Effect of Day-length Extension Treatment Using LED on Growth and Flowering of Freesia hybrid 'Yvonne' (LED를 이용한 장일 처리가 프리지아(Freesia hybrida) '이본느'의 생육 및 개화에 미치는 영향)

  • The aim of this study was to determine the effect of day-length extension treatment using LED lighting (blue, green, red, or 3 mixed) on vegetative growth and flowering of freesia 'Yvonne', in comparison to that using glow lamps and metal halide lamps. Lighting treatments were imposed from 5 PM to 8 PM for 150 days from after flower bud differentiation to flowering end. For the period from leaf emergence to floral bud formation, no light source affected plant height but the number of leaves was decreased by the metal halide lamp. The highest SPAD value in the flower bud developing period occurred in the metal halide lamp treatment. The time of flowering was advanced by blue or green LED treatment compared to the no lighting control. The green LED lighting advanced flowering by 6 and 8 days compared to no lighting and metal halide lamp treatment, respectively. The blooming period tended to be shortened by the red LED treatment. As for the flower characteristics of 'Yvonne', floret length and width, and the weight of cut flowers were highest in the metal halide lamp treatment. Red LED decreased corm width and weight of 'Yvonne' while glow lamp decreased height and weight. Starch contents in corm were not influenced by the lighting source. Our results indicate that the green LED lighting advanced the time of flowering and the metal halide lamp was good for cut-flower quality.

In Vitro Mass Propagation and Economic Effects of Bioreactor Culture in Ever-bearing Strawberry 'Goha' (Bioreactor를 이용한 사계성 딸기 기내대량증식과 경제성)

  • This study was conducted to compare various culture methods and evaluate economic feasibility of each method for mass propagation of new ever-bearing strawberry 'Goha'. Four different methods such as semi-solid culture, solid culture, liquid suspension culture and bioreactor culture were compared. The solid culture and bioreactor culture showed the shortest and longest root length, such as 3.6 cm and 8.3 cm, respectively. Fresh weights of plants cultured in bioreactor were 2,261 mg, which were heavier than those of cultures. Dry weights of plants cultured in bioreactor were the heavier compared to those in other cultures. The number of axillary bud developed in bioreactor was seven, but axillary bud was not developed in other cultures. Production cost through bioreactor culture was calculated to be 303 won per plant which was 542 won less than that of solid culture. As a result, we found that the bioreactor culture was the most cost effective culture method for in vitro mass propagation in new ever-bearing strawberry 'Goha'.

Mass Propagation by In Vitro Culture of Bupleurum latissimum Nakai (섬시호(Bupleurum latissimum Nakai)의 조직배양을 통한 대량생산)

  • This study was carried out to establish the micropropagation system of Bupleurum latissimum Nakai that is a Korean native endangered species. Callus were induced from the leaf, petiole and floral bud and the percentage of callus formation was highest in the floral bud on the MS medium containing 2.0 mg ${\cdot}$ $L^{-1}$ 2,4-D. Especially, callus induced from floral bud was formed 77.8% and the percentage of shoot formation was 42.6% on the MS medium containing 2.0 mg ${\cdot}$ $L^{-1}$ 2,4-D plus 1.0 mg ${\cdot}$ $L^{-1}$ TDZ. For simultaneously callus formation and shoot regeneration, 1/2 MS medium was more effective than MS medium. The percentage callus formation, shoot regeneration and rooting were 46.3%, 13.0%, 13.0% in 1/2 MS medium, respectively. Soot regeneration from callus was good in 1/2 MS medium supplemented with 2.0 mg ${\cdot}$ $L^{-1}$ 2,4-D plus 1.0 mg ${\cdot}$ $L^{-1}$ BA where percentage of shoot regeneration was 74.1 %, and the number of shoot per explant was 2.4. The percentage of rooting was lowest (57.8%) in control while it was highest (97.8%) in 1.5 mg ${\cdot}$ $L^{-1}$ NAA. In acclimatization of regenerated plantlets, the percentage of survived plantlets was highest (86.1%), and plant height, root length and fresh weight were good in the soil for horticulture.

