A New Salt-Tolerant Thermostable Cellulase from a Marine Bacillus sp. Strain

  • A salt-tolerant cellulase secreted by a marine Bacillus sp. SR22 strain with wide resistance to temperature and pH was purified and characterized. Its approximate mass was 37 kDa. The endoglucanase, named as Bc22Cel, was purified by ammonium sulfate precipitation, gel filtration chromatography, and extraction from the gel after non-reducing sodium dodecyl sufate-polyacrylamide gel electrophoresis. The optimal pH value and temperature of Bc22Cel were 6.5 and $60^{\circ}C$, respectively. The purified Bc22Cel showed a considerable halophilic property, being able to maintain more than 70% of residual activity even when pre-incubated with 1.5 M NaCl for 1 h. Kinetic analysis of the purified enzyme showed the $K_m$ and $V_{max}$ to be 0.704 mg/ml and $29.85{\mu}mol{\cdot}ml^{-1}{\cdot}min^{-1}$, respectively. Taken together, the present data indicate Bc22Cel as a potential and useful candidate for industrial applications, such as the bioconversion of sugarcane bagasse to its derivatives.

Effect of Probiotics on Water Quality in the Shrimp (Fenneropenaeus chinensis) Ponds (대하 (Fenneropenaeus chinensis) 양식장 사육수에 미치는 Probiotics의 효과)

  • Applications of probiotics to shrimp ponds were carried out to determine their effects on water quality. Fermented solutions consisting of Bacillus spp. and Nitrosomonas spp. were applied to a 4 ha shrimp (Fenneropenaeus chinensis) pond from July to September, 2000. In the pond treated with probiotics, daily variations of DO and pH, and concentrations of DIN and DIP were lower than those in the ponds without probiotic treatment. Concentration of phytoplankton was less variable and the number of species was more variable in the probiotic-treated pond than those in the control pond. Variation of bacterial numbers and the number of Vibrio spp. were lower in the treated pond than those in the control pond. It is confirmed that the probiotics can be used to improve water quality of the shrimp ponds.

Improvement effect of total nitrogen and amino acid content in spent mushroom substrates by bacterial treatment (세균을 이용한 수확후배지의 총질소 및 아미노산 증진 효과)

  • This study was conducted to reuse spent mushroom substrates (SMS) of Pleurotus ostreatus and improve their nitrogen content by bacterial treatment. Two kinds of bacteria were used to investigate the increase in total nitrogen (T-N) content. Bacillus sp. (GM20-4) was isolated from SMS of oyster mushroom, and Rhodobacter sphaeroides (RS) was obtained from Gwangju Si Agricultural Technology Center. SMS samples were collected from three oyster mushroom cultivation farms located in Gyeonggi-do province, Korea. When dried SMS was inoculated with 30% culture broth of GM20-4 and RS and incubated at room temperature ($25{\pm}2^{\circ}C$) for 5 days, T-N content increased. To investigate the T-N content of other SMS, three dried SMS samples (A, B, and C) were treated by the same method using GM20-4 and RS. As a result, the T-N content of sample B was 20% higher than that of the control, whereas the T-N content of samples A and C increased to 17% and 12%, respectively. The change in T-N content by bacterial treatment of wet SMS was slightly higher than that of the control. The changes in amino acid content were also found to be higher than those in the control in all SMS samples by GM20-4 and RS treatment. Aspartic acid and glutamic acid contents were the highest among all amino acid compositions. Especially, the aspartic and glutamic acid contents of sample B increased by 2.9 folds higher than the control.

A Study on the Odor Characteristics and Identification of Microbial in Biological Swine Manure Treatment Process by Bioreactor (생물반응기를 도입한 돈분뇨의 생물학적 처리공정에서 악취발생 특성 및 미생물동정에 관한 연구)

  • A bioreactor filling with pellets and stones was equipped to the swine manure treatment system, which is expected to emit high concentration of odor in the process of the organic wastewater treatment system, and in comparison with the activated sludge process as the control process, the reactor operation state, treatment water quality and odor emission concentration were measured. The reactor using the bioreactor proved to be much more stable in the bubble condition, treatment water transparency, etc, and BOD removal efficiency was also much better. The removal efficiency of T-N and T-P, however, showed little difference in the two reactors. Odor, as a result of examining $NH_3-N$, $NH_3$ concentratio, and complex odor, was 4 times to 24 times less emitted in the system using bioreactor than in the activated sludge system. $H_2S$, methyl mercaptan, dimethyl sulfide, and dimethyl disulfide were not found or were found in only 5 ppbs in each reactor and showed little difference between the two reactors. In the bioreactor process, Bacillus sp./ Pseudomonas sp. species were mainly found and in the activated sludge process, acterium sp. Chryseobacterium sp. species were mainly found.

