Evaluation of Traits of Button Mushroom Agaricus bisporus (양송이버섯 수집균주의 다양한 형질특성평가)

  • White button mushroom (Agaricus bisporus) is the most widely consumed mushroom in the world, and domestic consumption is increasing owing to expansion of the westernized diet in recent years. Before 2000, most Korean mushroom farmers cultivated foreign varieties, but recently, Korean breeders are developing hybrid strains using molecular breeding tools. To produce a better mushroom cultivar, we evaluated some traits of button mushroom Agaricus bisporus strains. Mycelial growth rate at different culture temperatures was investigated by 25>20>15>30>$10^{\circ}C$. Recently developed domestic varieties had superior mycelial growth rate compared to the traditionally grown species. In particular, the ASI1338 strain showed excellent mycelial growth rate at different temperatures. Further, we confirmed that ASI1007, ASI1085, ASI1310, ASI1339 were strongly resistant to bacterial brown blotch, whereas ASI1053, ASI1103, ASI1140, ASI1146, ASI1177, ASI1183, ASI1195, ASI1321, ASI1331, ASI1336 were relatively weakly resistant. The average number of days for 16.5, and that for harvest was 5.7; the average yield was 142.7 g/2-kg bag. Mycelial growth rates at 10, 15, 20, $25^{\circ}C$ were similar, but that at $30^{\circ}C$ was lower. The mushroom yield was highly correlated with the number of days for pinheading and harvest.

Identification of Pseudomonas fluorescens antagonistic to Pseudomonas tolaasii and its cultivation (버섯의 갈변병 유발세균 Pseudomonas tolaasii의 길항세균인 Pseudomonas fluorescens의 분리동정 및 배양조건)

  • A Pseudomonas fluorescens was selected from mushrooms and studied in both batch and fed-batch cultures in order to get maximal biomass concentration. P. fluorescens is an aerobic bacterium and antagonistic to Pseudomonas tolaasii which causes blotch disease on the mushroom cap. P fluarescens and P. tolaasii were identified by Gram staining, gelatin liquefaction, oxidase test, etc. and were characterized by pigment production, temperature sensitivity, salt tolerance and rapid pitting test, etc., Celts of P. fluorescens well in medium containing 30g/L of glucose, whereas the growth was inhibited at the glucose levels at higher than 30g/L. The highest values of specific growth rate and productivity were obtained when using 10g/1 of yeast extract. Optimum concentrations of $NH_4Cl$ and ${(NH_4Cl)}_2SO_4$ for culture were found to be 1.0g/L and 0.1g/L respectively. Optimum concentration of $MgSO_4{\cdot}7H_2O$ used as a sulfursource was 1.0g/L. It was also found that the cell concentrations reached the maximum level when grown on the medium containing 1.0g/L of $KH_2PO_4$ and 0.1g/L of $CaCl_2$. Also, the optimum culture conditions were $30^{\circ}C$ and pH 6.0. Cultivation of P. fluarescens at high dissolved oxygen (DO) concentration led to a decrease of bacterial productivity in batch culture. Maximum productivity was achieved at 40% DO concentration.

Production of Flammulina velutipes by Using Culture Medium Containing Herb Medicine Refuse and Analysis of Characteristics of Its Fruiting Bodies (폐 한방슬러지 첨가배지를 이용한 팽이버섯의 생산 및 자실체 특성 검토)

  • This study aims to reduce the production cost of mushrooms and therefore boost the income of mushroom producers. The addition of a 10% concentration of herb medicine refuse was adequate for promoting mycelium growth in Flammulina velutipes. A moisture content of 60% in the culture medium was adequate for promoting hyphal growth. The optimum temperature and humidity were $16^{\circ}C$ and 70%-85%, respectively. The average yield of Flammulina velutipes fruting body per 1,100 ml bottle was 275-282 g, which is similar to that of the control. The outbreak of diseases such as mildew and bacterial blotch in mushroom was never observed in the bottle treated with herb medicine refuse. Fewer deformed and second-class quality mushrooms were observed than in the control group. Deviation reduced, and mushroom growth was observed to be significantly uniform. The winter mushroom had a white fruiting body with a hemispherical shape. The diameter of the mushroom cap was 9.2-9.3 mm, stipe length was 12.7-12.8 cm, and thickness was 3.3-3.4 mm; these were almost the same as those in the control group. The partial hardness of winter mushrooms in both the experimental and the control groups showed that the stipe ($54.6-57.3g/cm^2$) is a little harder than the mushroom cap ($46.8-47.6g/cm^2$). The calories and nutrients per 100 g of mushrooms in the control and samples were similar to each other.

Effect of Microwave Plasma on Sterilization of Acidovorax citrulli Infected Watermelon Seeds (저온플라즈마(Microwave Plasma)를 활용한 Acidovorax citrulli 감염 수박종자의 살균 효과 검정)

  • This study was conducted to check the sterilization efficacy of microwave plasma (MWP) against the watermelon seeds infected with Acidovorax citrulli 11-251. Watermelon seeds were artificially vacuum inoculated to produce A. citrulli 11-251 infected seeds. Aac ImmunoStrip and scanning electron microscope (SEM) results suggests that, seeds (coat and endosperm) were infected under the concentration of 1×107/30 min. MWP sterlization process was carried out at 50 W (3 min, 5 min, and 10 min), 80 W (3 min, 5 min, and 10 min), and 100 W (3 min, 5 min, and 10 min). According to the results, MWP sterilized the artificially inoculated seed coats by 95.96% at 80 W/10 min and seed endosperms by 100% at 100 W/10 min respectively. Although, seeds were sterlized by MWP, germination rate of seeds were low as compared to non treated (negative control) seeds. Moreover, cell membrane of A. citrulli 11-251 was damaged while observed in SEM after sterilized with MWP. Further studies regarding the appropriate sterilization condition by MWP against A. citrulli infected seeds for germination will be conducted in our next study.
