the subcutaneous injection site, abscess, and ulceration in the abdominal wall and legs. At 0.15 mg/kg, anorexia, salivation, and swelling of the injection site were observed. The food consumption was decreased with a statistical significance at 6 and 12 weeks treatment in males of 7.6 mg/kg. At 0.0375, 0.15 and 0.6 mg/kg, body weights were decreased significantly in a dose-related fashion after 17 weeks treatment. Total white blood cell counts for male dogs at 0.6 mg/kg were lower than those of control dogs after 13 weeks treatment, which appeared mainly due to decreased neutrophils. At 0.15 mg/kg, testicular atrophy was found in all males by gross pathology and the testicular weights were significantly decreased when compared to those of control males. Microscopically, the testis showed moderate atrophy of the seminiferous tubules and marked decrease in number of spermatozoa in the epididymal tubules. At 0.6 mg/kg, petechia or echymotic hemorrhage was observed in gastrointestinal tract, heart, lungs, and other organs at the necropsy, Marked atrophy of thymus were observed in both males and females. In addition, severe testicular atrophy was noted in all males. Microscopically, gastrointestinal tract showed hemorrhage, epithelial denudation, hypermucus secretion, and atrophy of intestinal villi. Seminiferous tubules of the atrophic testis were lined with Sertoli cells only and devoid of germ cells. Severe oligospermia or aspermia was present in the epididymal tubules. Bone marrow showed marked depletion of hemopoietic cells. In addition, marked atrophy was found in the lymphoid tissue of gastrointestinal tract, various Iymph nodes, and thymus. Injection sites showed marked inflammatory response with necrosis, necrotizing vasculitis, thrombus formation, and ulceration in the skin. According to the present results, no observed effect level appeared to be 0.0375 mg/kg. At 0.15 mg/kg, testis was a target organ, while at 0.6 mg/kg hemopoietic tissue, gastrointestinal tract, and testis were considered to be target organs. At 0.6 mg/kg the test compound seems to inflict a damage on the blood vessels causing hemorrhage in the various organs and tissues.

  • Ursodeoxycholic acid(UDCA) is a hydrophilic gall bladder acid and has been used as a effective drug for liver disease related to in1munity. This drug inhibits secretions of IL-2, IL-4, and $IFN-{\gamma}$ from T-cells and production of immunoglobulin from B-cells. Also it has been reported that UDCA inhibits production of IL-1 related to the progression of periodontal disease and activation of collagenases. The purpose of the present study was to elucidate the effects of UDCA on inhibition of periodontal disease progression using clinical, microbiological and histometrical parameters. Twelve pure bred, 16 month-old-beagle dogs were used in the study. After ligature-induced periodontal diseases were formed, experimental drugs were applied twice a day and then the results of clinical, microbiological, and histometrical parameters were measured at baselie(initiation of experiment) , 4weeks and 8weeks. The gel with UDCA(concentration 0.5%, 5% 3 dogs in each) was applied to experimental group, chlorhexidine to positive control group(3dogs) and the gel without UDCA(base) to negative control group. After induction of general anesthesia, the maxillary 2nd, 3rd premolars and 1st molar and the mandibular 2nd, 3rd, 4th premolars and 1st molar were ligated in one side selected randomly and were not ligated in the opposite side. The plaque index(PI), gingival index(GI), pocket depth(PD) and gingival crevicular fluid(GCF) volum were measured clinically. The PI and GI were measured at 3 buccal points of all experimental teeth and the GCF was measured only at the 3rd premolar in the maxilla and the 4th premolar in the mandible. In the microbiological study, the samples extracted from the 3rd premolar of the maxilla and the 4th premolar of the mandible at the center of buccal surface were analyzed aerobics, anaerobics and Streptococcus colony forming units, After clinical and microbiological examination at 8weeks, the dogs were sacrificed by carotid artery perfusion. The samples were fixed and sectioned including interproximal area, and the distance from cementoenamel junction(CEJ) to alveolar crest was measured. The results were that PI, GI and PD increased until 4 weeks and decreased at 8 weeks in three groups but the differences between all the groups were not significant. The 0.5% UDCA in non-ligated group showed remarkable decrease of GCF. The experimental group applied 5% UDCA decreased the number of aerobics and anaerobics. The distance from CEJ to alveolar crest was greater in the negative control group on both ligated and non-ligated sides, but the differences were not significant stastically.

