Subunit Assembly of Laminin Variants in Cultured BAEC (BAEC세포에서의 Laminin 이형체 Subunit의 회합에 관한 연구)

  • Bovine aortic endothelial cells(BAEC) produce two variant forms of laminin with a subunit composition of AB1B2 and A'B1B2. Analyses of the intracellular assembly of these subunits revealed that the B1B2 dimer formed first, and that A or A' joined to form the AB1B2 or A'B1B2 trimer. Angiostatic steroids shifted the relative size of the A and A' monomer pool in BAEC, and competition between the A and A' subunits in joining the B1B2 dimer produced AB1B2 and A'B1B2 in different ratios. This result suggests that subunit replacement is the general mechanism for producing laminin variants by various cells for tissue morphogenesis. When laminin subunits in BAEC were cross-linked with dithio-bis-succinimidylpropionate(DSP) and immunoprecipitated with anti-Iaminin antiserum, monomeric A,A',B1 and B2 monomers and the B1B2 dimer migrated as extremely large molecules in sodium dodecyl sulfate gel electrophoresis under nonreducing conditions. When the crosslinking disulfide bonds were cleaved under reducing conditions, they migrated as the usual subunits. This result suggests that molecular chaperones were involved in the process of the assembly and replacement of laminin subunits.

  • A local area network (LAN) is a computer network within a small geographical area such as a home, school, computer laboratory, office building or group of buildings. A LAN is composed of interconnected workstations and personal computers which are each capable of accessing and sharing data and devices, such as printers, scanners and data storage devices, anywhere on the LAN. LANs are characterized by higher communication and data transfer rates and the lack of any need for leased communication lines. Communication between remote parties can be achieved through a process called Networking, involving the connection of computers, media and networking devices. When we talk about networks, we need to keep in mind three concepts, distributed processing, network criteria and network structure. The purpose of this Network is to design a Local Area Network (LAN) for a BAEC (Bangladesh Atomic Energy Commission) Head Quarter and implement security measures to protect network resources and system services. To do so, we will deal with the physical and logical design of a LAN. The goal of this Network is to examine of the Local Area Network set up for a BAEC HQ and build a secure LAN system.

Recovery Effects from Oxidative Cell Damage by So-Hap-Hyang-Won on Bovine Aortic Endothelial Cells (BAEC)

  • So-Hap-Hyang-Won, a traditional oriental medicine used in the treatment of stroke patients, was examined for its ability to reverse the cell damage caused by lipid peroxidation products and oxidative stress in bovine aortic endothelial cells (BAEC). The effects of herbal medicine on cell proliferation and recovery of oxidative damaged situation were studied in BAEC, which was considered an appropriate in vitro model for stroke resulting from various vascular diseases prevalent in advanced age. In a clinical study of stroke patients, So-Hap-Hyang-Won appeared to improve considerably arm and leg movements as well as consciousness disturbance condition, compared with other traditional medicines used for stroke. When BAEC were treated with extracts of the lyophilized herbal medicines, only that of So-Hap-Hyang-Won stimulated cell proliferation and showed no toxicity even at high concentrations. In studies of BAEC treated with extracts of the lyophilized material of the 14 components of So-Hap-Hyang-Won, only the extract of Foeniculi Fructus stimulated cell growth at all concentrations tested. Moreover, when cells were treated with Foeniculi Fructus (10 and 100 mg/ml) extract after prior exposure to t-BHP ($l0\mu\textrm{M}$) or HNE ($0.2\mu\textrm{M}$), lipid peroxidation products which are known to be involved in aging and vascular diseases, or after the exposure to SIN-l ($500\mu\textrm{M}$), which generates nitric oxide (NO) and other reactive oxygen species, there was substantial recovery from the oxidative damage, presumably due to the radical-scavenging effect of Foeniculi Fructus extract. Foeniculi Fructus not only showed stimulatory effects on cell growth and cell damage repair in BAEC, but also appeared to show the most anti-aging activity among all the herbal components of So-Hap-Hyang-Won.

The status of NORMs in natural environment adjacent to the Rooppur nuclear power plant of Bangladesh

  • The Rooppur Nuclear Power Plant (RNPP), the first nuclear power plant in Bangladesh with a capacity of 2.4 GWe, is under construction on the bank of the river Padma, at Rooppur in Bangladesh. Measurement of background radioactivity in the natural environment adjacent to RNPP finds great importance for future perspectives. Soil and sediment samples collected from upstream and downstream positions of the Padma River (adjacent to RNPP) were collected and analyzed by HPGe gamma-ray spectrometry for primordial radionuclides. The average activity concentrations (in Bqkg-1) of 226Ra, 232Th and 40K radionuclides in soil samples were found to be 44.99 ± 3.89, 66.28 ± 6.55 and 553 ± 82.17 respectively. Respective values in sediment samples were found to be 44.59 ± 4.58, 67.64 ± 7.93, 782 ± 108. Relevant radiation hazard indices and dosimetric parameters were calculated and compared with the world average data recommended by US-EPA. Analytical results show non-negligible radiation hazards to the surrounding populace. Measured data will be useful to monitor any change of background radioactivity in the surrounding environment of RNPP following its operation for the generation of nuclear energy.

