낮은 농도의 Hypoxanthine과 FSH가 미성숙난자의 체외성숙에 미치는 영향 (The Effect of Low Concentrated Hypoxanthine and FSH in 10% FBS Supplemented Medium on Immature Oocyte in vitro Maturatio)

  • 목 적: Hypoxanthine (Hx)과 FSH가 미성숙난자의 배양에 미치는 영향을 관찰하기 위해 미성숙난자를 배양하여 GVBD, MII기 발생률을 비교 관찰하였다. 연구방법: 단순배양액인 BSAL-XI-HTF 배양액을 사용하여 (1) 0.3% BSA mBASAL-XI-HTF (0.3% BSA 배양액), (2) 0.1 IU/ml FSH를 첨가한 0.3% BSA mBASAL-XI-HTF (FSH 0.3% BSA 배양액), (3) 10% FBS mBASAL-XI-HTF (10% FBS 배양액), (4) 0.1 IU/ml FSH를 첨가한 10% FBS mBASAL-XI-HTF (FSH 10% FBS 배양액)의 4종류의 배양액을 만들었고, 각 종류의 배양액에서 생쥐 난구세포로 둘러싸인 미성숙난자의 성숙을 3, 6, 18시간별로 비교 관찰하였다. 각 배양액에 1 mM, 2 mM, 4 mM 농도의 미성숙난자성숙억제제인 Hx을 섞어 난자의 성숙억제양상을 관찰하였고 GVBD와 MII기 발생률을 비교 관찰하였다. 결 과: Hx을 첨가하지 않은 4종류의 배양액에서 미성숙난자의 자연성숙은 3시간 내에 대부분 GVBD가 발생하였고 MII기로의 발육은 6시간 이후 발생하였다. 18시간 후 각 군의 배양에서 모두 유사한 GVBD 발생률을 보였다. 1 mM, 2 mM, 4 mM Hx 농도의 0.3% BSA 배양액과 10% FBS 배양액에서 난자의 성숙억제양상을 비교해 보면 4 mM 농도의 Hx 배양액에서 18시간 동안 완전한 성숙억제를 보였다. 2 mM 농도의 Hx 배양액에서도 18시간 배양까지 억제를 보였으나 4 mM 농도의 Hx 배양액보다 작은 난자성숙억제양상을 보였다. 1 mM 농도의 Hx 배양액에서는 모두 난자성숙억제를 보이지 못했다. FSH를 첨가한 배양액에서는 2 mM 농도의 Hx 뿐만 아니라 1 mM 농도의 Hx 첨가에서도 초기 3시간까지 GVBD 성숙이 억제되었다. 또 같은 농도의 Hx을 함유하고 FSH를 첨가하지 않은 배양액에 비해 FSH를 첨가한 배양액에서 3시간, 6시간 동안 GVBD 발생을 더 억제하였다. 그러나 18시간 배양 후 난자성숙이 회복됨을 보였다. 이 결과로 FSH가 배양초기에 난자성숙을 억제하나 후기에는 성숙을 촉진함을 알 수 있었다. 18시간 후 MII기발육은 4 mM 농도의 Hx을 함유한 모든 배양액에서 낮은 발생률을 보였고, FSH를 함유한 10% FBS 배양액에 1 mM, 2 mM의 낮은 농도의 Hx을 첨가한 배양액은 다른 배양액에 비해 통계학적으로 높은 MII기 발생률을 보였다. 결 론: 본 실험에서 낮은 농도의 Hx과 FSH는 FBS를 함유한 배양액에서 미성숙난자의 초기배양 동안 성숙을 억제한 후 MII기 성숙을 촉진함을 알 수 있었다.

Effects of Squid Ink on Growth Performance, Antioxidant Functions and Immunity in Growing Broiler Chickens

  • This study was conducted to explore the effects of squid ink on growth performance, immune functions and antioxidant ability of broiler chickens during a period of six weeks. Either sex Arbor Acres broilers were equally allotted to 4 groups with 3 replicates of 20 chickens each. Broilers diets for the 4 test groups were prepared separately with starter and finisher phases. Control chickens were fed with basal diet and birds of group Exp 2, Exp 4 and Exp 6 were fed with the basal diet supplemented with 2%, 4% and 6% of squid ink, respectively. Broilers were sacrificed to investigate antioxidant parameters of sera, indices of thymus, spleen and bursa of fabricius and spleen lymphocyte proliferation, as well as growth performance on the $21^{th}$ and $42^{th}$ day. The results revealed that, i) squid ink promoted growth performance of broilers during days 22 to 42 and the whole trial period (p<0.05 or p<0.01); ii) squid ink elevated relative weight of the three immune organs during the starter phase and spleen lymphocyte proliferation throughout the experiment (p<0.05); iii) squid ink increased SOD activity and decreased MDA level in sera from broilers during the whole period (p<0.05). The above results suggest that squid ink could improve growth performance, antioxidant ability and immune functions of growing broiler chickens and be employed in the development of feed additives for animals.

