Efficacy of Diphenyleneiodonium Chloride (DPIC) Against Diverse Plant Pathogens

  • Many of the fungicides and antibiotics currently available against plant pathogens are of limited use due to the emergence of resistant strains. In this study, we examined the effects of diphenyleneiodonium chloride (DPIC), an inhibitor of the superoxide producing enzyme NADPH oxidase, against fungal and bacterial plant pathogens. We found that DPIC inhibits fungal spore germination and bacterial cell proliferation. In addition, we demonstrated the potent antibacterial activity of DPIC using rice heads infected with the bacterial pathogen Burkholderia glumae which causes bacterial panicle blight (BPB). We found that treatment with DPIC reduced BPB when applied during the initial flowering stage of the rice heads. These results suggest that DPIC could serve as a new and useful antimicrobial agent in agriculture.

Seed-born Burkholderia glumae Infects Rice Seedling and Maintains Bacterial Population during Vegetative and Reproductive Growth Stage

  • Rice world production is affected due to the growing impact of diseases such as bacterial panicle blight, produced by Burkholderia glumae. The pathogen-induced symptoms include seedling rot, grain rot and leafsheath browning in rice plants. It is currently recognized the entrance of this pathogen to the plant, from infected seeds and from environmental sources of the microorganism. However, it is still not fully elucidated the dynamics and permanence of the pathogen in the plant, from its entry until the development of disease symptoms in seedlings or panicles. In this work it was evaluated the infection of B. glumae rice plants, starting from inoculated seeds and substrates, and its subsequent monitoring after infection. Various organs of the plant during the vegetative stage and until the beginning of the reproductive stage, were evaluated. In both inoculation models, the bacteria was maintained in the plant as an endophyte between $1{\times}10^1$ and $1{\times}10^5cfu$ of B. $glumae.g^{-1}$ of plant throughout the vegetative stage. An increase of bacterial population towards initiation of the panicle was observed, and in the maturity of the grain, an endophyte population was identified in the flag leaf at $1{\times}10^6cfu$ of B. $glumae.g^{-1}$ fresh weight of rice plant, conducting towards the symptoms of bacterial panicle blight. The results found, suggest that B. glumae in rice plants developed from infected seeds or from the substrate, can colonize seedlings, establishing and maintaining a bacterial population over time, using rice plants as habitat to survive endophyticly until formation of bacterial panicle blight symptoms.

Development of Near-Isogenic Lines (NILs) Conferring Xa4, xa5 and Xa21 Genes Resistant to Bacterial Blight (Xanthomonas oryzae pv. oryzae) in japonica rice Genetic Background

  • Near-isogenic lines (NILs) carrying bacterial blight resistance genes (Xa4, xa5 and Xa21) were developed in japonica rice using Suweon345 as genetic background. NILs were selected by gene specific DNA markers and inoculation of K1 or K3a race. NILs conferring Xa4 were resistant to K1, K2, K3, and moderately resistant to K3a. NILs conferring xa5 were resistant to K1, K2, K3, and K3a. NILs having Xa21 were susceptible to K1, while resistant to K2, K3 and K3a. Target genes of NILs with the genetic background of Suweon345 were also confirmed by using eleven Philippines races and International Rice Bacterial Blight (IRBB) NILs carrying Xa4, xa5 and Xa21. All NILs had no significant difference from their recurrent parents in the major agronomic traits except for panicle length and brown rice 1,000 grain weight. Heading date of NILs ranged from Aug. 10 to Aug. 11, which was similar to that of recurrent parent, Suweon345. Culm length, number of grains per panicle and ratio of ripened grain of NILs were similar to those of Suweon345. Milled rice of NILs was ranged from 4.82 to 4.93MT/ha. These NILs will be useful for improving resistance to K3a race of bacterial blight pathogens in Korean japonica cultivars.

벼 생육시기별 흰잎마름병 발병이 쌀 수량감소 및 미질에 미치는 영향 (Effects of Bacterial Leaf Blight Occurrence on Rice Yield and Grain Quality in Different Rice Growth Stage)

  • 2002년부터 2005년까지 4년간 벼 흰잎마름병 초발병 조사결과 2002년보다 초발병일이 20$\sim$30일 이상 빨라지고 있음을 확인하였다. 벼 흰잎마름병 발병시기 및 병반면적율에 따라 수량이 감소하였는데 병반면적률이 10% 이하에서는 피해가 적었으나, 병반적율이 증가함에 따라서 수량감소가 점점 많아졌고, 25% 이상일 때부터 수량감소 및 현미천립중 등에서 현저히 낮아지고 병반면적율이 50% 이상이고 유수형성기 발병시 수량 29%, 현미천립중 2.3g, 현미완전립률 18%까지 감소되었다. 밥맛의 기준이 되는 식미치인 점도에서도 병반면적율이 증가함에 따라 낮은 식미치를 보였다.