Changes of Microbial Community Depending on Different Dissolved Oxygen in Biological Nitrogen Removal Process (생물학적 질소제거 공정에서 용존산소변화에 따른 미생물의 군집변화)

  • PCR-DGGE method was applied to analyze changes of microbial community in simultaneous nitrification and denitrification (SND) bioreactor with various DO concentrations. In the analysis of eubacterial community, band profiles of DGGE were similar with 2 or 1 mg/L DO concentrations in the reactor. Experimental results led to 16 different bacteria being identified, including 5 dominant strains(3 strains of Uncultured Bacterium, 1 strains of Bacillus, 1 strains of Uncultured Bacteroidetes). DGGE results at 0.5 mg/L DO concentration led to 12 strains being identified, including 7 dominant strains(5 strains of Uncultured Bacterium, 2 strains of Zoogloea sp.). DGGE results at 0.1 mg/L DO concentration led to 11 strains being identified, including 3 dominant strains(1 strains of Uncultured Bacterium, 2 strains of Zoogloea sp.). In DGGE band profiles of $\beta$-AOB($\beta$-Ammonia Oxidizing Bacteria), only one band was observed. This band had 97% similarity with Nitrosomonas sp. done DNB Y20. This band was clearly observed at the 2, 1 and 0.5 mg/L DO concentrations, while the brightness of the band at 0.1 mg/L DO concentration was mostly dimmed. In DGGE band profiles of denitrification process, 5 bands(3 strains of Uncultured organism containing nirS, 2 strains of Uncultured organism containing nirK) were observed. Among those bands, the brightness of one band was gradually increased at the lower DO concentrations. This band has 86% identity with Uncultured organism clone eS1 cd1 nirS gene, partial cds. Based on this result, it could be concluded that Uncultured organism clone eS1 cd1 nirS gene, partial cds is a predominant microorganism in the denitrification process.

Isolation and Identification of Bacillus sp. LAM 97-44 Producing Antifungal Antibiotics (항진균성 항생물질을 생산하는 Bacillus sp. LAM 97-44의 분리 및 동정)

  • In order to develop an effective antifungal antibiotics, over 700 isolates of bacteria, mold and actinomytes were screened from soil, and LAM 97-44 were selected as a strain producing the strong antifungal antibiotics against Candida albicans. Morphological, cultural and physiological characteristics of LAM 97-44 were investigated for the indentification. The cell size of LAM 97-44 was $2{\sim}3{\times}1{\sim}1.5\;{\mu}m$, and the shape of spore was of ellipsoidal. As a carbon source, LAM 97-44 utilized fructose, glucose, glycerol, maltose and raffinose but did not utilize arabinose, cellulose and xylose. The fatty acids of the cells included various iso-type and anteiso-type. Conclusively, the strain LAM 97-44 was proved to be Bacillus subtilis.

Biological Control of Plant Pathogens by Bacillus sp. AB02. (Bacillus sp. AB02를 이용한 식물 병원균에 대한 생물검정)

  • In the greenhouse fields for fruits and vegetables during the winter in Korea, there are serious damages by the sclerotium diseases due to the low temperature and humidity. This study was carried out to select an antagonic agent for the biological control of the sclerotium diseases. The 55 antagonic agents were selected from the rhizosphere in soil where the fruits and vegetables were cultivated in the green house fields, and strain AB02 among the tested isolates was estimated to be the strongest antagonist against the sclerotium disease. Using strain AB02, the antifungal spectrum was tested against 5 different plant pathogens. According to the results of the test, strain AB02 . showed the high antagonistic effect against Botrytis cinerea and Sclerotinia sclerotiorum. For the experiment of biological control against the sclerotium disease, it was estimated the suppression effect and the control effect by the strain in the pot experiment using the green perilla. According to the result of the pot experiments, the suppression effect was 40% and the control effect was 62%, respectively. For the stimulation effect of the tested plant growth by strain AB02 compared to the control, it was improved as 120% for the total length, 141% for the liveweight, 121% for the total number of leaves, 185% for the leaf area, and 327% for the liveweight of the root, respectively. Strain AB02 showing the antagonistic effect against the sclerotium disease and the stimulation effect for the plant growth was identified as Bacillus sp.

Structure and Regulation of a Complex Promoter Region from an Alkali-tolerent Bacillus sp.

