Isolation of Novel Alkalophilic Bacillus alcalophilus subsp. YB380 and the Characteristics of Its Yeast Cell Wall Hydrolase

  • An alkalophilic mi.croorganism (strain YB380), which produces yeast cell wall hydrolase extracellulary, was isolated from Korean soil. The rod-shaped cells were 0.3~0.4 by 2~4${\mu}{\textrm}{m}$ long, motile, aerobic, gram-positive, and spore-forming. The color of the colony was light yellow. The temperature range for growth at pH 9.0 was 25 to $45{\circ}C, with optimum growth at $35{\circ}C. The pH range for growth at $35{\circ}C was 8 to 11 with an optimum pH of 9.0. Therefore, the strain YB380 is an obligate alkalophile. The 16S rRNA of strain YB380 has a 99% sequence similarity with that of Bacillus alcalophilus. On the basis of physiological properties, cell wall fatty acid composition, and phylogenetic analysis, we propose that the isolated strain is Bacillus alcalophilus. The yeast cell wall hydrolase from Bacillus alcalophilus subsp. YB380 has been purified and partially characterized. The molecular weight was estimated to be 27,000 daltons with an optimum temperature and pH of $60{\circ}C and 9.0, respectively. The N-terminal amino acid sequence of the enzyme was analyzed as Gln- Thr- Val- Pro- Trp- Gly- Ile- Asn- Arg- Val.

Purification and Characterization of Cell Wall Hydrolase from Alkalophilic Bacillus mutanolyticus YU5215

  • Streptococcus mutans has the capacity of inducing dental caries. Thus, to develop a novel way of preventing dental caries, a cell wall hydrolase-producing strain was isolated and its characteristics were investigated. Among 200 alkalophilic strains isolated from soil, 8 strains exhibited lytic activities against Streptococcus mutans. However, strain YU5215 with the highest cell wall hydrolase activity was selected for further study. Strain YU5215 was identified as a novel strain of Bacillus based on analyzing its 16S rDNA sequence and Bergey's Manual of Systematic Bacteriology, and thus designated as Bacillus mutanolyticus YU5215. The optimal conditions for the production of the cell wall hydrolase from Bacillus mutanolyticus YU5215 consisted of glucose ($0.8\%$), yeast extract ($1.2\%$), polypeptone ($0.5\%$), $K_{2}HPO_{4}\;(0.1\%$), $MgSO_{4}{\cdot}7H_{2}O$ ($0.02\%$), and $Na_{2}CO_{3}\;(1.0\%$) at pH 10.0. Bacillus mutanolyticus YU5215 was cultured at 30^{circ}C for 72 h to produce the cell wall hydrolase, which was then purified by acetone precipitation and CM-agarose column chromatography. The molecular weight of the lytic enzyme was determined as 22,700 Da by SDS-PAGE. When the cell wall peptidoglycan of Streptococcus mutans was digested with the lytic enzyme, no increase in the reducing sugars was observed, while the free amino acids increased, indicating that the lytic enzyme had an endopeptidase-like property. The amino terminus of the cell wall peptidoglycan digested by the lytic enzyme was determined as a glutamic acid, while the lytic site of the lytic enzyme in the Streptococcus mutans peptidoglycan was identified as the peptide linkage of L-Ala and D-Glu.

Isolation and Characterization of a Protease-Producing Bacterium, Bacillus amyloliquefaciens P27 from Meju as a Probiotic Starter for Fermented Meat Products

  • This study was performed to select protease-producing Bacillus sp. as a potential probiotic starter for fermented meat products. In order to isolate protease-producing bacterium from meju, measured the diameter of the clear zone on agar plate (TSA, 1% (w/v) skim milk) and analyzed for intracellular protease activity, then 10 Bacillus-like strains were isolated. Three Bacillus-like strains (P19, P27, and P33) among 10 strains were able to tolerate in acidic condition (TSB, pH 2.5, 2 h incubation). These 3 strains were showed antimicrobial activity against food-borne pathogenic bacteria. These vegetative cells of 3 strains were showed a survival rate of 0.04% to 0.08% under the artificial gastric acidic condition (TSB, pH 2.5 with 1% (w/v) pepsin), but spore-forming cells were 56.29% to 84.77%. Vegetative cells of 3 strains were the least bile-resistant, while spore-forming cells of 3 strains showed higher survival rate more than 76% under artificial bile condition (TSB, 0.1% (w/v) oxgall bile). In these strains, P27 strain was finally selected as a good probiotic strain. P27 strain was tentatively identified as Bacillus amyloliquefaciens by API CHB kit and 16S rDNA sequence analysis. The results of this study suggest that B. amyloliquefaciens P27 can be used as a potential probiotic starter for fermented meat product.