개의 실험적 급성 괴사성 췌장염 평가에서 Balthazar Computed Tomographic Severity Index의 적용 (Balthazar Computed Tomographic Severity Index Application for Experimental Acute Necrotizing Pancreatitis in Dogs)

  • 컴퓨터 단층촬영술(computed tomography; CT)은 인의에서 췌장염의 진단 기준으로 이용되고 있으며, Balthazar CT severity index (CTSI)는 CT 영상을 바탕으로 췌장 괴사 부위를 측정하여 췌장염의 심각도를 평가하는 지수이다. 본 연구에서는, 25마리의 비글견을 정상 그룹과 대조군, 자가 담즙 주입을 통해 급성 괴사성 췌장염을 유발한 실험군으로 나누어 Balthazar CTSI 측정 결과와 조직 검사 결과를 비교하였다. 정상 개의 췌장의 Hounsfield unit (HU)은 $52.44{\pm}4.58$ 이었고, 급성 괴사성 췌장염에서는 유의적으로 감소하였다(P < .05). 조영 증강 CT 영상에서 확인한 췌장 실질의 괴사 부위를 찾아 Balthazar CTSI를 계산한 결과 조직 검사 결과와 밀접한 상관 관계를 보였으며, 민감도 100%, 특이도 88.89%로 확인되었다. Balthazar CTSI는 급성 췌장염이 발생한 개에서 심각도 평가에 유용한 방법으로 판단된다.

Evaluation of Skin Microcirculation by Laser Doppler Flowmeter in Healthy Beagle Dogs

  • The cutaneous microcirculation plays a role in various physiological processes and pathological conditions. Two non-invasive methods were used in this study to obtain reference values for cutaneous microcirculation in intact male beagles. Twenty intact male beagles were used. The experimental environments were standardized. Laser Doppler flowmetry was used to measure cutaneous blood flow, and an infrared thermometer was used to measure cutaneous temperature. The blood flow and temperature were measured from the right side of the subjects at 20 cutaneous sites. Based on the laser Doppler flowmetry, the region with the highest blood flow was the periocular region that with the lowest was the forelimb foot pad. In addition, the standard deviation of the chest wall was the highest while that of the periocular region was the lowest. For skin temperature, the inguinal region had the highest mean skin temperature and the forelimb foot pad had the lowest. The correlation coefficient between the two methods was 0.72. Similar to a previous study, the values derived from repeated measurements at the 20 regions are reproducible and can contribute to research. Compared to the results of a previous study, the temperatures of the two smallest skin regions were the same; however, no specific trend was observed. The correlation coefficient between the two methods was significantly comparable, and this good correlation can reduce their limitations and variables complementarily. In addition to possible use in human studies, accumulated resources on measurements of skin blood flow in the future will potentiate its use in the veterinary medicine field.

Effect of the respiratory rate on the pulse pressure variation induced by hemorrhage in anesthetized dogs

  • Background: Studies on anesthetized dogs regarding pulse pressure variation (PPV) are increasing. The influence of respiratory rate (RR) on PPV, in mechanically ventilated dogs, has not been clearly identified. Objectives: This study evaluated the influence of RR on PPV in mechanically ventilated healthy dogs after hemorrhage. Methods: Five healthy adult Beagle dogs were premedicated with intravenous (IV) acepromazine (0.01 mg/kg). Anesthesia was induced with alfaxalone (3 mg/kg IV) and maintained with isoflurane in 100% oxygen. The right dorsal pedal artery was cannulated with a 22-gauge catheter for blood removal, and the left dorsal pedal artery was cannulated and connected to a transducer system for arterial blood pressure monitoring. The PPV was automatically calculated using a multi-parameter monitor and recorded. Hemorrhage was induced by withdrawing 30% of blood (24 mL/kg) over 30 min. Mechanical ventilation was provided with a tidal volume of 10 mL/kg and a 1:2 inspiration-to-expiration ratio at an initial RR of 15 breaths/min (baseline). Thereafter, RR was changed to 20, 30, and 40 breaths/min according to the casting lots, and the PPV was recorded at each RR. After data collection, the blood was transfused at a rate of 10 mL/kg/h, and the PPV was recorded at the baseline ventilator setting. Results: The data of PPV were analyzed using the Friedman test followed by the Wilcoxon signed-rank test (p < 0.05). Hemorrhage significantly increased PPV from 11% to 25% at 15 breaths/min. An increase in RR significantly decreased PPV from 25 (baseline) to 17%, 10%, and 10% at 20, 30, and 40 breaths/min, respectively (all p < 0.05). Conclusions: The PPV is a dynamic parameter that can predict a dog's hemorrhagic condition, but PPV can be decreased in dogs under high RR. Therefore, careful interpretation may be required when using the PPV parameter particularly in the dogs with hyperventilation.

