New embryogenesis from atypical bodies and plant regeneration from long-term subcultured embryogenic callus in rose (장기간 계대배양 된 장미 배발생 캘러스로부터 식물체 재분화 및 비정형체로부터 새로운 배발생캘러스 재생)

  • Long-term subcultured rose embryogenic calluses, which had been maintained for more than 5 to 6 years since the first embryogenesis from calluses induced from in vitro roots of rose, were identified as potential material for the development of transgenic plants. The first embryogenic calluses from 'Sweet Yellow' and two breeding lines (KR056002 and KR056006) were obtained in 2007 and 2009, respectively. Subsequently, we found that plants regenerated from long-term embryogenic calluses (LEC). Whereas the LEC from 'Sweet Yellow' takes 3 to 4 months to regenerate plants, those of the two breeding lines take 4 to 5 months. This period of time is the same as that taken for plants to regenerate from the first embryogenic callus. New embryogenesis was observed from atypical bodies (ABs) that appeared during the process of long-term subculture. We found that it is possible to use the AB as a material for new embryogenesis.

Amphibian Biotope Planning to Research on Reproduction of Bombina orientalis (무당개구리(Bombina orientalis) 번식 연구를 위한 양서류 바이오톱(amphibian biotope) 조성)

  • Bombina orientalis, or Korean fire-bellied toad is belonging to Class Amphinia, Order Anura, Family Bombinatoridae, Genus Bombina. Ecological characteristics, such as small size, slow moving and swimming, gentle attitude to other species, under laboratory and/or wild condition lead the toad as suitable experimental animals with easy handling. Seasonal reproductive biology except breeding season (usually from Apr. to late Jun.) are unknown still caused by sampling methods for hibernating specimens have not been established yet. Most species of amphibian, include B. orientalis, are declined around the world related with global warming, therefore, wild caught specimen for any purposes must be limited controled soon. Even though, some pre-studies of biotope for increasing biodiversity have been reported, construction of an artificial biotope to get natural breeding cycling specimen for study of amphibian reproduction is unique modelling and only trial. In an effort to set year-round sampling for study of reproduction of B. orientalis, we suggest protocols for planning, construction manual and SOP(Standard Operation Process) to manage amphibian biotope. During pilot operation of a designed and constructed biotope, the seasonal acclimation during winter hibernation of B. orientalis were successful. It will be helpful for studying the reproductive biology of wild amphibians in temperate region and allow to get reasonable research conditions in amphibian laboratory.

Breeding Hybrid Rice with Good Quality and High Yield I. Breeding of Cytoplasmic-Genetic Male Sterile and Restorer Lines with Backgrounds of Korean Rice Varieties (양질 다수성 일대잡종 벼 육성 연구 I. 우리나라 품종 배경의 세포질-유전자적 웅성불임 및 임성회복 계통 육성)

  • This study was conducted to breed various cytoplasmic genetic male sterile (CGMS) and restorer lines with backgrounds of Korean japonica rice varieties. The CGMS line BT-CMS was crossed with the restorer line AR-3. The fertile $F_1 was emasculated and crossed with the Korean japonica rice varieties of the early maturing Sobaegbyeo, Odaebyeo, Gwanagbyeo and Daeseongbyeo, and of the medium maturing Hwajinbyeo, Paldal, Suwon 224 and Iri 386, and of the late maturing Nagdongbyeo, Palkweng, Hwacheongbyeo and Milyang 97. Each of the three way cross $F_{1S} was segregated into fertile and sterile individuals. The sterile individuals in each cross were discarded and the fertile individuals were emasculated and backcrossed with the Korean japonica rice varieties. The same process was applied from BC$_1$F$_1$ to BC$_4$F$_1$ generation. In the ${BC_4}{F_1}of each cross, the male sterile individual was crossed with the recurrent Korean japonica variety which was maintainer of male sterility. The male sterile lines of ${BC_6}{F_1}were named as Sobaegbyeo R, Odaebyeo R, Gwanagbyeo R, etc. The fertile individuals homozygous in pollen fertility were selected from the ${BC_4}{F_2}generation and named as Sobaegbyeo R, Odaebyeo R, Gwanagbyeo R, etc. Agronomic characteristics of the CGMS, restorer lines with backgrounds of Korean japonica, and the recurrent Korean japonica rice varieties grown in the field condition were compared. Culm length of the CGMS lines tended to be shorter than that of the recurrent parent, however no significant differences in heading date, panicle length and yield component were found among the CGMS, restorer and recurrent lines.

