6배체 트리티케일과 4배체호밀 교잡에서의 교잡율, 염색체수 변이 및 작물학적 특성 (Crossability, Variation in Chromosome Number, and Agronomic Characters in Hybrid of Hexaploid Triticale with Tetraploid Rye)

  • 6배체 트리티케일 신기호밀과 4배체 호밀 품종 두루호밀을 교잡하여 이들의 임실율, 잡종 후대들의 염색체수의 변이, 작물학적 특성등을 검토하므로써 트리티케일 품종육성의 기초자료로 제공코져 실시한 본 시험의 결과를 요약하면 다음과 같다. 1. 신기호밀(P$_1$)과 두루호밀(P$_2$)의 교잡에서 교잡율은 30.5%, 역교배에서는 3.26%였다. F$_1$/P$_1$(=BC$_1$)에서는 8.75%, F$_1$/P$_2$(=BC$_2$)에서 7.20%, 그리고 F$_1$/F$_1$(=F$_2$)은 1.53%의 교잡율을 보였다. 2. 교잡된 교배종자의 발아율은 F$_1$ 종자 37%, F$_2$ 43.0%, BC$_1$, 39.0%, BC$_2$, 50.0%로 나타났으며, 천립중은 각각 20.7g, 24.5g, 25.4g, 23.6g 이었다. 3. F$_1$(=P$_1$/P$_2$) 식물체의 화분임성은 30.2%였고, 69.8%는 비정상 화분이었다. 4. F$_1$식물체의 화분 모세포 감수분열에서 1가 염색체 13.5%개, 2가 염색체 8.89%, 3가 염색체 1.24개로 2개 염색체는 50.8%였다. 5. 체세포 염색체수는 F$_1$ 35개 였고, F$_2$는 32-33개, 35-36개를 갖는 개체가, BC$_1$은 35-39개를 갖는 개체가 많았다. 그리고 BC$_2$는 28-36개의 분포를 보였다. 이는 F$_1$ 식물체에서 14-18개의 염색체를 갖는 배우자와의 결합이 많음을 보여준다. 6. 임성 개체의 비율은 F$_1$, 100%, F$_2$, 4.5%, BC$_1$, 42.9%, BC$_1$, 50.0%였으며 임성개체의 평균수당 임실립수는 F$_1$, 1.17개, F$_2$, 13.3개, BC$_1$, 2.36개, BC$_2$, 3.75개 였다. 7. 출수기는 F$_1$이 양친의 중간을 보였고, BC$_1$과 BC$_2$는 양친의 영향을 보였으며, F$_2$는 만숙친 보다도 늦었다. 간장은 F$_1$이 장간친과 비슷 하였으나 F$_2$, BC$_1$, BC$_2$는 단간친보다 작았고 특히 BC$_1$은 단간친보다 35cm나 짧았다. 수장과 수당 소수수는 F$_1$이 양친보다 길거나 많았다. 소수당 영화수는 양친의 영향을 크게 받았다.

Anti-apoptotic effect by Bcl-2 in UVB-irradiated keratinocytes.

  • Bcl-2 is a member of large bcl-2 family and protects cells from apoptosis. Using bcl-2-expressing adenovirus vector (Ad-bc1-2), we investigated the effect of bc1-2 on UVB-induced apoptosis. Adenovirus vector efficiently introduced bc1-2 gene in cultured normal mouse keratinocytes (NMK cells); almost all NMK cells (lx10$^{6}$ ) were transfected at Ixl0$^{8}$ PFU/ml. Bcl-2-transfected NMK cells were significantly resistant to UVB-induced apoptosis with the suppressive effect dependent on bcl-2-expression level. Following UVB irradiation caspase 8, 3, 9 activities were stimulated in NMK cells, while in bc1-2-transfected cells, only caspase 8, but not caspase 3 or 9 activities were stimulated. In order to investigate the effect of bc1-2 in vivo, topical application of Ad-bc1-2 on tape-stripped mouse skin was performed. Following the application, Bc1-2 was efficiently overexpressed in almost all viable keratinocytes. The expression was transient with the maximal expression of Bc1-2 at 1st day following the application of lxl0$^{9}$ PFU in 200ml. The introduced Bc1-2 remained at least for 6 days. UVB irradiation (1250 J/m$^2$) induced apoptosis within 12 h and the maximal effect was observed at 24 h in control mouse skin. Bc1-2-transfected mice skin were resistant to UVB-induced apoptosis. Topical application of empty adenovirus vector alone had no effect on Bc1-2 expression or UVB-induced apoptosis. These results indicate that adenovirus vector is an efficient gene delivery system into keratinocytes and that Bcl-2 is a potent inhibitor of UVB-induced apoptosis both in vitro and in vivo.