Stimulatory Efect of Cinnamomum cassia Extract on Angiogenesis Through Regulation of VEGF (VEGF 조절을 통한 육계 추출물의 신생혈관 형성 촉진 작용)

  • 목적 : 한의학 분야에서 육계(CCE: Cinnamomum cassia)는 혈류 순환이 저하된 상태에 널리 쓰이는 약재이다. 이 연구에서는 CCE와 그 활성 혼합물인 cinnamic acid가 in vitro와 in vivo에서 발휘하는 신생혈관 형성 작용에 대해 알아보고자 한다. 방법 : CCE와 CA의 신생혈관형성 작용을 알아보기 위하여 Human unbilical endothelial cells(HUVECs)와 Bovine arterial endothelial cells(BAECs) 그리고 Matrigel assay를 이용하였다. 결과 : In vitro에서 CCE와 그 활성 혼합물은 HUVEC 증식, 이동, 모세혈관-유사 관 형성을 유도한다. CCE에서의 25-50% 증가(1 또는 10ng/ml 용량에서)는 CA에서 증식과 유사하게 나타났다. HUVEC에서는 증식이 뚜렷하게 나타났으나 BAEC에서는 증식이 뚜렷하게 나타나지 않아 증식 효과는 내피세포의 기원에 따라 다른 것으로 나타났다. BAEC는 CCE와 CA에서 증식은 보이지 않았지만 이동과 모세혈관-유사 관 형성 또는 분화를 보였다. 또한 CCE와 CA는 HUVEC에서 VEGF 발현과 Flk-1/KDR 발현을 각각 2.2배와 2.5배 증가시켰다. 결론 : CCE와 그 활성 혼합물은 in vivo와 in vitro에서 혈관 내피세포의 VEGF 발현을 통하여 신생혈관 형성 작용을 유도하며 이는 앞으로 다른 한약 추출물의 신생혈관형성 촉진 작용 연구에 모델을 제시할 것으로 보인다.

Effect of Trametes cubensis Extract on Vascular Function of Bovine Aortic Endothelial Cells (Trametes cubensis 버섯 추출물이 소의 대동맥 내피세포의 혈관 기능에 미치는 효능)

  • Mushrooms have been extensively used as traditional medicines to treat cancer and inflammatory diseases. In this study, we examined whether Trametes cubensis extract (TCE) exerted beneficial effects on cardiovascular function. First, we demonstrated that TCE was non-cytotoxic and enhanced cell proliferation of bovine aortic endothelial cells (BAEC). Moreover, TCE induced cell migration and blocked lipopolysaccharide-induced adhesion of monocytes to BAEC. We performed a variety of cell signaling studies, showing that TCE activates p38 MAPK and generates reactive oxygen species (ROS). Our results showed that TCE-induced vascular functions were mediated by p38 MAPK, but not by ROS. These results provide insights into bio-medical applications of TCE as a preventive or therapeutic agent for treating cardiovascular diseases including atherosclerosis.

Effects of Bambusae Caulis in Liquamen on the Stress Proteins Induced by Heating in Endothelial Cells (혈관내피세포에 열 충격 부과시 죽력이 stress proteins의 발현에 미치는 영향)

  • We have previously observed that Bambusae Caul is in Liquamen (BCL) stimulates the adipose conversion of 3T3-L1 cells and molecular chaperones were involved in the process of the assembly and replacement of laminin subunits in Bovine aortic endothelial cells(BAEC). Endothelial cells are exposed to continuous shear stress due to the blood flow. Heat shock protens(hsp) are a well-known stress response protein, namely, stress proteins. To investigate effects of BCL on the stress proteins induced by heating in endothelial cells, we have analyzed synthetic amounts of stress proteins in sodium dodecyl sulfate gel electrophoresis under reducing conditions. Under the condition of heating stress, BCL inhibited the synthesis of stress proteins in endothelial cells. These results suggest that BCL may have an important role for expression of stress proteins induced by heating in endothelial cells.

Regulation of NO from Endothelial Cells by the Decrease of Cellular cAMP Under Arsenite Exposure

  • In an attempt to delineate the direct effect of arsenite-induced endothelial dysfunction on nitric oxide (NO) production, confluent bovine aortic endothelial cells (BAEC) were incubated with arsenite, and endothelial NO synthase expression and NO production were measured. Exposure of arsenite decreased NO production for up to 24h. This decrease was accompanied by decreases in cAMP, protein kinase A (PKA) activity, and furthermore, significant reduction of pCREB. In conclusion, this study is the first to demonstrate that exposure of arsenite decreases NO production by a reduction of pCREB and PKA activity that may be mediated by cAMP, leading to endothelial dysfunction.

  • Cytosolic oxidation by 4-hydroxy-2-nonenal (4HNE) and tert-butyl hydroperoxide (t-BHP) results in cell death of bovine aortic endothelial cells (BAEC). In this study, we have investigated the roles of antioxidants such as 2,3,6-tribromo-4,5-dihydroxy benzyl methyl ether (TDB) and phloroglucinol in preventing cell death. After treatment with oxidants for 6h, cells became compact and showed nuclear condensation, which were characteristics of early apoptosis. After l2h treatment, morphologic features including severe cytoplasm condensation, membrane blebbing, and apoptotic bodies were prominent and these findings were interpreted as characteristics of late-apoptosis. When the apoptotic cells were treated with antioxidants for 12h, both early and late apoptotic cells did show no significant change. After oxidant treated cells were incubated with antioxidant for 24h, the characteristics of early-apoptosis were eliminated but cells in lateapoptosis could not return to normal cells. These results suggest that TDB and phloroglucinol prevent the cells from dying through apoptosis induced by 4HNE and t-BHP in early stage.