Speciation of the Two Species of the Genus Hyla (Anura) in Korea (한국산 청개구리속 2조의 종분화에 관한 연구)

  • A comparison between two species of Hyla japonica and H. suweonesis in Korea was carried out to clarify their morphological, genetic and ecological differences, to estimate presumed divergent time between them, and to elucidate their probable process and mechanism of speciation. The results are as follows; 1.The breeding season of H. japonica was earlier than that of H. suweonensis about one and half months. 2.H.japonica was distributed all over the country, while H. suweonensis was restricted in Kyonggi plains. 3.On the extemal morphometric characters, differences were hardiy seen between two species. But on the discriminant function analysis, there was apparent differences between them. 4.Genic variation based on 24 loci showed that H. japonica populations were about two fold more variable than that of H. suweonensis populations. 5.Genetic relatedness between two species was rather remote (S =0.520, D=0.634) 6.Their presumed divergent time was estimated to be about three and a quarter mfllon years. It is presumed that H. suweonensis was originated from H. japonica during the period of late pleocene and early pleistocene. 7.Premating isolating mechanisrns (breeding season, mating call, and microhabitat segregation) may be operating to prevent gene ifow between them.

Transplanted and many other machines were developed in the early 1990s. In ginseng breeding only pure line selection was of practical significance several verities were at the stage of seed propagation at ginseng plantations. Mutation breeding (${\gamma}$-ray. X-ray chemicals) was not successful. The research on plantlet formation through tissue culture was a little progressed but still far behind to vegetative propagation. Disease control research was concentrated in the isolation and identification of pathogans. their ecological charactelistics and biological control and soil humigation. Potato root rot nematodes was found and control method was established. Insect and small animal control research was greatly progresses in identification, ecological investigation, and ecological and physical control. Weed control was less important due to the development of mulching method of ridge and ditch. Quality factors of raw ginseng in relation to red ginseng process were extensively studied. Traditional quality measures were elucidated in accordance with modern analytical chemistry resulting in the importance of peptides in the centrat part rather than ginsenosides For large root production growth promoting rootzone micrcorganisms (PGPRM) were isolated and active compounds were identified. Field test on PGPRM was on going. Varictus methods formality improvement through cultivation were developed. Management research of ginseng production was rare Extension was active throuch official and private organizations and through workshop for the extension specialists, and direct lectures to grower's. Extension services made the researcher to understand the existing problems at grower's fields. Research environment for ginseng production was in prime time only for three years when Korea Ginseng Research Institute was established then gradually aggravated.

Breeding Process and Characteristics of Gopoong, a New Variety of Panax ginseng C.A. Meyer. (인삼(Panax ginseng C.A. Meyer) 신품종 고풍의 육성경과 및 생육특성)

  • To develop a new ginseng variety with good quality and high yielding, a lot of individual ginseng plant were selected in the farmers'fields in 1968. Among them, a promising line,680-83-4, has been developed through comparative cultivation of several lines selected with pure line separation from local races in KT&C Central Research Institute, preliminary and advanced yield trials were performed fir 8 years.680-83-4 was designated as KG103, which was then registered as a new variety “Gopoong” with the regional yield and adaptation trials for 10 years (1981-1990) on December 27,2000 in Korea. Gopoong has different phenotype of a dark violet stem and dark red fruit and inverted triangle shape of berries cluster as compared with other varieties. Taproot of Gopoong was longer than local race Jakyungjong, and root yield was 4.5% higher than local race Jakyungjong. In red ginseng quality, the rates of Chun-Jeesam grade(Chun and Jee means 1 st and 2nd grade, respectively) were 16.6% and 9.4% for Gopoong and Jakyungjong, respectively. In these results, it was that Gopoong was superior ginseng line with good quality far manufacture of red ginseng.

