Sodium hypochlorite alkaline was tested against Pseudomonas tolaasii causing bacterial blotch on cultivated oyster mushroom (Pleurotus ostreatus). The minimum inhibitory concentration of sodium hypochlorite against P. tolaasii contained active chlorine (AC) at 1.4 mg/l on plate assay. The highest cultivation yield was obtained from the treatment of AC 5.7 mg/l. Treatment of sodium hypochlorite at the rate of higher than AC 11.4 mg/l resulted in reduced yields at the harvest. However, the population of total bacteria on the bed surface treated with AC 5.7 mg/l of sodium hypochlorite was maintained to some extent. Inhibitory concentration against total bacteria on the bed surface was over AC 22.8 mg/l. Mushroom mycelium was damaged and its growth strongly inhibited at the concentration of AC 200 mg/l. Mushroom caps showed yellowish symptom by chemical injury by treatments of AC 74.1 mg/l or higher. Sporocarps infected by P. tolaasii were irrevocable at any concentration of sodium hypochlorite. Routine watering with AC 5.7 mg/l from mushroom initiation to the end of picking resulted in reduced bacterial blotch incidence of 40% and 86% at two mushroom farms. The treatment resulted in higher quality mushroom production compared to that conventionally watered with tap water alone.
During the period from 2013 to 2014, disease occurrences by various pathogens in apple cultivars have been investigated in northern Gyeongbuk province of Korea. Anthracnose, white rot, Alternaria leaf spot, Marssonina blotch, and bacterial shoot blight as major diseases have been observed. Pathogens isolated from the symptomatic plants were identified as Colletotrichum gloeosporioides for anthracnose, Botryosphaeria dothidea for white rot, Alternaria alternata for Alternaria leaf spot, Marssonina mali for Marssonina blotch, and Pseudomonas syringae pv. syringae for bacterial shoot blight. Of all diseases, the bacterial shoot blight has been severely increased in chronically infested fields in Gyeongbuk province.

In September and October 2005, melons(Cucumis melo L.) from the commercial greenhouses in Naju and Gwangju exhibited severe foliar necrosis and fruit rot. Leaf symptoms initially appeared as V-shaped, necrotic lesions and extending to the midrib. Symptoms on the fruit were occurred randomly as necrotic and sunken spots. Two isolates from diseased leaves and fruits were identified as Acidovorax avenae subsp. citrulli on the basis of bacteriological and genetic characteristics. Pathogenicity of the isolates was confirmed by inoculating on 3-week-old melon and cucumber seedlings. This is the first report of bacterial fruit blotch of melon in Korea.
Acidovorax citrulli causes bacterial fruit blotch in Cucurbitaceae, including watermelon. Although A. citrulli is a seed-borne pathogen, it can cause diverse symptoms in other plant organs like leaves, stems and fruits. To determine the infection routes of A. citrulli, we examined the virulence of six isolates (Ac0, Ac1, Ac2, Ac4, Ac8, and Ac11) on watermelon using several inoculation methods. Among six isolates, DNA polymorphism reveals that three isolates Ac0, Ac1, and Ac4 belong to Clonal Complex (CC) group II and the others do CC group I. Ac0, Ac4, and Ac8 isolates efficiently infected seeds during germination in soil, and Ac0 and Ac4 also infected the roots of watermelon seedlings wounded prior to inoculation. Infection through leaves was successful only by three isolates belonging to CC group II, and two of these also infected the mature watermelon fruits. Ac2 did not cause the disease in all assays. Interestingly, three putative type III effectors (Aave_2166, Aave_2708, and Aave_3062) with intact forms were only found in CC group II. Overall, our results indicate that A. citrulli can infect watermelons through diverse routes, and the CC grouping of A. citrulli was only correlated with virulence in leaf infection assays.
A DNA fragment which is involved in tolassin production was cloned to obtain a molecular marker of Pseudomonas tolaasii, a casual agent of bacterial brown blotch disease of oyster mushroom (Pleurotus ostreatus). Tolaasin is a lipodepsipeptide toxin and known as a primary disease determinant of the P. tolaasii. It is responsible for formation of white line in agar when P. tolaasii were cultured against white line reacting organisms (WLROs). White line negative mutants (WL-) were generated by conjugation between rifampicin resistant strain of P. tolaasii and E. coli carrying suicidal plasmid pSUP2021 : : Tn5. The ability of tolaasin production of the WL- mutants was examined by hemolysis test, pathogenicity test, and high pressure liquid chromatography (HPLC) analysis of culture filtrate. All of the WL- mutants were lost the ability of tolaasin production (Tol-). Genomic library of the Tol- mutant was constructed in pLAFR3 and the cosmid clone containing Tn5 was selected. DNA fragment fro franking region of Tn5 was cloned from the plasmid and used as a probe in Southern blot. DNA-DNA hybridization with the probe to total DNA from group of bacteria ecologically similar to P. tolaasii including WLORs, fluorescent Pseudomonads isolated from oyster mushroom, P. agarici, P. gingeri, and some of other species of Psedomonas showed that some of the tested bacteria do not have any hybridized band and others have bands sowing RFLP. The cloned DNA fragment or its nucleotide sequence will be useful in detection and identification of the P. tolaasii.