  • A DNA fragment from an alkali-tolerent Bacillus sp., conferring strong promoter activity, was subcloned into the promoter probe plasmid pPL703 and the nucleotide sequence of this promoter region was determined. The sequence analysis suggested that this highly efficient promoter region containing the complex clustered promoters comprised three kinds of promoters (P1, P2 and P3), which are transcribed by $\sigma^B (formerly \sigma^{37}), \sigma^E(formerly \sigma^{29}) and \sigma^A (formerly \sigma^{43})$ RNA polymerase holoenzymes which play major rules at the onset of endospore formation, during sporulation and at the vegetative phase of growth, respectively. S1 nuclease mapping experiments showed that all three promoters had staggered transcription initiation points. The results of chloramphenicol acetyltransferase assay after the subcloning experiments also indicated that the expression of these clustered promoters was correlated with the programs of growth and endospore development. Promoter P1, P2 and P3 were preceded by 75% AT, 79% AT and 81% AT regions, respectively, and a partial deletion of AT-rich region prevented transcription from promoter P1 in vivo. Two sets of 5 -AGTGTT-3 sequences and inverted repeat sequences located around the promoter P1 were speculated as the possible cis acting sites for the catabolite repression in B. subtilis. In vivo transcripts from these sequence regions may be able to form a secondary structure, however, the possibility that a regulatory protein induced by the excess amount of glucose could be bound to such a domain for crucial action remains to be determined.

Effect of Organic Amendments on Efficacy of Biological Control of Seedling Damping-off of Cucumber with Several Microbial Products (유기물 첨가가 오이 모잘록병에 대한 미생물 제제의 생물학적 방제 효과 증진에 미치는 영향)

  • Several microbial biocontrol products (Greenbiotech Co., Paju, Korea), Green-all T (Trichoderma harzianum), Green-all S(Bacillus sp.) and Green-all G (Streptomyces sp.) were supplemented with organic amendments such as sawdusts and rice hulls to study on efficacy of biological control of seeding damping-off of cucumber caused by Pythium ultimum. Sawdusts amended into potato dextrose agar alone could inhibit in vitro mycelial growth of P. ultimum. All there microbial products of Green-all T, Green-all G and Green-all S significantly reduced seeding damping-off (LSD, P=0.05). However, several amendments such as sawdusts and rice hulls into Green-all T and Green-all S products did not increase efficacy of biological control compared to non-amended treatment. In contrast, supplements of aminodoctor containing several amino acids (Greenbiotech Co., Korea) into Green-all G product significantly increased efficacy of biological control of seeding damping-off, resulting in from 42% to 2% disease incidence in relation to seedling emergence (LSD, P=0.05). Also, amendment of sawdusts into Tricoderma product significantly increased efficacy of biological control as disease index of 5.0 compared to non-amended control of 56.0 in Green-all T product alone. This indicates that organic amendments could increase efficacy of biological control of cucumber seedling damping-off.

Comparison of microbial community profiling on traditional fermented soybean products (Deonjang, Gochujang) produced in Jeonbuk, Jeonnam, and Jeju province area (제주·호남권 전통된장과 고추장의 미생물 군집구조의 분석)

  • In order to evaluate the diversity of microbial population of Korean traditional Deonjang and Gochujang produced in Jeju, Jeonnam, and Jeonbuk province area, microbial communities were analyzed using next generation sequencing. In this result, the dominant bacteria of Deonjang in three area were Bacillus amyloliquefaciens, Tetragenococcus halophilus, and Bacillus was major dominant bacteria in Jeonnam (43.16%) and Jeonbuk (64.54%) area. But in Jeju area, Bacillus was 0.22%, which was significantly different from the other two. Equally, the dominant fungi of Deonjang in 3 area were Candida versatilis. Common fungus in Jeonnam and Jeonbuk area was Candida sp., respectively, 64.22% and 33.68% and Micor sp. was a common fungus in Jeonnam (15.66%) and Jeonbuk area (36.73%). But in Jeju area, Candida sp. and Zygosaccharomyces rouxii were dominant than mold. Bacillus subtilis, Bacillus licheniformis, and B. amyloliquenfaciens were the preminant bacteria in the traditional Gochujang in three regions. But there were no common dominant fungi in the 3 regions. Aspergillus sp. and Rhizopus sp. prevailed in Jeju and Jeonnam region, and Zygosaccharomycess rouxii predominanted in Jeonbuk area. These results suggested that the difference in the samples collected for the study were classified into similar groups according to the characteristics of each sample rather than regional characteristics.