Study on the Genetic Characteristics of Waterlogging Tolerant Pepper (Capsicum annuum L.) for Breeding Tolerant Varieties against Flooding Stress (내습성 고추 품종 육성을 위한 선발계통의 유전적 특성 구명)

  • This study was conducted to select pepper lines that were tolerant to excessive water injury among the pepper germplasm and investigate the genetic characteristics of those lines to contribute to the breeding of pepper cultivars with stable productivity in abnormal weather. Each of the tolerant and susceptible lines went through immersion treatment, and differentially expressed genes between them were analyzed. The tolerant line showed increased expression of the CA02g26670 gene, which is involved in the CONSTANS protein pathway and regulation of flowering by day length, but it exhibited decreased expressions of CA01g21450, CA01g22480, CA01g34470, CA02g00370 and CA02g00380. The susceptible line showed increased gene expressions of CA02g09720, CA02g21290, CA03g16520, CA07g 02110, and CA12g17910, which are involved in the inhibition of proteolytic enzyme activity, DNA binding, inhibition of cell wall-degrading enzyme, and inhibition of nodulin, respectively. Meanwhile the expressions of CA02g02820, CA03g21390, CA06g17700 and CA07g18230 decreased in the susceptible line, in relation to calcium-ion binding, high temperature, synthesis of phosphocholine and cold stress, respectively. The expressions of genes related to apoptosis and peroxidase increased, while that of CA02g16990, which functions as a nucleoside transporter, decreased in both the tolerant and susceptible lines. Based on the different gene expressions between the tolerant and susceptible lines, further studies are needed on breeding abiotic stress-tolerant lines.

A New Sweetpotato Cultivar for Use of Bioethanol 'Daeyumi' (바이오에탄올용 고구마 신품종 '대유미')

  • A new sweetpotato variety, 'Daeyumi', was developed by Bioenergy Crop Research Center, National Institute of Crop Science (NICS), RDA in 2008. This variety was obtained from the cross between 'Jinhongmi' and 'Xusju 18' in 2000. The seedling and line selections were performed from 2001 to 2003, preliminary and advanced yield trials were carried out from 2004 to 2005, and the regional yield trials were conducted at six locations from 2006 to 2008. 'Daeyumi' has cordate leaf, green vine and petiole, elliptic storage root, red skin and yellow flesh color of storage root. This variety is also resistant to Fusarium wilt and nematode. The starch value was 25.9%, ethanol yield was 418 L/Ton, which was 7% higher than that of 'Yulmi' variety, and the total sugar content was 2.47 g/100g, dry weight. 'Daeyumi's initial temperature of starch gelatinization was lower, 76.2$^{\circ}C$, and the retrogradation process was earlier than 'Yulmi'. The average yield of storage root was 27.8 ton/ha in the regional yield trials, which was 36% higher than that of 'Yulmi' variety. Number of storage roots over 50 gram per plant was 3.0, and the average weight of one storage root was 152 gram. This variety can be used for the production of bioethanol and starch processing.

  • Recently, as the fishery resources are depleted, expectations for productivity improvement by 'rearing fishery' in land farms are greatly rising. In the case of land farms, unlike ocean environments, it is easy to control and manage environmental and breeding factors, and has the advantage of being able to adjust production according to the production plan. On the other hand, unlike in the natural environment, there is a disadvantage in that operation costs may significantly increase due to the artificial management for fish growth. Therefore, profit maximization can be pursued by efficiently operating the farm in accordance with the planned target shipment. In order to operate such an efficient farm and nurture fish, an accurate growth prediction model according to the target fish species is absolutely required. Most of the growth prediction models are mainly numerical results based on statistical analysis using farm data. In this paper, we present a growth prediction model from a stochastic point of view to overcome the difficulties in securing data and the difficulty in providing quantitative expected values for inaccuracies that existing growth prediction models from a statistical point of view may have. For a stochastic approach, modeling is performed by introducing a Gaussian process regression method based on water temperature, which is the most important factor in positive growth. From the corresponding results, it is expected that it will be able to provide reference values for more efficient farm operation by simultaneously providing the average value of the predicted growth value at a specific point in time and the confidence interval for that value.

  • As the consumption of fishery products continues to increase, aquaculture industry has emerged instead of fishing industry facing limitations of fish stock resources. Recently, smart fish farming industry has rapidly developed through convergence with 4th Industrial Revolution technology. Accordingly, it is important to derive a future model of smart fish farming platforms in order to secure the superiority of the aquaculture industry and the technology standard hegemony. In this study, the future direction of smart fish farm platform was derived through the analysis of environment related to politics, economy, society, and technology related to smart fish farming by applying PEST methodology of macro-environment analysis. It is expected that it will help the public and industrial circles in planning and implementing related projects by including the entire process of value chain of aquaculture industry of breeding, production, management and distribution, and by presenting advanced models based on artificial intelligence and digital twin.
