Cloning and Sequence Analysis of the Cellobiohydrolase I Genes from Some Basidiomycetes

  • Genes encoding the cellobiohydrolase enzyme (CBHI), designated as cbhI, were isolated from the basidiomycetes Auricularia fuscosuccinea, Pleurotus giganteus, P. eryngii, P. ostreatus, and P. sajor-caju. Initially, the fungal genomic DNA was extracted using a modified cetyltrimethyl ammonium bromide (CTAB) protocol and used as a DNA template. The cbhI genes were then amplified and cloned using the pGEM-T Easy Vector Systems. The sizes of these PCR amplicons were between 700~800 bp. The DNA sequences obtained were similar showing high identity to the cbhI gene family. These cbhI genes were partial consisting of three coding regions and two introns. The deduced amino acid sequences exhibited significant similarity to those of fungal CBHI enzymes belonging to glycosyl hydrolase family 7.

Enzyme activity of Basidiomycetes products in each cereals (곡물별 담자균 배양물의 효소활성)

  • In this study, growth rates and enzyme activities of mushroom mycelium were investigated in each cereals. Cultivation on hulled barley resulted in slightly faster mycelial growth as compared to other substrates. Enzyme activities were measured in different periods. In result, ${\alpha}$-amylase activity was higher in Pleurotus ostreatus and Phellinus linteus cultured cereals, whereas ${\beta}$-amylase activity was higher in Pleurotus ostreatus and Fomitella fraxinea cultured. Protease level did not affect kinds of cereal except Phellinus linteus. The color values lightness decrease by Basidiomycetes cultivation and redness and yellowness increased.

Characterization and Phylogenetic Analysis of Chitin Synthase Genes from the Genera Sporobolomyces and Bensingtonia subrorea

  • We cloned seven genes encoding chitin synthases (CHSs) by PCR amplification from genomic DNAs of four strains of the genus Sporobolomyces and of Bensingtonia subrosea using degenerated primers based on conserved regions of the CHS genes. Though amino acid sequences of these genes were shown similar as 176 to 189 amino acids except SgCHS2, DNA sequences were different in size, which was due to various introns present in seven fragments. Alignment and phylogenetic analysis of their deduced amino acid sequences together with the reported CHS genes of basidiomycetes separated the sequences into classes I, II and III. This analysis also permitted the classification of isolated CHSs; SgCHS1 belongs to class I, BsCHS1, SaCHS1, SgCHS2, SpgCHS1, and SsCHS1 belong to class II, and BsCHS2 belongs to class III. The deduced amino acid sequences involving in class II that were discovered from five strains were also compared with those of other basidiomycetes by CLUSTAL X program. The bootstrap analysis and phylogenetic tree by neighbor-joining method revealed the taxonomic and evolutionary position for four strains of the genus Sporobolomyces and for Bensingtonia subrosea which agreed with the previous classification. The results clearly showed that CHS fragments could be used as a valuable key for the molecular taxonomic and phylogenetic studies of basidiomycetes.

  • This study was carried out to elucidate phylogenetic relationship of a yellow lump, Phellinus linteus by comparing the nuclear ribosomal intergenic spacer (IGS) I region with that of other genera of basidiomycetes retrieved from Genbank. IGS I region of Phellinus linteus was 730 bp long and sequence homology was conserved in the 5' region, in particular $1{\sim}280\;bp$, and decreased in the direction toward the 3' end. ITS region was widely studied in phylogenies related to basidiomycetes, but IGS region was not well understood yet. Our study indicated that IGS region can be a good tool in phylogenetic study of basidiomycetes.

