Effect of Fine Copper Sulfides on the High Cycle Fatigue Properties of Bake Hardening Steels for Automotive (자동차용 소부경화형(BH) 강의 고주기 피로 특성에 미치는 미세 황화물의 영향)

  • Bake hardening steels have to resist strain aging to prevent the yield strength increment and stretcher strain during press process and to enhance the bake hardenability during baking process after painting. The bake hardening steels need to control the solute carbon and the solute nitrogen to improve the bake hardenability. Ti and/or Nb alloying for nitride and carbide precipitation and low carbon content below 0.003% are used to solve strain aging and formability problem for automotive materials. However, in the present study, the effect of micro-precipitation of copper sulfide on the bake hardenability and fatigue properties of extremely low carbon steel has been investigated. The bake hardenability of Cu-alloyed bake hardening (Cu-BH) steel was slightly higher (5 MPa) than that of Nb-alloyed bake hardening (Nb-BH) steel, but the fatigue limit of Cu-BH steel was far higher (45 MPa) than that of Nb-BH steel. All samples showed the ductile fracture behavior and some samples revealed distinct fatigue stages, such as crack initiation, stable crack growth and unstable crack growth.

Evaluation of Image Quality for 2D TSE(RT) and 3D GRASE in MRCP Study: Fast MRCP Method (췌담관자기공명영상에서 2D TSE(RT)와 3D GRASE(BH) 기법에 대한 영상의 질 평가 : Fast MRCP 기법)

  • In this study, we intend to evaluate image quality and provide to clinical basic data by applying to 2D TSE (RT) and 3D GRASE (BH) techniques using Fast MRCP testing methods for application to patients in poor patient condition. Data were analyzed for 30 patients (15 males, 15 females, and 64±4.26 average age) who underwent MRCP tests. The equipment used was Ingenia CX 3.0 T equipment and Ds anterior coil was used for data acquisition. SNR and CNR of each image were measured through quantitative analysis, and the quality of the image was evaluated by dividing it into 5 grades for qualitative evaluation. The image evaluation was performed on the paired t-test and the Wilcoxon test, and when the p value was 0.05 or less, it was considered to be significant. As a result of quantitative analysis of SNR and CNR, 3D GRASE (BH) was measured high when comparing the two techniques, 2D TSE (RT) MRCP and 3D GRASE (BH) (p<0.05). The qualitative analysis result is a sharpness of the bile duct: 3D GRASE(BH): 4.12±0.03, Overall image quality: 3D GRASE(BH): 4.21±0.91 was high (p=0.001). The motion artifact of the bile duct showed no significant difference with two techniques(2D TSE(RT): 4.41±0.04, 3D GRASE(BH): 4.53±0.14(p=0.067). However, the background suppression obtained significant results with 2D TSE(RT) of 4.14±0.55(p=0.001). In conclusion, as a result of using the Fast MRCP testing method, MRCP images obtained by 3D GRASE (BH) had an advantage over MRCP images using 2D TSE (RT). However, there will be useful results of 2D TSE(RT) MRCP technique in patients who have difficulty holding their breath.

Detoxification of Glutaraldehyde Treated Porcine Pericardium Using L-arginine & $NABH_4$

  • Background: Calcification is the most frequent cause of clinical failure of bioprosthetic tissues fabricated from GA-fixed porcine valves or bovine pericardium. A multi-factorial approach using different mechanisms was recently developed to reduce the calcification of bioprosthetic tissues. The purpose of the present study was to evaluate the synchronized synergism of using L-arginine and $NaBH_4$, compared with ethanol and L-lysine, in glutaraldehyde treated porcine pericardium from the standpoint of calcification and tissue elasticity. Materials and Methods: Porcine pericardium was fixed at 0.625% GA (7 days at room temperature after 2 days at $4^{\circ}C$). An interim step of ethanol (80%; 1 day at room temperature) or L-lysine (0.1 M; 2 days at $37^{\circ}C$) or L-arginine (0.1 M; 2 days at $37^{\circ}C$) was followed by completion of the GA fixation. A final step of NaBH4 (0.1 M; 2 days at room temperature) was followed. Their tensile strength, thickness, and thermal stability were measured. Treated pericardia were implanted subcutaneously into three-week-old Sprague-Dawley rats for 8 weeks. Calcium content was assessed by atomic absorption spectroscopy and histology. Results: L-arginine and $NaBH_4$ pretreatment ($1.81{\pm}0.39$ kgf/5 mm p=0.001, $0.30{\pm}0.08$ mm p<0.001) significantly increased tensile strength and thickness compared with the control ($0.53{\pm}0.34$ kgf/5 mm, $0.10{\pm}0.02$ mm). In a thermal stability test, L-arginine and $NaBH_4$ pretreatment ($84.25{\pm}1.12^{\circ}C$, p=0.023) caused a significant difference from the control ($86.25{\pm}0.00^{\circ}C$). L-lysine and $NaBH_4$ pretreatment ($183.8{\pm}42.6$ ug/mg, p=0.804), and L-arginine and $NaBH_4$ pretreatment ($163.3{\pm}27.5$ ug/mg, p=0.621) did not significantly inhibit calcification compared to the control ($175.5{\pm}45.3$ ug/mg), but ethanol and $NaBH_4$ pretreatment did ($38.5{\pm}37.3$ ug/mg, p=0.003). Conclusion: The combined pretreatment using L-arginine and $NaBH_4$ after GA fixation seemed to increase the tensile strength and thickness of porcine pericardium, fixed with GA. Additionally, it seemed to keep thermal stability. However it could not decrease the calcification of porcine pericardium fixed with GA. $NaBH_4$ pretreatment seemed to decrease the calcification of porcine pericardium fixed with GA, but only with ethanol.

