Physiological and Ecological Studies on Mycelia of Armillariella mellea (뽕나무 버섯 균사체의 생리.생태학적 연구)

  • To study physiological and ecological characters of the secondary mycelia of Armillariella mellea, it was cultivated on the various media. It was grown very well on the malt extract medium compared with its growth on yeast extract medium, potato dextrose medium and Hamada medium. The temperature of $27^{\circ}C$ gave the best condition for it to grow. The highest rate of growth was shown in medium of pH 6. Heteroauxin of 5 ppm concentration showed a increase as 26.2% in growth in compared with the control group. Gibberelline 10ppm, Tomatoton of 10,000 dilution and Adoton of 5,000 dilution showed positive effect. It was shown that the use of above hormones in appropriate concentration brought the effect of growth, but overuse of them brought inhibitory effect. Under the same condition of 200ppm concentration, vitamin A gave the highest growth of 18.2% than that of the control used.

  • The mycelial growth of Agaricus bitorquis and Pleurotus ostreatus in synthetic media were carried out by ordinary methods. The optimum pH and temperature for mycelial growth were from pH 6.0 to 6.5 and 25 to $30^{\circ}C$, and from pH 5.0 to 6.5 and $25^{\circ}C$ in A. bitorquis and P. ostreatus, respectively. Among the carbon and nitrogen sources, glucose, starch, and peptone showed the good result for the mycelial growth of A. bitorquis, and glucose, fructose, starch and peptone were good for the mycelial growth of P. ostreatus. The yield of mycelium decreased under lower or higher C/N ratio. Also, at the same C/N ratio, the higher the concentration of glucose and peptone, the more the yield was increased. Among various vitamins thiamine, Ca-pantothenate and folic acid were suitable for the mycelial growth of A. bitorquis, and thiamine, folic acid and ino­sitol for the mycelial growth of P. ostreatus. Although pH, total nitrogen and glucose contents of media decreased gradually during culture period the yield of mycelium increased.

Effects of Different Substrate on the Growth and Microstructure of Fruit Body in the Basidiomycetes, Pleurotus ostreatus. (느타리버섯 자실체 생육 및 미세구조에 미치는 배지의 영향)

  • Comparison of fruit bodies of Pleurotus ostreatus cultivar chunchu No .2 grown on the sawdust, rice straw and cotton waste substrates revealed differences in the pattern of differentiation of hyphal compartments. Required period for primordium induction of fruit bodies grown on sawdust substrate was 13 days. Physical structure shown as hardness of stipes grown on the sawdust substrate, fruit bodies were harden than control. Pileocystidia were well developed on the surface of pileus in the fruit body cultivated on rice straw. Microstructures of fruit body grown on the sawdust and cotton wastes substrates shown fast-discharge of basidiospore and sytoms ageing. Hyphae of fruit bodies formed on sawdust substrate had less stainable cytoplasmic material and many more vacuoles than hyphae of fruit bodies formed on synthetic substrate with 50% of pine sawdust, 30% of cotton seed hull and 20 of beet pulp(control).

Studies on Protoplast Isolation of Pleurotus cornucopiae (노랑느타리버섯의 원형질체(原形質體) 분리(分離)에 관한 연구(硏究))

  • The optimal conditions for high yields of mycelial protoplasts from P. cornucopiae were established. The concentraion of enzyme system containing Novozym 234, ${\beta}-D-glucanase$ and ${\beta}-glucuronidase$ was $5mg\;ml^{-1}$ each. The osmotic stabilizer most effective for protoplast isolation was O.6 M sucrose. The optimal reaction time of mycelium with the lytic mixture was 90 min in a shaking condition at 120 strokes $min^{-1}$. When the myelium of P. cornucopiae was cultured for 4 days on mushroom complete medium at $28^{\circ}C$, the formation of protoplast was effective. When the pH of the digestion mixture with O.6 M sucrose as stabilizer varied between pH 4.0 and 7.0, the production of protoplasts was effective in phosphate buffer (pH 6.2) and Na-maleate buffer (pH 5.0). Generally, phosphate buffer was more effective for protoplast isolation than Na-maleate buffer, but 0.6 M sucrose osmotic stabilizer without adjusting pH was most effective. Using these conditions, protoplasts from P. cornucopiae were obtained at a ratio $1{\times}10^7\;ml^{-1}$.

Studies on Protoplast Formation and Regeneration of Lyophyllum decastes (Lyophyllum decastes의 원형질체 분리와 재생에 관한 연구)

  • This experiment was carried out to investigate proper conditions for protoplast isolation and regeneration from mycelia of Lyophyllum decastes. Novozym 234(10 mg/ml) with 0.6 M $MgSO_4$ in phosphate buffer(pH 4.0) was proper for protoplast isolation. The optimal reaction time of the mycelium with the lytic enzyme was four hours in shaking condition at 120 strokes per min. When the mycelium of L. decastes was cultured at $24^{\circ}C$ for 5 days, the formation of protoplasts was effective. The liquid medium was more effective for protoplast isolation than the solid medium. In the liquid medium, high yields of protoplasts were obtained from 0.6 M $MgSO_4$ osmotic stabilizer. Protoplasts of L. decastes were regenerated to normal hyphal growth and the regeneration frequency of the protoplasts in the complete agar medium containing Triton X-100(0.0025%) was $5.94{\sim}8.32%$. The regeneration medium stabilized with 0.6 M sucrose was the best for regeneration of the protoplasts. In contrast to protoplast formation, regeneration was inhibited by the inorganic salts used as osmotic stabilizer.

