Effect of Stewartia koreana Nakai Branch Extracts on the Biofilm Formation of Porphyromonas gingivalis (노각나무 가지 추출물이 Porphyromonas gingivalis의 바이오필름 형성에 미치는 영향)

  • This study was investigated to evaluate the potential of Stewartia koreana Nakai as an oral healthcare material. The inhibitory effects of extracts on the biofilm formation and fimbriae genes expression of Porphyromonas gingivalis were determined by minimal inhibitory concentrations (MIC), biofilm biomass staining, SEM, and qRT-PCR analysis. The S. koreana Nakai branch was extracted into 70% ethanol, and bacteriostatic MIC of extracts against P. gingivalis were 0.6 mg/ml. In P. gingivalis cultures treated with 0.2-2.0 mg/ml of extract, biofilm production rate was significantly decreased in a concentration-dependent manner. The morphology of treated and untreated samples was observed by SEM, and cell aggregation and biofilm were only observed in those treated with extract. Subsequently, qRT-PCR analysis showed that the mRNA expression on fimbriae genes fimA and fimB was suppressed in a concentration-dependent manner. Based on these results, it can be suggested that S. koreana branch extract has the potential to be used as naturally derived oral healthcare material because of its bacteriostatic action and inhibition of P. gingivalis biofilm formation.

  • The physical and chemical characteristics of bacteriostatic water filter media are compared to granular activated carbon upon which a silver impregnated. Silver ion that has been chemically impregnated the activated carbon was the bactericidal agent in water filter media. Also, silver Ion that has teen chemically impregnated onto the activated acetate resin was the bactericidal agent in ultrasonic humidifier. Silver impregnated active carbon and silver cartridge will be able to the bactericidal agents for statical water.

  • Eighteen 2-aryl-3-(5-nitro-2-furyl) acrylamides were synthesized as antimicrobial agent, by the condensation of 2-(p-sulbstituted phenyl)-3-(5-nitro-2-furyl) acryloyl chloride with amino component, such as sulfisozxazole, sulfamethoxypyridazine and sulfadimethoxine. All compounds showed fair bacteriostatic activity against staphylococcus aureus 6538p, Bacillus subtilis, Escherichia coli, and Klebsiella pneumoniae. 2-(p-Hydroxyphenyl)-and 2-(p-acetoxyphenyl) compounds also showed antimicrobial activity against Saccharomyces cerevisiae 396 and Aspergillus oryzae.

Genomics and LC-MS Reveal Diverse Active Secondary Metabolites in Bacillus amyloliquefaciens WS-8

  • Bacillus amyloliquefaciens is an important plant disease-preventing and growth-promoting microorganism. B. amyloliquefaciens WS-8 can stimulate plant growth and has strong antifungal properties. In this study, we sequenced the complete genome of B. amyloliquefaciens WS-8 by Pacific Biosciences RSII (PacBio) Single Molecule Real-Time (SMRT) sequencing. The genome consists of one chromosome (3,929,787 bp) and no additional plasmids. The main bacteriostatic substances were determined by genome, transcriptome, and mass spectrometry data. We thereby laid a theoretical foundation for the utilization of the strain. By genomic analysis, we identified 19 putative biosynthetic gene clusters for secondary metabolites, most of which are potentially involved in the biosynthesis of numerous bioactive metabolites, including difficidin, fengycin, and surfactin. Furthermore, a potential class II lanthipeptide biosynthetic gene cluster and genes that are involved in auxin biosynthesis were found. Through the analysis of transcriptome data, we found that the key bacteriostatic genes, as predicted in the genome, exhibited different levels of mRNA expression. Through metabolite isolation, purification, and exposure experiments, we found that a variety of metabolites of WS-8 exert an inhibitory effect on the necrotrophic fungus Botrytis cinerea, which causes gray mold; by mass spectrometry, we found that the main substances are mainly iturins and fengycins. Therefore, this strain has the potential to be utilized as an antifungal agent in agriculture.

Bactericidal action by complement of fish serum (어류혈청의 보체에 의한 살균작용)

  • In order to know the defense mechanism of fish, bactericidal activity was examined into the sera of carp Cyprinus carpio, tilapia Oreochromis niloticus and flounder Paralichthys olivaseus. Each examined normal serum of fishes showed the bactericidal activity against avirulent Escherichia coli but it wasn't appeared against virulent Edwardsiella tarda. When the normal serum of each fish was treated with zymosan, it lost the bactericidal activity completely. After addition of EGTA into the normal serum of each fish, the sera of tilapia and flounder still exhibited the bactericidal activity but the serum of carp lost it. In the presence of specific antibody and complement, bactericidal activity of each antiserum was exhibited high level with in one hour incubation. On the other hand, heat inactivated antiserum showed bacteriostatic reaction. From the above results, although the activity of alternative or classical pathway of each fish complement is different, it is an important function in fish defense mechanism.