Hydrolysis Reaction of NaBH4 Using Activated Cabon Supported Co-B/C, Co-P-B/C Catalyst (활성탄 담지 Co-B/C, Co-P-B/C 촉매를 이용한 NaBH4 가수분해 반응)

  • Sodium borohydride, $NaBH_4$, shows a number of advantages as hydrogen source for portable proton exchange membrane fuel cells (PEMFCs). Properties of $NaBH_4$ hydrolysis reaction using activated carbon supported Co-B/C, Co-P-B/C catalyst were studied. BET surface area of catalyst, yield of hydrogen, effect of $NaBH_4$ concentration and durability of catalyst were measured. The BET surface area of carbon supported catalyst was over $500m^2/g$ and this value was 2~3 times higher than that of unsupported catalyst. Hydrogen generation of activated carbon supported catalyst was more stable than that of unsupported catalyst. The activation energy of Co-P-B/C catalyst was 59.4 kJ/mol in 20 wt% $NaBH_4$ and 14% lower than that of Co-P-B/FeCrAlloy catalyst. Catalyst loss on activated carbon supported catalyst was reduced to about 1/3~1/2 compared with unsupported catalyst, therefore durability was improved by supporting catalyst on activated carbon.

  • Sodium borohydride($NaBH_4$) known as the material of hydrogen generation and storage can produce the hydrogen via catalytic hydrolysis. This protide chemical could be used in the hydrogen supply system for residential and mobile fuel cells, and thus many researches and developments regarding to these chemicals and decomposition reactions have been implemented. We experimented the hydrolysis of $NaBH_4$ alkaline solution by metal oxide-supported PGM(platinum group metal) catalysts and measured the generation rate of hydrogen which is product of decomposition reaction. We compared oxides as catalyst supports, and the precious metals, Pt and Ru for the catalysts and studied the effects of amounts of catalyst added and $NaBH_4$ concentrations on the hydrogen generation rates and patterns.

Study on the Protective Effects of 6R-Tetrahydrobiopterin on the Oxidative Neuronal Injury in Mouse Cortical Cultures (배양된 대뇌피질세포에서 산화성 손상에 대한 6R-Tetrahydrobiopterin의 억제작용)

  • Objective : 6R-Tetrahydrobiopterin(BH4) is a cofactor for the aromatic amino acid hydroxylases which is essential for the biosynthesis of catecholamines and serotonin. It also acts as a cofactor for nitric oxide synthase, and stimulates the release of some neurotransmitters such as dopamine, serotonin, acetylcholine and glutamate. Recently, it has been reported that BH4 could induce cellular proliferation and enhance neuronal survival. This study was performed to investigate the antioxidative effect of BH4 on the various oxidative insults in mouse cerebral cortical cell cultures. Methods : Iron ion(FeCl2), zinc ion(ZnCl2), sodium nitroprusside(SNP) and buthionine sulfoximine(BSO, a glutathione depletor) were used as oxidants. Cell death was assessed by measurement of lactate dehydrogenase efflux to bathing media at the end of exposure. Result : All 4 oxidants induced neuronal cell death associated with cell body swelling, which was markedly inhibited by trolox($100{\mu}M$), a vitamin E analog. BH4($10-100{\mu}M$) markedly inhibited the neuronal cell death induced by all 4 oxidants($20{\mu}M\;Cu^{2+}$, $20{\mu}M\;Zn^{2+}$, $1{\mu}M$ SNP or 1mM BSO). However, BH4 failed to inhibit the neuronal cell death induced by 24hr exposure to $20{\mu}M$ NMDA. Conculsion : These results suggest that BH4 has antioxidative action independently of any actions of enzyme cofactor.