Acidovorax citrulli is a causal agent for bacterial fruit blotch on watermelon. Here, we report the complete genome sequence of A. citrulli strain KACC17005. The genome contains 5,349,924 bp with G + C contents of 68.54%, including 4,520 protein coding genes in a circular chromosome. It also possesses at least 15 genes encoding putative type III effector proteins, which may contribute to promoting virulence in susceptible hosts or triggering immune responses in resistant hosts.
Studies were conducted to determine the potential of sodium hypochlorite(SHC) on the control of bacterial yellow blotch in cultivated oyster mushroom, Pleurotus ostreatus. SHC at the concentration of 80 ppm was effective on the control of Pseudomonas agarici causing yellow blotch in oyster mushroom except number 916 isolate. In vitro the mycelial growth was slightly inhibited at the concentration higher than 100 ppm of sodium hypochlorite, but retardation of the mycelial growth was soon recovered. Spray of SHC solution at the concentration of 40-50 ppm per day significantly reduced the incidence of the yellow blotch without impairing the growth of oyster mushroom in field culture. However, the higher concentration of SHC(67 ppm) induced yellow brown or dark gray in color and deformed cap and elongated stripe in morphology of fruiting body. Results indicate that periodical spray of sodium hypochlorite seems to be the recommendable method for protection against bacterial yellow blotch disease in oyster mushroom without reducing food quality.
The 2 strains of Pleurotus ostreatus showing very severe and mild symptom of brown blotch were selected, and their F1 hybrid strains were made by mating of their single spore isolates. Hydrophobicity of fruiting body surface and brown blotch severity of the parental and their 11 F1 hybrid strains showing different level of severity of brown blotch were determined. Correlation coefficient (R) between disease severity and hydrophobicity were 0.68 and 0.70 on two independent experiments. Correlation coefficient between disease severity and disease incidence that was determined in the oyster mushroom farm were 0.46 and 0.52 on two independent experiments. When GFP-tagged Pseudomonas tolaasii was monitored with confocal microscope on cap surface of fruiting body, more cells of the pathogen were clustered on the severe strain than the mild strain, which indicates that the bacterial pathogen proliferates more on the severe strain. These results suggest that hydrophobicity of oyster mushroom fruiting body affects disease severity of the brown blotch disease; the longer the bacterial suspension stays on fruiting body surface more severe symptom of the blotch develops.
Bacterial fruit blotch (BFB) caused by Acidovorax avenae subsp. citrulli is defective disease to watermelon cultivated areas. To control of BFB, we investigated control efficiency to use commercial antibacterial pesticides. Growth inhibition zone on medium were formed as oxolinic acid WP and oxytetracycline WP. Control efficacy of four anti-bacterial pesticides on seed and seedling stage were performed. As a results, oxytetracyclin WP is shown over 90% control efficiency on seed and acibenzolar-S-methyl + mancozeb WP shown over 90% control efficiency on seedling stage Hot-water treatment method could be possible to reduced infection rate on seed. The conditions of hot-water treatments are $50{\sim}55^{\circ}C$ on 20~30 minutes. These results suggested that the methods were helpful watermelon seedling nursery to control of the bacterial fruit blotch by A. avenae subsp. citrulli.

Bacterial fruit blotch (BFB) caused by Acidovorax citrulli is a devastating disease found in many cucurbits cultivation fields. The genetic diversity for 29 strains of A. citrulli collected from various cucurbits in South Korea was determined by DNA fingerprinting with a pathogenicity test, multi locus analysis, Rep-PCR (repetitive sequence polymerase chain reaction), and URP (universal rice primers) PCR bands. Two distinct groups (Korean Clonal Complex, KCC1 and KCC2) in the population were identified based on group specific genetic variation in the multi locus phylogeny using six conserved loci and showed a very high similarity with DNA sequences for representative foreign groups [the group I (CC1-1 type) and the group II (CC2-5 type)] widely distributed worldwide, respectively. Additionally, in the case of phaC, a new genotype was found within each Korean group. The KCC1 was more heterogeneous compared to the KCC2. The KCC1 recovered mainly from melons and watermelons (ratio of 6 : 3) and 15 of the 20 KCC2 strains recovered from watermelons were dominant in the pathogen population. Accordingly, this study found that two distinct groups of differentiated A. citrulli exist in South Korea, genetically very similar to representative foreign groups, with a new genotype in each group resulting in their genetic diversity.
