Characterization of PUL Haplotypes and Its Evolutionary Analyses in Korean Rice Accessions

  • Pullulanase (PUL), a debranching enzyme, has been utilized in hydrolyzing the a-1,6 glucosidic linkages in starch, amylopectin, pullulan, as well as related oligosaccharides. It has also been indicated that PUL is a novel indicator of inherent RS (Resistant Starch) formation in rice. In this study, we performed haplotype analysis on 320 bred rice accessions, and additional 54 wild accessions were added to study genetic diversity along with other population-based analyses of the PUL gene. Through these investigations, we summarized a total of 10 functional (non-synonymous) SNPs from 7 different exons on chromosome 4. There were 10 haplotypes, of which only six haplotypes were functional, implicating different subpopulations. Diversity reduction was noticed in temperate japonica (0.0005) compared to the highest one (aus, 0.0154), illustrating their higher genetic differentiation by FST-value (0.926). The highest Tajima^ D value was observed in indica (3.6613), indicating PUL gene domestication signature under balancing selection, while the lowest Tajima's D value was found in temperate japonica (-2.2191) which might have undergone under positive selection and purified due to the excess of rare alleles. PCA, population structure, and phylogenetic analyses provide information on the genetic relatedness between and or among the cultivated subpopulations and the wild based on PUL genomic region.

Virus-induced Silencing of the WRKY1 Transcription Factor that Interacts with the SL1 Structure of Potato virus X Leads to Higher Viral RNA Accumulation and Severe Necrotic Symptoms

  • $Potato$ $virus$ $X$ (PVX) replication is precisely regulated by regulatory viral sequences and by viral and/or host proteins. In a previous study, we identified a 54-kDa cellular tobacco protein that bound to a region within the first 46 nucleotides (nt) of the 5' non-translated region (NTR) of the viral genome. Optimal binding was dependent upon the presence of an ACCA sequence at nt 10-13. To identify host factors that bind to 5' NTR elements including AC-rich sequences as well as stemloop 1 (SL1), we used northwestern blotting and matrixassisted laser desorption/ionization time-of-flight mass spectrometry for peptide mass fingerprinting. We screened several host factors that might affect PVX replication and selected a candidate protein, $Nicotiana$ $tabacum$ WRKY transcription factor 1 (NtWRKY1). We used a $Tobacco$ $rattle$ $virus$ (TRV)-based virus-induced gene silencing (VIGS) system to investigate the role of NtWRKY1 in PVX replication. Silencing of $WRKY1$ in $Nicotiana$ $benthamiana$ caused lethal apical necrosis and allowed an increase in PVX RNA accumulation. This result could reflect the balancing of PVX accumulation in a systemic $N.$ $benthamiana$ host to maintain PVX survival and still produce a suitable appearance of mosaic and mottle symptoms. Our results suggest that PVX may recruit the WRKY transcription factor, which binds to the 5' NTR of viral genomic RNA and acts as a key regulator of viral infection.

  • This paper proposes an attitude control system to keep the balance for a two-wheeled mobile manipulator which consists of a mobile platform and a three D.O.F. manipulator. In the conventional control scheme, complicated dynamics of the manipulator need to be derived for balancing control of a mobile manipulator. The method proposed in this paper, however, three links are considered as one body of mass and the dynamics are derived easily by using an inverted pendulum model. One of the best advantage of a sliding mode controller is low sensitivity to plant parameter variations and disturbances, which eliminates the necessity of exact modeling to control the system. Therefore the sliding mode control algorithm has been adopted in this research for the attitude control of mobile platform along the pitch axis. The center of gravity for the whole mobile manipulator is changing depending on the motion of the manipulator. And the orientation variation of center of gravity is used as reference input for the sliding mode controller of the pitch axis to maintain the center of gravity in the middle of robot to keep the balance for the robot. To confirm the performance of controller, MATLAB Simulink has been used and the resulting algorithms are applied to a real robot to demonstrate the superiority of the proposed attitude control.

증기터빈용 Synchro Clutch Coupling에서 발생하는 진동에 관한 연구 (A study on Mass Unbalance Vibration Generated from 200MW Steam Turbine Synchro Clutch Coupling)

  • The vibration of steam turbine is caused by Mass Unbalance, Shaft Misalignment, Oil Whip and Rubbing etc. but in turbine which is normally operated and maintained, the Mass Unbalance component possesses the greatest portion. Our power plant has two steam turbines in capacity of 200MW and 135MW respectively and each turbine is supported by 6 journal bearings. However, we had many difficulties because the vibration amplitude of No 3 and 4 Bearings was high during the start-up and operation mode change of steam turbine. But, with this study, we completely solved the vibration problem caused by the mass unbalance of No 1 steam turbine. Until a recent date, No 3 and 4 bearings which support high pressure turbine for No 1 steam turbine had shown about 135${\mu}$m in vibration amplitude (sometimes it increased to 221${\mu}$m maximum. alarm: 6mils, trip: 9mils) at base load. After applying the study, they decreased to about 40${\mu}$m maximum. It is a result from that we did not change the setting value of Bearing Alignment and only changed the assembly position of internal parts in Synchro Clutch Coupling Rachet Wheel which links between high pressure turbine and low pressure turbine, and increased the internal gap and machining of the Pawl stopper surface. In the operation of steam turbine, if the vibration value increases by 1X, we should reduce the vibration of bearing by weight balancing. However, unless the vibration of bearing is declined by the balancing, we will have to disassemble and check the component and find the cause. In this study, We researched the way to lower mass unbalance that is 1X vibration component which has the greatest portion of vibration generated by steam turbine and We got good result by applying the findings of this study.

