Determination of Alisol B 23-acetate and Alisol C 23-acetate in Alismatis Rhizoma by HPLC-ESI-MS

  • An HPLC-ESI-MS method has been developed to identify and quantify two main tetracyclic triterpenes, alisol B 23-acetate and alisol C 23-acetate in the Alismatis Rhizoma (Taeg-Sa). The relative distribution of the two triterpenes in the methanolic extract of commercially available Alismatis Rhizoma was established by selective ion monitoring (SIM) mode via electrospray ionization (ESI) source. Regression equations revealed good linear relationship, and the correlation coefficients were 0.999 and 0.998 for alisol B 23-acetate and alisol C 23-acetate, respectively, between the peak areas of the components and their concentration in a range of $0.06-2.0{\mu}g/mL$. It was found that there were significant differences in the amount of alisol B 23-acetate and alisol C 23-acetate between Korean and Chinese origins. The results showed that this method could be used to identify the two components in Alismatis Rhizoma with high sensitivity and selectivity.

The Depressive effect of Depletion in Hypertentive Stroke Patients (고혈압(高血壓) 뇌졸중(腦卒中) 환자(患者)에 대한 사혈(瀉血)의 강압효과(降壓效果))

  • Objective : The aim of this research was to investigate the depressive effect of depletion of B15(心兪), B23(腎兪), L5(尺澤), B40(委中) in hypertentive stroke patients. Methods : Depletion on B15(心兪), B23(腎兪), L5(尺澤), B40(委中) in hypertentive stroke patient who were admitted in Dong-Seo Oriental Medical Hospital every other day form 4 weeks, without western antihypertensive drug. Results : After treatment in 2nd week, depletion on B15(心兪), B23(腎兪), L5(尺澤), B40(委中) without western antihypertensive drug increased the systolic and diastolic blood pressure. After treatment in 4th week, depletion on B15(心兪), B23(腎兪), L5(尺澤), B40(委中) without western antihypertensive drug decreased the systolic and diastolic blood pressure. After treatment in 4th week, values of RBC and hemoglobine were not significantly changed. Conclusions : Continous depletion on B15(心兪), B23(腎兪), L5(尺澤), B40(委中) in hypertentive stroke patient without western antihypertensive drug implies decreasement of blood pressure, without change of values of RBC and hemoglobine.

Protective Effects of Methanol Extract and Alisol B 23-acetate of Alisma orientale on Acetaminophen-Induced Hepatotoxicity in Rats

  • Hepatoprotective effects of methanol extract and alisol B 23-acetate of Alisma orientale were studied in acetaminophen (APAP)-treated rats. APAP increased hepatic content of lipid peroxide, which was suppressed by methanol extract and alisol B 23-acetate. The liver of rats treated with APAP had higher P-450, aminopyrine N-demethylase and aniline hydroxylase activities than those of normal control rats. The increases in hepatic drug metabolizing enzymes by the i.p. injection of APAP were significantly alleviated by the administration of methanol extract or alisol B 23-acetate. The injection of APAP also resulted in a substantial reduction of hepatic glutathione content and glutathione S-transferase activity, and the decreases were partially, but significantly, restrained by the oral administration of methanol extract prior to the i.p. injection of APAP. Hepatic activities of glutathione reductase (GR) and ${\gamma}$-glutamylcystein synthetase ${\gamma}$-GCS) were also decreased significantly in APAP-treated rats. The decreases in hepatic GR and ${\gamma}$-GCS activities by APAP injection were improved partially, but significantly, with administration of methanol extract of A. orientale. Treatment with alisol B 23-acetate also improved the hepatic ${\gamma}$-GCS activity significantly, but not GR.

  • In order to observe the effects of moxibustion on anemic condition as a acute loss of blood, white rats were induced loss of blood by cadiac puncture. And then morphologically variation of The Bone Marrow was observed the following points of each rat were used ; Shinsu($B_{23}$ or $VII_{23}$), Hyeon jong($GB_{39}$ or $XI_{39}$), and complex of Shinsu($B_{23}$ or $VII_{23}$) and Hyeon Jong($GB_{39}$ or $XI_{39}$) Comparing these above methods with the control group the results were as follow : 1. Selection of Brevibacillus brevis B23 and Bacillus stearothermophilus B42 as Biological Control Agents against Sclerotinia Rot of Lettuce (상추 균핵병 생물적방제를 위한 Brevibacillus brevis B23과 Bacillus stearothermophilus B42의 선발)