  • 2004년은 전년에 비해 기온이 높았고 강수량이 적었다. 벼 못자리 병, 잎도열병 발생 필지, 잎집무늬마름병, 흰잎마름병과 고추 역병, 탄저병 및 바이러스병, 시설참외의 급성 시들음 증상, 흰가루병 등이 2003년에 비해 증가하였다. 특히 벼잎집무늬마름병과 흰잎마름병은 전년에 비해 2배 이상 병 발생이 증가하였다. 벼 이삭도열병이 전년 $3\%$에서 2004년 $0.7\%$로 감소된 것은 출수기 동안 평균기온은 높았으나 강수일수, 강수량 및 습도가 낮아 잎도열병균이 이삭으로의 전파를 억제한 것으로 추정되었다. 고추역병은 전년에 비해 크게 감소되었다. 그리고 발표하지 않은 다른 병에 의한 피해는 다소 감소 혹은 유사한 수준이었다.

Genomics-based Sensitive and Specific Novel Primers for Simultaneous Detection of Burkholderia glumae and Burkholderia gladioli in Rice Seeds

  • Panicle blight and seed rot disease caused mainly by Burkholderia glumae and Burkholderia gladioli is threatening rice cultivation worldwide. The bacteria have been reported as seed-borne pathogens from rice. Accurate detection of both pathogens on the seeds is very important for limiting the disease dissemination. Novel primer pairs targeting specific molecular markers were developed for the robust detection of B. glumae and B. gladioli. The designed primers were specific in detecting the target species with no apparent cross-reactions with other related Burkholderia species at the expected product size. Both primer pairs displayed a high degree of sensitivity for detection of B. glumae and B. gladioli separately in monoplex PCR or simultaneously in duplex PCR from both extracted gDNA and directly preheated bacterial cell suspensions. Limit of detection was as low as 0.1 ng of gDNA of both species and $3.86{\times}10^2cells$ for B. glumae and $5.85{\times}10^2cells$ for B. gladioli. On inoculated rice seeds, the designed primers could separately or simultaneously detect B. glumae and B. gladioli with a detection limit as low as $1.86{\times}10^3cells$ per rice seed for B. glumae and $1.04{\times}10^4cells$ per rice seed of B. gladioli. The novel primers maybe valuable as a more sensitive, specific, and robust tool for the efficient simultaneous detection of B. glumae and B. gladioli on rice seeds, which is important in combating rice panicle blight and seed rot by early detection and confirmation of the dissemination of pathogen-free rice seeds.

Development of a Selective Medium for Surveillance of Fusarium Head Blight Disease

  • Fusarium head blight (FHB), predominantly caused by Fusarium graminearum and F. asiaticum, is a significant fungal disease impacting small-grain cereals. The absence of highly resistant cultivars underscores the need for vigilant FHB surveillance to mitigate its detrimental effects. In 2023, a notable FHB outbreak occurred in the southern region of Korea. We assessed FHB disease severity by quantifying infected spikelets and grains. Isolating fungal pathogens from infected samples often encounters interference from various microorganisms. We developed a cost-effective, selective medium, named BGT (Burkholderia glumae Toxoflavin) medium, utilizing B. glumae, which is primarily known for causing bacterial panicle blight in rice. This medium exhibited selective growth properties, predominantly supporting Fusarium spp., while substantially inhibiting the growth of other fungi. Using the BGT medium, we isolated F. graminearum and F. asiaticum from infected wheat and barley samples across Korea. To further streamline the process, we used a direct PCR approach to amplify the translation elongation factor 1-α (TEF-1α) region without a separate genomic DNA extraction step. Phylogenetic analysis of the TEF-1α region revealed that the majority of the isolates were identified as F. asiaticum. Our results demonstrate that BGT medium is an effective tool for FHB diagnosis and Fusarium strain isolation.