Granule-Bound Starch Synthase I (GBSSI): An Evolutionary Perspective and Haplotype Diversification in Rice Cultivars

  • Granule-bound starch synthase I (GBSSI), encoded by the waxy gene, is responsible for the accumulation of amylose during the development of starch granules in rice endosperm. Despite many findings on waxy alleles, the genetic diversity and evolutionary studies are still not fully explored regarding their functional effects. Comprehensive evolutionary analyses were performed to investigate the genetic variations and relatedness of the GBSSI gene in 374 rice accessions composed of 54 wild accessions and 320 bred cultivars (temperate japonica, tropical japonica, indica, aus, aromatic, and admixture). GBSS1 coding regions were analyzed from a VCF file retrieved from whole-genome resequencing data, and eight haplotypes were identified in the GBSSI coding region of 320 bred cultivars. The genetic diversity indices revealed the most negative Tajima's D value in the tropical-japonica, followed by the aus and temperate-japonica, while Tajima's D values in indica were positive, indicating balancing selection. Diversity reduction was noticed in temperate japonica (0.0003) compared to the highest one (wild, 0.0044), illustrating their higher genetic differentiation by FST-value (0.604). The most positive Tajima's D value was observed in indica (0.5224), indicating the GBSSI gene domestication signature under balancing selection. In contrast, the lowest and negative Tajima's D value was found in tropical japonica (-0.5291), which might have experienced a positive selection and purified due to the excess of rare alleles. Overall, our study offers insights into haplotype diversity and evolutionary fingerprints of GBSSI. It ako provides genomic information to increase the starch content of cooked rice.

Control of Single Propeller Pendulum with Supervised Machine Learning Algorithm

  • Nowadays multiple control methods are used in robot control systems. A model, predictor or error estimator is often used as feedback controller to control a robot. While robots have become more and more intensive with algorithms capable to acquiring independent knowledge from raw data. This paper represents experimental results of real time machine learning control that does not require explicit knowledge about the plant. The controller can be applied on a broad range of tasks with different dynamic characteristics. We tested our controller on the balancing problem of a single propeller pendulum. Experimental results show that the use of a supervised machine learning algorithm in a single propeller pendulum allows the stable swing of a given angle.

  • In this paper, we have proposed a new adaptive fuzzy control algorithm based on Takagi-Sugeno fuzzy model. The regulation problem for the uncertain SISO nonlinear system is solved by the proposed algorithm. Using the advanced stability theory, the stability of the state, the control gain and the parameter approximation error is proved. Unlike the existing feedback linearization based methods, the proposed algorithm can guarantee the global stability in the presence of the singularity in the inverse dynamics of the plant. The performance of the proposed algorithm is demonstrated through the problem of balancing and swing-up of an inverted pendulum on a cart.

  • The purpose of this paper is to provide technical background, techniques and actual diesel engine signature analysis evaluation result. Engine signature analysis(ESA) is a process for monitoring the material condition of diesel engine using external sensors, eliminating the need to periodically disassemble the engine. ESA is also used to balanced the engine. Engine balancing is the process of tuning the engine so that all cylinders carry equal load. ESA is a useful tool to non-intrusively determine the operability and performance and assessment of the material condition of internal component of a diesel engine.

  • This paper considers an integrated inventory-distribution system with a fleet of heterogeneous vehicles employed where a single warehouse distributes a single type of products to many spatially distributed retailers to satisfy their dynamic demands and the product is provided to the warehouse via procurement ordering from any manufacturing plant or market The Problem is formulated as an Mixed Integer Programming with the objective function of minimizing the sum of inventory holding cost (at the warehouse and retailers), and transportation cost and procurement ordering cost at the warehouse, subject to inventory-balancing constraints, ordering constraints, vehicle capacity constraints and transportation time constraints. The problem is Proven to be NP-hard. Accordingly, a Lagrangean heuristic procedure is derived and tested for its effectiveness through computational experiments with some numerical instances.

  • A design modification was made on the 9-th stage wheel dovetail of a high-intermodiate pressure (HIP) turbine rotor for a fossil power plant that necessitates the use of new long-shank buckets for the row. A bucket vibration test is necessary to verify that the new 9-th stage buckets have adequate frequency margin from a nozzle passing frequency when running at speed. A finite element analysis (FEA) has been performed using a commercial S/W to approximately estimate bucket natural frequencies, and thus to help the vibration test. A row of the new buckets has been assembled on the HIP rotor for the vibration tests using dynamic balancing facilities. The tests have been done during deceleration run with air excitation. The test results are compared with the calculation using our empirical formula, and show that the modified design meets the frequency-margin requirements.
