YEp 13 vector를 이용한 Bacillus amyloliquefaciens amylase gene의 cloning I. Escherichia coli에서의 발현 (Cloning of Bacillus amyloliquefaciens amylase gene using YEp13 as a vector I. Expression of cloned amylase gene in Escherichia coli)

  • E. coli-Yeast shuttle vector YEp 13에 B. amyloliquefaciens의 $\alpha$-amylase gene을 cloning하여 얻은 hybrid plasnidlasmid를 E. coli를 숙주세포로 하여 형질을 발현시켰다. 형질전환주의 $\alpha$-amylase 활성 측정 결과, B. amyloliquefaciens에 대해 20-30%의 상대 활성도를 나타내었다. 형질전환주의 경우 생성된 $\alpha$-amylase의 60-65%가 periplasm에 축적되었으며 세포 외부로의 분비는 없었다. Hybrid DNA를 agarose-gel 전기영동으로 조사한 결과 그 크기가 다른 다수의 hybrid DNA가 확인되었으며 pHA28의 plasmid (a)(Fig.3)에서만 $\alpha$-amylase 활성을 나타내는 것으로 밝혀졌다 pHA28의 plasmid(a)의 크기가 YEp 13 plasmid DNA보다 작은 것은 YEp13 plasmid에서 yeast gene부분이 deletion된 결과로 추측되었다.

13~18세 청소년기 여학생의 체형 특성에 관한 연구 (A Study on the Somatotype Characteristics of Adolescent Girls between the Ages of 13 and 18)

  • This study analyzes the characteristics of somatotype changes of girls from the ages of 13 to 18 that provide basic data on clothing construction for middle and high school girls. We use data obtained from body measurements of 1,590 adolescent girls 13 to 18 years of age provided in the 6th national anthropometric survey report. The differences among the age groups for 56 direct measurements were analyzed by ANOVA, and the structures of the differences by SNK (Students-Newman-Keuls test) were analyzed. The two major growth ages were 'ages 13-14' and 'ages 15-16'. The first major growth ages focused on all measurement items of height, length, circumference, breadth, depth and other items. The second one focused on measurement items of height and length of torso, circumference, and breadth of lower body. The major growth pattern of the age groups was 'ages 13<14&15<16&17&18' which appeared focused on the height and length items of upper body and torso, the width items of lower body and the circumference items of upper body and lower body. The measurement items of front length like waist front length, N.P. to B.P to waistline, N.P. to B.P., and B.P. to B.P. showed the growth pattern for 'ages 13<14<15<16<17&18'.

고온성이며 호알칼리성인 Bacillus cereus B-13으로부터 내열성, 호알카리성 Cyclodextrin Glucanotransferase의 생산과 특성 (Production and Characterization of Thermo-alkalotolerant Cyclodextrin Glucanotransferase from Thermo-alkalophilic Bacillus cereus B-13)

  • 내열성의 cyclomaltodextrin glucanotransferase (CGTase)를 이용하여 열에 안정한 싸이클린 텍스트린 (CD)을 생산하기 위하여 매우 높은 CGTase 활성을 보이는 고열성이며 호알카리성인 B-13세균을 분리하여 형태적, 생리학적 특성과 16S 리보솜 RNA 서열분석 등을 통하여 Bacillus cereus B-13으로 동정하였다. Bacillus cereus B-13 2% 가용성 전분, 1% 효모 추출물, 1% $Na_2CO_3$ 등을 함유하는 SYC 배지 (pH 8.5)에 접종하여 $50^{\circ}C$에서 24시간 배양하였을 때 최고의 CGTase 활성(130 U/ml)을 보였다. 또한 부분 정제된 CGTase의 작용 최적 온도와 pH는 각각 $60^{\circ}C$, pH 8.5-9.0 이었고 $80^{\circ}C$이하와 pH5.0-10.0에서 안정하였다. 1% 가용성 전분을 부분 정제한 CGTase와 작용시켰을 때 49%의 CD 수율을 보였다.

옥수수 사일리지용 미생물의 발효능력 평가 (Evaluation of Fermentation Ability of Microbes for Corn Silage Inoculant)

  • 옥수수는 우리나라에서 매우 중요한 사료작물이다. 대부분 사일리지 형태로 이용이 되고 있으며 많은 비율의 곡실을 함유하고 있어 반추가축의 우수한 사료로 이용된다. 본 시험은 옥수수 사일리지용 미생물 첨가제 개발을 위해 수집된 균주의 발효능력을 평가하기 위해 수행되었다. 양질의 옥수수 사일리지에서 선발한 젖산균을 0.02%의 Sodium azide가 들어있는 MRS 배지에 접종을 한 후 균을 분리하여 MRS broth에 접종하여 산 생성 능력을 조사하였다. 총 6종의 젖산균을 선발하였고 동정결과 C3-2, B13-1 및 CC9-1은 Lactobacillus plantarum으로 판명되었고 C11-4는 Lactobacillus fermentum, B14-1은 Lactobacillus paracasei, 그리고 A3-1은 Leuconostoc lactis로 동정되었다. 선발된 5종의 균주와 시판되는 2종 첨가제 그리고 무처리구를 두고 황숙기에 수확된 옥수수에 접종하여 사일리지를 제조하고 2개월 후에 개봉하여 분석한 결과 B13-1 및 CC9-1 처리구의 사일리지가 pH가 낮았고 젖산함량도 높았다. 프리그 점수와 품질등급에 있어서도 B13-1 및 CC9-1 처리구에서 가장 높게 나타났다. 이상의 결과를 종합하여 볼 때 Lactobacillus plantarum B13-1 및 CC9-1는 사일리지 접종제로 사용이 권장되었다.