  • Bacillus spp. isolated from mushroom medium wastes were evaluated for their biocontrol potentials on control of Sclerotinia rot of lettuce. The Bacillus isolates were more effectively obtained from waste suspension when directly added into nutrient agar(NA) medium than plating on the agar medium. Totally 42 isolates obtained from the wastes B23 and B42 showed highest antifungal activity against eight fungal pathogens such as Sclerotinia sclerotiorum, Rhizoctonia solani, Pythium ultimum, Phytophthora capsici, Fusarium oxysporum, Colletotrichum gloeosporioides, Cladosporium cucumerinum, and Botrytis cinerea and B23 and B42 were finally selected for further studies. Optimal concentration of the isolates was $10ml(10^7cfu/ml)$ to suppress the Sclerotinia rot of lettuce. Supplements such as starch, glycerol, and egg-yolk successfully maintained the bacterial population for 30 days in vitro and increased bio-control potentials against the disease. The bacterial isolate B23 alone showed 72% control value, furthermore it presented 95% control value when supplemented with 0.2% of starch, glycerol, and egg-yolk. The promising Bacillus isolates B23 and B42 were identified as Brevibacillus brevis and Bacillus stearothermophillus, respectively, based on morphological and physiological characteristics according to API database.

Design and Fabrication of Isolator for Mobile Repeater (이동통신 중계기용 아이솔레이터의 설계 및 제작)

  • The lumped element isolators with temperature compensation magnetic circuits were designed in this paper. Isolators for mobile repeater at each band of K-PCS, GSM1900, W-CDMA, WiBro were fabricated by using optimum parameters of EM simulation, Form the results of this study, first of all, insertion loss at K-PCS band was below 0.2 dB and return loss was over 25dB. The bandwidth in isolation 23dB was found 30 MHz. Secondly, the figures of both losses at GSM1900 were 0.25dB and 23dB, respectively. The measured bandwidth in isolation 30dB was 60MHz. Thirdly, the losses at W-CDMA band were 0.15dB and 25dB. The bandwidth in isolation 24.8dB was found 60Mhz. Finally, the figures of both losses at Wibro band were 0.25dB and 23dB, respectively. The measured bandwidth in isolation 22.1dB was 100Mhz. In addition, the results of measured IMD were shown from 76.4dBc to 80.1dBc.

FPTase Inhibition Effect of Protostanes from Alismatis Rhizoma and Derivatives from Alisol B 23-acetate (택사 (Alismatis Rhizoma)에서 분리한 Protostane계 화합물과 그 유도체의 FPTase 억제활성)

  • The purpose of this research is to study of inhibitory activity of protostane type triterpens against farnesyl-protein transferase (FPTase). The ingredients of Alismatis Rhizoma, alisol B 23-acetate, C 23-acetate, alisols B and A 24-acetate, and thirteen synthetic analogues from alisol B 23-acetate exhibited inhibition activity against FPTase by scintillation proximity assay method. As a result, alisol C 23-acetate, one of the constituents of Alismatis Rhizoma, the synthetic analogues carboxylated and hydroxylated on branch chain of protostane exhibited a significant inhibitory activity. However, the compounds significantly lowered the inhibitory activity, when there is no 3 position keto on protostane skeletone.

Alisol B 23-Acetate Ameliorates Ovalbumin-Induced Allergic Asthma during Sensitization and Challenge Periods

  • Rhizome of Alisma orientale has been used as a traditional medicine for treating kidney diseases in East Asian countries. Its inhibitory effects on hypersensitivity responses have been reported for methanol extracts, with alisol B 23-acetate (AB23Ac) being the most active constituent among six terpenes in inhibiting the direct passive Arthus reaction. However, whether AB23Ac has efficacy against allergic asthma has not been tested to date. The in vivo efficacy of AB23Ac in an ovalbumin (OVA)-induced allergic asthma mouse model was evaluated by administrating AB23Ac before OVA sensitization or OVA challenge in BALB/c mice. AB23Ac suppressed antigen-induced degranulation of RBL-2H3 mast cells in a concentration-dependent manner. The administration of AB23Ac both before OVA sensitization and OVA challenge greatly lowered pulmonary resistance and the increase in immune cell counts and inflammatory responses around the peribronchial and perivascular regions. In addition, the inflammatory cytokine levels of Th1/Th2/Th17 cells in the bronchoalveolar lavage fluid decreased in the AB23Ac-treated groups. AB23Ac reduced the number of PAS-stained cells in the lungs. Furthermore, a computer modeling study indicated that AB23Ac can bind tightly to spleen tyrosine kinase (Syk). These results suggest that AB23Ac may ameliorate allergic asthma by suppressing immune responses in dendritic cells during sensitization and in mast cells during challenge periods.
