Dyeability and Functionality of Dried Dendropanax morbiferus Extracts (건조 황칠나무 부위별 추출물을 이용한 견직물의 염색성과 기능성)

  • This study aims to examine the possibility of using dried Dondropanax morbiferus extract as a functional dye. The leaves and branches of were extracted with distilled water and 30% ethanol, and the dyeability and functionality of silk fabrics were examined according to the color characteristics of the extract and dyeing conditions. As a result of analyzing the ultraviolet and visible light absorption spectrum of the extract, it was possible to confirm the peak of flavonoid belonging to polyphenol, and the peak of riboflavin expressing yellow color was confirmed. Adsorption equilibrium was observed at 4% dyeing concentration and 60 minutes of dyeing time, and as the temperature increased, dyeing amount increased without color change of Y-series. Aluminum mordanting also increased the yellow color. The color fastness of washing and UV irradiation was low, but the color fastness of rubbing was evaluated as relatively good. The silk fabric dyed with the distilled water extract of the leaves showed a 99.9% bacteriostatic reduction against Staphylococcus aureus and Klebsiella pneumoniae, showing excellent antibacterial properties.

A Study on Analysis of Residual Antibiotics and Prop Acid in Honey (벌꿀중의 잔유항생물질 및 Propionic Acid 분석011 관한 조사연구)

  • A sensitive and simple analytical system for the simultaneous determination of residual oxytetracycline, tetracycline, and doxycycline in honey was described, and that the analytical method for determination of residual propionic acid in honey was established. Experimental subjects were purchased four kinds of honey, native kind honey, acaccia honey, mixed floral honey, chestnut honey in Kyung Sang Nam Do. Several microbiological methods are available to determine tetracycline antibiotecs(TCs) in foods but their precision apears to be variable and the specificity is questionable. These methods are considered to be not suitable for analysis of tetracycline antibiotics in honey because honey itself has bacteriostatic action. For determination of tetracycline antibiotics in honey, therefore the High Performance Liquid Chromatography(HPLC) method was applied, and the propionic acid were determined by Gas Chromatography(5.C). Ethylacetate, as an extract solvent, was found to be suitable for seperation of TCs in honey, but methanol and acetone were not. The recoverly rate of Oxytetracycline(OTC), Tetracycline(TC), Doxycycline(DC) from honey spiked at a level of 10 $\mu $g/g were 97%, 89%, and 91%, respectively. The cailbration curve in TCs was linear expression from 2$\mu $g/ml to 10$\mu $g/ml. As the results of analysis, the residual tetracycline antibiotics were not detected in the 100 samples of honey. The recovery rate of propionic acid from honey spiked at level of 10$\mu $g/g was 98.3% , and the calibra lion curves were linear expression from 21$\mu $g/ml to 101$\mu $g/ml. As the results of analysis, the residual propionic acid was not detected in the 100 samples of honey. Retention time(min) of OTC, DC, and TC were 3.35, 4.61, and 5.30 minutes at the conditions of table 2, respectively, and retention time(min) of propionic acid was 3.50 minutes at the conditions of table 3. The residual TCs and propionic acid were not detected in the 100 samples of honey, but there is a possibility that antibiotics or propionic acid will be to remain in honey if they are used during product period in order to prevent putrefaction of honey-bee.

  • Vibriosis has caused severe losses among cultured yellowtail (Seriola quinquerediata) at some cage farms in Korea in recent years. Major object of this study was to investigate the causative organism of the diseased cultured yellowtail. The samples were collected from the aquarium of Fisheries Research & Development Agency during the period from November 1984 to January 1985. The experimental results are summarized as follows ; Among the isolated bacteria from the diseased yellowtail, Vibrio sp. isolated from the kidney was considered to he the causative organism. Tetracycline, chloramphenicol and gentamycin were observed as bacteriostatic agents to the pathogenic strain, but sulfisomezole and sulfisoxasole were not. When the isolated strain was injected intramuscularly to yellowtail, rid sea-bream, rock-bream and common carp, it had virulence at $25^{\circ}C$ to all fish examined but to virulence at $15^{\circ}C$ except for yellowtail.

