  • Various bacterial strains that secret extracellular fibrinolytic enzyme were screened from Doen-Jang, a traditional soybean fermented food in Korea. Five microbes of them were identified to be Bacillus sp. strains according to Bergey's manual of systematic bacteriology. The culture filtrates of B. amyloliquefaciens (2.46 plasmin unit/ml) and B. pantothenticus (3.82 plasmin unit/ml) showed a level of fibrinolytic activity that was about three times higher than that of plasmin 1.0 unit and Bacillus subtilis showed the highest fibrinolytic activity (4.94 plasmin unit/ml). All of the extracellular proteases showing the fibrinolytic activity are confirmed by SDS-PAGE followed by reverse fibrin zymogram activity assay and we proposed that some of the fibrinolytic enzymes from this work are novel enzymes.

  • Emerging of antibiotic resistance of pathogenic bacteria is now a very serious problem in the clinics to treat the diseases, which have been easy to cure by antibiotic treatments before. Unfortunately, antibiotics developed till now are not effective any more against the resistant bacteria. Lots of efforts to discover new antibiotics having novel and unique structures and functions are really urgent and undergoing in the whole world. In this study, we tried to screen and isolate Same unique bacterial strains producing antibacterial substances from the sea water, which is the poor environment for bacteria 10 make their growing. Three bacterial strains among 916 strains isolated showed inhibition clear zone on the marine agar plate growing pathogenic bacteria including Acinetobacter baumannii, Edwardsiella tarda, Pseudomonas aeruginosa, Staphylococcus aureus, Salmonella enterica. One of them, which was identified as Bacillus sp. DH-9 from 16S rRNA gene analysis, showed especially considerable antibacterial activity against S. aureus which is notorious for methicillin resistant S. aureus (MRSA). The growth of S. aureus was totally inhibited when the supernatant of Bacillus sp. DH-9 culture was treated on.

  • In this study, established soil-borne Bacillus sp. FF-9 with strong antifungal activity was isolated for identification and to determine optimal culture condition. By using 16s rDNA sequencing method, FF-9 of the selected bacteria was identified as genus Bacillus sp., Bacillus sp. FF-9 was cultured at $30^{\circ}C$, for 24 h in the LB medium. Cell growth increased quickly after 6 h and the highest cell growth was indicated at 12 h. The most antifungal activity against Rhizoctoina solani AGII-II appeared at 18 h and the optimal temperature and pH were 30 and pH 8.0, respectively. A testing of carbon and nitrogen sources showed the highest antifungal activity at 1% lactose and 1% yeast extract Furthermore an addition of salt showed the most antibiotic activity in the 0.15% $K_2HPO_4$.

Characterization of the Functional Properties of Soy Milk Cake Fermented by Bacillus sp.

  • The mucilage production and tyrosine content in soy milk cake (SMC) fermented by Bacillus firmus NA-1, Bacillus subtilis GT-D, and B. subtilis KU-A was improved by fortification with 10% defatted soybean flour. The fibrinolytic activity and consistency of the SMC were drastically increased by solid-state fermentation for 1 day. However, the consistency of the fermented SMC gradually decreased during fermentation for 3 days. Furthermore, the tyrosine content of the freeze-dried powder of SMC fermented by three Bacillus sp. was 9 times higher than that of unfermented SMC. The soybean proteins, including the 7S and 11S subunits, were partially digested during alkaline fermentation, producing lower molecular-weight peptides. The fibrinolytic enzyme produced in SMC fermented by B. firmus NA-l and B. subtilis KU-A exhibited higher thermal stability than that of B. subtilis GT-D fermentation. The powder obtained from B. subtilis GT-D fermentation had an ${\alpha}$-amylase activity and lower consistency compared to those of B. firmus NA-1 and B. subtilis KU-A. In addition, this powder contained 6.3% moisture content, 27% crude protein content and 9 units of fibrinolytic activity and proteolytic activity.

Production and Characterization of Cyclodextrin Glucanotransferase fronm Bacillus sp. JK-43 Isolated from Kimchi (김치 분리균인 Bacillus sp. JK-43이 생산하는 Cyclodextrin Glucanotransferase의 생산 및 특성)

  • A bacterial strain, designated as JK-43, producing extracellular cyclodextrin glucanotransferase (CGTase)[EC 2.4.1.19] was isolated from kimchi. The CGTase from isolated strain JK-43 showed the transglucosylation activity from soluble starch to L-ascorbic acid(AA) compared to those obtained from other strains. A main product formed by this reaction was identified as $2-O-{\alpha}-glucopyranosyl$ L-ascorbic acid(AA-2G) by testing its susceptibility to ${\alpha}-glucosidase$ hydrolysis, the HPLC profiles, and through the elementary analysis. the ${\beta}-CD,\;{\gamma}-CD$, potato starch and corn starch were identified to be suitable glucosyl donor for transglucosylation reaction on AA by CGTase. Acceptor specificity on AA-2G production was examined by use of AA, Iso-AA and AA-2P. Transglucosylation was observed toward AA-2P as well as AA and Iso-AA. The microorganism isolated from kimchi was identified as a strain of Bacillus sp. JK-43 based on the morphological, cultural, biochemical characteristics and partial 16SrDNA sequence analysis. The maximal CGTase production was observed in a medium containing 1.0% soluble starch, 1.0% yeast extract, 1.0% $Na_2CO_3\;0.1%\;K_2HPO_4,\;and\;0.02%\;MgSO_4{\cdot}7H_2O$ with initial pH 7.0. The strain was cultured at $37^{\circ}C$ for 26 hrs with reciprocal shaking.