Effects of γ-aminobutyric acid (GABA) on growth performance, nutrient digestibility, blood profiles, meat quality, and backfat in finishing pigs

  • This experiment was conducted to estimate the influence of ${\gamma}$-aminobutyric acid (GABA) on growth performance, nutrient digestibility, blood profiles, meat quality, and backfat in finishing pigs. A total of 100 mixed finisher pigs [(Landrace ${\times}$ Yorkshire) ${\times}$ Duroc] were randomly allotted to two dietary treatments for 10 weeks, the average initial body weight was $52.03{\pm}1.08kg$. Dietary treatments consisted of supplementation of GABA to 0.05% of the basal diet (GAB) and a CON (basal diet) without supplementation. Each treatment had 10 replicates with 5 pigs per pen (three gilts and two barrows). Measurements were made at three time period: weeks 1 - 5, weeks 5 - 10 and overall experimental period. Compared to CON, the pigs fed GABA had average daily gain (ADG) increases among phases (p < 0.05). Average Daily Feed Intake ADFI increased during the overall experimental period (p < 0.05). The digestibility of Dry Matter (DM) increased. The concentration of epinephrine decreased at 5th and 10th weeks (p < 0.05), meanwhile, cortisol decreased at 10th week (p < 0.05) in GAB treatment. Otherwise, the pigs' GABA supplementation had no effect on meat quality and backfat thickness (p > 0.05). We conclude that a dietary supplementation of GABA at 0.05% improves growth performance and the digestibility of dry matter, while reducing the stress response of finishing pigs.

정맥 주입한 Catecholamine이 흰쥐의 기초상태시 위산, Gastrin 및 Secretin 분비에 미치는 영향 (Effects of Intravenous Catecholamine on Gastric Acid, Gastrin and Secretin Secretion in Basal State of the Rat)

  • This study was conducted to investigate the effects of epinephrine and norepinephrine on basal gastric acid secretion and plasma gastrin and secretin concentration in the conscious rat. One hundred and eighty-four albino rats with gastric cannula were used after 18 hours or more of fast, with water ad libitum. In a restraint cage for collection of gastric juice, physiological saline (0.9% NaCl) was continuously infused into the jugular vein through a catheter for one hour at a rate of 1 ml/hr (control period). Immediately after the control period, epinephrine (1, 2, 4, 8 and $16{\mu}g/ml/hr)$, norepinephrine (1, 2, 4, 8 and $16{\mu}g/ml/hr)$ or physiological saline (1 ml/hr) was infused for another one hour. Gastric juice was collected at one hour interval for two hours infusion period. Adrenergic antagonists, phentolamine and propranolol were injected into the jugular vein 5 min prior to the infusion of epinephrine or norepinephrine at a dose of 0.2 mg/0.1 ml. Blood was sampled from the jugular vein for the radioimmunoassay of plasma gastrin and secretin after the collection of gastric juice. The results were as follows: 1) Both epinephrine and norephinephrine significantly increased gastric acid output in a dosedependent manner. 2) The effects of epinephrine and norepinephrine on the gastric acid secretion were antagonized by the pretreatment with phentolamine and propranolol. 3) Plasma gastrin and secretin concentrations were not significantly affected by the intravenous infusion of epinephrine and norepinephrine. It can be inferred from the above results that epinephrine and norepinephrine facilitate gastric acid secretion in conscious rats and the mechanism of which is attributed to ${\alpha}\;and\;{\beta}$ adrenergic receptors rather than gastrin and secretin.

Amino acid digestibility in diets containing copra meal with β-mannanase fed to growing pigs

  • Objective: The objective of this study was to estimate standardized ileal digestibility (SID) of amino acids (AA) in growing pigs fed diets containing increasing levels of copra meal (CM) with β-mannanase supplementation. Methods: Twenty barrows (initial body weight: 34.43±0.11 kg) surgically fitted with T-cannulas at the distal ileum were individually housed in metabolism crates. Pigs were allotted to 5 dietary treatments in a completely randomized design with 4 replicates per treatment. The dietary treatments were: i) NC, negative control, corn-soybean meal (SBM) based diet, ii) PC, positive control, basal diet + 0.10% β-mannanase supplementation (800 IU/kg), iii) CM6, PC diet with 6% CM supplementation, iv) CM12, PC diet with 12% CM supplementation, and v) CM18, PC diet with 18% CM supplementation. A nitrogen-free diet was used to estimate basal endogenous losses of AA for SID calculation. All experimental diets contained 0.5% chromic oxide as an indigestible marker. Each period consisted of a 4-d diet adaptation period and a 3-d ileal digesta collection period. Results: There were no differences in apparent ileal digestibility (AID) and SID of all AA between the NC and PC treatments except that the PC treatment had lower AID and SID of glycine than the NC treatment (p<0.05). There were linear decreases in AID and SID of lysine (p<0.05) and aspartic acid (p = 0.06; tendency) with increasing levels of CM in the diets with β-mannanase. Conclusion: The β-mannanase supplementation had no effect on AA digestibility in pigs fed the corn-SBM based diet but increasing levels of CM reduced SID of lysine and aspartic acid.

