The Bacterial Biological Response Modifier Enriched Chungkookjang Fermentation (면역증강물질 강화 청국장 발효)

  • The aim of this study was to produce Chungkookjang-a food produced through fermentation with Bacillus licheniformis E1-that contains an increased concentration of a bacterial biological response modifier (B-BRM). Unfortunately, sensory studies have indicated that B. licheniformis E1-fermented Chungkookjang is unacceptable for commercial use. We isolated another bacterial strain from this food product: B. subtilis S2. The optimum time and temperature for Chungkookjang fermentation with B. licheniformis E1 and B. subtilis S2 were 48 hr and $40^{\circ}C$, respectively. Sensory studies showed that Chungkookjang fermented by both B. licheniformis E1 and B. subtilis S2 was more acceptable than B. licheniformis E1 only. The amino nitrogen and crude protein content of the product were 359 mg% and 45.6% respectively. Additionally, it was confirmed that the proliferation of mouse splenic lymphocytes increased significantly, when the cells were treated with the BRM from Chungkookjang fermented using the mixture of bacterial strains in vitro. These results suggest that the enriched Chungkookjang may help patients who are medically in need of potentiation of lymphocytes proliferation.

Comparative Study of Toxicological Methods and Field Resistance to Insecticides in Diamondback moth(Lepidoptera: Plutellidae) (배추좀나방(Plutella xylostella L.)의 독성시험방법 비교와 지역별 약제저항성에 관한 연구)

  • These studies were conducted to evaluate the five comparative test methods for detecting chemical resistance and to investigate resistant level of field populations of diamondback moth (Plutella xylostella L.). Leaf disc method was practically rocomrnendable because of its rapidity and low CV(l1.4%). Topical application method was a precise replicabiliLy(CV=8.00/0) but it was time consuming and difficult in mampulation. The other 3 methods showed higher CV ranging from 14.9% to 21.4%. Based on $LC_{50}$ values by topical application method, field populations of diamondback moth collected from 4 different regions, Kwangju, Kimhae, Jeju, and Inje to prothiofos showed from 3.3 to 61.1 times higher resistance than the susceptible strain, whereas to cypermethrin, Lhey were from 7.5 to 141.7 times higher than the susceptible. To cartap hydrochloride, they showed from 10.5- to 33.3-fold resistant levels as high as the susceptible. Finally, based on $LC_{50}$ values to Bacillus thuringiensis by leaf disc technique, the resistant levels of the field populations were from 1.9 Lo 8.1 times as compared to the susceptible.

Biofilm Formation Characteristics of Major Foodborne Pathogens on Polyethylene and Stainless Steel Surfaces

  • This research was investigated the effects of temperature and time against the formation of biofilms by foodborne pathogens on surfaces of polyethylene and stainless steel. After preliminary experiments with 32 strains from 6 species of foodborne pathogens (Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Salmonella Typhimurium), one strain from each species with the highest biofilm formation efficiency was selected. All foodborne pathogens showed a tendency toward an increased ability for biofilm formation with increasing temperature, but there was no consistency between the two materials and between foodborne pathogens. At all tested temperatures, the biofilm formation ability of E. coli and P. aeruginosa on the polyethylene surface was higher than that on the stainless steel surface with significant differences. The foodborne pathogens all formed biofilms immediately upon inoculation, and biofilm formation by E. coli, P. aeruginosa, and S. Typhimurium increased on both the polyethylene and stainless steel surfaces at 1 h after inoculation compared to at 0 h. At 7 days after biofilm formation, the other strains except S. aureus showed no difference in survival rates on polyethylene and stainless steel. The ability of these 6 foodborne pathogens to form biofilms showed different trends depending on the type of bacteria and the instrument material, i.e., polyethylene and stainless steel.

The Effects of Dietary Soybean Fermented with Aspergillus oryzae or Bacillus natto on Egg Production and Egg Lipid Composition in Layer

