Studies on Effect of Temperature and Relative Humidity on Aspergillosis in Silkworm, Bombyx mori L.

  • The growth and multiplication of Aspergillus flavus Link and A. tamarii Kita were observed in vitro under variable temperatures of $22-31^{\circ}C$. The matte weight of mycelium and number of conidia/ml were significantly higher (P < 0.01) at the higher temperature than the lower temperature in both the species of Aspergillus. In vivo the mortality in silkworm, Bombyx mori L. with the infection of Aspergillus species was significantly (P < 0.01) different at different temperature and relative humidity conditions.

  • As the Selenium is known to be growth promoter in sheep and cattle, the efficacy of selenium has been tested in silkworm Bombyx mori L. The V instar larvae were fed with lethal and sub-lethal doses of selenium treated leaves. The larvae fed with lethal dose showed a significant decrease in growth and Cocoon commercial characters. The treatment with sub lethal dose exhibited a significant increase in the growth of the silkworm. Correspondingly, the cocoon commercial characters on exposure to the lethal dose showed significant decrease and sub lethal dose showed a significant increase. In the light of similar findings reported earlier in other cocoon crops and vertebrates, it can be inferred that selenium at lower doses acts as a growth stimulator, resulting in the higher yield of cocoon crop.

Hemocyte Changes after the Extirpation of the Hemopoietic Organ-wing Disc Complexes in the Silkworm, Bombyx mori (Lepidoptera: Bombycidae)

  • We successfully extirpated all four hemopoietic organ-wing disc complexes of the fifth instar larvae of Bombyx mori, and found that most of the treated silkworms could still develop into the moths. We investigated the changes of the circulating hemocytes and evaluated the effects of extirpation on the hemopoiesis. The results showed that proliferation of circulating hemocytes was sufficient to allow development of the silkworms which complexes were totally extirpated. We also found that hemopoietic organ-wing disc complexes extirpation might cause a certain hemopoietic compensation of the remainder complexes during early spinning. Exogenous hormones such as $20-{\beta}-hydroxyecdysone$ and juvenile hormone analog had a positive effect on hemocytes proliferation.

Relation among Food Consumption, Conversion and Cocoon Production in Silkworm, Bombyx Mori L. Reared at Different Temperatures

  • Rearing temperature influences food consumption, conversion and cocoon production in silkworm Bombyx mori L. Young age $({1^st}to{3^rd} instar)$ worms of Pure Mysore, a multivoltine were reared at 26, 28, 30 and $32^{\circ}C$ followed by combinations of these temperature levels during late age $({4^th} and {5^th} instar)$. Food consumption, conversion, larval duration and growth for each instar were recorded. The influence of combinations of these temperature levels during rearing on survival and cocoon production was assessed. Food consumption increased with the temperature up to $4^th$ instar and decreased during $5^th$ instar. Temperature beyond $28^{\circ}C$ during young age followed by higher temperature ($30^{\circ}C$ and above) has shown deleterious effect on cocoon production. The paper discusses the rearing performance in relation to food intake and conversion under the tested temperature levels.

Studies on the Evaluation of Some Three-Way Cross Hybrids of the Silkworm, Bombyx mori L.

  • Eight new three-way cross heterozygotic hybrid genotypes of the mulberry silkworm Bombyx mori, resulted from hybridization of multivoltine as female and bivoltine Fl hybrid as male component, have been evaluated for the cumulative effect of nine traits of commercial economic importance with the objective of selecting out the most prospective ones for their exploitation as an intermediate technology during unfavorable season. Five of them namely, G(P5${\times}$NBl8) (E.I. =57.23); S(NB18${\times}$P5) (E.I. : 55.48); S(P5${\times}$NBl8) (E.I. = 52.44); S(KBN${\times}$B7) (E.I. = 50.42) and S(NB7${\times}$KB) (E.I. : 50.16) have scored higher values of the Evaluation Index (E.I.) and could be selected in the same order for exploitation during unfavorable seasons in tropical climates.

  • A comprehensive study on daily pupal weight loss due to histolysis and histgenesis in Bombyx mori Linn., has been undertaken. The percentag of pupal weight loss in the male pupa is higher than the female, conforming that the female pupa require more energy in the form of less pupal weight loss for ovipositional activities. The regression equation clearly shows higher percentage loss of daily pupal weight in male than female with respect to age and, this is also evident from the slope of the regression line. Moreover, analysis of the coefficient of correlation shows that the weight loss of pupa is directly correlated with the age rather than climatic parameters under which the pupa dwells.