산란계 사료내 Conjugated Linoleic Acid(CLA)와 불포화지방산 함유 Oil의 첨가가 산란율과 계란의 품질에 미치는 영향 (Effects of Dietary Conjugated Linoleic Acid (CLA) and Oil Containing Unsaturated Fatty Acid Supplementation on Egg Production Rate and Quality in Laying Hens)

  • 본 시험은 산란계 사료내 conjugated linoleic acid(CLA)와 불포화지방산 함유 oil의 첨가가 산란율과 계란 품질에 미치는 영향을 알아보고자 실시하였다. 시험 동물은 36주령 ISA Brown 갈색계 288수를 공시하였으며, 사양 시험은 5주간 실시하였다. 시험 설계는 1) BO(basal diet + 1% soybean oil + 1% oat), 2) BS(basal diet + 1% safflower oil + 1% oat), 3) BF (basal diet + 1% free fatty acid + 1% oat), 4) CD(basal diet + 1% CLA containing diglyceride + 1% oat), 5) CT(basal diet + 1% CLA containing triglyceride + 1% oat), 6) CP(basal diet + 1% CLA by-product + 1% soybean oil)로 6개 처리를 하여 처리당 8반복, 반복당 6수씩 완전 임의 배치하였다. 산란율에 있어서는 처리구간에 유의적인 차이를 보이지 않았다(P>0.05). 난각 강도에 있어서는 3주째에 CP 처리구가 BS, BF, CD, CT 처리구와 비교하여 유의적으로 낮았으며(P<0.05), 4주째에는 BO와 CP 처리구가 다른 처리구들과 비교하여 유의적으로 낮았다(P<0.05). 난각 두께에서는 3주와 4주째 모두에서 CP 처리구가 BS, BF, CD, CT 처리구와 비교하여 유의적으로 낮았다(P<0.05). 난중에 있어서는 3주와 4주째 모두에서 CP 처리구가 다른 처리구들과 비교하여 유의적으로 낮았다(P<0.05). 시험 4주째의 난황고에 있어서는 BF와 CT 처리구가 BO, CD, CP 처리구와 비교하여 유의적으로 가장 높았으며(P<0.05), BO와 CD 처리구는 CP 처리구와 비교하여 유의적으로 높았다(P<0.05). 또한, 4주째의 난황색에 있어서는 CP 처리구가 BO와 BS 처리구와 비교하여 유의적으로 높았다(P<0.05). 호우 유닛(Haugh unit)에 있어서는 3주와 4주째 모두에서 BO와 CP 처리구가 다른 처리구들과 비교하여 유의적으로 낮았다(P<0.05). 본 시험 결과 산란계 사료내 CLA와 불포화지방산 함유 oil을 첨가.급여하면 산란율에는 아무런 영향을 미치지 않으나, 계란 품질에 있어서는 향상되는 결과를 보였다.

  • Beta-aminopropionitrile (BAPN)-a Lathyrus toxin was fed to laying chicken to investigate effects on egg weight, production and malformity. Two feeding trials were conducted where the levels of BAPN varied from 0.1 to 0.6 g per kg diet. The responses with regard to egg weight, egg production and the incidence of malformed eggs due to varying concentrations of the toxin were recorded and the results on weekly changes in these important variables were evaluated by plotting graphs. The results showed that there were increasing trends in egg weight and egg shell abnormalities and a decreasing trend in egg production as the dietary level of BAPN was increased indicating that the responses of layers were dose dependent. The effects on egg weight, egg production and egg malformity disappeared following withdrawal of the toxin from the diets.