Yield Improvement by Two Cycles of Mass Selection in Two Sweet Corn Populations

  • Mass selection (MS) is an efficient selection method to directly improve highly heritable traits. In the present study, two cycles of MS for ear length were conducted on two sweet corn populations, BC2-l0 and $BC1-10{\times}Syn-II$ after introgression of exotic germplasm. The improved populations generated from these selections were evaluated in comparison with the base populations at two locations, to determine the genetic gains and performance of the improved populations. The two base populations showed varied average realized responses to MS. In BC2-l0 derived population, the realized responses were $9.1\%$ in BC2-l0 C1 and $1.2\%$ in BC2-l0 C2, whereas in$BC1-10{\times}Syn-II$ derived population, the realized responses were$ 5.6\%$ in $BC1-10{\times}Syn-II$ C1 and $2.9\%$ in $BC1-10{\times}Syn-II$ C2. All the improved populations showed longer ears than their respective base populations and the check varieties. Ear length, which was used as the selection criterion in this study, showed high broad-sense heritability in the BC2-l0 and $BC1-10{\times}Syn-II$ derived populations, while fresh ear yield revealed low heritability, indicating that selection for ear length in these populations would be more effective than direct selection for yield. Results of this study indicate that MS conducted on BC2­10 and $BC1-10{\times}Syn-II$ had significantly increased ear length and fresh ear yield in both populations. The improved populations obtained would serve as better germplasm sources and further selection in these populations could offer better responses.

양파와 파간의 종간잡종 F1과 여교잡계통의 핵 DNA 함량 (Variation of Nuclear DNA Content in Interspecific Allium cepa L.×A. fistulosum L. hybrids and Their Successive Backcross Lines)

  • Allium 속 근연종인 파와 양파간 종간교잡을 이용하여 양파로부터 새로운 형질을 도입하는 과정에서 유래되는 종간교잡 F1과 여교잡 세대들의 게놈크기의 변화를 flow cytometry를 이용하여 측정한 결과 다음과 같은 결과를 얻었다. 1. 양파와 파의 종간교잡 F1의 식물학적 특성은 양친의 중간형을 보였으나 여교잡이 진전됨에 따라 반복친인 파의 표현형이 우세하였다. flow cytometry를 이용하여 2C nuclear DNA content을 측정한 결과 $F_1$은 양친에 비해 오히려 증가하였으나 여교잡이 진행됨에 따라 급속히 감소하여 $BC_2F_1$에서는 반복친과 비슷한 수준으로 감소하였다. 여교잡세대의 자식인 $BC_1F_2$은 반복친과 비슷한 $23.1{\pm}1.0pg$으로 측정되었으나 $BC_2F_2$$22.9{\pm}0.4pg$으로 반복친의 2C nuclear DNA content인 $23.2{\pm}0.2pg$ 보다 오히려 감소하였다.

임의의 경계조건을 갖는 철근 콘크리트 사판의 해석 (An Analysis of the R/C Skew-Plates With Arbitrary Boundary Conditions)

  • This study was carried out to investigate mechanical characteristics of the uniformly loaded skew-plate at 4 kinds of boundary condition : i) all edges are clamped (BC-1) , ii) all edges are simply supported (BC- 2), iii) two opposite edges are clamped and the other two edges are free (BC-3), and iv )two opposite edges are simply supported and the other two edges are free (BC-4). Various skew angles, 0$^{\circ}$, 10$^{\circ}$, 15$^{\circ}$, 30$^{\circ}$, 40: 45: and 60, of the plate were tested for the above boundary conditions. Resutts obtained from the study are summarized as follows ; 1.The lateral displacement at the center of a skew- plate was decreased as the skewangle increased at all of the boundary conditions. The decrements of the conditions of BC-3 and BC-4 were considerable. And, difference of the displacement between the boundary conditions was decreased as the skew-angle was increased. 2. X-moments (to the Y-axis) at the center of a skew- plate and the minimum principal moments were shown as a similar pattern of change with respect to the skew-angle variation between BC-i and BC-2 and between BC-3 and BC-4, and the pattern of change at the conditions of BC-3 and BC-4 were shown higher rates than those for the conditions of BC-i and BC-2 3.Y-moments (to the X- axis) at the center of a skew-plate and the maximum principal moment were decreased as the skew-angle increased in a similar pattern at all of the boundary conditions. 4.X-moments at the obtuse angle side of a skew-plate were shown as a parabolic pattern of change (frist increased after then decreased) as the skew-angle increased, and a skew-angle resulting the maximum absolute moment was depended on the boundary conditions. 5.Y-moments at the obtuse angle side of a skew-plate were affected by the skewangle much more at the boundary condtions of BC-2 and BC-4 than at the conditions of BC-i and BC-3. 6.Maximum principal moments at the obtuse angle side of a skew-plate at the skew angle of 40$^{\circ}$- 45$^{\circ}$ were resulted almost the same value at all of the boundary conditions .

