Possible Application of Artificial Insemination Buffer for Increasing Production Efficiency of Female Cow Offspring

  • The present research was carried out to evaluate the possibility of increasing female offspring production ratios using artificial insemination buffer (AIB) before artificial insemination (AI). In this experiment, we optimized AIB composition, made an AIB gun and analyze factors affecting AI non-return rate after AIB treatment. The AIB was made with the base of Tris-buffer supplemented with L-arginine and several other chemicals that might reduce the motility of male sperm compared to the female counterpart, therefore, increasing the possibility of fertilization by female sperm. AIB must be deposited into $2^{nd}$ to $4^{th}$ cervix by AIB gun. After 15 min of AIB deposition, frozen semen was deposited into the same place. A total of 348 cattle were inseminated with AIB insemination, and there were no significant differences between AIB and traditional AI non-return rates (56.8% vs. 55.7%). The AI non-return rate in AIB group, however, differed significantly among 7 Hanwoo farms. The parturition numbers ($1^{st}$ to $7^{th}$) of cows did not affect AIB AI rate. The proportion of AIB AI success rates was significantly higher in Hanwoo cows than in dairy cows (61.0% vs. 48.7%), but the average AI success rate did not differ significantly between AIB and conventional AI (56.8% vs. 55.7%). The female offspring production rate in $2^{nd}$ to $4^{th}$ cervix deposition place was significantly higher than that in the uterus body (77.7% vs. 59.6%, p<0.05). The injection volume of AIB in 5 and 10 ml was significantly higher than that in 2 ml (77.7%, 78.7% vs. 51.8%, p<0.05), but there were no differences in AIB injection volume between 5 and 10 ml. The best exposure time of AIB in the cervix was 10 to 15 min rather than 5 min (79.2%, 77.2% vs. 52.6%, p<0.05). AIB therefore needs to have an exposure time of at least over 10 min for a higher production rate of female offspring. In conclusion, AIB could be used in AI industry to increase the female offspring ratio and AIB AI can increase the AI success rate.

Human Acyl-CoA: Cholesterol Acyltransferase (hACAT) Inhibitory Activities of Triterpenoids from Roots of Glycine max (L.) Merr

  • Eight triterpenoids, six lanostanes 1-6, one lupenane 7, and one oleanane 8, were isolated by bioactivity-guided fractionation of the ethylacetate extract from roots of Glycine max (L.) Merr. All isolated compounds were examined for their inhibitory activities against human ACAT-1 (hACAT-1) and human ACAT-2 (hACAT-2). Among them, three triterpenoids showed potent hACAT inhibitory activities, (24R)-ethylcholest-5-ene-3,7-diol (1) and 3b -hydroxylup-20(29)-en-28-oic acid (7) exhibited more potent inhibitory activity against hACAT-1 (1: IC50 = 25.0 1.2 and 7: IC50 = 11.5 0.4 m M) than hACAT-2 (1: IC50 = 102.0 5.4 and 7: IC50 = 33.9 3.7 m M), respectively. Interestingly, 5a ,8a -epidioxy-24(R)-methylcholesta-6,22-diene-3b -ol (4) has proven to be a specific inhibitor against hACAT-1 (IC50 = 38.7 0.8 m M) compared to hACAT-2 (IC50 >200). In conclusion, this is the first study to demonstrate that triterpenoids of G. max have potent inhibitory activities against hACAT-1 and hACAT-2.

Biphenyl의 Sphingobium yanoikuyae BK-10에 의한 분해 특성 (Biodegradation of Biphenyl by Sphingbium yanoikuyae BK-10)