  • This study was conducted to determine the effects of dietary low grade soybean, fermented with Aspergillus oryzae (FSB 1) or Bacillus subtilis var. natto (FSB 2), on egg production and quality, fat and cholesterol content, and the fatty acid (FA) profile of eggs by lipid layer. A total of 18 Hi-Line strain layers, 22 wk of age, were randomly assigned to three dietary treatments: no fermented soybean (control), control with 15% FSB 1 (C + FSB 1), and control with 15% FSB 2 (C + FSB 2). The rate of egg production and egg weight were evaluated between two periods: one was from the 1st to 4th wk and the other was from the 5th to 8th wk. At the 8th wk, a total of 30 eggs were randomly selected from each treatment group and analyzed for physical quality, fat content, fatty acid composition and cholesterol content. The results showed that egg production was increased in hens fed with diets containing fermented soybeans from the 5th to 8th wk period (p<0.01). A similar tendency was observed through eight weeks' cumulative egg production (p<0.05). There were no significant differences in egg production between the C + FSB 1 and C + FSB 2 treatment groups (p>0.05). Egg weight and other physical properties did not vary between treatment groups (p>0.05). Egg yolks among different treatment groups were similar in fat content, but egg yolks in the C + FSB 1 and C + FSB 2 groups had lower oleic acid (p<0.05), higher linoleic, ${\alpha}$-linolenic, and arachidonic acids (p<0.01), and lower cholesterol content (p<0.05) than those in the control group. In conclusion, supplementation of fermented low grade soybeans might be useful as a functional feedstuff to improve egg production and quality for a healthy human diet.

Interactions between Biosynthetic Pathway and Productivity of IAA in Some Rhizobacteria (근권에서 분리한 세균의 IAA 생합성 경로와 IAA 생성능과의 관계)

  • This study explores the interaction between the production of indole-3-acetic acid (IAA), a typical phytohormone auxin and the role of IAA biosynthetic pathways in each IAA producing rhizobacterial strain. The bacterial strains were isolated from rhizosphere of wild plants and identified as Acinetobacter guillouiae SW5, Bacillus thuringiensis SW17, Rhodococcus equi SW9, and Lysinibacillus fusiformis SW13. A. guillouiae SW5 exhibited the highest production of IAA using tryptophan-dependent pathways among the 4 strains. When indole-3-acetamide (IAM) was added, Rhodococcus equi SW9 showed the highest IAA production of $3824{\mu}g/mg$ protein using amidase activity. A. guillouiae SW5 also showed the highest production of IAA using two pathways with indole-3-acetonitrile (IAN), and its nitrile hydratase activity might be higher than nitrilase. B. thuringiensis SW17 showed the lowest IAA production, and most of IAA might be produced by the amidase activity, although the nitrilase activity was the highest among 4 strains. The roles of nitrile converting enzymes were relatively similar in IAA synthesis by Lysinibacillus fusiformis SW13. Tryptophan-independent pathway of IAA production was utilized by only A. guillouiae SW5.

Antibacterial Activities of Suaeda maritima Extract. (제주도 해홍나물(Suaeda maritima) 추출물의 항균활성)

  • To develop natural food preservatives, methanol and water extracts were prepared from the Suaeda maritima and their antibacterial activities were examined against 12 microorganisms which were food borne pathogens bacteria, food poisoning microorganisms and food-related bacteria. Methanol extracts exhibited antibacterial activities for the 5 Gram positve and 7 Gram negative bacteria by agar diffusion method, The antibacterial activities and cell growth inhibition were investigated on each strain with the different concentrations of Suaeda maritima extracts. Antibacterial activities were shown in root, stem, furit extracts of Suaeda maritima. However stem and fruit extracts showed weak antibacterial activity against the tested microorganisms. Root extracts showed the highest antibacterial activities against microorganisms tested, such as Bacillus cereus,Bacillus subtilis, Vibrio parahaemolyticus, Staphylococcus aureus. The highest antibacterial activity against bacteria test was found in the methanol extract.

Inhibitory Effects of Candidate Probiotic Bacteria on the Growth of Fish Pathogenic Bacteria, Streptococcus sp. (Candidate Probiotic Bacteria의 어류병원성 Streptococcus sp. 성장에 대한 억제 효과)

  • For the treatments and protection of bacterial fish disease, many requirements are needed for aquatic probiotics so that they are effective in aquaculture animals but are also harmless to humans. In the present study, among 17 candidate probiotic bacteria (CPB) obtained from the edible part of the shellfish, Bacillus sp. CPB-St (CPB-St) were selected and in vitro evaluated for the possibility as a probiotic strain for the control of fish streptococcosis which frequently occurs in the olive flounder farms. CPB-St showed inhibitory effects on the growth of various fish pathogenic bacteria, Streptococcus sp., S. parauberis, S. iniae, Lactococcus garvieae and L. piscium by the double layer test ranging about 18~26 mm of clear zone. Inhibitory activity of CPB-St to Streptococcus sp. was observed 6 hours after and the growth of Streptococcus sp. was decreased to 8~55 folds in the co-culture of CPB-St with Streptococcus sp.. The safety of CPB-St to fish and survival of CPB-St in the intestine were assessed in the olive flounder, Paralichthys olivaceus. Fish mortality was not observed in artificial infection with CPB-St for 2 weeks. CPB-St was entirely excreted from the stomach and intestine 24 hours after oral injection. This results indicate that CPB-St has potential applications as a probiotic for the control of fish streptococcosis in aquaculture.