Factorial design에 의한 Acetobacter xylinum KJ1의 Bacterial cellulose 생산조건의 최적화 (Optimization for the Bacterial Cellulose Production of Acetobacter xylinum KJ1 by Factorial Design)

  • Factorial design model을 이용하여 A. xylinum KJ1의 BC생산을 위한 최적 배양조건을 결정하였다. 요인 분석을 위한 실험 계획법으로는 부분요인 분석을 통한 factorial model을 이용하였으며, 주요 실험인자인 탄소원 농도, 교반속도(rpm), 산소분압, CSL의 농도의 네 가지 factor의 영향에 의한 BC생산량의 변화를 측정하였다. SAS 프로그램을 이용하여 전체 24개의 실험계획에서 각각의 factor의 조합에 의한 BC 생산성에 관한 결과 및 예측값을 비교한 결과 BC 생산량 면에서는 상관계수($R^2$)가 0.91으로 이었고, 수율면에서는 상관계수($R^2$)는 0.81이다. 최적 BC 생산을 위한 각 factor들이 탄소원의 농도 4%, 교반속도 460 rpm, 산소분압 0.28 atm, CSL 농도 6%일 때, 이때의 BC 생산량은 11.67 g/L로 예측되었다. 그리고 BC 생산 수율면에서 최적 배양조건이 탄소원 농도 4%, 교반속도 564 rpm, 산소분압 0.21 atm, CSL 농도 2%일 때 최적의 수율 0.42 g/g를 얻을 수 있을 것으로 예측되었다. 결정된 최적 조건에서의 실증 실험 결과 11.47 g/L의 BC 생산량을 얻을 수 있었으며, 이는 fermentor상에서의 기본실험에서 얻은 4.83 g/L보다 2.4배 이상 향상된 결과이다.

Pilot Scale의 박테리아 셀룰로오스 생산 및 그의 물성 (Production of Bacterial Cellulose by Pilot Scale and Its Properties)

  • 본 연구에서는 음식폐기물 효소당화액을 BC생산 배지원으로 이용하여, flask정치배양과 50 L로 규모확대한 공기순환배양기에서 BC생산성을 검토하고, 생산된 BC의 제반물성을 조사하였다. 전단력을 낮추기 위해 당화액배지에 agar를 첨가하여 10 L, 50 L 공기순환배양기에서 배양한 결과, 50 L 규모로 scale-up한 반응기에서 5.64 g/L로 10 L 공기순환배양기 (5.84 g/L)에서와 동등한 BC 농도가 얻어져 BC 대량생산에 효과적인 배양방법임이 입증되었다. 50 L 규모로 확대된 공기순환배양기를 이용하는 본 배양 방법은 BC의 저비용 대량생산을 위한 효율적인 생산 방법 중 하나가 되리라 사료된다. 또한, 당화액을 배지로 하여 공기순환 방법으로 제조한 BC의 물성을 정치배양 방법으로 제조한 BC의 물성과 비교 검토한 결과, 공기순환법으로 제조하였을 때 중합도 및 역학적 성질이 약간 저하하였으나 결정성은 비슷하였으며, 배양기 용량의 대소에 따른 물성의 변화는 거의 없었다.

육계 생산성 및 맹장내 미생물에 대한 유산균의 첨가 효과 (Effects of Supplemental Lactobacillus spp. on Performance and Cecum Microflora in Broiler)