  • Bacillus sp. SP-KSW3 is an auxothroph bacteria that is being used for starter in fermentation. Physico-chemical characteristics, enzyme activities, ACE inhibitor and antimutagenicity in fermented soybean inoculated with Bacillus sp. SP-KSW3 starter was investigated for the ripening duration of fermentation. Tyrosinase and ACE showed 10% higher activity degree on test field than control. for antimutagenicity using S. enterica serovar Typhimurium TA100 against MNNG and NPD showed 86.24% and 75.63% Similarly, S. enterica serovar Typhimurium TA98 was used against NPD and NQO showed 60.28% and 50.92% respectively. Hydrogen donating ability increased compared to the control having 81.7% and 80.1% respectively. Daidzin of isoflavone in fermented soybean showed higher concentration in control than in the test field. Genistein from two years of ripening test field contained 11.67 mg/kg compared to the test field. The initial test field for daidzin contained 389.96 mg/kg which increased to 453.67 mg/kg after two years and the initial genistein contained 402.68 mg/kg which also increased to 556.86 mg/kg.

  • Approximately 80 species of bacteria were isolated from the fermented mushroom first and the HJ-57 antibacterial micro-organism was selected to the final isolation bacteria. It has a high degree of CMCase, amylase, and protease activity as well as high antibacterial activity against mushroom pathogenic bacteria without affecting the growth and development of Flammulina velutipes and Lentinus edodes mushrooms. The finally selected HJ-57 antibacterial micro-organism was identified as Bacillus sp. HJ-57. The initial pH for culture was pH7 and its optimum culture temperature was $35^{\circ}C$. The antibacterial material produced by Bacillus sp. HJ-57 showed a little antibacterial activity even in the 12 hr of culture, but showed the highest antibacterial activity in the 36~48 hr of culture. The HJ-57 antibacterial micro-organism also showed a high antibacterial activity against mushroom pathogenic bacteria and molds in the corn cob contained culture medium is used in Flammulina velutipes cultivators.

Isolation and Characterization of a Xylanolytic Bacterium, Bacillus sp. MX47 (Xylanase 생산균 Bacillus sp. MX47의 분리 및 동정)

  • A xylanolytic bacterial strain, MX47, was isolated from rotting plant matter in soil. The strain was aerobic and gram positive, and grew between pH 6.0 and 11.0. Cells were susceptible to thiostrepton and chloramphenicol. The major fatty acids (>3%) comprised 64.55% of iso-$C_{15:0}$, 22.76% of anteiso-$C_{15:0}$, and 3.92% of iso-$C_{17:0}$. The G/C content of the DNA was 44.15 mol%. The predominant respiratory quinone was menaquinone 7 (MK-7). Searches for 16S rRNA gene sequence similarity as well as phylogenetic analyses strongly suggested that the strain should be classified to the genus Bacillus. However, its biochemical characteristics, including acid production and enzyme activities, are different from those of other Bacillus strains in the same clade, and therefore, we propose the name Bacillus sp. MX47.

Antifungal Activity of Bacillus sp. AM-651 Against Phytophthora capsici (고추역병 유발병원균 Phytophthora capsici에 대한 Bacillus sp. AM-651의 항진균활성)

  • Biological antagonists of Phytophthora capsici were isolated from soil in Gyeongbuk, Korea. Among the isolated bacteria, a Bacillus sp. was identified from l6S rDNA sequence analysis and named Bacillus sp. AM-651. Bacillus sp. AM-65l strain which can strongly a antifungal activity against Phytophthora capsici. Culture conditions for the maximum production of the antagonistic substance were optimized. The production of antibiotic were high on modified Davis mineral medium pH 7 at $30^{\circ}C$. The medium for highest production of the agonistic substance optimized. It is composed the best activity on glucose, $(NH_4)_2SO_4$ and $K_2HPO_4$ at 0.5%, 0.1%, and 0.7%, respectively. By time course of culture solution selected Bacillus sp. AM-65l, the culture solution after 48hrs had strongly growth inhibition rate against P. capsici. And culture solution of Bacillus sp. AM-651 was stable within a pH range $5{\sim}11$ and temperature range $4{\sim}70^{\circ}C$. Bacillus sp. AM-651 cultured broth shown fungal growth inhibitory activity against B. sorokiniana, B. cinerea, R. solani avove and beyond P. capsici and comparatively showed a high activity against C. gloeosporioides, B. dothidea, B. cinerea and F. graminearum by agar diffusion method.

Purification and Characterization of Extracellular Inulinase from Bacillus sp. (Bacillus sp.가 세포외로 생산하는 Inulinase의 정제 및 특성)

  • The extracellular inulinase from Bacillus spp. was purified to a single protein through a sequence of operations including ammonium sulfate fractionation, heat treatment, DEAE Sepharose C1-6B ion exchange chromatography, Sephadex 6-100 and Sephadex 6-150 gel filtration. The purified enzyme was confirmed to be a $\beta$ -D-fructofuranosidase(EC 3.2.1.26) which was much more active on sucrose than on inulin(I/S = 0.2). The maximal inulinase activity was observed at pH 6.0 and at the temperature of $50^{\circ}C$. The mo1ecular weight of the enzyme was about 56, 000. Tryptophan and histidine residues of the enzyme molecule were found to be essential for its catalytic activity.