Studies on the Constityents and Cultite of Kotean Basidiomycetes Anitumor Polysaccharides from the Cultured Mycelia of Some Basiodiomycetes

  • To develop potent anitumor polysac-charides from higher hungi of Korea the alcohol precipitated polysaccharides, which were obtained from hot water extracts of shake-cultured mycelia of eight species of basidiomycetes, were subjected to antitumor screening tests, using sarcoma 180 as a tumor cell. When administered i. p, at the dose level of 20mg/kg/day for ten days into the mice which were implanted with $1{times}10^{8}$ cells of sarcoma 180 twenty-four hour before the start of polysaccharide injection, the polysaccharides of Plutcus cervinus ISA-Pc-1004, Pleurotus pulmonarius, Hydnum repandum ISA-Hr-1006, and Laccaria laccata ISA-LI-1008 respectively showed inhibition ratios of 62.0%, 51.7%, 64.8%, and 57.8%. They were selected for further study, while those of Pancillus serotina ISA-Ps-1012, Laccaria amethystina ISA-La 1001, Pholiota adiposa ISA-Pa-1010, and Lepista spISA-Ls-1007 respectively showed inhibition ratios of -9.3%, 25.3%, 27.3% and 33.0%.

A Preliminary Screening of 46 Korean Basidiomycetes Including Hebeloma crustulineforme for Their Hemolytic Activities (무우자갈버섯(Hebeloma crustulineforme)을 위시한 한국산 담자균류 46종의 용혈활성 검색)

  • To elucidate the hemolytic toxicities of Korean basidiomycetes, the cold-water extracts of 46 wild mushrooms were tested for hemolytic activity on mouse erythrocytes. Twelve species including Amanita esculenta, A. griseofarinosa, A. longistriata, A. melleiceps, A. phalloides, A. rubescence, A. spissacea, A. vaginata, Hebeloma crustuliniforme, Lepiota cygnea, Macrolepiota procera, and Oudemansiella platyphylla showed strong hemolytic activities. Of these, A. griseofarinosa and H. crustuliniforme showed no decrease in their hemolytic activity even after five minutes' boiling.

  • Spore production mechanism of selected lignicolous basidiomycetes was inhibited by soaking their sporocarps in water for varying periods. Depending on the duration of immersion, some species resumed spore discharge, while in others it was completely ruined. This was related to the nature and consistency of these sporocarps. All the species studied absorbed water quite readily into their sporocarps. The loss of this water through dehydration was more rapid in smaller and delicate species than the bulky and robust ones.

  • Mechanism of lignin biodegradation caused by basidiomycetes and the history of lignin biodegradation studies were briefly reviewed. The important roles of fungal extracellular ligninolytic enzymes such as lignin and manganese peroxidases (LiP and MnP) were also summarized. These enzymes were unique in their catalytic mechanisms and substrate specificities. Either LiP or MnP system is capable of oxidizing a variety of aromatic substrates via a one-electron oxidation. Extracellular fungal system for aromatic degradation is non-specific, which recently attracts many people working a bioremediation field. On the other hand, an intracellular degradation system for aromatic compounds is rather specific in the fungal cell. Structurally similar compounds were prepared and metabolized, indicating that an intracellular degradation strategy consisted of the cellular systems for substrate recognition and metabolic response. It was assumed that lignin-degrading fungi might be needed to develop multiple metabolic pathways for a variety of aromatic compounds caused by the action of non-specific ligninolytic enzymes on lignin. Our recent results on chemical stress responsible factors analyzed using mRNA differential display techniques were also mentioned.

Antitumor Components of Korean Basidiomycetes

  • To find antitumor components in the shake-cultured mycelia of Volvariella bombycina, the mycelia were extracted with hot water. After the extract was dialyzed and freeze-dried, a protein-polysaccharide fraction was obtained and examined for antitumor activity against the solid form of sarcoma 180 in ICR mice. It showed 60.3% inhibition ratio at a dose of 20mg/kg/day for 10 days. It was found to consist of a polysaccharide moiety and a protein moiety. After gel filtration on Sepharose 4B, Fraction B was obtained and showed the highest inhibition ratio of 71.1%. When the antitumor component was examined for immunopotentiating activity, it was found to increase the macrophage accumulation in the peritoneal cavity as well as the antibody production of the spleen cells of the mice.

  • Among the higher fungi collected from 5 different locations in Korea, 37 species of Basidiomycetes were demonstrated to be new to Korea. These included 10 edible species and 27 inedible species. Korean common names of all the fungi were designated by the authors.
