Effect of Saururus chinensis BAILL Extract for Pharmacopuncture on the melanogenesis in B16F10 cells (삼백초 약침액이 B16F10 흑색종세포의 멜라닌 합성에 미치는 영향)

  • Objectives : The purpose of this study was to investigate the melanogenesis inhibition effect of Saururus chinensis BAILL (SC) on in B16F10 melanoma cells. Methods : SC was fractionated ethanol extract by the hexane, ethyl acetate, butanol and water. We confirmed the inhibitory effect of tyrosinase activity and melanogenesis of all fraction samples. Results : Hexane fraction of Saururus chinensis BAILL (HSC), ethyl acetate of SC (ESC), and butanol of SC (BSC) were discovered to inhibit tysoinase activity and melanogenesis in the absence or presence of ${\alpha}$-MSH. However, water fraction of SC (WSC) did not affect tyrosinase activity and melanogenesis. In addition, all fractions did not inhibit the catalytic activity of cell-free tyrosinase from B16F10 melanoma cell lines. Conclusions : These results suggest that HSC, ESC and BSC reduce pigmentation by indirectly regulating tyrosinase.

Effect of Myricetin Combined with Vitamin C or Vitamin E on Antioxidant Enzyme System in Murine Melanoma Cells (B16F10 세포에서 Flavonoid인 Myricetin과 Vitamine C, Vitamine E의 병용 투여가 항산화 효소계에 미치는 영향)

  • Flavonoids are class of polyphenolic compounds widely distributed in the plant kingdom, which display a variety of biological activities, including antiviral, antithrombotic, antiiflammatory, antihistaminic, antioxidant and free-radical scavenging abilities. To determined flavonoid, myricetin in the presence of other antioxidants - vitamin C and vitamin E - can exert antioxidative properties not only directly by modulating the AOE system but also scavenging free radical, we investigated cell viability, antioxidant enzyme activities and ROS level in B16F10 murine melanoma cell. B16F10 cells were exposed to medium containing myricetin in the presence or absence of vitamin C or vitamin E for a period of 24 hr. Cell viability was measured by MTT assay. In co-treating myricetin with other antioxidants, CAT activities were increased, compared with control, but SOD and GPx activities were decreased, compared with each antioxidant treated groups . In the group of myricetin or myricetin present with other antioxidants, ROS levels were decreased dose-dependently. Especially, myricetin present of other antioxidants were decreased compared with myricetin.

Constituents of Cynanchum auriculatum and their Inhibitory Effect on Melanogenesis in B16 Mouse Melanoma Cell Lines (이엽우피소의 성분 및 멜라닌 생성 억제활성)

  • Fourteen compounds were isolated from the roots of Cynanchum auriculatum and their chemical structures were identified as ${\beta}$-sitosterol (1), acetovanillone (2), p-hydroxyacetophenone (3), 2,4-dihydroxyacetophenone (4), 2,5-dihydroxyacetophenone (5), cynandione A (6), methyleugenol (7), daucosterol (8), Succinic acid (9), cynauriculoside A (10), wilfoside C3N (11), wilfoside C1N (12), wilfoside K1N (13) and wilfoside C1G (14). Among them, compounds 2-5 were isolated from this plant for the first time. And 2, 5-dihydroxyacetophenone (5) showed the most potent inhibitory effect on melanogenesis in B-16 mouse melanoma cell lines with $IC_{50}$ value of $20\;{\mu}M$.

The Effects of Soybean Protopectinase on Melanin Biosynthesis (효소(Protopectinase) 처리한 대두가 세포내 멜라닌 생성에 미치는 영향)