활성오니에서 분리한 pentachlorophenol 내성균주의 pentachlorophenol 제거에 관한 연구 (Removal of pentachlorophenol by pentachlorophenol resistant strains isolated from activated sludge)

  • 서울시 중량천 하수처리장의 활성오니로부터 총 20주의 pentachlorophenol(PCP) 내성균주들을 분리하여 B. sphaericus B-2와 B. schlegelii B-13를 포함한 Bacillus spp. 9주, Corynebacterium spp. 4주, Staphylococcus aureus S-1 1주, Aeromonas spp. A-19 1주, Arthrobacter LN-3 1주로 동정하였다. 이 균주들은 $200{\sim}300\;ppm$ PCP와 0.1% glucose가 포함된 mineral salt 배지에서 생육하였으며 PCP를 분해하는 종류 13주와 분해하지 못하고 흡착 혹은 흡수하는 종류 7주로 구분되었다. PCP 분해균주들은 대부분 배양 24시간후 생육이 최고에 도달하여 PCP를 약 $75{\sim}90%$ 분해하였으며 PCP 흡착 혹은 흡수하는 균주들은 흡착한 PCP를 배양 24시간 이후 또는 72시간 이후에 방출하였다. PCP 분해균주 B. sphaericus B-2, B. schlegelii B-13의 분해 산물을 TLC를 이용하여 검출한 결과 B. sphaericus B-2의 배양액에서 2-chlorophenol, B. schlegelii B-13에서는 2,4-dichlorophenol로 추정되는 물질과 4종의 분해산물로 보이는 물질들을 검출하였다.

Application of Collagenolytic Proteases from Bacillus subtilis B13 and Bacillus siamensis S6 for Tenderizing Goat Meat during Wet Aging

  • This research aimed to assess the effect of collagenolytic proteases from Bacillus subtilis B13 and Bacillus siamensis S6 for tenderizing goat meat during wet aging. Collagenolytic proteases B13 and S6 were prepared at 5 U/mL of collagenolytic activity before injecting into goat meat with 10% (v/w) of initial weight. The control sample was injected with distilled water and used as a negative control. The injected meats were placed in vacuum-sealed bags and wet aged at 4℃ for 0, 3, 5, 7, 14, and 21 days. Thereafter, total aerobic count and physicochemical quality were elucidated. Both enzyme-treated samples from B13 and S6 aged for 5 days showed an acceptable microbial quality with lower than 5.7 Log CFU/g. These conditions produced the tender meats by the reduction in shear force accounting for 30% for B13 and 26% for S6 as compared to the control. Moreover, the enzyme-treated samples showed lower values of hardness, gumminess, and chewiness, with higher springiness and trichloroacetic acid-soluble peptides than the control (p<0.05). The detrimental impact on cooking loss and lipid oxidation was not found. Enzyme-injected meat had a lower cooking loss than the control (p<0.05) with no significant difference in lipid oxidation (p>0.05). Notably, meats treated with B13 and S6 were lower in CIE L* value as compared to the control (p<0.05) with no significant impact on CIE a* and CIE b* (p>0.05). These results suggested that these two collagenolytic proteases could enhance the quality of goat meat in terms of tenderness and reduce the aging time for meat tenderization.

한국인에서 중합효소반응을 이용한 Short Tandem Repeat(STR)유전좌위 F13B분석 (Analysis of Short Tandem Repeat(STR) Locus F13B by Polymerase Chain Reaction in Korean)

  • In order to be utilized as a database in forensic identification and parentage test, allelic frequency and genotype distribution of short tandem repeat(STR) F13B locus was analysed by polymerase chain reaction in 210 Korean adults who are not related. The results were as follows. 1. 3 alleles and 56 genotypes of F13B locus were detected and heterozygosity value was 48.6% and allelic diversity value was 0.639 and the power of discrimination was 0.804. 2. The observed each alleles and allelic frequency was 8(0.069), 9(0.193), 10(0.738). In conclusion, the allelic frequency of STR F13B locus in the Korean is considered as an useful DNA allelic profile for forensic identification, but it should be used with several other STR locus to get definitive conclusion of analysis for individual identification and parentage testing.

  • In this paper, we investigated the recognition range according to modulation index for the ISO(International Standards Organization) 14443 13.56MHz contactless Type B RFID(Radio Frequency IDentification) system. We measured recognition range with changing modulation index step by step from 5% to 24% where we used 4 samples of Type B transponder with different resonance frequencies between 13.838MHz and 17.200MHz. While gradually increasing a distance in vertical direction from the center of the reader antenna, we measured the distance where the transponder's PUPI(Pseudo Unique PICC(Proximity IC Card) Identifier) is recognized continuously during 10 seconds and the distance where the transponder's PUPI is recognized at least two times during 5 seconds. From the measurement results, we found that the best recognition ranges were achieved when the reader had modulation index between 11% and 14%.

  • Effect of isothermal aging on the crystallization of $Fe_{78}B_{13}Si_{9}$ metallic glass has been investigated by electrical resistivity, X-ray measurements, bending test, thermal analysis and transmission electron microscopy. Amorphous $Fe_{78}B_{13}Si_{9}$ alloy was annealed isothermally for 5 to 1200 mon. between 300 C and 540 C. It has been found that close relation between relative resistivity and X-ray diffraction pattern showed. The crystalline peak of $\alpha$-(Fe, Si) and Fe$_2$B are detected by X-ray experiment. The crystalline phases observed by TEM show $\alpha$-(Fe, Si) and Fe$_3$B with dendritic and cylindrical morphology, respectively. It has been also found that the embrittleness of aged samples rapidly increased with the crystallization and was shown before the crystallization.th the crystallization and was shown before the crystallization.
