Removal of Heavy Metals, Cd(II) and Pb(II) Ions in water by Sargassum Herneri (괭생이 모자반에 의한 수중 중금속 Cd(II), Pb(II) 이온의 제거)

  • Brown algae, Sargassum horneri, was used as an adsorbent to remove heavy metal Cd(II) and Pb(II) ions. Sargassum horneri picked in East Coast was formulated into the grain with the size of 40-60 mesh after wind dry. Batch method was used in order to investigate the adsorption rate by measuring the adsorption amounts with shaking time. In the column method, the adsorption amounts were measured by flowing metal solutions into the algae-packed column at the rate of 1 ml/min. Adsorption amounts in both batch method and column method were in the following order : pH 10.5>7.0>3.5. It was found that Pb(II) was more adsorbed on the algae grain than Cd(II). It was also revealed that the adsorption amounts reached the maximum within 5 minutes irrespective of pH condition in the batch method. It was concluded that the batch method was more effective than the column method in terms of recovery rate.

Production of Mycelia and Water Soluble Polysaccharides from Submerged Culture of Lentinus lepideus in Bioreactor (생물반응기를 이용한 잣버섯(Lentinus lepideus)의 균사체 및 수용성 다당체 생산특성)

  • The mushroom Lentinus lepideus was used to produce mycelial as well as soluble polysaccharides in bioreactor cultures. To determine optimal submerged culture conditions, both growth characteristics and water soluble polysaccharides production were compared among four different types of bioreactor and culture conditions. For the production of mycelial biomass, the following bioreactors were proven to be effective in decreasing order: an external-loop type air-lift bioreactor (ETAB; 7g/l), a balloon type air bubble bioreactor (BTBB; 6.2g/l), a stirrer type bioreactor (STB; 6g/l), and a column type air bubble bioreactor (CTBB; 5g/l). Maxiaml production of water soluble exopolysaccharides (EPS; 0.62g/l) and endopolysaccharides (PPS; 7.7%) could also be obtained from BTBB. The mycelial biomass increased with increase in glucose concentration from 15g/l to 75g/l in the media. In contrast, PPS contents in the cells decreased with increase in glucose concentration in the media, showing the highest PPS content (7%) at 15g/l. Among different medium feeding types, fed-batch culture based on concentration control in media (10g/l) produced higher mycelia than fed-batch culture based on volume control of media (5.8g/l) or batch culture (3.4g/l). EPS production was also higher in fed-batch culture based on medium concentration control than that in other feeding types.

Effect of Ozone Treatment for Nakdong River Raw Water I. Efficiency of Water Treatment by Conducting Batch Test of Ozonation Experiments (낙동강 상수원수의 오존처리 효과 I. 회분식 오존처리에 의한 수처리효과)

  • This study was carried out to evaluate the pollutant removal efficiencies of the advanced drinking water treatment using ozonation. for raw water, Nakdong River was used. By conducting batch ozonation test, the following results were obtained. when ozone dosage of 5mg/$\ell$was used, preozonation of raw water reduced turbidity, KMnO$_4$consumption, DOC(dissolved organic carbon), UV254 absorbance, THMFP(trihalomethane formation potential) as much as 3.9NTU, 5.5mg/$\ell$, 1.15mg/$\ell$, 0.112 and 0.065mg/$\ell$, respectively. In case of postozonation of sand filtered water, water quality was also improved with decrease in turbidity, KMnO$_4$consumption, DOC, UV254 absorbance and THMFP at the amount of 0.08NTU, 2.6mg/$\ell$, 0.88mg/$\ell$, 0.042 and 0.018mg/$\ell$, respectively. On the other hand, contents of dissolved oxygen increased at the level of 1.3mg/$\ell$ after preozonation process and 1.0mg/$\ell$ after postozonation process. The effect of ozone dosage was higher than that of its contact time for the removal of the pollutants.

  • In the present paper, batch and semi-continuous settling characteristics of a binary calcium carbonate-water system were investigated. Using experimental data of batch settling characteristics, a graphical analysis for a semi-continuous thickening column was developed and compared with experimental results on the basis of Kynch theory, only where the feed velocity line was within the limits of Kynch theorem III. The analysis showed good agreement with experiments. Quite erroneous results, however, was observed for the analysis of a sludge region on the underloaded operation, which was considered due to the deviation from the limits of theory.