Development of Optimal Pruning Method on Okra (Abelmoschus esculentus L. Moench) Production (오크라 생산에 있어서 적정 적심방법 개발)

  • This study was conducted to develop an optimal pruning method for okra production. Three pruning methods were tested including apical bud removing, one-third removing from the top of plant, and no pruning as a control with 3 kinds interval for 15 days after 2 months sowing. The growth and development of okra was better at the treatment of one-third removing of plant than the others. The number of branches was 0.7 in control, 3.7~4.0 in apical bud removing, and 3.0~6.0 in one-third removing treatment. In summary, one-third pruning of plant from the top of plant at 30 days after starting of pruning treatment, which showed the highest yield by 12,910 kg/10a.

Some Chip Budding Techniques for Improving the Nursery Performances in Apple Trees (사과나무의 묘목생산성 향상을 위한 몇 가지 깎기눈접 기술)

  • This experiment was carried out in order to develop some chip budding techniques related to the production of high quality maiden apple trees. Optimum chip budding time for apple trees appeared to be from mid-August to early September. Untying wrapping tapes after 6 8 weeks from budding led to higher graft success and better shoot growth than removing the tapes earlier or later than the 6-8 weeks. Dipping pre-cut bud chips into ordinary water of $25^{\circ}C$ for 5 hours did not hamper the graft-success nor the early season growth of the graft compared with non-treated control. Dipping the pre-cut bud-chips into the $35^{\circ}C$ water for 3 hours or for 5 hours hampered the union formation and shoot growth of the 'Fuji' apple trees.

  • Objectives: Antibiotics help treat Vaginitis, and prolonged usage of antibiotics can lead to resistance. Methods: This study investigates the antimicrobial activity of two types of lactic acid bacteria using essential oils. After cultivation by adding grapefruit seed extract (GSE), eucalyptus, tea tree, clove bud, cinnamon, lemongrass, thyme, and ginger oils in a specific ratio, pathogenic microorganisms, namely E. coli, C. albicans, and lactic acid bacteria were released. The number of bacteria was measured using a medium suitable for the strains. Results: The essential oils and GSE inhibited pathogenic microorganisms, and the inhibitory concentration of GSE against pathogenic bacteria (E. coli, C. albicans) was confirmed. The non-inhibitory mixing ratio was also confirmed (50 μl of eucalyptus globulus (EG) oil and 50 μl of melaleuca alternifolia oil (tea tree oil, TTO) at 200 ppm GSE (pH 5.0, 5.5, 6.0)). Conclusion: Essential oils can be considered as an alternative to antibiotics because of their antibacterial properties. They are useful as auxiliary antibacterial agents for patients under long-term antibiotic treatment.

  • To establish direct multipropagation through organogenesis from nodal explants of Anoectochilus formosanus Hayata, the nodes were cultured on LS medium containing various concentrations of 6-benzyladenine(BA). High plant regeneration and adventitious bud formation were obtained from supplemented with 4.0mg/l of BA. Plant height was promoted by adding 0.3% activated charcoal. Plantlet regeneration capacity from nodes was depended on nodal parts on the stem, upper position was the best comparing with intermediate and lower.

The Effects of the Expression of GATA Binding Protein 6 on Heart and Brain Development (심장과 뇌 발달에서 GATA6 유전자 발현 감소가 미치는 영향)

