A Study on Substrate Pre-treatment for Mathane Production Performance (메탄생성 효율증진을 위한 소화원료 전처리 연구)

  • During the process of anaerobic digestion, hydrolysis of substrate is the key factor determining methane production efficiency. The hydrolysis efficiency are directly affected by biodegradability of substrate. In this study, three types of pre-treatment methods were tested to enhance biodegradability effectiveness. By the application of blender and the Cavitation treatment, the SCODcr increased from 24,723 mg/l to 24.726~29,175 mg/l and to 25,589~26,456 mg/l, respectively. In case of ozone treatment, the SCODcr decreased from 24,723 mg/l to 22.540~23,988 mg/l. In batch experiment, there was a little improvement of the biochemical methane potential (BMP) by the blender and the cavitation treatment. In contrast, the BMP somewhat decreased by ozone treatment. This result shows that higher anaerobic digestion efficiency of livestock manure could be obtained through pre-treatment of substrate.

Gene expression pattern during osteogenic differentiation of human periodontal ligament cells in vitro

  • Purpose: Periodontal ligament (PDL) cell differentiation into osteoblasts is important in bone formation. Bone formation is a complex biological process and involves several tightly regulated gene expression patterns of bone-related proteins. The expression patterns of bone related proteins are regulated in a temporal manner both in vivo and in vitro. The aim of this study was to observe the gene expression profile in PDL cell proliferation, differentiation, and mineralization in vitro. Methods: PDL cells were grown until confluence, which were then designated as day 0, and nodule formation was induced by the addition of 50 ${\mu}g$/mL ascorbic acid, 10 mM ${\beta}$-glycerophosphate, and 100 nM dexamethasone to the medium. The dishes were stained with Alizarin Red S on days 1, 7, 14, and 21. Real-time polymerase chain reaction was performed for the detection of various genes on days 0, 1, 7, 14, and 21. Results: On day 0 with a confluent monolayer, in the active proliferative stage, c-myc gene expression was observed at its maximal level. On day 7 with a multilayer, alkaline phosphatase, bone morphogenetic protein (BMP)-2, and BMP-4 gene expression had increased and this was followed by maximal expression of osteocalcin on day 14 with the initiation of nodule mineralization. In relationship to apoptosis, c-fos gene expression peaked on day 21 and was characterized by the post-mineralization stage. Here, various genes were regulated in a temporal manner during PDL fibroblast proliferation, extracellular matrix maturation, and mineralization. The gene expression pattern was similar. Conclusions: We can speculate that the gene expression pattern occurs during PDL cell proliferation, differentiation, and mineralization. On the basis of these results, it might be possible to understand the various factors that influence PDL cell proliferation, extracellular matrix maturation, and mineralization with regard to gene expression patterns.

Role of MAP kinase on MMP-13 expression in rat periodontal ligament cells (백서 치주인대세포에서의 MMP-13 mRNA에 대한 MAP kinase의 역할)

  • Matrix metalloproteinases (MMPs)는 치주조직내에 존재하는 세포외기질의 유지와 분해에 중요한 역할을 담당하고 있으며 이중 MMP-13은 치주질환의 진행과 깊은 관계가 있다고 알려져 있다. 이번 연구는 치주질환의 진행에 있어서 MMP-13의 활성에 대한 mitogen activated protein(MAP) Kinase의 역할을 구명하기 위해 시행되었다. 백서 치주인대세포에서의 MMP-13 mRNA의 발현은 RT-PCR에 의하여, 그리고 MAP Kinase의 발현은 Western blot에 의하여 측정하였다. $Interleukin-1{\beta}$(IL $-1{\beta}$), Tumor necrosis $factora(TNF-{\alpha})$와 parathyroid hormon(PTH)는 MMP- 13 mRNA 발현을 각각 320%, 180%, 380% 증가시켰으나 bone morphogenetic protein-7(BMP-7)은 MMP-13 mRNA의 발현을 증가시키지 않았다. p38 MAP Kinase 억제제인 SB203580은 IL $-1{\beta}$ 유도 MMP-13의 발현을 약 40% 정도 억제시켰으나, PTH-유도 MMP-13 mRNA의 발현은 억제하지 못했다. IL $-1{\beta}$는 MMP- 13 mRNA의 반감기를 약 2시간 정도로 증가시켰으나, p38 MAP Kinase 억제제로 전처치한 경우에는 반감기가 60분으로 줄어들었다. $IL-1{\beta}$는 p38 MAP kinase와 JNK의 인산화 활성을 증가시켰으나 PTH, $TNF-{\alpha}$와 BMP-7은 p38, JNK, ERK의 활성을 증가시키지 못했다. 이상의 연구결과는 p38 MAP Kinase가 백서 치주인대세포에서의 MMP-13 mRNA 발현을 조절하는데 중요한 역할을 담당함을 시사하였다.