Characterization of Bacterial Community in the Ecosystem Amended with Phenol (페놀이 첨가된 생태계에서 세균 군집구조 변화의 분석)

  • The effect of phenol on the change of bacterial community in the effluent water from a wastewater treatment plant was analyzed by PCR and terminal restriction fragment length polymorphism (T-RFLP). The fragments of 16S rDNA were amplified by PCR with bacterial primers, where one of the primers was biotinylated at the 5'-end. After digestion with restriction enzymes, HaeIII and AluI, the biotinylated terminal restriction tragments (T-RFs) of the digested products were selectively isolated by using streptavidin paramagnetic particles. The single-stranded DNA of T-RFs was separated by electrophoresis on a polyacrylamide gel and detected by silver staining technique. When 10 standard strains were analyzed by our method, each strain had a unique T-RF which corresponded to the calculated size from the known sequences of RDP database. The T-RFLP fingerprint generated from the effluent water was very complex, and the predominant T-RFs corresponded to members of the genus Acinetobacter, Bacillus and Pseudomonas. In addition, the perturbation of bacterial community was observed when phenol was added to the sample at the final concentration of 250 $l^{-1}$. The number of T-RFs increased and the major bacterial population could be assigned to the genus Acinetobacter, Comamonas, Cytophaga and Pseudomonas. A intense band assigned to the putative genera of Acinetobacter and Cytophaga was eluted, amplified, and sequenced. The nucleotide sequence of the T-RF showed close relationship with the sequence of Acinetobacter junii.

Selection of Biogenic Amine-reducing Microorganisms from a Traditional Korean-style Fermented Food, $Cheonggukjang$ (전통 발효 청국장으로부터 biogenic amine 저생성 미생물의 선발)

  • Microorganisms, having the lower decarboxylase activity, among the isolated strains from $cheonggukjang$ and rice-straw in this study were selected by using biogenic amine (BA) media. The selected strains were identified as $Bacillus$ $subtilis$ HH12, $B.$ $subtilis$ HR254, and $Paenibacillus$ $barcinonensis$ KR97, by using 16S rRNA analysis. PCR analysis showed that the histidine decarboxylase ($hdc$) gene was absent in the HH12, HR254, and KR97 strains. However, PCR analysis showed that the tyrosine decarboxylase ($tdc$) gene was present in the HH12, HR254, and KR97 strains. Quantitative analysis of the selected strains by using high-performance liquid chromatography showed that histamine was absent in the HH12, HR254, and KR97 strains. However, these 3 strains showed tyramine concentrations of 6.09, 3.68, and 6.30 mg/L, respectively. These strains produced lower concentrations of amines (approximately 7.9, 0, and 9.3% amines in the HH12, HR254, and KR97 strains, respectively) than the $B.$ $subtilis$ MC138 strain, which showed the higher protease activity.

Characterization of Lactobacilli Isolated from Chicken Ceca as Probiotics (닭의 맹장에서 분리된 유산균의 생균제적 특성)

  • This experiment was conducted to investigate enzyme activity, antimicrobial activity, and antibiotics susceptibility of Lactobacilli strain(Lactobacillus reuteri BLA5, Lactobacillus crispatus BLA7, Lactobacillus reuteri BLA9, Lactobacillus amylovorus LLA7, Lactobacillus crispatus LLA9, Lactobacillus vaginalis LLA11) isolated from chicken ceca and were selected by organic acid synthesis, acid tolerance, bile salt tolerance. The enzymes activities were different among strains of Lactobacilli. The amylase activity and lipase activity of Lactobacillus were high but cellulase activity and protease activity of that were low. Lactobacillus culture showed high antimicrobial activity against E. coli but low antimicrobial activity against Salmonella. The inhibitory factor of Lactobacilli isolated from chickens’ cecum on E. coli was low pH by organic acid. All of Lactobacillus isolated from chicken’s cecum were susceptible to ampicillin and amoxicillin but weren’t susceptible at the optimum level of feed additive antibiotics(virginiamycin and salinomycin).