  • GATA binding protein 6 (GATA6) is a transcription factor that is expressed in the early blastocyst stage and controls the expression of important genes in the differentiation and development of the heart, pancreas, and intestine. This study confirmed the role of GATA6 in cell differentiation and organ development using mouse embryonic stem cells and zebrafish, respectively. First, the mouse embryonic stem cells were differentiated into pacemaker cardiomyocytes. An RT-PCR analysis revealed that the expression of the GATA6 gene was greatly increased from day 4 of differentiation. The expression of GATA6 was upregulated prior to increased expression of NK2 homeobox 5 (Nkx2.5) and myocyte enhancer factor 2C (MEF2C), which are critical transcription factors involved in regulating heart formation. To examine the role of GATA6 in development, GATA6 morpholino was microinjected into zebrafish embryos. Knockdown of GATA6 expression significantly decreased the heart size and heart rate in the zebrafish compared to a control. In addition, the brains were degenerated in the GATA6 morpholino-injected zebrafish. Acridine orange staining showed that knockdown of GATA6 expression increased apoptotic cells in the brain. Interestingly, knockdown of GATA6 expression decreased apoptotic cells in the early bud stage. This study points to the importance of the GATA6 gene in heart and brain development.

Effect of GA3 Treatment on Bud Formation, Fruit Set, and Enlargement in Ardisia pusilla (GA3에 의한 산호수의 화아형성과 착과 및 비대 증진효과)

  • The objective of this study was carried out to investigate the proper plant growth regulator for increasing the number of flower, fruit set, and to enlarge the size of the berries in Ardisia pusilla. Flower bud formation was used rooted cutting, and fruit set, enlargement, and coloration of fruit were used with two years-old. $GA_3$ concentrations were treated with 0, 100, 200, or $400mg{\cdot}L^{-1}$. Flower bud formation was effective in $400mg{\cdot}L^{-1}$ $GA_3$ and it was 1.8 times greater than control. Plant growth regulators were applied by foliar spray at full bloom stage to increase the fruit set. As a result, $GA_3$ was the most effective for increasing fruit set. Also, auxins of 4-CPA (Tomatotone, Donbu hitech Co., Korea) and dichloprop triethanol amine (Antifall, Bayer Crop Science Co., Ltd., Korea) were effective. When $GA_3$ concentrations of 0.5 and $1.0mg{\cdot}L^{-1}$ were used, fruit set (%) reached to 70% and 77%, respectively. Effectiveness of $GA_3$ was 1.8 times greater than control. Also, auxins, dichloprop triethanol amine increased to about 7-12% during fruit setting, but cytokinin and anti-gibberellin were ineffective. To investigate the fruit enlargement and coloration, $GA_3$ was treated with 0.3, 0.6, and $1.2mg{\cdot}L^{-1}$. Fruit enlargement was achieved to about 15% by $GA_3$ $0.6mg{\cdot}L^{-1}$ when $GA_3$ was treated 3 times at the interval of 1 month per treatment when fruit size was about 2-3mm (after full-blooming two months). But anthocyanin contents for coloration of fruit skin were not significant according to $GA_3$ concentration. The results showed that $GA_3$ enhanced bud formation, fruit set and enlargement of fruit size in Ardisia pusilla.

Flower Bud Chloranthy Disease Caused by Caeoma makinoi on Prunus mume (Caeoma makinoi에 의한 매실나무 꽃눈녹병의 발생 보고)

  • Abnormal chloranthy was observed on Prunus mume trees grown in an orchard in Uiseong, Gyeongbuk province, Korea in 2015. Flower buds showing chloranthy were covered with numerous orange-colored sori, which were confirmed to contain spermogonia and aeciospores by stereo microscopy and light microscopy. When observed using scanning electron microscope, the aeciospores were globose, ellipsoid, or ovoid, and measured $22.8{\sim}35.6{\times}19.8{\sim}36.8{\mu}m$, with a verrucae wall ornamentation. To construct a phylogeny, the large and small subunit regions of rDNA were sequenced and compared with those of allied species by the maximum likelihood method. The isolated fungus species showed high similarities with Caeoma makinoi (Teleomorph: Blastospora smilacis), which was previously isolated from P. mume in Japan. Therefore, we initially report this fungal agent parasitic on P. mume as C. makinoi in Korea and name the disease it causes as "flower bud chloranthy disease".