SLA로 표면처리한 microthreaded crest module 임프란트의 임상적, 조직학적, 조직계측학적 분석 (Clinical, histologic, and histometric evaluation of implants with microthreaded crest module and SLA surface)

  • Background: Crestal bone loss known as saucerization is a frequently observed phenomenon. Recent studies have shown that implants with micothreaded crest module reduced the force concentration in the crestal region thus resulting in no or reduced crestal bone loss. This study presents a clinical, histologic, and histometric evaluation of implants with microthreaded crest module and SLA surface. Methods: The implants were placed in the mandible of 5 beagle dogs weighing 10-15kg. Four premolars were bilaterally extracted 8 weeks prior to implant placement. Mucoperiosteal flap was elevated and drilling with increasing diameter was performed under saline irrigation. After countersinking 2 implants were placed in each side resulting in 4 implants per dog. Healing period of 8 weeks was allowed before sacrificing the animals. Histologic preparation was performed for histologic and histometric analysis. Bone to implant contact as well as percentage of bone area inside threads were measured. T-test was used for statistical analysis with pvalue p<0.05. Results: 1. Healing was uneventful without any cover screw exposure. New bone formation around the implants was observed without any inflammatory infiltration. 2. Bone to implant contact in the microthread and thread were 43.90 ${\pm}$ 20.30 %, and 53.19 ${\pm}$ 20.97 % respectively. The overall bone to implant contact was 48.54 ${\pm}$ 20.95 %. 3. Percentage of bone area inside threads were 54.43 ${\pm}$ 10.39 %, and 38.44 ${\pm}$ 16.44 % for the microthread and thread respectively. There was statistically significant difference(p<0.05). The overall percentage of bone area inside threads was 46.67 ${\pm}$ 15.68 %.

  • 인간의 감성을 자극하는 대상에는 여러 가지가 있을 수 있을 수 있는데, 첫 째가 바로 우리와 같은 사람이고 그 중에서도 아리따운 여성과 귀여운 아기들이다. 그 다음 순위가 바로 인간의 동반자로 자리매김한 개과 동물이다. 이는 광고에서 많이 쓰이고 있는 3B법칙(미인 Beauty, 아기 Baby, 동물 Beast - 긍정적인 이미지 덕분에 광고나 선거에서 많이 사용)인데, 이러한 관계는 미술사에서도 오랜 기간 표현되고 있다. 인간과 함께 1만년 이상의 역사를 살아 온 개는 사냥의 임무이자, 목동의 양 떼를 지키거나 농부의 집과 재산을 지키는 임무를 지녔던 인간의 조력자에서, 동반자로서 반려동물이라는 주요한 역할과 함께 현대 도시 문화 안으로 들어오게 되었다. 이처럼 개는 유목과 목축, 시골과 도시 생활 등 여러 가지 상황에 끊임없이 적응해왔는데, 이 논문은 인간과 반려동물이 맺고 있는 밀접한 관계를 수많은 예술가들의 손을 거쳐 나타난 개와 강아지의 다양한 도상들을 통하여 탐구하고자 한다. 여기서 반려동물이라는 뜻은 사람과 함께 더불어 살아가는 동물(companion animal)이라는 뜻인데, 서로가 도움을 주는 일방통행이 아닌 왕복의 관계를 나타낸다. 따라서 필자는 각 시대별로 생존을 위한 가장 훌륭한 사냥꾼의 모습부터, 왕실의 품격으로 우대 받았던 모습, 그리고 기쁨을 선사하며 현대인들에게 일상의 행복감을 전하는 마스코트와 같은 모습을 개, 강아지의 시각적 신호인 '표정'이라는 논의로 담론화 해보고자 한다. 20세기에 들어서는 종종 스크린과 대중매체의 영향을 받아왔는데, 비글, 달마티안이 유행한 시기에는 <피너츠>의 주인공 스누피와 영화 <101 달마티안>이 영화관과 텔레비전에서 성공을 거둔 때와 일치한다. 이런 주인공들은 관객들로 하여금 절로 행복감에 젖어들게 하는데, 이처럼 현대 형상 예술에서도 작가와 독자 간의 소통으로 이루어진 개, 강아지의 도상을 통하여 인간과 개가 나눈 끈끈하고 오랜 우정을 다시 확인해 보고, 각박한 현대 사회를 살아가는 현대인들에게 심리적 안정효과를 기대해 볼 수 있는 계기가 되길 희망해본다. 따라서 인간과 개의 교감을 연구하는데 있어서 본 연구 논문이 새로운 텍스트로 거듭나고 향후 개와 인간과의 여정에서 효과적인 커뮤니케이션으로 확대되기를 기대해본다.