Flavonoids as Substrates of Bacillus halodurans O-Methyltransferase

  • Bacillus halodurans O-methyltransferase (BhOMT) is an S-adenosylmethionine dependent methyltransferase. In our previous study, three dimensional structure of the BhOMT has been determined by comparative homology modeling and automated docking study showed that two hydroxyl groups at 3'- and 4'-position in Bring and structural rigidity of C-ring resulting from the double bond characters between C2 and C3 of flavonoid, were key factors for interaction with BhOMT. In the present study, BhOMT was cloned and expressed. Binding assay was performed on purified BhOMT using fluorescence experiments and binding affinity of luteolin, quercetin, fisetin, and myricetin were measured in the range of $10^7$. Fluorescence quenching experiments indicated that divalent cation plays a critical role on the metal-mediated electrostatic interactions between flavonoid and substrate binding site of BhOMT. Fluorescence study confirmed successfully the data obtained from the docking study and these results imply that hydroxyl group at 7-position of luteolin, quercetin, fisetin, and myricetin forms a stable hydrogen bonding with K211 and carboxyl oxygen of C-ring forms a stable hydrogen bonding with R170. Hydroxyl group at 3'-and 4'-position in the B-ring also has strong $Ca^{2+}$ mediated electrostatic interactions with BhOMT.

Effects of $NaBH_4$ and laponite on the stability of colloidal Ag nanoparticles (나노 은 콜로이드 입자의 안정성에 대한 $NaBH_4$ 및 Laponite의 영향)

  • The synthesis and characterization of silver colloidal nanoparticles by chemical reduction of silver ions in aqueous $AgNO_3$ using sodium borohydride $(NaBH_4)$ as the reducing agent are described. The experimental conditions for aggregation and paricle size of nanosilver particles in water is investigated in terms of concentration of $NaBH_4$, reaction temperature, dropping rate of $AgNO_3$ and concentration of laponite. Stable nanosilver sol is obtained at three molar ratio of $NaBH_4/AgNO_3$ in conditions of without laponite. The size of nanosilver particles is increased as the reaction temperature is increased. The large size of nanosilver sol is obseved as the dropping rate of $AgNO_3$ is increased due to the aggregation of initial high local concentration of nanosilver particles. Stable nanosilver sol at high temperature $(>\;100^{\circ}C)$ can be prepared when laponite is used as protective colloid.

Clinical Findings and Gene Analysis of BH4 Responsive PKU Patients in Korea (BH4 responsive PKU 환자들의 임상적 특성과 유전자분석)

  • Purpose: Phenylketonuria (PKU) is the first inherited metabolic disease of which treatment is known. We performed this study to find out clinical symptoms and prognosis of tetrahydrobiopterin (BH4) responsive PKU patients and genetic relation. Methods: Clinical, biochemical, genetic analysis were done retrospectively in 23 patients diagnosed BH4 responsive PKU in Soonchunhyang University Hospital from March 2000 to September 2012. Results: Patients were classified to mild hyperphenylalaninemia and mild phenylketonuria with initial plasma phenylalanine level below 20 mg/dL. After BH4 loading, blood phenylalanine decrease level ranged between 37% and 99%. Initial treatment with low phenylalanine formula or BH4 was started before 2 month after birth except 2 patients. And one of them resulted in developmental delay in language and social activity. The others showed satisfactory progress without developmental delay. In genetic analysis, of 46 allele, R241C allele mutation was identified most commonly (41%). R241C/A259T, R241C/R243Q, R241C/V388M, R241C/T278I was detected in 5 (21.7%), 3 (13%), 2 (8%), 2 (8%) patients, respectively. Conclusion: R241C mutation was detected most frequently in this study group and R243Q mutation which is known to be prevalent in Korean PKU patients was found in 4 patients (8.6%). Early diagnosis and treatment is important in PKU patients.

Comparative optimization of Be/Zr(BH4)4 and Be/Be(BH4)2 as 252Cf source shielding assemblies: Effect on landmine detection by neutron backscattering technique

  • Monte Carlo simulations were used to model a portable Neutron backscattering (NBT) sensor suitable for detecting plastic anti-personnel mines (APMs) buried in dry and moist soils. The model consists of a 100 MBq 252Cf source encapsulated in a neutron reflector/shield assembly and centered between two 3He detectors. Multi-parameter optimization was performed to investigate the efficiency of Be/Zr(BH4)4 and Be/Be(BH4)2 assemblies in terms of increasing the signal-to-background (S/B) ratio and reducing the total dose equivalent rate. The MCNP results showed that 2 cm Be/3 cm Zr(BH4)4 and 2 cm Be/3 cm Be(BH4)2 are the optimal configurations. However, due to portability requirements and abundance of Be, the 252Cf-2 cm Be/3 cm Be(BH4)2 NBT model was selected to scan the center of APM buried 3 cm deep in dry and moist soils. The selected NBT model has positively identified the APM with a S/B ratio of 886 for dry soils of 1 wt% hydrogen content and with S/B ratios of 615, 398, 86, and 12 for the moist soils containing 4, 6, 10, and 14 wt% hydrogen, respectively. The total dose equivalent rate reached 0.0031 mSv/h, suggesting a work load of 8 h/day for 806 days within the permissible annual dose limit of 20 mSv.