양송이 배지로부터 분리된 Bacillus subtilis NO12의 특성 (Isolation and characterization of Bacillus subtilis NO12 from button mushroom substrates)

  • Cellulase와 xylanase 분비능이 우수한 세균을 분리하기 위하여 부여군 석성면 지역의 양송이 재배농장으로부터 수확후배지를 수집하였다. 양송이 수확후배지로부터 12종의 균주를 분리하였으며 이 중 cellulase와 xylanase 활성이 가장 우수한 균주 NO12를 최종 선발하였다. 분리균 NO12의 생리적 생화학적 특성은 Bacillus ID kit와 MicroLog system을 이용하여 조사하였으며 분리균 NO12는 Bacillus subtilis와 유사한 특징을 나타내었다. 분리균 NO12의 16S rDNA 염기서열도 B. subtilis와 99.2%의 상동성을 나타내었다. 이와 같은 결과를 종합하여 분리균 NO12는 B. subtilis NO12로 동정되었다. 분리균이 분비하는 cellulase와 xylanase 활성은 분리균이 증식함에 따라 대수증식기 중반부터 급격히 증가하였고 정지기에 진입하면 효소활성이 더 이상 증가하지 않는 것으로 나타났으며 xylanase 활성은 대수증식기 초기부터 지속적으로 증가하여 대수증식기 중반에 최대활성을 나타내었다.

Bacillus cereus에 대한 길항적 저해 작용과 biogenic amines 분해 능력을 지닌 Bacillus licheniformis SCK A08 균의 특성 (Characterization of Bacillus licheniformis SCK A08 with Antagonistic Property Against Bacillus cereus and Degrading Capacity of Biogenic Amines)

  • Biogenic amine은 주로 단백질이 풍부한 전통장류 중 된장과 간장에서 높게 발생하고 있다. 현재까지 국내외 식중독 사고를 보면 biogenic amine중 tyramine, histamine과 putrescine에 의한 보고가 있다. 전통장류의 biogenic amine에 대한 위생적인 문제를 해결하기 위해 전통장류 제조에 적합한 발효미생물 중에서 biogenic amine 비생성 및 분해하는 미생물을 선발하여 종균으로 적용한다면 biogenic amine에 대한 문제를 해결할 수 있을 것이다. 이러한 문제의 해결을 위해 본 연구에서는 우수한 발효능력과 함께 유해균에 대해 강한 길항 능력을 지니며, biogenic amine을 생산하지 않지만 높은 분해능력을 보이는 균주를 선발하여 장류제조 종균으로의 이용가능성을 실증하였다. 결론적으로 선발된 장류발효균주를 종균으로 이용하여 메주제조 및 청국장 제조 등에 이용한다면 biogenic amine과 Bacillus cereus를 안전한 수준으로 낮출 수 있을 것으로 사료되었고, 전통장류의 HACCP적용을 위한 HAZARD를 관리하는 우수한 수단으로 이용될 수 있을 것이다. 또한 향후에는 단일 균종만 종균으로 사용할 경우 전통장류의 우수한 풍미를 재현하는데 한계가 있기 때문에 종균들을 적절하게 조합한 혼합발효를 통해 제조한다면 풍미가 좋을 뿐만 아니라, 위생문제를 해결한 우수한 장류를 생산할 수 있을 것으로 판단되었다.

산업폐수로부터 분리한 인제거 미생물의 인 제거 특성 (Phosphorus Removal Characteristics by Bacteria Isolated from Industrial Wastewater)

  • 본 연구에서는 산업폐수로부터 분리한 3가지 종의 인 제거율을 확인하였다. 3 가지 분리된 균을 동정한 결과 A1균은 Stenotrophomonas maltophilia strain CUPS 3, A2균은 Rhodococcus erythropolis strain Sco-C01, A3균은 Bacillus sp. 3434BRRJ이었다. 3 가지 분리된 균의 인 제거 및 경유분해를 확인한 결과 인 제거율은 Bacillus sp. 3434BRRJ > Stenotrophomonas maltophilia strain CUPS균 > Rhodococcus erythropolis strain Sco-C01균 순으로 높은 것으로 나타났으며 제거율은 약 99%, 50%, 20%로 확인되었다. 따라서 이 후 실험은 인 제거율이 가장 높았던 Bacillus sp. 3434BRRJ를 가지고 실험하였다. Bacillus sp. 3434BRRJ의 인 제거를 확인하기 위하여 온도별(15, 25 그리고 $30^{\circ}C$), 인 농도별(20, 30 그리고 40 mg/L) 그리고 산소조건(호기, 혐기/호기조건) 및 탄소원을 달리하여 실험하였다. 그 결과Bacillus sp. 3434BRRJ는 $30^{\circ}C$에서 인 제거가 가장 좋았으며 20 mg/L의 인은 약 99% 처리하였고 혐기조건을 거치고 탄소원으로 acetate와 glucose를 혼합하여 처리하였을 때 가장 효율이 좋은 것으로 나타났다. 본 연구에서는 Bacillus sp. 3434BRRJ의 인 제거를 확인 하였다. 균의 생장이 높을수록 인의 제거율은 높았으며, 미생물을 이용한 환경오염물질의 제거에 있어서 가장 중요한 것은 미생물의 활성을 높이는 환경조건을 제공하는 것이다.