  • The effects of cultural conditions on the mycelial growth and fruit-body formation and the yield of Pleurotus sajor-caju grown in rice straw medium were examined. The change of content of chemical components of mushroom and the medium during the cultivation period was also examined. The growth of mycelium was best in the medium sterilized for 60 min at $1.2kg/cm^2$ with a moisture content of 60%. The optimum pH and temperature for mycelial growth were 6.0 and $25^{\circ}C$, respectively, whereas the optimum pH and temperature for fruit-body formation ranged from 5.0 to 6.0 and from 20 to $30^{\circ}C$, respectively. Rice bran and starch cake were better than the other additives used for both mycelial growth and fruit-body formation. The optimum concentration of rice bran in the medium was 10%. The total yield of mushroom was $20.2kg/m^2$, 66% of which was prouduced from the first and second cropping period. As the harvest proceeded, the crude fiber cotent increased in the mushroom, whereas it decreased in the medium. The content of free sugar and crude ash, on the other hand, decreased in the mushroom, whereas it increased in the medium. The content of the other chemical components examined decreased in both the mush­room and the medium.

Anti-Varicella Zoster Virus Activity of Water Soluble Components of Elfvingia applanata Alone and in Combinations with Interferons (잔나비걸상 수용성성분의 항-Varicella Zoster Virus 작용과 Interferon과의 병용효과)

  • To search for less toxic antiviral agents from Basidiomycetes, the water soluble components (=EA), were isolated from the carpophores of Elfvingia applanata (Pers.) Karst. Anti-varicella zoster virus (Oka strain; anti-VZV/Oka) activity of EA was examined in MRC-5 cells by plaque reduction assay in vitro. And the combined antiviral effects of EA with interferon (IFN) alpha or IFN gamma were examined on the multiplication of VZV/Oka. EA exhibited a dose-dependent reduction in the plaque formation of VZV/Oka with 50% effective concentration $(EC_{50})$ of $464.14\;{\mu}g/ml$. The results of the combination assay were evaluated by the combination index (CI) that was calculated by the multiple drug effect analysis. The combination of EA with IFN alpha showed partially synergistic or additive effects with CI values of $0.83{\sim}1.09$ for 50%, 70%, 90% effective levels, and those with IFN gamma showed antagonism with CI values of $1.20{\sim}1.24$.

  • Interspecific fusion products were obtained from fusion of Ganoderma applanatum and Ganoderma luridum. Frequency of fusion was 0.77-1.38%. Fusion products were selected by the comparision of morphology and color of colony. Fusion Products had chracteristics of two parental strains and generally grew faster than the parents. Some fusants were segregated on GCM. Fusion products were confirmed by mycelial morphology and electrophoretics pattern of esterase isozyme from mycelium. Most of fusion products were lack in clamp connection but those fusion products, that were segregated produced mycelium with and without clamp connection. Some of the fusion products produced fruit body on sawdust medium.

  • Antitumor components were obtained from the cultured mycelia of Pleurotus pulmonarius by ethanol precipitation. The protein-bound polysaccharide was subjected to DEAE-­Sephadex column chromatography and Sephadex G-200 gel filtration. The antitumor fraction $C_1$ was isolated. The inhibition ratio of fraction $C_1$ was 81.8 % in the doses of 10 mg/kg/day for 10 days. The antitumor fraction $C_1$ consisted of a polysaccharide and a protein. The protein-moiety was composed of 14 amino acids. From the peritoneal cell populations in the mice given antitumor fraction $C_1$, the injection of the fraction caused the influx of peritoneal macrophages at two days when compared with those of soluble starch. This was named pulmonaran after its species name.

Protoplast Regeneration and Reversion in Pleurotus cornucopiae (노랑느타리버섯의 원형질체(原形質體) 재생(再生) 및 환원(還元)에 관한 연구(硏究))

  • Protoplasts of P. cornucopiae were reverted to normal hyphal growth and reversion frequency was $0.04{\sim}19%$. The complete medium stabilized with 0.6 M sucrose was most effective for regeneration of protoplasts. When hypertonic mushroom complete medium not containing agar was overlaid, regeneration frequency of protoplasts was the highest rate among the others of topagar. The protoplast reversion frequency and mycelial growth of P. cornucopiae were increased when various amino acids, nucleic acid components and vitamin compound were added to the hypertonic minimal medium. The relation between sources increasing reversion frequency and sources accelerating mycelial growth was similar in amino acids and nucleic acid components but it was different in vitamins. The protoplast reversion frequency showed the highest rate when all sources were added to the regeneration minimal medium. Microscopically, regeneration patterns of protoplasts showed formation of a bud-like structure, direct germination, yeast-like cell chain of the protoplast, and the production of both direct germ tube and yeast-like cell chain from a protoplast.