개에서 Pentoxifylline 첨가에 따른 동결정액 성상과 인공수정에 관한 연구 (Studies on Frozen Semen Characteristics Following Pentoxifylline Treatment and Artificial Insemination in Dog)

  • 본 연구는 개 정액 채취방법에 따른 정액성상과 동결전 pentoxifylline의 첨가가 미치는 영향을 조사하였고, CASA를 이용하여 정자의 운동성 측정 및 생존율, 첨체 온전성, 저삼투성 팽창화를 조사하였다. 그리고 자연발정과 발정유기된 암캐에서 동결정액 주입의 임신율을 조사하였다. 1.품종별 정액성상을 조사한 결과 Beagle, English cockers spanie, Collie 및 Grey hound의 정액량과 정자 수에서 큰 차이가 없었으나, Shihtzu에서 정액량과 정자수가 유의적은 아니나 다소 적었다. 2.동결희석액에 pentoxifylline 0, 1 및 3mM 첨가에서 융해 후 정자활력은 각각 46.4, 56.4 및 47.2%였으며 1mM 처리구가 다른 처리구보다 유의적으로 높았다. 3.동결희석액에 pentoxifylline 첨가는 융해 후 정자의 생존성, 첨체막 온전성 및 원형질막 온전성(HOST) 모두 유리한 결과를 나타냈으며, 1mM의 첨가에서 정자의 생존성과 기능성이 향상되었다.4.자연발정에서 발정개시 후 11일, 13일 및 15일째 초음파기로 측정한 난포 크기는 각각 6.5mm, 11.5mm 및 8.0mm이였다. 배란은 13-15일 사이에 일어났다. 5.발정유기한 개체에서 수정후 임신 20일째부터 5일 간격으로 3회 측정한 태낭의 크기는 13.7mm, 28.5mm 및 40.5mm 이었다. 6.자연발정과 발정을 유기한 개체에서 pen- toxifylline 1mM이 첨가된 정액을 동결융해하여 인공수정 후 임신율은 71.4%와 75%로 두 방법간에 차이가 없었으나, 자연발정군에서 평균산자수는 6.6두였고, 발정유군의 평균산자수는 2.6두였다.

척수손상 모델 견 표준화를 위한 방사선, 자기공명영상 및 조직 병리 평가 (Radiographic, MRI and Histopathologic Assessment to Standardize Canine Spinal Cord Injury Model)

  • 대부분 척수손상 모델에서의 척수 손상 정도 평가는 자기공명 영상 등을 통한 유발 후 평가를 실시하고 있으며, 유발 전 평가를 위해 풍선 카테타에 주입된 공기 양을 기준한 예가 있으나 종 특이성과 개체 차이를 고려하지 못하는 단점이 있다. 이러한 단점을 극복한 척수손상 기준모델을 제시하기 위해 본 실험을 실시하였다. 방사선 평가를 통해 요추 1 번 척수강 높이가 8 mm로 측정된 임상적으로 건강한 비글견 8 마리를 풍선카테타의 직경과 척수 압박시간을 기준으로 4개 군 (4 mm/3 시간, 4 mm/6 시간, 4 mm/12 시간 그리고 6 mm/3 시간)으로 구분하였다. 손상 정도는 행동 관찰, 자기공명영상 해석, 체성감각유발전위평가 그리고 병리조직검사를 실시하여 평가하였다. 실험결과, 행동평가와 체성감각유발전위평가는 단지 손상 유발 여부만 지시할 뿐 정도 평가에는 유용하지 못하였다. 자기공명영상 평가에서 척수손상 부위는 단시간반전회복영상과 T2강조영상에서 불균질한 고신호강도 영역으로 관찰되었다. 고신호강도 영역은 삽입된 풍선 직경과 압박시간 증가에 따라 보다 확장되어 관찰되었으며, 이러한 소견은 공포화 등의 손상부위 증가와 카스파제-3 및 PARP 면역반응 세포의 수적 증가로 나타난 병리조직검사 결과와 일치하였다. 이러한 결과로 미루어 정형화된 척수손상 모델 유발을 위한 척수강 직경과 풍선카테타 직경 그리고 압박시간의 변수 이용과 손상 정도 평가를 위해 자기공명영상은 매우 유용할 것으로 판단된다.