  • This study was performed to assess the antioxidant activities and whitening effects of protopectinase enzymes and mechanical maceration from soybeans on melanin synthesis. The whitening effects of enzyme treatment and mechanical maceration were examined by an in vitro mushroom tyrosinase assay and by assessing markers in B16BL6 melanoma cells. We assessed inhibitory effects on the expression of melanogenic enzymes, including tyrosinase, tyrosinase-related protein 1 (TRP-1), and tyrosinase-related protein 2 (TRP-2) in B16BL6 cells. Inhibitory effects on free radical generation were determined by measuring DPPH and hydroxyl radical scavenging activities. In DPPH radical scavenging activity, enzyme treatment and mechanical maceration had a potent anti-oxidant activity in a dose-dependent manner and significantly inhibited tyrosinase activity in vitro and in B16BL6 melanoma cells. There was also an inhibition in the expression of tyrosinase, TRP-1, and TRP-2 in B16BL6 melanoma cells. Our results show that soybean protopectinase treatment inhibits melanogenesis, with the underlying mechanism possibly due to the inhibition of tyrosinase activity and tyrosinase, TRP-1, and TRP-2 expression. We suggest that soybean protopectinase should be contained as natural active ingredients for antioxidant and whitening cosmetics.

Effects of Hizikia fusiforme Fractions on Melanin Synthesis in B16F10 Melanoma Cells (톳 분획물이 B16F10 흑색종 세포에서의 멜라닌합성에 미치는 영향 연구)

  • The objective of this study was to evaluate the anti-melanogenic effects of Hizikia fusiforme (HF) fractions in ${\alpha}$-melanocyte stimulating hormone-induced B16F10 mouse melanoma cells. Ethanol extractions of Hizikia fusiforme (EEHF) were subjected to fraction by using dichloromethane (CFHF), ethyl acetate (EAFHF), butanol (BFHF), and water (WFHF). EEHF, CFHF, and EAFHF inhibited tyrosinase activity and melanin synthesis in B16F10 mouse melanoma cells in a dose-dependent manner. The melanin contents were inhibited by 40.5% and 33.2% in response to treatment with 50 ${\mu}g/ml$ of EEHF and CFHF, respectively. In addition, tyrosinase activities showed a 53.3% and 54.1% reduction in treatment with 50 ${\mu}g/ml$ of EEHF and CFHF. Western blotting analysis showed that EEHF, CFHF, and EAFHF inhibited tyrosinase, TRP-1, TRP-2, and MITF expression in a dose-dependent manner. In conclusion, these findings indicate that ethanol and dichloromethane fractions of Hizikia fusiforme, which inhibit melanin synthesis and tyrosinase activity, are effective skin-whitening agents.

Influence of Herb-combined Remedies Based on "Yooam" Prescription of Dongeuibogam on Migration and Invasion of B16F10 Melanoma Cells (B16F10 흑색종 세포의 이동과 침윤에 미치는 동의보감 "유암" 처방에 근거한 한약복합처방들의 영향)

  • Objectives : We compared the inhibitory effects of herb-combined remedies, which were recorded on "Yooam" prescription of Dongeuibogam, on cell migration and invasion, two critical cellular processes that are often deregulated during metastasis, in B16F10 melanoma cells. For this purpose, water extracts of Sipyukmiryukieum (SYMRKU), Danjacheongpitang (DJCPT), Cheongganhaeultang (CGHUT) and Jipaesan (JPS) were used. Methods : Cytotoxicity was assessed by an MTT assay. Wound healing and matrigel transwell assays were used to examine on B16F10 cell migration and invasion. The levels of mRNA and protein expression of matrix metalloproteinases (MMPs) and tissue inhibitor of metalloproteinases (TIMPs) were analyzed by RT-PCR and Western blotting. Results : Our data showed that DJCPT showed the strongest inhibitory effect among the four prescriptions in inhibiting cell motility of B16F10 melanoma cells within the concentration range that was not cytotoxic. The inhibitory potential of colony formation was higher in DJCPT and SYMRKU compared to the other two types of prescriptions, and the inhibitory effect of invasiveness is shown in order of DJCPT, SYMRKU, CGHUT and JPS. DJCPT, and SYMRKU strongly inhibited the activity and expression of MMP-2 and MMP-9, which are important mediators in cancer invasion, compared to CGHUT and JPS, and the increased expression of TIMP-1 and TIMP-2 was also more effective in these two prescriptions. In conclusion, DJCPT is expected to exhibit the most potent blocking effect on migration and invasion among four herb-combined remedies compared in B16F10 melanoma cells. Conclusion : Overall, the results of this study will be used as an important source to validate these prescriptions in animal models and to understand the mechanism of action of herbal remedies recorded in Dongeuibogam.

