Determine the effects of drying temperature on the quality change and antioxidant activity characteristics of blueberry (건조 온도에 따른 블루베리의 품질변화 및 항산화특성)

  • This study was performed to investigate the effect of drying temperature on the quality characteristics and antioxidant activity of blueberry. Blueberries were dried at different times and temperatures, including A ($40^{\circ}C$, 72 hr), B ($40^{\circ}C$, 48 h and $50^{\circ}C$, 24 hr), C ($50^{\circ}C$, 72 hr), and D ($60^{\circ}C$, 72 hr). The yield and pH ranges of blueberry were determined to be 17.73~31.17% and 3.20~3.25, respectively. The yield rate of A treatments, soluble solid of D treatments, was the highest. The yield rate and soluble content was the highest in the treatment of A and D, respectively. The moisture content and water activity were significantly decreased with the increase in drying temperature (p<0.05). The L value of A treatment, and a and b values of D treatment were the greatest. In the analysis of texture analyzer, hardness, springiness, cohesiveness, gummiess and chewiness were significantly increased with the increase in drying temperature (p<0.05). In the sensory evaluation of blueberry, the appearance and color were the highest in the A treatment. The moisture, texture and taste was highest in the B treatment. The overall acceptability was in the order of B > A > C >D. The total polyphenol content and DPPH radical-scavenging activity were 9.21~13.05 mg/GAEg and 61.90~81.42%, respectively, which were significantly decreased with the increase in drying temperature (p<0.05). Therefore, the optimum time and temperature for blueberry drying was founded to be B treatment ($40^{\circ}C$, 48 hr and $50^{\circ}C$, 24 hr) among other treatments.

Effect of Commercial Antimicrobials in Combination with Heat Treatment on Inactivation of Bacillus cereus Spore

  • Thirteen commercial antimicrobial products were examined to assess the sporicidal activity against Bacillus cereus spores at room temperature, 60 and $85^{\circ}C$. Neither the antimicrobials showed detectable antimicrobial activity against the B. cereus spores nor induced spore germination after the treatment at 0.5 or 1.0%(w/v, v/v) commercial antimicrobial agents at room temperature for 0.5 to 4 hr. However, when the antimicrobials such as chitosan, lactic acid, fermented pollen, grapefruit extract were applied with heat at $85^{\circ}C$ for 30 min, more than 1 log CFU/mL spores were additionally inactivated compared to only heat treatment without antimicrobials. Imposition of $60^{\circ}C$ to B. cereus spores with the higher concentration of 5.0%(v/v) lactic acid or 2.5%(w/v) thiamine dilaurylsulfate for the longer time incubation of 24 hr resulted in 3 log CFU/mL spore inactivation. This work showed that low concentrations of commercial antimicrobials by themselves did not inactivate B. cereus spores. However, when physical processes such as heat were combined together, antimicrobials showed a synergistic effect against B. cereus spores.

Effects of Cyclic Nucleotides and Glipizide on the Cardiovascular Response of Baclofen in the Rats (흰쥐의 척수에서 Cyclic Nucleotides 및 Glipizide가 Baclofen의 심혈관반응에 미치는 영향)

  • The purpose of present study is to investigate the influence of a spinal gamma-aminobutyric acid B($GABA_B$) receptor on a central regulation of blood pressure(BP) and heart rate(HR), and to define its mechanism in the spinal cord. In urethane-anesthetized, d-tubocurarine-paralyzed and artificially ventilated male Sprague-Dawley rats, intrathecal administration of drugs were carried out using injection cannula(33-gauge stainless steel) through the guide cannula(PE 10) which was inserted intrathecally at lower thoracic level through the puncture of a atlantooccipital membrane. Intrathecal injection of an $GABA_B$ receptor agonist, baclofen(30, 60, 100 nmol) decreased both BP and HR dose-dependently. Pretreatment with 8-bromo-cAMP(50 nmol), a cAMP analog, or glipizide(50 nmol), a ATP-sensitive $K^+$ channel blocker, attenuated the depressor and bradycardic effects of baclofen(100 nmol), but not with 8-bromo-cGMP(50 nmol), a cGMP analog. These results suggest that the $GABA_B$ receptor in the spinal cord plays an inhibitory role in central cardiovascular regulation and that this depressor and bradycardic actions are mediated by the decrease of cAMP via the inhibition of adenylate cyclase and the opening of $K^+$ channel.