  • This study was carried out to examine the effects of Lactobacillus spp. on performance and cecal microflora in broiler chicks. Eight diets consisting of basal diet(C), C+Lactobacillus crispatus BC7 (T2), C+Lactobacillus reuteri BC9 (T3), C+L.reuteri BC5+L.crispatus BC9 (T4), C+L. reuteri BC5 (T1)+L.reuteri BC9 (T5), C+L.crispatus BC7+L.reuteri BC9 (T6) and C+L.reuteri BC5+L.crispatus BC7+L.reuteri BC9 (T7), were fed to Ross male broiler chicks for 5 weeks. The level of supplemented Lactobacillus spp. was 107 cfu/g diet. Body weight and feed intake were measured every week, and cecal microfla was counted at 1 and 5 weeks for Lactobacillus and yeast. Body weight increased signigicantly in supplemental Lactobacillus treatments from 2 weeks of age (p<0.05). Chicks in T2, T5 and T5 of treatments were heavier than those of other treatments at 5 weeks of age(p<0.05). Viability was not different significantly. Feed intake and feed conversion also were not different, although feed conversion improved slightly in supplemental Lactobacillus treatments. Lactobacillus spp. of cecal content was increased in supplemental Lactobacillus treatments at 1 week, but no significance was found. The number of yeast in cecum was not different from that of supplemental Lactobacillus. In conclusion, Lactobacillus, L.crispatus BC7 and L.reuteri BC9 from broiler cecum could contribute to the increase in body weight with supplemented mono-or mixing of di-lactobacilli. The results indicate that Lactobacillus feeding can benefit to intestinal lactobacillus at early growing broiler.

대구지역 택배서비스업 종사자의 디젤엔진배출물 노출 평가 (Exposure Assessment of Diesel Engine Exhaust among Door-to-door Deliverers in Daegu)

  • Objectives: This study evaluated the diesel engine exhaust (DEE) exposure levels of door-to-door deliverers in Daegu from July to September. Methods: We measured exposure levels of DEE surrogates for the same door-to-door deliverers who joined the particulate matter 2.5 exposure study previously published in this journal. Black carbon(BC) concentrations were measured using real-time BC monitoring devices with 1 minute interval. $NO_2$ concentrations were monitored using passive badges. DEE exposure data were analyzed using the same characteristics and GPS information as the first study. Results: A total of 40 measurements of BC concentrations and $NO_2$ concentrations were collected during delivery of parcels. The average exposure levels to BC, and $NO_2$ were $2.23{\mu}g/m^3$ ($0.001-350.85{\mu}g/m^3$) and 21.26 ppb(3.3-61.37 ppb), respectively. Exposure levels to BC according to the day of a week and coverage areas were not significantly different(p>0.05). Delivery trucks manufactured before 2006 caused significantly higher exposure to BC than the trucks manufactured after 2006(p<0.05). Exposure levels of BC integrated for each time in residential area and roadsides were $1.96{\mu}g/m^3$ and $3.46{\mu}g/m^3$, respectively, and the difference was statistically significant(p<0.001). The Pearson correlation coefficients between the ambient $PM_{2.5}$ and BC was significant, r=0.26(p<0.01); however, the correlations between $PM_{2.5}$ and ambient $PM_{2.5}$, and between BC of DEE and $PM_{2.5}$ of DEE did not show a significant correlation Conclusions: BC and $NO_2$ exposure levels were significantly lower when door-to-door deliverers drove newer trucks. BC exposure levels of deliverers were higher in roadsides than in residential area. DEE from nearby vehicles through open windows might be the main source of BC exposure.

Perturbation of Background Atmospheric Black Carbon/PM1 Ratio during Firecracker Bursting Episode

  • Perturbation in ambient particulate matter ($PM_1$, $PM_{2.5}$, $PM_{10}$) and black carbon (BC) concentrations was studied during a firecracker bursting episode in Diwali (Festival of Lights) celebrations in Nagpur, India. Firecracker bursting resulted in greater escalation in fine particulates over coarse particulates while $PM_{2.5}$ was found to be dominated by $PM_1$ concentration. On the Diwali day, daily mean concentration of $PM_{2.5}$ and $PM_{10}$ exceeded Indian National Ambient Air Quality Standards by over 1.8 and 1.5 times, respectively, while daily mean BC concentration on the same day was almost two times higher than the previous day. The BC/$PM_1$ ratio reduced remarkably from about 0.26 recorded before fire-cracker bursting activity to about 0.09 during fire-cracker bursting on Diwali night in spite of simultaneous escalation in ambient BC concentration. Such aberration in BC/$PM_1$ was evidently a result of much higher escalation in $PM_1$ than BC in ambient air during firecracker bursting. The study highlighted strong perturbations in ambient $PM_1$, $PM_{2.5}$, $PM_{10}$ concentrations and BC/$PM_1$ during the firecracker bursting episode. Altered atmospheric BC/$PM_1$ ratios could serve as indicators of firecracker-polluted air and similar BC/$PM_1$ ratios in local and regional air masses might be used as diagnostic ratios for firecracker smoke.
