5'-Fluoro-orotic acid를 이용한 여름느타리버섯의 pyrimidine 영양요구성 균주의 positive 선발 (A Positive Selection for Pyrimidine Auxotrophic Mutants from Basidiospores of Pleurotus sajor-caju Using 5'-Fluoro-orotic acid)

  • 여름느타리버섯의 담자포자에 자외선을 조사하여 5'-fluoro-orotic acid(FOA) 선발배지에서 pyrimidine 영양요구성 균주를 직접 선발하였다. 24개의 5'-FOA 저항성 균주중 13개의 pyrimidine 영양요구성 균주를 선발할 수 있었으며, 이들의 mating type group과 성장속도를 측정하여 형질전환을 위한 최적의 DNA수용체를 선발할 수 있었다.

Extracellular Enzyme Activities of the Monokaryotic Strains Generated from Basidiospores of Shiitake Mushroom

  • To obtain basic information on the biochemical property of basidiospores of shiitake mushroom (Lentinula edodes), the ability of producing extracellular enzyme was assessed using a chromogenic plate-based assay. For the aim, amylase, avicelase, $\beta$-glucosidase, CM-cellulase, pectinase, proteinase, and xylanase were tested against monokaryotic strains generated from forty basidiospores of two different parental dikaryotic strains of shiitake mushroom, Sanjo-101Ho and Sanjo-108Ho. These two parental strains showed different degree of extracellular enzyme activity. No identical patterns of the degree of enzyme activity were observed between monokaryotic strains and parental strains of the two shiitake cultivars. The degree of extracellular enzyme activity also varied among monokaryotic strains of the two shiitake cultivars. Our results showed that dikaryotic parental strains of shiitake mushroom produce monokaryotic basidiospores having very diverse biochemical properties.

Morphological Discretion of Basidiospores of the Puftball Mushroom Calostoma by Electron and Atomic Force Microscopy

  • Comparative morphology among species of the genus Calostoma, including C. cinnabarina, C. ravenelii, and C. japonicum, was investigated by scanning electron microscopy and atomic force microscopy. Spore morphology of C. cinnabarina and C. ravenelii showed no dramatic differences by light microcopy and scanning electron microscopy. To differentiate these species, atomic force microscopy was employed. Quantitative analysis of the surface roughness of basidiospores revealed subtle differences in height fluctuation at the nanometer scale between the species of Calostoma. Basidiospores of C. cinnabarina had a relatively rougher surface than those of C. ravenelii at $2.0{\times}2.0\;{\mu}m^2$ scan areas.

  • Ganoderam lucidum은 비자실체성(非子實體性) 담자포자(擔子胞子)를 형성(形成)하여 다른 담자균류(擔子菌類)에서 볼 수 없는 특이(特異)한 생활환(生活環)을 보였다. G. lucidum은 배지상(培地上)의 기중균사(氣中菌絲)에서 Basidial strom상(狀)의 균사조직(菌絲組織)을 형성(形成)하고 조직표면(組織表面)에 있는 균사(菌絲)의 정단세포(頂端細胞)가 담자기(擔子器)로 분화(分化)되어 그 위에 비자실체성(非子實體性) 담자포자(擔子胞子)를 형성(形成)하였다. 자실체(子實體)없이 Basidial stroma에서 형성(形成)된 비자실체성(非子實體性) 담자포자(擔子胞子)는 정상 담자포자(擔子胞子)와 유사한 형태(形態)를 보이고 있었으나 크기가 약간 작고 포자표면(胞子表面)의 hollow수(數)가 현저히 적었다. 비자실체성(非子實體性) 담자포자(擔子胞子)의 형성(形成)은 균(菌)의 계통(系統)에 따라 뚜렷한 차이(差異)를 보였고 광(光) 및 환기조건하(換氣條件下)에서만 형성(形成) 되었으며 배지(培地)에 따른 차이(差異)는 거의 없었다.

  • 노랑 느타리버섯의 유전 연구 및 다른 느타리종과의 융합을 위해서 노랑 느타리의 담자포자에 자외선을 조사하여 영양요구성 균주를 선발하였다. 노랑 느타리 포자에 UV를 조사한 결과 UV조사시간이 $40{\sim}70$초이고 생존율 $0.4{\sim}1.4%$ 일때 많은 mutants를 얻을 수 있었고, 선발된 14 auxotrophic mutants의 유전 marker는 amino acid-requiring strain 3균주, nucleic acid base-requiring strain 1 균주, vitamin-requiring strain 3 균주, amino acid/nucleic acid base-requiring strain 2 균주, amino acid/vitamin-requiring strain 5 균주로 되었다.

