Protoplast Formation and Regeneration of Bacillus spp. (Bacillus spp.의 원형질체 형성 및 재생)

  • This study was to provide the basic data in improving protoplast formation and regeneration of antagonistic bacteria against phytopathogenic fungi and pest. The antagonistic rhizobacterium, BS 101, against Rhizoctonia solrmi and Fusurium oxyspomm was isolated and identified as Bacillus subtilis. Another bacterium for protoplast formation and regeneration was B. thuringiensis subsp. kurstcJtiHD-l (BT 37669) which have insectcidal toxin in the orders Coleopteria, Dipteria etc.. Auxotrophic mutants, BS 1013 and BT 69, were isolated by treating with NTG 300 ug/ml for 40 min. at $37^{\circ}C$, and with NTG 300 ug/ml for 30 min. at $37^{\circ}C$, respectively. The BS 1013 and BT 69 were converted to protoplas by treating with lysozyme 300 ugh1 for 30 min. at 37C, and lysozyme 9 mglml for 60 min. at $37^{\circ}C$, respectively. The fequencies of the protoplast formation of BS 1013 and BT 69 were 90.00 and 92.83% respectively, after 1~2 day at $37^{\circ}C$. The regeneration kequencies of the protoplasts BS 1013 and B T 69 were 0.52 and 0.10%, respectively, after 4~6 days at $37^{\circ}C$.

Modeling Survival in Patients With Brain Stroke in the Presence of Competing Risks

  • Objectives: After heart disease, brain stroke (BS) is the second most common cause of death worldwide, underscoring the importance of understanding preventable and treatable risk factors for the outcomes of BS. This study aimed to model the survival of patients with BS in the presence of competing risks. Methods: This longitudinal study was conducted on 332 patients with a definitive diagnosis of BS. Demographic characteristics and risk factors were collected by a validated checklist. Patients' mortality status was investigated by telephone follow-up to identify deaths that may be have been caused by stroke or other factors (heart disease, diabetes, high cholesterol, etc.). Data were analyzed by the Lunn-McNeil approach at alpha=0.1. Results: Older age at diagnosis (59-68 years: adjusted hazard ratio [aHR], 2.19; 90% confidence interval [CI], 1.38 to 3.48; 69-75 years: aHR, 5.04; 90% CI, 3.25 to 7.80; ≥76 years: aHR, 5.30; 90% CI, 3.40 to 8.44), having heart disease (aHR, 1.65; 90% CI, 1.23 to 2.23), oral contraceptive pill use (women only) (aHR, 0.44; 90% CI, 0.24 to 0.78) and ischemic stroke (aHR, 0.52; 90% CI, 0.36 to 0.74) were directly related to death from BS. Older age at diagnosis (59-68 years: aHR, 21.42; 90% CI, 3.52 to 130.39; 75-69 years: aHR, 16.48; 90% CI, 2.75 to 98.69; ≥76 years: aHR, 26.03; 90% CI, 4.06 to 166.93) and rural residence (aHR, 2.30; 90% CI, 1.15 to 4.60) were directly related to death from other causes. Significant risk factors were found for both causes of death. Conclusions: BS-specific and non-BS-specific mortality had different risk factors. These findings could be utilized to prescribe optimal and specific treatment.

Detection of Antibiotic Residues in Meats and Internal Organs of Cattle and Pigs (소 및 돼지의 정육과 내부장기중의 항생물질 잔류 조사)

  • The purpose of the present survey was to evaluate the antibiotic residues in meats and internal organs such as muscle, liver, heart, kidney and spleen of cattle (n=59) and pigs (n=115). The EEC-4-plate-method were employed. The results were obtained as follows ; 1. In BS 6.0, BS 7.2 and BS 8.0 used as media to detect antibiotic residues, the zone($M{\pm}SD,$ cm) of bacterial growth inhibition was narrow($1.40{\pm}0$) in meats, whereas the zone was wide($1.69{\pm}0.25-1.88{\pm}0.23$ and $1.58{\pm}0.18-1.86{\pm}0.15$ in cattle and pigs, respectively) in internal organs. But in SL 8.0, it was difficult to detect the zones ($0-1.40{\pm}0$) of both meats and internal organs. 2. Residues of antibiotic in beef and pork were rarely detected in BS 6.0, BS 7.2 and BS 8.0 (range 1.7-11.9% and 2.6-4.3%, respectively), whereas residual percentages of internal organs were relatively higher(range 69.5-96.6% and 43.5-84.3%, respectively). But in SL 8.0, it was not detected in both beef or pork, whereas they were 0-13.6% and 0-4.3% in interanal organs.