Antimicrobial Effects of Natural Flavonoids and a Novel Flavonoid, 7-O-Butyl Naringenin, on Growth of Meat-borne Staphylococcus aureus Strains

  • The antimicrobial effects of the natural flavonoids kaempferol, quercetin, apigenin, and naringenin as well as a novel flavonoid 7-O-butyl naringenin against the growth of four meat-born Staphylococcus aureus strains were evaluated. First, the flavonoids were screened for inhibitory effects against the growth of each strain using the paper disc diffusion method. Second, the growth inhibitory effects of flavonoids that showed antimicrobial activity were measured using the microplate method. Third, the bactericidal effects of flavonoids were evaluated in a 0.8% (w/v) NaCl solution. All flavonoids showed bacteriostatic effects at >20 mM. Among the flavonoids studied, quercetin was more effective than the others tested. However, the inhibitory effect of 7-O-butyl naringenin on growth of S. aureus KCCM 32395 was greater than that of quercetin at the same concentration. Additionally, 7-O-butyl naringenin exhibited significant bactericidal effects at >25 ${\mu}M$. When bacterial cells were examined using scanning electron microscopy, it appeared that the S. aureus membranes were damaged or morphologically changed when treated with quercetin and 7-O-butyl naringenin at 200 ${\mu}M$.

Effect of Enrobing and Adding Antioxidants on the Quality of Pork Patties

  • The effect of applying edible coating and addition of antioxidants (butylated hydroxyanisole (BHA) plus butylated hydroxytoluene (BHT) in a 1:1 ratio) on pork patties were investigated prior to achieve desired physico-chemical, microbiological and sensory qualities. For this, five treatments were conducted as (i) control (neither coated nor antioxidants treated); (ii) coated without antioxidants treated; (iii) coated and antioxidants added in the batter mix only (100 ppm); (iv) coated and antioxidants added in meat mix (100 ppm) only; and (v) coated and antioxidants added both in the batter mix (50 ppm) and the meat mix (50 ppm). Addition of antioxidants both in the batter mix and the meat mix significantly (p<0.05) reduced the microbial loads and thiobarbituric acid (TBA) values. The TBA values significantly (p<0.05) increased up to day 14 and then progressively increased with the advancement of each interval of storage days up to 28 days. Total plate count significantly (p<0.05) increased with the increase in storage days.Coliform and Staphylococcus aureus were absent throughout the storage days in all samples. Staphylococcus aureus however, were present in the control group at day 14 and in enrobed (coated) patties (without antioxidants treated) at 28th day. Addition of antioxidants to batter mix and meat mix did not substantially enhance bacteriostatic activity. Application of coatings and antioxidants retarded the loss of firmness, flavor, changes in appearance and color, and also other sensory attributes. Control patties were better with respect to microbial quality and TBA values but had poorer sensory quality than coated patties.

Sensitivity of Escherichia coli to Seaweed (Ascophyllum nodosum) Phlorotannins and Terrestrial Tannins

  • Pure culture experiments were conducted to assess the bacteriostatic and bactericidal effects of phlorotannins (PT) isolated from Ascophyllum nodosum (brown seaweed) on Escherichia coli O157:H7. In Exp. 1, one non-O157:H7 strain (25922) and three strains of E. coli O157:H7 (3081, EDL933 and E318N) were cultured in M9 medium with PT included at 0 (control), 25, 50 or $100{\mu}g/ml$ (n = 3). Bacterial growth was monitored by $OD_{600}$ at 0, 4, 6, 12 and 24 h, and by dilution plating at 0, 4, 6 and 24 h. All strains were inhibited (p<0.001) by PT to varying degrees. At 50 or $100{\mu}g/ml$, PT prevented growth of all four strains. At $25{\mu}g\;PT/ml$, growth of 25922, 3081, E318N and EDL933 was inhibited for 6, 12 and 24 h, respectively, but 25922 and 3081 resumed growth by 12 and 24 h. Direct plating confirmed bactericidal effects of PT on all four strains at $100{\mu}g/ml$, and on EDL933 and E318N at $50{\mu}g/ml$. In Exp. 2, strains 25922 and 3081 were incubated with no tannins or with $50{\mu}g/ml$ of PT, purified condensed tannins (CT) from Quebracho (Schinopsis balansaei), or purified tannic acid from Rhus semialata (Anacardiaceae) as hydrolysable tannins (HT). Strain 3081 was unaffected by HT or CT, but was completely inhibited (p<0.001) by PT at 4, 6 and 24 h. Strain 25922 was unaffected by HT, slightly inhibited by CT, and almost eradicated by PT at 4 and 6 h. Transmission electron microscopy revealed tannin-mediated alterations to bacterial cell walls. Phlorotannins from A. nodosum exhibit growth-inhibiting and bactericidal effects in vitro against the strains of E. coli O157:H7 investigated. Anti-E. coli efficacy of A. nodosum PT is superior to that of terrestrial tannins purified from Quebracho and from Rhus semialata.