Morphological characteristics of fruit bodies and basidiospores of Wolfiporia extensa

  • Wolfiporia cocos is a well-known traditional medicine in China, Japan, Korea, and other Asian countries owing to its numerous therapeutic properties. With the aim to determine the morphology and genetic characteristics of W. cocosten strains of W. cocos were cultivated in vitro, and subsequently, rapid amplification of polymorphic DNA was performed. To the best of our knowledge, this is the first study to examine the morphology of fruit bodies of W. cocos in Korea. W. cocos were cultured on PDA agar at different temperatures (12, 16, 20, 24, and $28^{\circ}C$) under 12-hour light (600 Lux) / 12-hour dark photoperiod condition for 1 month. Appearance of fruit body was the highest at $28^{\circ}C$ condition in all the strains investigated. Honeycomb-like structure on sclerotia was observed in Andong 01, Andong 02, Andong 03, KFRI 1104, KFRI 1105, KFRI 1106, KFRI 1107, KFRI 1108, and ASI 13007 strains of. The KFRI 1103 strain formed cosmos petal-like structure on sclerotia. The average size of basidiospores was recorded as $7.55{\mu}m$ in height and $3.35{\mu}$ in width.

자외선(紫外線) 조사(照射)에 의한 표고버섯 담자포자(擔子胞子)의 영양요구성(營養要求性) 균주(菌株) 선발(選拔)에 관한 연구(硏究) (Isolation of Auxotrophic Mutants from Basidiospores of Lentinus edodes)

  • 식용버섯의 유전 연구와 균주 개발을 위하여 표고버섯의 담자포자에 자외선을 조사하여 영양요구성 균주를 유발하였는바, 자외선을 담자포자에 40초 조사했을때 약 5 %의 포자 생존율을 보였으며 120초 조사에서 완전히 발아력을 상실하였다. 30초에서 100초까지 10초 간격으로 자외선을 조사하여 7088 colonies를 검정한 결과 71개의 auxotrophs를 선발하여 1.0%의 유발율을 얻었으며, 60초 조사에서 1.35 %의 가장 높은 유발 빈도를 나타내었는데 이때의 포자 생존율은 1.3% 이었다. 71개의 auxotrrophs 중에서 29개 균주의 영양요구성을 검정한 결과 nucleic acid-requiring strain이 3, nucleic acid/vitamin-requiring strain이 13, nucleic acid/amino acid-requiring strain이 1, vitamin-requiring strain이 7, 그리고 amino acid-requiring strain이 5이었다.

Two Inonotus species newly found in Japan, Inonotus formosanus and Inonotus nodulosus

  • Two Inonotus species newly found in Japan were described. I. formosanus T.T. Chang & W.N. Chou was identified with the following characters; thin basidocarps with hispid when young and later the erect hyphae agglutinate to scrupose tuft, pore surface light yellow to rusty brown later, absence of setal hyhpae, ventricose hymenial setae, and small ellipsoid, hyaline to yellowish basidiospores. I. nodulosus (Fr.) P. Karst. was identified with the following characters; basidocarps nodulose, wart-like shape, scrupose to warted by agglutinated hayphae, margin up to 5 mm, pore surface cinnamon to rusty brown when dry with a whitish or silvery shine, absence of setal hyphae, acute straight hymenical setae, ellipsoid to subglobose basidiospores, which are weakly dextrinoid in Melzer's reagent.

  • Two new species of Fulvifomes are described from specimens collected in rainforests of Nonggang Nature Reserve of southern China, based on morphological characteristics and molecular phylogenetic analysis of the internal transcribed spacer (ITS) and nuclear large subunit ribosomal DNA (nLSU) sequences. Fulvifomes nonggangensis sp. nov. is characterized by perennial, sessile and solitary basidiocarps, applanate pileus, small cystidioles of 9.9-15.4 × 2.9-3.5 ㎛, large pores of 5-6 per mm, a dimitic hyphal system, and broadly ellipsoid basidiospores of 4.3-5.3 × 3.3-4.2 ㎛. F. tubogeneratus sp. nov. is characterized by perennial, sessile, and imbricate basidiocarps, a duplex context, small pores of 7-8 per mm, a dimitic hyphal system, and ovoid to subglobose basidiospores of 5.72 × 5.00 ㎛.

Induction of Sexual Stage and Colony Morphology of Some Isolates of Sclerotium rolfsii Causing Spotted Leaf Rot in Plants

  • Twenty-two isolates of Sclerotium rolfsii causing spotted leaf rot from Varanasi, India were grown on 6% Cyperus rotundus rhizome meal agar (CRMA) medium for the induction of athelial stage (Athelia rolfsii). Only one isolate obtained from Sphaeranthus indicus formed basidial stage on CRMA medium while the other 21 isolates did not. Basidial stage was also produced in S. indicus isolate at different concentrations (5.5, 6.0 and 6.5% w/v) of CRMA medium. Size of basidia, sterigmata and basidiospores of this isolate was measured. Basidia clavate, hyaline and measured $10{\sim}12{\times}4{\sim}5\;{\mu}m$ in size, basidiospores hyaline, unicellular, subglobose to ellipsoid produced on sterigmata and measured $3{\sim}5{\times}2{\sim}4\;{\mu}m$ in size, sterigmata hyaline and measured $4{\sim}5{\times}1.5{\sim}2\;{\mu}m$ in size. The results of the present study revealed wide variation in spotted leaf rot isolates of S. rolfsii. A reddish zone around the colony of S. rolfsii isolate from Vernonia sp. was observed on CRMA medium. HPLC analysis of the zone revealed the presence of gallic and ferulic acid which were also thought to be responsible for reduced mycelial growth of the isolate on CRMA medium.