7×7 MIMO System Using Extended 13-Element ESPAR Antenna (확장 13-Element EPSAR 안테나를 사용한 7×7 MIMO 시스템)

  • Multiple-input and multiple-output (MIMO) technique is used in many communication fields in order to increase the channel capacity. However, this MIMO system has difficulty of miniaturization of antenna size due to the multiple RF chains Also, multiple RF chain raises some problems which increase power consumption at RF circuit and degrade the system performance due to the interference between RF chains. Because of these reasons, beamspace MIMO (BS-MIMO) technique with only single RF chain was proposed for MIMO transmission. This BS-MIMO system basically uses electronically steerable parasitic array radiator (ESPAR) antenna. Existing ESPAR antenna has a 5-element structure. So, it is possible to do only $3{\times}3$ MIMO transmission. Therefore, in order to extend BS-MIMO dimension, extension of ESPAR antenna structure is essential. In this paper, we show that BS-MIMO dimension can be increased according to the extension of structure of the ESPAR antenna, as in the conventional MIMO techniques. For example, we show that it is possible to design the $7{\times}7$ BS-MIMO transmissions with the 13-element ESPAR antenna. Also, when the number of parasitic elements of ESPAR antenna increases by two elements, MIMO dimension is expanded by 1.

RTP1, a Rat Homologue of Adenovirus ElA-associated Protein BS69, Interacts with DNA Topoisomerase II

  • Topoisomearse II is an essential enzyme in all organisms with several independent roles in DNA metabolism. Recently, it has been demonstrated that the C-terminal region of topoisomerases II is associated with hetero-logous protein-protein interactions in human and yeast. In this study, we identified that RTP1, a rat homologue of EIA binding protein BS69, is another topoisomerae II interacting protein by yeast two-hybrid screening. RTP1 has an E1A-binding domain and a MYND motif, which are known to be required for transcriptional regulation by binding to other proteins and interaction with the leucine zipper motif of topoisomerase II. The physical interaction between RTP1 and topoisomerase ll$\alpha$ was examined by GST pull-down assay in vitro. The expression level of RTP1 peaks in S phase as that of topoisomerase ll$\alpha$. These results suggest that the interaction between topoisomerase ll$\alpha$ and RTP1 might play an important role in regulating the transcription of genes involved in DNA metabolism in higher eukaryotes.

Biogenic Amine Formation in "Bez Sucuk," a Type of Turkish Traditional Fermented Sausage Produced with Different Meat: Fat Ratios

  • This study aims to evaluate biogenic amine levels of bez sucuks (BS) produced with different meat:fat ratios. For this, three BS groups were manufactured with meat:fat ratios of 90:10 (BS10), 80:20 (BS20), and 70:30 (BS30). The pH and water activity values and biogenic amine amounts of sucuk samples were determined during processing and storage periods and the pH values of the initial mixtures of BS samples were in the range 5.51-5.74, decreasing to 4.72-4.94 by the $14^{th}$ day. The water activity values of BS samples showed significant decreases as a result of the drying stage and reached to range 0.913-0.935 on the $14^{th}$ day of processing (p<0.05). Although BS10 had the highest tyramine (434.12 mg/kg), histamine (5.69 mg/kg), cadaverine (12.48 mg/kg), putrescine (17.83 mg/kg), 2-phenylethylamine (15.43 mg/kg), and tryptamine (122.41 mg/kg) levels at the end of processing stage (p<0.05), spermine and spermidine levels did not differ between the BS samples due to their utilization of different meat:fat ratios (p>0.05). Similarly, the tryptamine (205.11 mg/kg), putrescine (43.57 mg/kg), and tyramine (766.23 mg/kg) levels of BS10 were higher than BS20 and BS30 samples at the end of storage (p<0.05). The results showed that BS10 with the highest meat ratio had the highest tryptamine, putrescine, and tyramine levels at the end of the processing and storage period.

Nitrogen Balance and Biological Nitrogen Fixation of Soybean in Soybean-Barley Cropping System

  • This experiment was conducted to investigate the soil nitrogen credit of biological nitrogen fixation (BNF) and the nitrogen balance of soybean in soybean-barley cropping systems. Soybean cultivar, Shinpaldalkong2 and barley cultivar, Olbori, were used in soybean mono-cropping (SM), barley monocropping (BM), and barley­soybean double cropping system. The barley-soybean double cropping system was treated with two different levels of nitrogen fertilizers, 0 nitrogen fertilizer (BS-F0), and standard nitrogen fertilizer (BS-F1). Nitrogen and organic matter concentrations in soil of BS-F1 plot on October, 2001 were increased $4.8\%\;and\;5.9\%$, respectively, compared with those on October, 2000. The ranges of BNF rate in soybean were $69.1\~ 88.2\%$ in two years, and the rate was the highest in BS-F0 plot and the lowest in SM plot. The ranges of nitrogen harvest index (NHI) in all treatments were $83.9\~86.7\%$. The yield was 270 kg/10a in BS­F1 plot and 215 kg/10a in BS-F0 plot. However, the nitrogen balances were +0.6 kg/10a of gain of soil nitrogen in BS-F0 plot and -0.4 kg/10a of loss of soil nitrogen in BS-F1 plot. In comparisons of SM and BS-F1 plots, although the seed yields were similar in two plots, the loss of soil nitrogen was higher in SM than BS-F1 plot. Overall, our results suggest that barley-soybean double cropping system was more effective in respect to seed productivity and soil nitrogen conservation than soybean monocropping system, and the N credit to following crops by soybean cultivation was identified in soybean double cropping system.