  • Extracellular matrix component is degraded by enzymes of thematrix metalloproteinases(MMPs). MMPs are produced by both hemopoietic and structural cells. Increased activity of MMP-3 in periodontium is strongly associated with inflammatory periodontal disease. The purpose of the present study was to estimate the effect of BMP-7 on regeneration of periodontium. The optical density was measured by microwell plate reader at 450 nm.The difference of the optical density and the relative activity ofMMP-3 according to the concentration were statistically analyzed by one way ANOVA. The results were as follows: 1. Tetracycline-HCl showed the tendency to inhibit the activity of MMP-3 at the concentration lower than $25{\mu}g/ml$. 2. Doxycycline-HCl inhibited significantly the activity of MMP-3 at the concentration lower than $100{\mu}g/ml$. 3 . Minocycline-HCl inhibited the activity of MMP-3 at the concentration in the range of 10 to 200${\mu}g/ml$. Within the limit of the present study, the above results suggested that bone morphogenetic protein-7 may play a important role in development of periodontium.

  • As Graphic Information is a intuitive, simple and definite media to transfer information to human, It is widely used and deserved as a information exchange and store media. But the graphic information is so abstract that a human may misunderstand that's real meaning. So In order to transfer a Exact information to a human, a additional picture and description is needed. So, in this paper, A authoring system is suggested that it can be able to give a additional information at each region in graphic information, and to structurize relations between each region and to give linking information between each region using XML Linking Notation. In this paper, An suggested authoring system is implemented to support a authoring facility which is to display graphical information of CGM(Computer Graphic Metafile) and BMP, and to define structural information by selecting each region of graphics image on screen directly, and to save that structural information and linking information with XML Linking notation, and to support a browsing facility which to find hot spot using a structurized graphic information and then to show a proper graph.

  • 기후변화로 인하여 전 세계적으로 이상기후가 진행되고 있다. 우리나라의 경우 홍수와 가뭄 등의 이상 강수가 빈번히 발생하고 있으며, 이로 인한 비점오염물질의 유출과 집적이 발생하고 있다. SWAT (Soil and Water Assessment Tool)과 HSPF (Hydrological Simulation Program)등의 다양한 모델이 비점오염의 유출을 모의하기 위하여 활용되고 있으나, 농장 단위에서 유역 단위까지 다양한 대상지역에 대한 장기 모의와 BMP적용에 따른 효율평가가 어려운 실정이다. Texas A&M Agrilife Research에서 개발한 APEX(Agricultural Policy Environmental eXtender model) 모형의 경우, 농장 단위에서 유역 단위의 대상지역에 대하여 최대 100년까지 일단위 모의가 가능한 특징이 있다. 하지만 APEX 모형은 개발된지 상당한 시간이 지났으며, 데스크탑 기반의 모형이므로 다양한 플랫폼에서의 활용이 어려운 한계점이 있다. 따라서 본 연구에서는 1. 기존 데스크탑 기반의 APEX 모형을 웹에서 구동할 수 있도록 웹 기반 시스템을 구축하였으며, 2. 양분유출에 대한 모의 평가가 가능하도록 결과를 시스템화하는 모듈을 추가하였다. 또한, 3. 웹 기반 APEX 모형의 활용도를 위하여 최적관리기법을 적용할 수 있는 모듈을 개발 및 추가하였다. 본 연구의 결과는 향후 농업비점오염원 지역의 양분유출을 확인·예측하고 관리하기 위한 기초자료로 활용될 것으로 기대하며, 최적관리기법을 활용한 저감 정책을 수립하기 위한 평가도구로 활용될 수 있을 것이다. 또한, 기존 데스크탑 기반의 모형을 웹 기반으로 이전하였으므로, APEX 모형 혹은 다른 데스크탑 기반 모형들의 웹 이전을 통한 사용자 편의성 증대를 위한 기초자료로 활용될 수 있을 것이다. 아울러 본 연구의 결과는 APEX 모형의 작물 매개변수가 국내 환경에 최적화 되어있지 않으므로 정확한 BMP 저감효과 모의에는 한계가 있으므로, 이에 대한 추가적인 연구가 필요할 것으로 판단된다.

Assessing the impacts on runoff and irrigation water by controlling the growing period of rice for future climate scenarios using APEX model (APEX 모형을 이용한 미래 기상시나리오별 벼 생육기간 조절을 통한 유출 및 관개량 영향 평가)