Bacillus sp. LSC11가 생산하는 biosurfactant의 특성 (Characteristics of Biosurfactants produced by Bacillus sp. LSC11)

  • 원유 분해능이 강력한 해양균주를 얻고자 유류오염 지역으로부터 crude oil을 탄소원으로 이용하는 수십 종을 분리하였다. 분리된 균주중 원유분해능 및 biosurfactant 생성능이 우수한 균주를 선별하여, 형태학적, 생화학적 및 생리학적 특성을 조사한 후 Bacizzus sp. LSC11로 동정하였다. Bacillus sp. LSC11 균주 배양액의 표면장력은 최저 32mN/m까지 감소되었다. Biosurfactant의 CMC값은 0.0035% (w/v) 인 것으로 나타났다. Bacillus sp. LSC11가 생산하는 biosurfactant의 유화활성은 대두유에서 최대였으며, 원유에서도 높은 편이였다. 유화안정성은 합성계면활성제와 비슷하거나 우수하였다.

유기용매내성세균 Bacillus sp. BCNU 5006의 유용성 (Potential of Organic Solvent Tolerant Bacillus sp. BCNU 5006)

  • In the screening process of organic solvent tolerant bacteria showing good growth in media containing several kinds of organic solvents, one strain was isolated and identified as Bacillus sp. BCNU 5006. The strain was able to tolerate many organic solvents including benzene, toluene, xylene, octane, dodecane, butanol and ethylbenzene. Likewise, it could also utilize these solvents as the sole source of carbon with significant enzyme production. The lipolytic enzyme stability of Bacillus sp. BCNU 5006 was studied in the presence of several kinds of solvents at a 25% (v/v) concentration. The highest enzyme stability was observed in the presence of octane (107%), followed by ethylbenzene (88%), decane (86%), and chloroform (85%). Especially, BCNU 5006 lipase was determined to be more stable than immobilized enzyme (Novozyme 435) in the presence of octane, chloroform and xylene. This organic solvent tolerant Bacillus sp. BCNU 5006 could be expected as a potential bioremediation agent and biocatalyst for biodegradation and provide on organic-solvent-based enzymatic synthetic method in industrial chemical processes.

Antimicrobial Activities and Probiotic Properties of Bacillus sp. Strains Isolated from Fermented Cooked Rice

  • Fermented cooked rice is known to have remedial properties and good for health, but there is a lack of scientific knowledge to prove their beneficial effects. In this study, we investigated the presence of antibiotic-producing bacteria in fermented cooked rice. The probiotic properties of the bioactive isolates were also investigated. A total of seven pure isolates were isolated from fermented cooked rice prepared from parboiled cooked Aus rice in the lab following traditional methods. All the isolates were gram-positive, can grow at thermophilic range of temperatures, and all but CRS9 were able to coagulate milk. Three strains exhibited moderate to high broad-spectrum antibacterial activity against the test bacteria including Shigella brodie, Bacillus cereus, Escherichia coli, Pseudomonas sp., Staphylococcus aureus, and Proteus sp. Analysis of 16S rDNA sequences of the strain CRS8 and CRS11 showed that they belong to the genus Bacillus as they exhibited >99% identity to several strains of Bacillus. Both strains could survive the highly acidic conditions and can tolerate bile acid indicating their potential to be the candidate probiotic strains. The strain CRS8 was resistant to penicillins and cephalosporins including amoxicillin, penicillin, cephalothin, however, the CRS11 was sensitive to all the antibiotics tested. This is the first report that fermented cooked rice is a source of antibiotic-producing Bacillus sp. The probiotic properties of the Bacillus isolates from fermented cooked rice were also investigated for the first time.

Complete Genome Sequencing of Bacillus velezensis WRN014, and Comparison with Genome Sequences of other Bacillus velezensis Strains

  • Bacillus velezensis strain WRN014 was isolated from banana fields in Hainan, China. Bacillus velezensis is an important member of the plant growth-promoting rhizobacteria (PGPR) which can enhance plant growth and control soil-borne disease. The complete genome of Bacillus velezensis WRN014 was sequenced by combining Illumina Hiseq 2500 system and Pacific Biosciences SMRT high-throughput sequencing technologies. Then, the genome of Bacillus velezensis WRN014, together with 45 other completed genome sequences of the Bacillus velezensis strains, were comparatively studied. The genome of Bacillus velezensis WRN014 was 4,063,541bp in length and contained 4,062 coding sequences, 9 genomic islands and 13 gene clusters. The results of comparative genomic analysis provide evidence that (i) The 46 Bacillus velezensis strains formed 2 obviously closely related clades in phylogenetic trees. (ii) The pangenome in this study is open and is increasing with the addition of new sequenced genomes. (iii) Analysis of single nucleotide polymorphisms (SNPs) revealed local diversification of the 46 Bacillus velezensis genomes. Surprisingly, SNPs were not evenly distributed throughout the whole genome. (iv) Analysis of gene clusters revealed that rich gene clusters spread over Bacillus velezensis strains and some gene clusters are conserved in different strains. This study reveals that the strain WRN014 and other Bacillus velezensis strains have potential to be used as PGPR and biopesticide.