절식요법이 2명의 비만청소년의 체성분 변화에 미치는 영향 (The Effect of the Fasting for Body Composition Changes in Two Obese Adolescents)

  • Objectives : The purpose of this study is to investigate the effect of fasting therapy on the changes of body composition in obese adolescents. Methods : We examined the body composition of obese adolescents three times; before fasting, on the second refeeding day, two or four weeks later. Results : Body weight, body mass index(BMI), basal metabolic rate(BMR) and lean body mass (LBM) were decreased during the fasting, whereas percent-fat was increased. After fasting, patients have the dietary period. During this period BMR and LBM were increased and the decrease of percent-fat was continued. Conclusions : It seems that fasting therapy with keeping up well-controlled dietary period and receiving oriental medical management can be useful to decrease percent-fat in obese patients.

Effects of Additives on Laying Performance, Metabolic Profile, and Egg Quality of Hens Fed a High Level of Sorghum (Sorghum vulgare) during the Peak Laying Period

  • This experiment was conducted to determine the effects of supplemental methionine, lysine, choline, and sulfur on laying performance, metabolic parameters, and egg quality of hens fed diets containing sorghum (Sorghum vulgare) during the peak laying period. Lohman layers (n = 144), 30-wk of age as 6 replicate cages of 4 hens, were allocated randomly to receive basal diets containing either 22% corn (B) or 22% sorghum (BS) and diets BS plus 0.57% methionine, 0.66% lysine, 0.47% choline, or 0.05% sulfur for 98 d. Feed intake (FI) and egg production (EP) were recorded daily, egg weight (EW) was measured bi-weekly, and body weight (BW) was measured monthly. A sample of 12 eggs from each experimental group was collected every month to evaluate egg quality. At the end of the experiment, blood samples were collected for metabolite concentrations. Data were analyzed using one-way ANOVA as repeated measures and significant differences between the experimental groups were assessed using Duncan's Multiple Range test. Partial replacement of corn with sorghum in the basal diet did not affect BW, EP, and FCR but increased FI by 5.7% and EW by 2.4%. The effects of additives on laying performance were variable. Except for serum total protein (STP) concentration, other metabolic parameters were not affected by partial replacement of corn with sorghum in the basal diet. Hens fed diet BS had lower SPT concentration than hens fed diet B. Except for methionine supplementation, other supplements ameliorated depression in STP concentration. The additives did not affect other metabolic parameters. Egg quality responses to the experimental diets were also variable. Partial replacement of corn with sorghum in the basal diet did not affect eggshell characteristics (both thickness and stiffness), whereas it had variable effects on inner egg quality parameters (increased yolk index, depressed yolk color, and unaltered albumen index and Haugh unit). In conclusion, laying hen diets could include low-tannin sorghum (0.26%) up to 22% without necessitating extra supplements to overcome compromised performance.

The Effects of Different Copper (Inorganic and Organic) and Energy (Tallow and Glycerol) Sources on Growth Performance, Nutrient Digestibility, and Fecal Excretion Profiles in Growing Pigs

  • This study was conducted to determine the effects of different copper (inorganic and organic) and energy (tallow and glycerol) sources on growth performance, nutrient digestibility, gas emission, diarrhea incidence, and fecal copper concentration in growing pigs by using a 2${\times}$2 factorial design. In this trial, 96 pigs (63 d of age) were employed, with an average initial weight of 28.36${\pm}$1.14 kg. The dietary treatments were i) basal diet with 134 ppm copper (Korea recommendation) as $CuSO_4$+tallow; ii) basal diet with 134 ppm Cu as $CuSO_4$+glycerol; iii) basal diet with 134 ppm copper as CuMet+tallow; and iv) basal diet with 134 ppm copper as CuMet+ glycerol. Throughout the entire experimental period, no differences were noted among treatment groups with regard to the magnitude of improvement in ADG (average daily gain), ADFI (average daily feed intake) and G/F (gain:feed) ratios. The nitrogen (N) digestibility of pigs fed on diets containing organic copper was improved as compared with that observed in pigs fed on diets containing inorganic copper (p<0.05). An interaction of copper${\times}$energy was observed in the context of both nitrogen (p<0.05) and energy (p<0.01) digestibility. Ammonia emissions were significantly lower in the organic copper-added treatment groups than in the inorganic copperadded treatment groups (p<0.05). Mercaptan and hydrogen sulfide emissions were reduced via the addition of glycerol (p<0.05). No significant effects of copper or energy source, or their interaction, were observed in reference to diarrhea appearance and incidence throughout the entirety of the experimental period. The copper concentration in the feces was significantly lower in the organic copper source treatment group than was observed in the inorganic copper source treatment group (p<0.05). The results of this experiment show that organic copper substituted for inorganic copper in the diet results in a decreased fecal copper excretion, but exerts no effect on performance. The different energy (tallow and glycerol) sources interact with different copper sources and thus influence nutrient digestibility. Glycerol supplementation may reduce the concentrations of odorous sulfuric compounds with different Cu sources.