  • 최근 기상 이변으로 인한 재해 피해에 따라 기후변화에 대한 경각심이 높아지고 있으며, 국내 농업분야에서는 식량안보와 물이용에 대한 기후변화 영향을 완화할 실질적인 대책이 필요하다. 기후변화 적응 전략으로 생육기간 조절(TDS, Transplanting date shift)이 평가되었으나 유출, 관개량, 쌀 생산량 등에 미치는 영향에 대한 연구는 아직 미흡한 실정이다. 본 연구는 기후 변화에 대한 실질적인 적응 전략으로서 TDS뿐만 아니라 대표적인 논 BMP(Best management practice)인물꼬높이 조절(DOR, Drainage outlet rasing)도 평가하고자 하였다. 기후변화 적응 전략 평가를 위한 시나리오는 10개의 GCM과 APEX-Paddy 모형을 사용하여 TDS와 DOR을 조합하여 총 12개 시나리오를 구성하고 기후변화가 유출, 관개량 및 용수생산성에 미치는 영향을 분석하였다. 유출은 각 미래기간 (2020s, 2030s, 2040s) 동안 다르게 나타났으나 현재기간보다 증가하는 추세를 나타냈다. 쌀 생산량은 CO2 상승으로 인해 온도가 상승하면서 지속적으로 감소하였으나, TDS로 인해 상당한 양의 관개량이 절약되는 것을 나타냈다. 또한, DOR에 따라 미래기간의 유출량, 관개 요구량은 크게 감소된 반면 쌀 생산량은 소폭 증가하는 것을 나타냈다. DOR과 40일이 지연된 TDS를 포함하는 RM5 시나리오는 유출 감소 및 용수생산성 증가에 대한 영향이 가장 높은 시나리오로서 본 연구에서 가장 효과적인 BMP(Best Management Practice)로 평가되었다. 본 연구의 결과는 기후 조건을 고려하여 TDS와 DOR의 적절한 조합이 기후 변화의 부정적인 영향을 완화할 수 있을 것으로 시사한다.

Discovery and characterization of berberine derivatives as stimulators of osteoblast differentiation

  • Berberine has been used clinically for more than a decade to treat various diseases, has been shown to be effective in osteoblast differentiation, and is a potential treatment option for osteoporosis. However, compared with existing osteoporosis drugs, berberine is somewhat less effective. This study aimed to identify a new compound with efficacy superior to that of berberine. The osteogenic activities of various berberine derivatives were evaluated via cell differentiation in C2C12 preosteoblast cell lines. Alkaline phosphatase (ALP) staining assay and structure-activity relationship demonstrated that compound 2b had a potent osteogenic effect. Furthermore, compound 2b dose dependently increased ALP activity and showed no toxicity at the effective concentration, indicating its efficacy. Additionally, compound 2b upregulated BMP2-induced transcriptional activity in a promoter activity assay using ALP, BSP, and OC promoters.

Characterization Analysis for Cardiogenic Potential of Three Human Adult Stem Cells (세 종류 줄기세포의 특성 분석과 지방유래 줄기세포의 심근세포로의 분화)

  • In the present study, we isolated three human adult stem cells including adipose tissue-derived stem cells(HAD), umbilical cord-derived stem cells(HUC), and amnion-derived stem cells(HAM) and analysed their characteristics. And we examined whether HAD could be used as therapeutical cells for the heart diseases. Both HAM and HUC appeared very similar morphology but HAD was different. Doubling time of HUC was most fast, but total doubling numbers of HUC was same with HAM. Total doubling numbers of HAD was much more than others. Expression patterns of genes and proteins of three human adult stem cells were very similar. Also they were differentiated into adipocytes, osteocytes, and chondrocytes. In addition, they expressed many cardiomyocyte-related genes. But expression pattern of genes is a little different. When HAD were cultivated in the presence or absence of various combinations of BMP and FGF after 5-azacytidine expose for 24 h, expression of Cmlc-1, and ${\alpha}1c$ genes was significantly increased. However, expression of troponin T, troponin I and Kv4.3 genes was not changed. Based on these observations, it is suggested that HAD, HUC, and HAM might be used as potentially therapeutical cells for clinical application.

The Process Efficiency Evaluation of the Food Supernatant Using A/G (Acid/Gas) Phased Anaerobic Digestion (산/가스 분리 혐기소화공정을 이용한 음식물 탈리액의 처리효율 평가)

  • Several acidogenesis batch tests, and BMP (Biochemical Methane Potential) with food waste leachate was tested at various organic loading rates (OLRs) on the mesophilic ($35^{\circ}C$) and thermophilic ($55^{\circ}C$) conditions. In acidogenesis batch test, VS removal efficiencies were 27.3% and 30.6% at $35^{\circ}C$ and $55^{\circ}C$, respectively. Removal efficiency of VS at $55^{\circ}C$ was higher than that at $35^{\circ}C$. With decrease in VS, SCOD increased as reaction time increased. Solubilization efficiency of VS were 27.4% and 33.4% at each reaction temperature within 4 days acid fermentation. Methane yield were 461 and 413 $mLCH_4/gVS$ at mesophilic and thermophilic BMP test, respectively. SCOD solubilizations in the themophilic acid fermenter showed 8~17% higher than those in the mesophilic fermenter. COD removal efficiency showed higher in the mesophilic acid fermenter at low organic loading rate. While at high organic loading rate, it was higher in the thermophilic acid fermenter. VS removal efficiency was higher at the mesophilic temperature, however, it decreased at OLR higher than 6 kg $COD/m^3{\cdot}day$. On the contrary, VS removal efficiency did not decrease but maintain at thermophilic temperature. The amount of methane gas generated from mesophilic methanogenesis digester was 12.6, 21.6, 27.4 L/day at OLR of 4, 5, 6 $COD/m^3{\cdot}day$, respectively. The amount of methane gas generated from themophilic methanogenesis digester was 14.3, 20.6, 25.2 L/day at each OLR, respectively, which is about 15~20 L/day lower than those generated at mesophilic digester.