Two-Stage Fed-Batch Culture of Candida magnoliae for the Production of Erythritol using an Industrial Medium (산업용 배지를 이용한 Candida magnoliae의 2단계 유가식 배양에서 에리스리톨의 생산)

  • Experiments were carried out to select an industrial nitrogen source and optimize erythritol production by Candida magnoliae in fed-batch culture. Among the industrial nitrogen sources tested, light steep water (LSW) was found to be the best nitrogen source for producing erythritol, based on erythritol yield and raw material price. The maximum erythritol concentration obtained a 131.6 g/L, with a 52.6% yield and 0.52 g/L-hr productivity from a 250 g/L glucose and 43.3 mL/L LSW in batch culture. Two-stage fed-batch culture was chosen to improve the volumetric productivity and the yield of erythritol. High cell density culture in cell growth stage was achieved by batch type culture containing 100 g/L glucose and 500 mL/L LSW. The cell concentration was 71.0 g/L after 23 hours of culture. Erythritol productivity was decreased by increasing glucose concentration in the production stage. But 37.3% of the maximum erythritol yield was obtained with 185.5 g/L of erythritol and 1.66 g/L-hr of productivity when 820 g of glucose powder was directly added to a concentration of 450 g/L glucose in production stage.

Enhancement of ${\beta}$-1,3-Glucan Functionality by Batch and Semi-continuous Typed Specific Carboxylation (회분식 및 반연속식 특이적 카르복실화 반응에 의한 ${\beta}$-1,3-glucan의 기능성 향상)

  • In this study, batch or semi-continuous reactions, introducing site-specific carboxylic acids in ${\beta}$-1,3-glucan structures, were performed to increase water solubility and gel forming ability, using TEMPO/hypobromite with or without NaBr as catalysts. Regio-selective carboxylic acid formations were determined with infrared (IR) and $^{13}C$ nuclear magnetic resonance (NMR) spectroscopic analyses. The regio-selective reactions with and without NaBr gave oxidation yields of 92.5 and 85.6%, respectively, in the batch type, and yields of 93.9 and 86.4%, respectively, in the semi-continuous type. The reaction times in the batch and semi-continuous reactions without NaBr were delayed by 100 and 150%, respectively, as compared to those with NaBr. A combination of IR and $^{13}C$ NMR analyses were used to confirm the formation of carboxylic acids in ${\beta}$-1,3-glucan. From the batch reactions with and without NaBr, the water solubilities of oxidized products were 50.0 and 55.6%, respectively, and in the semi-continuous reactions they were 52.6 and 53.5%, respectively; while the water solubility of the native ${\beta}$-1,3-glucan was less than 1.0%. Finally, as compared to the native ${\beta}$-1,3-glucan, the gel forming ability of the reaction products was greatly increased irrespective of the presence of NaBr or the reaction type.

The Effect of Sludge Settling Characteristics on Sludge-Water Interface Height Change Model (슬러지계면층 높이변화모델에서 슬러지 침전특성에 대한 영향)

  • For the further study of the solids flux theory, several researchers have proposed models to predict sludge settling velocity for each different concentration by using sludge indexes, SVI, SSVI and $SSVI_{3.5}$. It is difficult to apply the above models to predict sludge-water interface height in a batch column because sludge settling velocity changes while sludge settle down. While sludge settle down in a batch column, sludge concentration becomes high. The sludge concentration change is one of the most critical causes of the change of sludge settling velocity. Also, sludge concentration change causes of sludge index to change. SVI is more sensitive than SSVI or $SSVI_{3.5}$ to the change of sludge concentration. Each sludge has physical characteristics of its own which makes the settling velocity for each sludge different. The purpose of this study is to establish the correction factors that are able to compensate the errors derived from each different sludge settling characteristic by using sludge indexes, therefore the correction factors are applicable to the model for the change of sludge-water interface height.

Effect of Ozone Treatment for Nakdong River Raw Water - II. Removal of VOCs and Algae in Raw Water by Conducting Batch Test of Ozonation Experiments - (낙동강 상수원수의 오존처리 효과 - II. 회분식 오존처리에 의한 휘발성유기화합물 및 조류제거 효과 -)

  • This study was carried out to evaluate the pollutant removal efficiencies of the advanced drinking water treatment using ozonation process. For raw water, Nakdong River was used. By conducting batch test of ozonation, the following results were obtained. When ozone dosage of $5 mg/{\ell}$ was used, ozone transfer and utilization efficiencies of the ozonation were 94 to 92%, respectively. Removal efficiencies of single VOC compound or mixed VOC compounds in the raw water were 80% to 90% by the ozonation with $2 mg/{\ell}$ dosage and 10 minutes contact time. Removal efficiencies of ABS by the ozonation with $1 mg/{\ell}$, $3 mg/{\ell}$ dosage and 20 minutes contact time were 83% to 96%, respectively. Almost 67% of chlorophyll-a at the concentration of $38.4\mu\textrm{g}/{\ell}$ was removed by ozonation at ozone dosage of $1 mg/{\ell}$ for 20 min. Considering the efficiency of ozone utilization and water treatment, the most effective ozonation could be obtained with high ozone dosage and short contact time.