Bacillus oryzicola sp. nov., an Endophytic Bacterium Isolated from the Roots of Rice with Antimicrobial, Plant Growth Promoting, and Systemic Resistance Inducing Activities in Rice

  • Biological control of major rice diseases has been attempted in several rice-growing countries in Asia during the last few decades and its application using antagonistic bacteria has proved to be somewhat successful for controlling various fungal diseases in field trials. Two novel endophytic Bacillus species, designated strains YC7007 and $YC7010^T$, with antimicrobial, plant growth-promoting, and systemic resistance-inducing activities were isolated from the roots of rice in paddy fields at Jinju, Korea, and their multifunctional activities were analyzed. Strain YC7007 inhibited mycelial growth of major rice fungal pathogens strongly in vitro. Bacterial blight and panicle blight caused by Xanthomonas oryzae pv. oryzae (KACC 10208) and Burkholderia glumae (KACC 44022), respectively, were also suppressed effectively by drenching a bacterial suspension ($10^7cfu/ml$) of strain YC7007 on the rhizosphere of rice. Additionally, strain YC7007 promoted the growth of rice seedlings with higher germination rates and more tillers than the untreated control. The taxonomic position of the strains was also investigated. Phylogenetic analyses based on 16S rRNA gene sequences indicated that both strains belong to the genus Bacillus, with high similarity to the closely related strains, Bacillus siamensis KACC $15859^T$ (99.67%), Bacillus methylotrophicus KACC $13105^T$ (99.65%), Bacillus amyloliquefaciens subsp. plantarum KACC $17177^T$ (99.60%), and Bacillus tequilensis KACC $15944^T$ (99.45%). The DNA-DNA relatedness value between strain $YC7010^T$ and the most closely related strain, B. siamensis KACC $15859^T$ was $50.4{\pm}3.5%$, but it was $91.5{\pm}11.0%$ between two strains YC7007 and $YC7010^T$, indicating the same species. The major fatty acids of two strains were anteiso-$C_{15:0}$ and iso $C_{15:0}$. Both strains contained MK-7 as a major respiratory quinone system. The G+C contents of the genomic DNA of two strains were 50.5 mol% and 51.2 mol%, respectively. Based on these polyphasic studies, the two strains YC7007 and $YC7010^T$ represent novel species of the genus Bacillus, for which the name Bacillus oryzicola sp. nov. is proposed. The type strain is $YC7010^T$ (= KACC $18228^T$). Taken together, our findings suggest that novel endophytic Bacillus strains can be used for the biological control of rice diseases.

2001년 농작물 병해 발생개황 (Review of Disease Incidence of Major Crops in 2001)

  • 2001년 기상의 특징은 3월부터 5월까지 계속된 극심한 가뭄과 생육기간 내내 계속된 고온현상 그리고 별다른 태풍의 내습 없이 9, 10월 수확기 맑은날의 지속으로 요약할 수 있다. 벼는 순조로운 기상 때문에 잎, 이삭도열병, 세균성벼알마름병, 흰잎마름병, 잎집무늬마름병의 발생이 예년에 비해 경미하였으나, 감비추진 정책에 의해 생육후기 깨씨무늬병이 발생하여 전국적으로 문제시되었으며 그동안 발생이 적었던 줄무늬잎마름병이 경기, 충청, 호남 서해안지역을 중심으로 많이 발생하였다. 고추는 생육기 비가 많이 왔던 전남북 지역에서 역병 발생이 심하였으며 토마토와 오이는 대부분 예년에 비해 병발생이 적었다. 수박의 경우는 꽃자리 부분이 썩는 푸른곰팡이병이 전국적으로 발생하였으며 CGMMV는 전년에 비해 상당폭 발생면적이 감소하였고 노지재배의 경우 탄저병 발생이 예년에 비해 심하였다. 참외와 딸기도 전년에 비하여 병발생이 경미하였으며 예년에 발생이 많았던 참외의 급성위조증상도 발생이 대폭 감소하였다. 마늘·양파의 흑색썩음균핵병은 봄철의 가뭄과 고온현상으로 병발생의 전년의 절반수준으로 감소하였다. 감자의 풋마름병은 발병포장이 증가하는 경향이었으며 고구마의 덩굴쪼김병도 발생이 감소하였다. 사과의 경우 전반적으로 병 발생이 예년에 비해 경미하였으며 배는 흰가루병이 많이 발생하였다. 맥류의 붉은곰팡이병은 예년처럼 거기 발생이 없었다.

  • 2005년은 전년에 비해 기온이 낮았고 강수량이 적었다. 벼 흰잎마름병, 세균성벼알마름병, 벼 깨씨무늬병, 벼 이삭도열병, 배나무 겹무늬병 등이 2004년에 비해 증가하였다. 특히 벼 깨씨무늬병은 전년에 비해 4배 이상 병 발생이 증가하였다. 벼 이삭도열병이 전년 0.7%에서 2005년 2%로 증가된 것은 출수기 동안 평균기온은 높았고 강수일수, 강수량 및 습도가 높아 잎도열병균이 이삭으로의 전파를 촉진한 것으로 추정되었다. 기타 조사된 병들은 전년에 비해 감소 혹은 유사한 수준이었다. 벼 잎도열병, 시설참외 시들음증상, 흰가루병 및 노균병은 전년에 비해 크게 감소되었다.
