황련의 증량에 따른 전탕액이 Pseudomonas aeruginosa에 미치는 항균효과에 대한 실험적 연구

  • Objective : This experimental study was performed to investigate the effect of Coptidis rhizoma extract compared with quantity on Pseudomonas aeruginosa that induce keratitis. Methods : MIC was measured by dropping to 50 ${\mu}l$ according to density Coptidis rhizoma extract(100%, 10%, 1%, 0.1%) compared with quantity(40g, 80g, 120g, 160g). Anti-bacterial potency was measured by the size of inhibition zone with change of volume. Results : MIC on Pseudomonas aeruginosa in Coptidis rhizoma extract compared with quantity(40g, 80g, 120g, 160g) was not showed all samples(100%, 10%, 1%, 0.1%). Conclusions : Coptidis rhizoma extract compared with quantity was not showed anti-bacterial potency on Pseudomonas aeruginosa. The results differ that Coptidis rhizoma extract was showed anti-bacterial potency in 1999 year. Anti-bacterial Potency of Coptidis rhizoma extract compared with quantity on Pseudomonas aeruginosa must be studied.

  • Objective : This experimental study was performed to investigate the effect of Soyum Herbal-acupuncture, Hwangryunhaedoktang & Coptidis rhizoma on Staphylococcus epidermidis that induce keratitis. Methods : MIC was measured by dropping to $50{\mu}l$ according to density Soyum Herbal-acupuncture, Hwangryunhaedoktang & Coptidis rhizoma(100%, 10%, 1%, 0.1%). Anti-bacterial potency was measured by the size of inhibition zone with change of volume. Results : 1. MIC on Staphylococcus epidermidis in Soyum Herbal-acupuncture dippear all sample. 2. MIC on Staphylococcus epidermidis in Hwangryunhaedoktang disappear all sample. 3. MIC on Staphylococcus epidermidis in Coptidis rhizoma was 100%, $50{\mu}l$ and anti-bacterial potency increase compare with quantity. Conclusions : The present author think that Soyum Herbal-acupuncture must be extracted a new method for anti-bacterial potency and Coptidis rhizoma was made Herbal-acupuncture for eye drops.

  • Bacterial leaf spot by Xanthomonas campestris pv. vesicatoria has been known to cause serious problem in red pepper in Korea. However recent survey showed that most smptoms in the leaves were mixed with two different symptoms one was leaf spot and the other was canker. bacteria isolated from canker were identified as Clavibacter michiganensis subsp. michiganensis on the basis of biochemical and physiological characteristics. The causal bacteria were non-motile rod-shaped and Gram-positive. The lesions on pepper leaves appeared at first as small blisters or pimple-like white spots which enlarged in size at a later stage. The centers of some of the spots became necrotic and brown and were surrounded by a white halo. Pathogenicity tests were performed on pepper cv. Alchan seedling by spraying of bacterial suspension. During 1997 and 1998 total 17% of 527 fields surveyed were infected by C. michiganensis subsp. michiganensis. The canker of red pepper caused by C. michiganensis subsp. michiganensis was first identified in this study in Korea, and new name "gueyangbyung" was tentatively given to the disease.

  • In the case of occlusive dressings currently used in dressings for burn treatment, it is impossible to confirm the replacement time, so replacement is delayed, resulting in additional infection. To solve this problem, liposomes capable of bacterial sensing were prepared using 5(6)-Carboxyfluorescein, Phosphatidylcholine, 1,2-Dipalmitoyl-sn-glycero-3-phosphocholine, Cholesterol, and 10,12-Tricosadiynoic acid. In this study, evaluation of changes in drug encapsulation rate in liposomes according to changes in three types of phosphatidylcholine phospholipids during liposome production, high-performance phosphatidylcholine phospholipids selected through vesicle size analysis, low and high temperature stability evaluation, bacterial sensitization ability evaluation, animals cell responses were assessed.