Effects of Ibuprofen on Bioavailability of Ciprofloxacin (이부프로펜이 씨프로플록사신의 생체이용률에 미치는 영향)

  • The effects of ibuprofen on bioavailability of ciprofloxacin were studied in rabbits. Animals were divided into three groups ; group A received 60 mg/kg of ciprofloxacin; group B and C received 60 mg/kg of ciprofloxacin with 60 and 240 mg/kg of ibuprofen, respectively. Ciprofloxacin and ibuprofen were given by single oral administration. Serum concentrations of ciprofloxacin were measured by high performance liquid chromatography with UV detector and pharmacokinetic parameters were calculated. Area under the serum concentrations versus time curve (${\pm}\;S.E.$) of ciprofloxacin were decreased in group B and C compared with group A ($12.26\;{\pm}\;0.94$ and $12.57\;{\pm}\;0.94$ vs. $15.71\;{\pm}\;1.06\;{\mu}g{\cdot}hr/ml$, p<0.05), whereas total clearances were increased ($1.81\;{\pm}\;0.13$ and $1.76\;{\pm}\;0.12$ vs. $1.40\;{\pm}\;0.09\;l/hr/kg$, p<0.05). No significant differances in these parameters were observed between group B and C. Relative bio-availability of group B and C to group A were 78 and 80%, respectively. These results suggest that the coadministration of ibuprofen with ciprofloxacin may reduce the bioavailability of ciprofloxacin.

  • The Photosynthetic bacterium, Rhodopseudomonas sphaeroides strain B6 was irnmobilized on agar gel. The optimum concentration of agar for hydrogen production was 2% (w/v). Maximum rates of hydrogen production by immobilized (300 ml of gel; 2.85 rng dry cells/ml) and free cells (1l culture; 0.87 mg dry cells/ml) were 47.5 and 48.0 ml/hr/culture, respectively. However, when both cultures were fed by 10 mmoles of lactate as limited electron donor at the later period of incubation, the activity of hydrogen production by free cells was significantly decreased but, immobilized cells continued hydrogen production with almost the same initial rate. Wc examined hydrogen production by immobilized cells of strain B6 under periodic illumination for 12 hr-intervals. When the culture was periodically fed by basal medium containing 9.3 rnmoles of DL-lactate and 1.86 mmoles of L-glutamate as consumed electron donor and nitrogen source, respectively, for every one liter of hydrogen produced, hydrogen was evolved continuously with the average rate of 510 ml/day/300 ml gel (2.9 rng dry cellslml) during the incubation time for 228 hr.

Domestic Ozone Sensitivity to Chinese Emissions Inventories: A Comparison between MICS-Asia 2010 and INTEX-B 2006 (중국 배출량 목록에 대한 국내 오존 민감도 분석: MICS-Asia 2010와 INTEX-B 2006 비교사례)

  • CMAQ (Community Multiscale Air Quality)-HDDM (High-order Direct Decoupled Method) simulations with MICS-Asia 2010 and INTEX-B 2006 emissions inventories were performed to investigate the impact of Chinese $NO_x$ and VOC emissions on 1-hr ozone concentrations over South Korea during May to July in 2014. Chinese $NO_x$ and VOC emissions in MICS-Asia 2010 were 60% higher and 100% lower than those in INTEX-B 2006 during the simulation period. It makes the ratio of Chinese VOC to $NO_x$ emissions in INTEX-B 2006 (Case 1) is 3.2 times higher than that in MICS-Asia 2010 (Case 2). When the observed period mean 1-hr ozone concentration averaged across 106 air monitoring sites in the SMA (Seoul Metropolitan Area) was 37.6 ppb, the modeled values were similar to each other; 37.3 ppb for Case 1, and 40.4 ppb for Case 2. Both cases show that daily maximum 1-hr ZOC (Zero-Out Contribution) of Chinese $NO_x$ and VOC emissions were as high as 55 ppb and 35 ppb for the episode respectively. Correlation coefficients between ZOC of Chinese $NO_x$ and VOC emissions and the SMA daily maximum 1-hr ozone were 0.49~0.69. It indicates that Chinese emissions occasionally affect the SMA daily ozone peaks. On the other hand, Case 2 predicted 7 ppb and 1 ppb higher ZOC of Chinese $NO_x$ and VOC emissions than Case 1, when simulated ozone in the SMA is over 80 ppb. It implies that upwind $NO_x$ emissions would be more important than upwind VOC emissions for the long-range transport of ozone in Northeast Asia.

Characterization of the Strong Proteolytic Bacteria Isolated from Low Salt Fermented Anchovy and of Protease Produced by that Strain (저식염멸치젓에서 분리한 단백질분해력이 강한 세균 및 생산된 단백분해효소의 특성)

  • For the purpose of producing low salt fermented anchovy by accelerated method with a strong proteolytic bacteria, in this study, a strong proteolytic bacterium was isolated from low salt fermented anchovy and its bacteriological characteristics and properties of protease were experimented. The results obtained were as fellows : three proteolytic bacteria, Aeromonas anaerogenes Barillus subtilis and Staphylococcus saprophyticus were isolated from low salt fermented anchovy($4\%\;of\;salt,\;4\%\;of\;KCl,\;0.5\%\;of\;lactic\;acid,\;6\%$of sorbitol and $4\%$ of alcohol extract of red pepper) after 40 days fermentation. Among these strains, which grow best at $30^{\circ}C$, pH 7.0, B. subtilis was found the best proteolytic strain and benefit for industrial use as shown $0.95\;hr^{-1}$ of specific growth rate, $89{\mu}g-Tyr/hr.ml$ of maximum activity after 12 hrs culture in TPY broth. The protease produced by by B. subtilis showed maximum activity at $35^{\circ}C$, pH 7.0, and molecular weight was estimated to be 23,000 by Sephadex G-100 filtration, and it was supposed to be a kind of metal chelator sensitive neutral protease from the results of strong sensitivity against EDTA, o-phenanthroline and metal ions such as $Cu^{2+},\;Ni^{2+},\;Fe^{2+}.Km$ value of that by method of Lineweaver-Burk was determinded to be $0.73\%$ for casein as a substrate.