Use of Bioluminescent Indicator Acinetobacter Bacterium for Screening and Characterization of Active Antimicrobial Agents

  • Because of the need for new antimicrobial substances with novel mechanisms of action, we report here the use of an Acinetobacter reporter system for high-throughput screening of active antimicrobial agents. The bioreporter Acinetobacter strain DF4/PUTK2 carrying luciferase genes luxCDABE was chosen because of its ecological importance and it is widespread in nature. This bioreporter is genetically engineered to emit light constitutively that can be measured in real time by luminometry. Hence, this reporter system was employed to determine the bacteriostatic actions of spent-culture supernatants derived from twelve bacterial isolates. Out of the results, the strongest bioluminescence inhibitory effect of the supernatants was recorded with Bacillus cereus strain BAC (S5). Subsequently, ethyl acetate extracts of extracellular products of strain BAC (S5) were separated by a thin-layer chromatography (TLC). Based on the bioluminescence inhibitory assay, three fractions were found to have antimicrobial activity. One fraction (C) having the strongest antimicrobial activity was further purified using TLC and characterized by IR, $^1H$ NMR, mass spectrometry, SDS-PAGE, and amino acid composition analysis. The results predicted the presence of 2-pyrrolidone-S-carboxylic acid (PCA) and the octadeconic-acid-like fatty acid. Fraction C also demonstrated a broad inhibitory activity on several Gram-negative and Gram-positive bacteria. In conclusion, the Acinetobacter reporter system shows great potential to be a reliable, sensitive, and real-time indicator of the bacteriostatic actions of the antimicrobial agents.

Bacteriostatic Effect of Condensed Phosphate on the Growth of Bacteria (중합인산염의 항균효과에 관한 연구)

  • Condensed phosphates have been used in various meat products to enhance the water holding capacity, to improve texture and to prevent the development of off flavors and off odors. This study was intended to observe the effects of poly - and pyrophosphate on the growth of sanitary indicative bacteria and food poisoning bacteria. The bacteriostatic effect of poly - and pyrophosphates against Gram positive bacteria was much stronger than that of against Gram negative organisms. The effective inhibitory concentration of sodium polyphosphate on the growth of bacteria was varied by species such as $0.3\%$ to Staphylococcus aureus, $0.9\%$ to Salmonella, and more than $1.0\%$ to Escherichia coli in nutrient broth. When Staphylococcus aureus suspension was treated with $0.5\%$ sodium polyphosphate at $35^{\circ}C$ for 1 hour, the release of UV-absorbing substances from the organism was confirmed. However no significant effect was observed in Escherichia coli under the similar condition. When alaska pollack fillets were dipped in $ 3.5\%$ sodium polyphospahate at $2^{\circ}C$ for 1 min. prior to freezing, the viable cell count and coliform MPN's of the frozen product were decreased with the range of 30 to $50\%$ in comparison with those of control.

  • Porous glass ceramics composed of $Ag_2-Li_2O-CaO-TiO_2-P_2O_5$-CaO with 0.05-1.5 mole CuO were prepared by melting and 2 step heat treatment for nucleation at $610^{\circ}C$ and crystallization at $840^{\circ}C$. $\beta$-$Ca_3(PO_4)_2$crystal phase was selectively leached out in 1N-HC1 solution for 3 days, leaving $AgTi_2(PO_4)_3$and $LiTi_2(PO_4)_3$crystal phases. Antibacterial effects and characterizations of the porous glass ceramics were investigated. Staphylococcus aureus and Salmonella typhi bacteroa were used in this study. It was found that the resultant porous glass ceramics show excellent bacteriostatic properties.

  • In order to find 2-(5-nitro)furylacrylic acid derivatives possessing antimicrobial activity, nine new 2-(5-nitro)furylacrylamino acids were synthesized which were obtained by the action of 2-(5-nitro)furylacryla chloride on amino acids, such as $_{L}$-phenylalanine, glycine, $_{L}$-isoleucine, $_{L}$-glutamic acid, $_{DL}$-methionine, $_{L}$-threonine, $_{L}$-valine, $_{L}$-tryptophan and $_{DL}$-alanine, according to Schotten-Baumann method. These compounds generally showed a good bactericidal and bacteriostatic activity against Bacillus subtilis but were less effective against staphylococcus aureus, Proteus vulgaris and Escherichia coli. Of the above nine compounds, 2-(5-nitro)furylacryl glycine exhibited a good bactericidal activity.