Effects of Dietary Bacillus subtilis and Oregano Oil Supplementation on Performance, Egg Quality, and Intestinal Morphology in Late-Phase Laying Hens (산란말기 사료 내 Bacillus subtilis와 오레가노 오일 첨가가 산란계의 계란 생산성, 계란품질 및 장의 형태학적 특성에 미치는 영향)

  • This study aimed to investigate the impact of Bacillus subtilis-based probiotics and oregano essential oil on the production performance, egg quality, and intestinal morphology of late-phase laying hens (69-weeks). A total of 150 laying hens of 69-week-old were randomly allotted into 6 treatment groups with 5 replicates of 5 birds in each replicate. Laying hens were divided into high (H) and average (A) egg production groups prior to the trial. The hens in each group were supplemented with Bacillus subtilis, or oregano essential oil: CON, a basal diet; BS, basal diet plus 3 × 108 CFU/kg feed Bacillus subtilis; OEO, basal diet plus 0.3 g/kg feed oregano essential oil. Egg performance, blood characteristics, egg quality, and intestinal morphology of the late-phase laying hens were evaluated. Both BS and OEO significantly enhanced (P<0.05) egg production compared to CON in high egg production. The blood characteristics indicated no significant differences based on the egg production and the supplementation of BS and OEO in the late-phase laying hens. The eggshell strength was significantly improved (P<0.05) in both OEO compared to both CON. A significantly decreased (P<0.05) the villus height to crypt depth ratio (VH/CD) in the ileum compared to H, and in the treatment groups supplemented with BS and OEO, VH/CD showed a significant increase (P<0.05) compared to both CON. These results suggest that the supplementation of Bacillus subtilis and oregano essential oil in the diet of late-phase laying hens could serve as a potential strategy to enhance egg production, egg quality, and gut health.

Properties of Doenjang (Soybean Paste) Prepared with Different Types of Salts

  • Samples of doenjang (a fermented soybean paste) were prepared with different types of salts; purified salt (PS), 3-year-aged solar salt (SS3), 1-year-aged solar salt (SS1), and bamboo salt (BS, 3rd processing product). For starter doenjang samples, selected starters comprising two bacilli, one yeast, and one fungus were inoculated, whereas for non-starter doenjang samples, microorganisms present in rice straw were inoculated after enrichment. The doenjang samples were fermented for 13 weeks at 25℃. During the fermentation period, SS and BS doenjang samples showed higher bacilli counts as well as much lower yeast counts than PS doenjang. At 13 weeks, yeast counts of starter doenjang samples were 7.75, 5.69, 6.08, and 4.74 log CFU/g for PS, SS3, SS1, and BS doenjang, respectively. For non-starter doenjang samples, counts were 7.17, 5.05, 5.92, and 4.54 log CFU/g for PS, SS3, SS1, and BS doenjang, respectively. SS and BS promoted growth of bacilli but inhibited growth of yeasts compared with PS. Debaryomyces hansenii was the dominant yeast in PS doenjang, whereas Candida guilliermondii and Pichia sorbitophila were dominant in SS and BS doenjang. In the sensory evaluation, SS and BS doenjang scored better than PS doenjang. In conclusion, SS and BS seem better than PS for production of high-quality doenjang.

the Antibacterial Activities of Persimmon Juice and Persimmon Juice Dyed Cotton Fabrics (감즙과 감즙염색 면직물의 항균성)

  • The antibacterial activities against Staphylococcus aureus of persimmon juice and persimmon juice dyed cotton fabrics were eveluated. We investigated the effects in pursuit of the proper treating conditions to use persimmon juice dye for comfortable and hygienic doffing. The antibacterial activities were estimated by test method KS K 0693 and the percentage reduction of bacteria at various contact conditions of test materials and bacteria. The results were as follows. no bacteriostatic activities (BS) of persimmon Juice were more than $99.98\%$ and the increase was dose-dependent according to concentration from $1\%$ to $10\%$. However, bactericidal activities (BC) were not found BS increased from $99.97\%$ to $100\%$ and BC from $79.74\%$ to $100\%$ with increasing persimmon juice concentration of cotton fabrics from $10\%$ to $100\%$. Antibacterial effects were significantly shown at $10\%$ of persimmon juices. BS increased from $75.69\%$ to $100\%$ and BC from $65.89\%$ to $100\%$ over 18 hours of contact time and continued for 50 hours. BS of $100\%$ persimmon juice dyed cotton fabrics increased from $96.56\%$ to $98.20\%$ and BC from $-136\%$ to $65\%$ at different incubation temperatures of $17\pm1^{\circ}C$, $27\pm1^{\circ}C$ and $37\pm1^{\circ}C$. BS ranged from $99.06\%$ to $99.99\%$ and BC from $84.31\%$ to $95.10\%$, according to the initial bacterial density from $4.08\times10^7$ to $4.08\times10^5$. Sixty-seven percent of subjects of answered not to smell any odor from persimmon juice dyed cotton fabrics by subjective test.