  • Novel glucoconjugates phenolic moiety, 3-(methoxycarbonyl)-4-(hydroxyphenyl)-$\beta$-D-glucopyranoside(4), 3-(methoxyacetyl)-4-(hydroxyphenyl)-$\beta$-D-glucopyranoside(7), 4-(hydroxyphenyl)-$\beta$-D-ribofuranoside(11), were synthesized. In order to investigate their depigmentation effect, inhibitory activity against mushroom tyrosinase and inhibitory activity of melanin synthesis in B16 melanoma cell were evaluated in vitro. Compound 11 showed 92.0$\mu\textrm{g}$/㎖ of tyrosinase inhibitory activity whereas compound 4 and 7 showed very low activity not less than 300$\mu\textrm{g}$/㎖. Inhibitory activities of melanin synthesis in B16 melanoma cell of compound 4, 7, and 11 were 8.7, 15.1, and 36.0%, respectively, at the concentration of 100$\mu\textrm{g}$/㎖. Inhibitory activity of compound 11 was much higher than that of arbutin at the same concentration.

  • To identify inhibitory effect of Bamboo extract on melanogenesis, the effect was compared with arbutin, ascorbic acid, hydroquinone, and kojic acid on the melanin biosynthesis in B16 mouse melanoma cells and cultured human melanocytes. The cell viability of the agent was tested on cultured human melanocytes. We also examined its. free radical scavenging activity on 1,1-diphenyl-2-picrylhydrazyl(DPPH). Bamboo extract showed considerable effect against melanin production and did not reduce cell viability at the concentration tested. It also showed potent free radical scavenging activity.

  • Magnoliae Flos(‘shin-j’). the flower buds of Magnolia kobus, is acrid to taste with a ‘warm’ property. It is a ‘wind-cold’ discutient and nasal decongestant and is principally used in the treatment of nasal congestion with headache, sinusitis and allergic rhinitis. By screening inhibitory activities on the melanin polymer biosysthesis in B-16 mouse melanoma cell lines, methylene chloride extract of Magnoliae Flos was found to have inhibitory effect on melanin polymer biosynthesis.   • Purpose ; In order to study the effect of Houttuyniae herbal acupuncture on growth of melanoma B 16 and immunity in mice, the Control group with normal saline acupunchure after subcutaneous inoculation of B16BL6 cells, the Sample I with Houttuyniae herbal acupuncture manufactured by distilled water extraction method after subcutaneous inoculation of B16BL6 cells and the Sample II with Houttuyniae herbal acupuncture manufactured by alcohol extraction method after subcutaneous inoculation of B16BL6 cells were divided. Methods ; To evauluate the effect of Houttuynias herbal acupuncture on growth of melanoma B16 in mice, the weight of mouse melanoma, body weight, spleen index( $\sqrt(Weight of spleen/Body weight){\times}100$), lymphocytes numbers in mouse peripheral blood and spleen tissue, the percentage of CD4+ T cells, CD8+ and the ratio of CD4+/CD8+ in mouse peripheral blood and spleen tissue are measured. Results ; In study of the weight of mouse melanoma, Houttuyniae herbal acupuncture groups(Sample I, Sample II) showed statistically significant inhibitory effect. And in study of effect of reduction of change in body weight of mouse, Sample I showed statistically significant inhibitory effect, too. In study of reduction of spleen index, lymphocytes numbers in mouse spleen tissue, and the percentage of CD4+ and the ratio of CD4+/CD8+ T cells in mouse peripheral blood, Houttuyniae herbal acupuncture groups(Sample I, Sample II ) showed with statistically significant inhibitory effect. Also, Sample I showed inhibitory effect of reduction of the percentage of CD8+ T cells in mouse peripheral blood and spleen tissue and Sample II showed inhibitory effect of reduction of the percentage of CD4+ and the ratio of CD4+/CD8+ T cells in mouse spleen tissue with statistical significance. Conclusions ; The inhibitory effect of reduction of lymphocytes numbers in mouse peripheral blood and IL-2 productivity, Houttuyniae herbal acupuncture groups(Sample I, Sample II) didn't show significant effect.