  • PCBs(polychlorinated biphenyl)는 난분해성 물질로써, 환경호르몬으로 분류된 유독한 화합물이다. 이런 유독한 화합물인 PCBs 화합물이 오염된 토양 및 수계를 회복하기 위해 PCBs의 모체인 biphenyl을 효과적으로 분해하는 미생물을 토양으로부터 분리 선별하여 S. yanoikuyae BK10 (AF406817)와 같이 분해능이 우수한 균주를 분리하였다. 분리된 S. yanoikuyae BK10의 특성을 조사하기 위하여 자연계의 토양 조건인 pH 5.0$\sim$8.0에서 99%이상의 높은 biphenyl 분해효율을 보였다. 또한, 온도를 달리하여 실험 한 결과, 10$\sim$50$^{\circ}C$의 범위에서 모두 70%이상의 높은 분해효율을 보여줌으로써 실제 biphenyl/PCBs로_오염된 토양에서 온도의 영향을 덜 받고 biphenyl을 효과적으로 분해 할 수 있을 것으로 생각된다. S. yanoikuyae BK10는 biphenyl이 500 $\mu$g/ml으로 처리된 mineral salt 배지에서 48시간동안 99% 이상의 biphenyl을 분해하는 높은 분해활성을 보이며, biphenyl을 mineralization 시키는 것으로 판단된다. 또한 biphenyl 분해효소 유도 실험결과는 기질을 biphenyl로 사용하여 증식한 균체가 다른 기질을 사용해서 증식한 균체보다 약 2배가량 biphenyl을 빨리 분해시켰다. 그렇지만, cell-mass를 많이 얻을 수 있는 당을 탄소원으로 사용하여 배양하였을 때에도 단시간 내에 biphenyl분해 효소를 분비하여 biphenyl을 분해하는 것으로 보아, S. yanoikuyae BK10는 실제 biphenyl/PCBs에 오염된 토양 적용 할 경우 안정적으로 균주의 제공이 가능하다고 판단된다. 이상의 결과를 토대로, 토양에서부터 분리한 S. yanoikuyae BK10는 자연계에서 유해화합물인 biphenyl/PCBs을 효과적으로 분해 할 수 있다고 생각되며, 분리균주인 S. yanoikuyae BK10의 분자 생물학적 특성을 조사하여 biphenyl과 PCBs를 분해하는 유전자 탐색에 유용한 정보를 얻을 수 있을 것으로 사료된다.

딸기재배를 위한 생물비료로 Chlorella fusca CHK0059의 시비효과 (Application Effect of Chlorella Fusca CHK0059 as a Biofertilizer for Strawberry Cultivation)

  • BACKGROUND: Green algae (Chlorella spp.) has been widely used as a biofertilizer to improve the crop yield and quality. However, available information is not enough to verify the mechanism of green algae's beneficial impact on strawberry. This study was conducted to investigate the effect of Chlorella fusca CHK005 application on the growth and nutrient status of strawberry plant and fruit characteristics. METHODS AND RESULTS: A total of 800 seedlings of strawberry variety 'Kuemsil' were planted. Once a week, C. fusca culture solution (1.0 × 107 cells mL-1) was applied into soil via irrigation in four different concentrations: no application (control), 1/1000 times (× 0.5), 1/500 times (× 1), and 1/250 times (× 2). Result showed that growth of strawberry plant was enhanced by Chlorella application and the highest impact on fresh weight (FW) and chlorophyll content of the plants were observed in × 2 treatment, followed by × 1, × 0.5, and control treatments. The phosphorus (P) concentration in the plant was significantly higher in × 1 and × 2 treatments compared to control. In case of fruit quality, sugar content (°Brix), hardness, and FW were lowest in control, but these values increased as application levels of Chlorella were higher. Also, P and K contents in the fruits increased with increasing the application levels and significant correlation between P content and oBrix in the fruits was found. CONCLUSION(S): Overall, Chlorella application seemed to improve plant growth and fruit quality by increasing the utilization efficiency of P and K in strawberries.

Synthesis of Novel H8-Binaphthol-based Chiral Receptors and Their Applications in Enantioselective Recognition of 1,2-Amino alcohols and Chirality Conversion of L-Amino acids to D-Amino acids

  • Novel $H_8$-binaphthol-based chiral receptors appended with an uryl moiety (2a) and a guanidinium moiety (2b) have been designed and synthesized for the enantioselective recognition of 1,2-amino alcohols via reversible imine formation. The selectivities ($K_R/K_S$ = 9.8 ~ 19.4) of 2b in imine formation with 1,2-amino alcohols are higher than those of 2a ($K_R/K_S$ = 1.8 ~ 4.5). Similar efficiency trend have been observed in the conversion of L-amino acids to D-amino acids, i.e., the efficiency of the receptor 2b (D/L ratio: 4.3 ~ 10.1) is superior to 2a (D/L ratio: 4.0 ~ 8.7).