The Experimental study of safety and efficacy in using Soyeom Pharmacopuncture solution as eyedrops (점안용 소염약침액의 안전성 및 유효성 평가)

  • Objective : This experimental study was performed to investigate the safety of Soyeom Pharmacopunture solution manufactured by extraction of alcohol and water. To identify the use of it as eyedrops, the eye irritation test of rabbits and antibacterial test of Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, and Candida albicans was performed. Methods : 1. The eye irritation test of this material was performed according to the Regulation of Korea Food & Drug Administration(2005. 10. 21, KFDA 2005-60). After Soyeom pharmacopuncture solution was administered in the left eye of the rabbits, eye irritation of the cornea, iris and conjunctiva was observed at 1, 2, 3, 4 & 7day. 2. After administering Soyeom Pharmacopuncture solution on bacterial species (Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis, MIC(Minimum Inhibition Concentration) and the size of inhibition zone were measured. Anti-bacterial potency was also measured using the size of inhibition zone. Results : 1. After Soyeom pharmacopuncture solution was administered in the left eye of the rabbits, it was found that none of nine rabbits have abnormal signs and weight changes. 2. After Soyeom pharmacopuncture solution was medicated in the left eye of the rabbits, no eye irritation of the cornea, iris and conjunctiva was observed at 1, 2, 3, 4 & 7day. 3. There was no response to MIC on bacterial species (Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) after Soyeom pharmacopuncture solution was medicated. Conclusions : The present study suggests that Soyeom pharmacopuncture solution is a non-toxic and non-irritant medicine, which does not cause eye irritation in rabbits, but dosen't have anti-bacterial effects on bacterial species which cause Keratitis. These study result recommends that more research on other herbal medicines of eye drop for Keratitis are required.

A Study on the Antibiosis and Physical & Mechanical Characteristics of Dental Resin (치과용 레진의 항균성 및 물리·기계적 특성에 대한 연구)

  • A series of experiments were conducted on dental denture base resins for their antibiosis. They were also compared in terms of physical and mechanical characteristics, which led to the following results: 1. As for bacterial deposition, the old products showed a wide range of distribution, while those products containing C. S. had restraints. 2. Among the products containing C. S., those ones of 7, 12 and 20% all had restraints of similar size to slop further bacterial growth. In the meanwhile, the old products had only small-size restraints for bacterial growth. 3. The following changes were observed in the aspect physical characteristics: There were no significance between the old products and the C. S. products. Thus, as for compressive force, the average${\pm}$standard deviation of the old products was $27.4083.{\pm}1.7397$, and those C. S. products of 20% showed the similar $27.5600{\pm}.1976$. In addition, there were no significance between the old products and those ones containing C. S. in terms of tensile force. The tensile force of the old products was $114.5600{\pm}.8916$ in average${\pm}$standard deviation, while there was no differences among the C. S. products of 7, 12, and 20% in that matter.

  • Animal cells show different behaviors in response to the mechanical properties of the substrates. We hypothesize that the rigidity of the substrates also affects the bacterial motility and controls the colony dynamics. It is found that the colony size of Escherichia colis and Bacillus subtilis grown on the agar plates is correlated with agarose gel concentrations and thus with the substrate rigidity. High- resolution microscopic imaging reveals that bacteria in single colonies form different aggregation patterns on the agar plates with varying gel concentration. We measured the apparent diffusion coefficients in the agarose gel plates made with different gel concentrations. Mathematical modeling and quantitative imaging of dye dispersion in the agar plates suggest that there is a close connection between the diffusion rate and the colony size. Nanoscale pore structures and kinetic constraints in the porous media may have an effect on bacterial colony dynamics.

Influence of Diet Induced Changes in Rumen Microbial Characteristics on Gas Production Kinetics of Straw Substrates In vitro

  • The effect of diets varying in level and source of nitrogen (N) and fermentable organic matter on dynamic characteristics of microbial populations in rumen liquor and their impact on substrate fermentation in vitro was studied. The diets tested were straw alone, straw+concentrate mixture and straw+urea molasses mineral block (UMMB) lick. The same diets were taken as substrates and tested on each inoculum collected from the diets. Diet had no effect on the amino acid (AA) composition of either bacteria or protozoa. Differences among the diets in intake, source of N and OM affected bacterial and protozoal characteristics in the rumen. Upper asymptote of gas production (Y$\alpha$) had a higher correlation with bacterial pool size and production rate than with protozoal pool size and production rate. Among the parameters of the gas production model, Y$\alpha$ and lag time in total gas has showed significant (p<0.01) correlation with bacterial characteristics. Though the rate constant of gas production significantly differed (p<0.01) between diet and type of straw, it was least influenced by the microbial characteristics. The regression coefficient of diet and type of straw for Y$\alpha$ indicated that the effect of diet on Y$\alpha$ was threefold higher than that of the straw. As microbial characteristics showed higher correlation with Y$\alpha$, and diet had more influence on the microbial characteristics, gas production on a straw diet could be used effectively to understand the microbial characteristics.