Effects of Chlorhexidine Digluconate on Rotational Rate of n-(9-Anthroyloxy)stearic Acid in Porphyromonas ginginvalis Outer Membranes

  • The aim of this study was to provide a basis for studying the molecular mechanism of pharmacological action of chlorhexidine digluconate. Fluorescence polarization of n-(9-anthroyloxy)stearic acid was used to examine the effect of chlorhexidine digluconate on differential rotational mobility of different positions of the number of membrane bilayer phospholipid carbon atoms. The six membrane components differed with respect to 2, 3, 6, 9, 12, and 16-(9-anthroyloxy)stearic acid (2-AS, 3-AS, 6-AS, 9-AS, 12-AS and 16-AP) probes, indicating different membrane fluidity. Chlorhexidine digluconate increased the rate of rotational mobility of hydrocarbon interior of the cultured Porphyromonas gingivalis outer membranes (OPG) in a dose-dependent manner, but decreased the mobility of surface region (membrane interface) of the OPG. Disordering or ordering effects of chlorhexidine digluconate on membrane lipids may be responsible for some, but not all of its bacteriostatic and bactericidal actions.

Development of new antibacterial materials for manufacturing functional corrugated board for agricultural products (농산물용 기능성 골판지 제조를 위한 신규 항균재료 개발에 대한 연구)

  • In this study, new antibacterial materials were developed to manufacture a functional corrugated board. Sulfur solution, a new antibacterial solution made from inorganic sulfur in the laboratory, and other antibacterial mat erials were adopted to treat the surface of a linerboard. We measured the antibacteriocidal and bacteriostatic activities, as well as the fungal resistance of the surface-treated linerboards, to identify the antibacterial properties. The mechanical properties of the surface-treated linerboard were also determined in order to identify the effects of the antibacterial materials on linerboard properties. Linerboard treated with sulfur solution, PVOH, and sodium metasulfite showed the highest antibacterial activity, while linerboard treated with sulfur solution and nano sulfur showed the highest fungal resistance. It was identified that sulfur solution has effective antibacterial properties. The antibacterial materials did not affect the mechanical properties of the surface-treated linerboard, but the binder showed significant effects in terms of the burst strength, the compressive strength, and the stiffness of the linerboard.

Comparative Antimicrobial Susceptibility by the Methods of Test (검사방법(檢査方法)에 따르는 항균제감수성(抗菌劑感受性)의 비교(比較))

  • The difference in antimicrobial susceptibility of multiply drug-resistant Salmonella typhi by the methods of test was tested against chloramphenicol(Cm), tetracycline(Tc), ampicillin(Ap), kanamycin(Km), and rifampicin(Rif), and the results were compared by the minimum inhibitory concentration(MIC). No appreciable difference was noted between MICs of Cm, Tc, and Rif measured by agar plate dilution(plate) method and broth dilution(broth) method. However, MICs of Km to about a half of test strains were 4 to 8-fold higher by broth method than plate method, and MICs of Ap by broth method were also a little higher than plate method in some strains. Cm and Rif were bacteriostatic rather than bactericidal to the most of test strains in high concentrations. Tc, Ap and Km were exclusively bactericidal to the test strains.

the Antibacterial Activities of Persimmon Juice and Persimmon Juice Dyed Cotton Fabrics (감즙과 감즙염색 면직물의 항균성)

  • The antibacterial activities against Staphylococcus aureus of persimmon juice and persimmon juice dyed cotton fabrics were eveluated. We investigated the effects in pursuit of the proper treating conditions to use persimmon juice dye for comfortable and hygienic doffing. The antibacterial activities were estimated by test method KS K 0693 and the percentage reduction of bacteria at various contact conditions of test materials and bacteria. The results were as follows. no bacteriostatic activities (BS) of persimmon Juice were more than $99.98\%$ and the increase was dose-dependent according to concentration from $1\%$ to $10\%$. However, bactericidal activities (BC) were not found BS increased from $99.97\%$ to $100\%$ and BC from $79.74\%$ to $100\%$ with increasing persimmon juice concentration of cotton fabrics from $10\%$ to $100\%$. Antibacterial effects were significantly shown at $10\%$ of persimmon juices. BS increased from $75.69\%$ to $100\%$ and BC from $65.89\%$ to $100\%$ over 18 hours of contact time and continued for 50 hours. BS of $100\%$ persimmon juice dyed cotton fabrics increased from $96.56\%$ to $98.20\%$ and BC from $-136\%$ to $65\%$ at different incubation temperatures of $17\pm1^{\circ}C$, $27\pm1^{\circ}C$ and $37\pm1^{\circ}C$. BS ranged from $99.06\%$ to $99.99\%$ and BC from $84.31\%$ to $95.10\%$, according to the initial bacterial density from $4.08\times10^7$ to $4.08\times10^5$. Sixty-seven percent of subjects of answered not to smell any odor from persimmon juice dyed cotton fabrics by subjective test.