Growth of an Obligatory Methanotroph Methylosinus trichosporium OB3b on Methanol (메탄자화균 Methylosinus trichosporium OB3b의 메탄올 기질에서의 성장)

  • An obligatory type II methanotroph Methylosinus trichosporium OB3b was cultivated on methanol as a sole carbon and energy source. The effects of methanol concentration, pH, temperature, nitrogen source and phosphate concentration on cell growth were investigated and the results were compared with the growth on methane, which had been studied previously. When $(NH_4)_2SO_4$ was used as a nitrogen source, the maximal specific growth rate (${\mu}max$) on methanol was $0.20hr^{-1}$ and the carbon conversion efficiency(CCE) was 43%. In comparison, on methane, ${\mu}max$ and CCE were $0.08hr^{-1}$ and 32%, respectively. Ammonia was found to be a better nitrogen source for methanol-growing cells. Cell yield on nitrogen (YX/N) was the same regardless of nitrogen source as 7.14g dry cells/g N, but the yield on methanol(YX/N) was higher with ammonia(0.8g dry cells/g MeOH) than with nitrate(0.64g dry cells/g MeOH). Optimal pH and temperature were 7.0 and $30^{\circ}C$, respectively. Methanol inhibition on cell growth was observed at above 0.5%(v/v). Inhibition by phosphate was observed at above 60mM, although the inhibition on methanol dehydrogenase activity started at a much lower level of 20mM. Based on the experimental findings, the cellular physiology of M. trichosporium OB3b growing on the two closely-related carbon sources were discussed extensively.

  • This research was carried out to offer the basis data for development of optimum drying schedule for a domestic oak species (Quercus grosseserrata B1.) by investigating drying rate. stress, defect, and moisuture gradient with board thicknesses and drying schedules (code number T4-C2 and T3-B1). The results were obtained as follows: 1. Average drying rate and total drying time from 52.2% to 5.8% were 0.105%/hr and 486 hours for drying schedule T4-C2 and those from 62.1% to 8.3% were 0.070%/hr. and 811 hours for drying schedule T3-B1. 2. Drying rates for 28mm- and 31mm-thick boards showed similar tendency, but were significantly different from 25mm- thick board in drying schedule T4-C2 and those for 22mm-, 25mm- 28mm- and 31mm-thick boards showed similar tendency but were significantly different from 19mm- thick boards in drying schedule T3-B1. 3. The moisture gradients for drying schedule T4-C2 were steeper than those for drying schedule T3-B1 during drying period. and especially in early drying stage slow slope of moisture gradients of drying schedule T3-B1 was effective in preventing serious problem of surface checks. 4. Drying stresses were lower in drying schedule T3-B1 than in drying schedule T4-C2 during drying period. 5. Drying schedule T4-C2 was appropriate for 25mm-thick board but not for 28mm- and 31mm-thick board because of strong drying condition. Drying schedule T3-B1 was appropriate for 28mm- and 31 mm-thick board but not for 19mm-, 22mm-, and 25mm-thick board because of weak drying condition.

Characterization of ${\gamma}$-Polyglutamic Acid Produced from the Solid-state Fermentation of Soybean Milk Cake Using Bacillus sp.

  • In this study, we optimized the production of ${\gamma}-polyglutamic$ acid (PGA) in soybean milk cakes (SMC) fermented with Bacillus subtilis GT-D and B. subtilis KU-A, to be utilized as a functional food ingredient. PGA production was dependent upon the glutamate content, fermentation time, and type of Bacillus sp. The consistencies of the SMCs fermented by B. subtilis GT-D and B. subtilis KU-A were highest after 36 hr of fermentation, and then decreased gradually. The SMC fermented by B. subtilis KU-A had a higher consistency than the SMC fermented by B. subtilis GT-D. In the presence of 10% defatted soy flour (DFS), 5% glutamate in the SMC was efficiently converted into polyglutamic acid (PGA) for 24 hr, indicating a conversion yield above 96%, but its conversion then decreased with higher concentrations of glutamate. The soluble solid content (mucilage) of the SMC fermented with B. subtilis KU-A was 9.5%(w/w), and composed of 65.6% PGA (Mw 1,536 kDa) and some polysaccharides. However, the SMC fermented with B. subtilis GT-D had a mucilage content of 7.8%(w/w), and was composed of 66.4% PGA (Mw 1,409 kDa), 11.5% levan, and some polysaccharides. The viscoelastic values of the mucilage obtained using B. subtilis KU-A were much higher than those of mucilage obtained using B. subtilis GT-D. Also, the G'-value (elastic modulus) was higher than the G"-value (viscous modulus).