Characterization of an Aminopeptidase A from Tetragenococcus halophilus CY54 Isolated from Myeolchi-Jeotgal

  • In this study, a pepA gene encoding glutamyl (aspartyl)-specific aminopeptidase (PepA; E.C. 3.4.11.7) was cloned from Tetragenococcus halophilus CY54. The translated PepA from T. halophilus CY54 showed very low similarities with PepAs from Lactobacillus and Lactococcus genera. The pepA from T. halophilus CY54 was overexpressed in E. coli BL21(DE3) using pET26b(+). The recombinant PepA was purified by using an Ni- NTA column. The size of the recombinant PepA was 39.13 kDa as determined by SDS-PAGE, while its optimum pH and temperature were pH 5.0 and 60℃, respectively. In addition, the PepA was completely inactivated by 1 mM EDTA, indicating its metallopeptidase nature. The Km and Vmax of the PepA were 0.98 ± 0.006 mM and 0.1 ± 0.002 mM/min, respectively, when Glu-pNA was used as the substrate. This is the first report on PepA from Tetragenococcus species.

Mitochondrial Genetic Diversity and Phylogenetic Relationships of Siberian Flying Squirrel(Pteromys volans) Populations

  • Siberian flying squirrel, an endangered species in South Korea, is distributed through major mountain regions of South Korea. The number of Siberian flying squirrel(Pteromys volans) in South Korea has decreased and their habitats are fragmented and isolated because of anthropogenic activities. So far no molecular genetic data has, however, been available for their conservation and management. To obtain better information concerning genetic diversity and phylogenetic relationships of the Siberian flying squirrel in South Korea, we examined 14 individuals from South Korea, 7 individuals from Russia, and 5 individuals from northeastern China along with previously published 29 haplotypes for 1,140 bp of the mtDNA cytochrome b gene. The 14 new individuals from South Korea had 7 haplotypes which were not observed in the regions of Russia and Hokkaido. The level of genetic diversity(0.616%) in the South Korean population was lower than that in eastern Russia(0.950%). The geographical distribution of mtDNA haplotypes and reduced median network confirmed that there are three major lineages of Siberian flying squirrel, occupying; Far Eastern, northern Eurasia, and the island of Hokkaido. The South Korean population only slightly distinct from the Eurasia, and eastern Russian population, and is part of the lineage Far Eastern. Based on these, we suggest that the South Korean population could be considered to belong to one partial ESU(Far Eastern) of three partial ESUs but a different management unit. However, the conservation priorities should be reconfirmed by nuclear genetic marker and ecological data.

Pharmacophore Identification for Peroxisome Proliferator-Activated Receptor Gamma Agonists

  • Peroxisome proliferator-activated receptors (PPARs) are members of nuclear receptors and their activation induces regulation of fatty acid storage and glucose metabolism. Therefore, the $PPAR\gamma$ is a major target for the treatment of type 2 diabetes mellitus. In order to generate pharmacophore model, 1080 known agonists database was constructed and a training set was selected. The Hypo7, selected from 10 hypotheses, contains four features: three hydrogen-bond acceptors (HBA) and one general hydrophobic (HY). This pharmacophore model was validated by using 862 test set compounds with a correlation coefficient of 0.903 between actual and estimated activity. Secondly, CatScramble method was used to verify the model. Hence, the validated Hypo7 was utilized for searching new lead compounds over 238,819 and 54,620 chemical structures in NCI and Maybridge database, respectively. Then the leads were selected by screening based on the pharmacophore model, predictive activity, and Lipinski's rules. Candidates were obtained and subsequently the binding affinities to $PPAR\gamma$ were investigated by the molecular docking simulations. Finally the best two compounds were presented and would be useful to treat type 2 diabetes.