Functional analysis of Bombyx mori Decapentaplegic gene for bone differentiation in a mammalian cell

  • Bone morphogenetic proteins (BMPs) belong to the transforming growth factor (TGF-${\beta}$) superfamily and are involved in osteoblastic differentiation. The largest TGF-${\beta}$ superfamily subgroup shares genetic homology with human BMPs (hBMPs) and silkworm decapentaplegic (dpp). In addition, hBMPs are functionally interchangeable with Drosophila dpp. Bombyx mori dpp may induce bone formation in mammalian cells. To test this hypothesis, we synthesized the 1,285-base pairs cDNA of full-length B. mori dpp using total RNAs obtained from the fat body of 3-day-old of the $5^{th}$ instar larvae and cloned the cDNA into the pCEP4 mammalian expression vector. Next, B. mori dpp was expressed in C3H10T1/2 cells. The target cells transfected with the pCEP4-Bm dpp plasmid showed biological functions similar to those of osteogenic differentiation induction growth factors such as hBMPs. We determined the relative mRNA expression rates of Runt-related transcription factor 2 (RUNX2), osterix, osteocalcin, and alkaline phosphatase (ALP) to validate the osteoblast-specific differentiation effects of B. mori dpp by performing quantitative real-time RT-PCR. Interestingly, mRNA expression levels of the 3 marker genes except RUNX2, in cells expressing B. mori dpp were much higher than those in control cells and C3H10T1/2 cells transfected with pCEP4. These results suggested that B. mori dpp signaling regulates osterix expression during osteogenic differentiation via RUNX2-independent mechanisms.

누에에의 차별화 선별을 통한 면역 관련 유도 유전자의 분리와 특성 (Isolation and Characterization of Inducible Genes from Bombyx mori Injected with E. coli by Differential Screening)

  • 누에에서 생체 방어에 관련된 새로운 항 세균성 펩타이드 유전자를 탐색 분리하기 위하여 누에 체강에 비 병원성 세균인 Escherichia coli를 주사하여 면역 반응의 일환으로 발현량이 증가하는 유도 유전자 종류를 조사 하였다. 체강 주사 8시간 후 누에에서 cDNA 유전자 은행을 만들고, 정상 및 유도 누에에서 분리한 각각의 mRNA를 탐침으로 차별화 선별을 하였다. 차별화 선별 결과 정상보다도 유도 누에의 탐침을 사용한 막에서 강도가 높은 클론 32개를 선발하였고, 29개 클론에 대해 전체 또는 부분 염기 서열을 분석하여 DNA 상동성을 조사하였다. DNA 상동성 비교를 통해 생산한 발현 유전자 꼬리표 중에는 비교적 상동 유의성이 인정되어 그 실체를 추정할 수 있는 19개의 클론이 있었다. 특히 곤충의 면역 작용에 직접적으로 관계하는 항세균성 펩타이드 유전자, hemolin 유전자, transferrin 유전자 등 4종의 유전 자원을 확보할 수 있었다.

Cloning, Sequencing and Characterization of Mitochondrial Control Region of the Domestic Silkwom, Bombyx mori

  • The nucleotide sequence of the domestic silkworm (Bombyx mori) mitochondrial (mt) control region and its flanking genes was determined from PCR clones. The control region of the silkworm mt genome was located between the small ribosomal RNA gene and transfer RN $A^{Met}$. This 499 bp control region hale 95.4% A+T content. Extensive comparative analysis studies performed with similar control region of other insect genomes could not reveal a highly conserved region containing conserved motifs of animal mito-chondrial genome. The remarkable feature that found in this control region was the presence of tandem motifs containing nine repetitive sequences. The potential usefulness of this motif sequences for Bombyx species or their taxonomically related species is enhanced by its unique localization in the maternally inheritance mitochondrial molecule.e.

  • 숙주범위가 서로 다른 Autographa californica NPV(AcNPV)와 Bombyx mori NPV(BmNPV)를 Spodoptera frugiperda(Sf9) 또는 Bombyx mori(BmN-4)의 세포에 동시감염(coinfection)시킨 후, 숙주범위가 확장된 재조합 바이러스를 Sf9세포에서 10종 BmN-4세포에서 2종씩 플라크 순화하여 선발하였다. 각 재조합 바이러스 DNA의 제한효소 분석결과는 한번 이상의 재조합이 일어났음을 보여주었다. 재조합 바이러스 RecB-8의 전자현미경 관찰결과는 다각체의 모양이 모바이러스인 AcNPV나 BmNPV와는 전혀 다른 정사면체 모양이었으며 또한, 모바이러스와는 달리 virion이 다각체에 거의 매립되어 있지 않은 특징을 보였다.
