Inhibition of mouse SP2/0 myeloma cell growth by the B7-H4 protein vaccine

  • B7-H4 is a member of B7 family of co-inhibitory molecules and B7-H4 protein is found to be overexpressed in many human cancers and which is usually associated with poor survival. In this study, we developed a therapeutic vaccine made from a fusion protein composed of a tetanus toxoid (TT) T-helper cell epitope and human B7-H4IgV domain (TT-rhB7-H4IgV). We investigated the anti-tumor effect of the TT-rhB7-H4IgV vaccine in BALB/c mice and SP2/0 myeloma growth was significantly suppressed in mice. The TT-rhB7-H4IgV vaccine induced high-titer specific antibodies in mice. Further, the antibodies induced by TT-rhB7-H4IgV vaccine were capable of depleting SP2/0 cells through complement-dependent cytotoxicity (CDC) in vitro. On the other hand, the poor cellular immune response was irrelevant to the therapeutic efficacy. These results indicate that the recombinant TT-rhB7-H4IgV vaccine might be a useful candidate of immunotherapy for the treatment of some tumors associated with abnormal expression of B7-H4.

Expression of Escherichia coli Heat-labile Enterotoxin B Subunit (LTB) in Saccharomyces cerevisiae

  • Heat-labile enterotoxin B subunit (LTB) of enterotoxigenic Escherichia coli (ETEC) is both a strong mucosal adjuvant and immunogen. It is a subunit vaccine candidate to be used against ETEC-induced diarrhea. It has already been expressed in several bacterial and plant systems. In order to construct yeast expressing vector for the LTB protein, the eltB gene encoding LTB was amplified from a human origin enterotoxigenic E. coli DNA by PCR. The expression plasmid pLTB83 was constructed by inserting the eltB gene into the pYES2 shuttle vector immediately downstream of the GAL1 promoter. The recombinant vector was transformed into S. cerevisiae and was then induced by galactose. The LTB protein was detected in the total soluble protein of the yeast by SDS-PAGE analysis. Quantitative ELISA showed that the maximum amount of LTB protein expressed in the yeast was approximately $1.9\%$ of the total soluble protein. Immunoblotting analysis showed the yeast-derived LTB protein was antigenically indistinguishable from bacterial LTB protein. Since the whole-recombinant yeast has been introduced as a new vaccine formulation the expression of LTB in S. cerevisiae can offer an inexpensive yet effective strategy to protect against ETEC, especially in developing countries where it is needed most.

Effect of Nigella sativa Oil on Melanogenesis (니겔라 사티바 오일의 미백 효능에 관한 연구)

  • In order to investigate the potential of Nigella sativa (N. sativa) oil as an active ingredient for whitening cosmetics, we prepared N. sativa oil. We measured its inhibitory effects on mushroom tyrosinase activity, cellular tyrosinase activity, and melanin synthesis inhibitory activity in B16 melanoma cells. N. sativa oil and its components showed inhibitory activity against mushroom tyrosinase and melanin synthesis. In a melanin synthesis inhibition assay using mouse B16-F10 melanoma cell, it reduced melanin production up to 86 % at a concentration of 10 mg/mL without cytotoxicity. In the study on the melanogenic protein expressions by using RT-PCR and Western blot, N. sativa oil and its components inhibited expression of tyrosinase protein, which is a well-known key protein on melanogenesis, and tyrosinase expression was gradually decreased in a dose-dependent manner. Therefore, this result suggests that N. sativa oil could be used as an active ingredient for whitening cosmetics.

Physico-chemical Properties of Soybean Curd Whey Concentrated by Reverse Osmosis (역삼투법으로 분리, 농축한 두부순물의 이화학적 특성)

  • Several Physico-chemical properties of the retentate obtained from reverse osmosis of soybean curd whey were studied. The contents of sucrose, raffinose and stachyose in the retentate were 32.59%d.b., 4.76%d.b. and 9.99%d.b., respectively. Potassium (5.23%d.b.), in the retentate was a dominant element in ash. Protein content (18.69%d.b.), amino acid composition and subunit pattern of protein in PAG-electrophoresis were somewhat different from those of the soybean protein. Emulsification activity, emulsification stability and viscosity of protein isolated from soybean curd whey (WPI) were slightly inferior compared to protein isolated from soybean (SPI), with the exception of solubility that depended on the pH of WPI.

Effect of Batryticatus Bombycis Extract on Apoptosis in B16F10 cells (백강잠 추출액의 피부암 세포주에서 세포사멸 유도 작용에 대한 연구)

  • This study was performed to investigate the anti cancer effect of Batryticatus Bombycis extract(BBE) in B16F10 cells. The cell viability after BBE treatment was quantified by MTT assay. The results showed that BBE inhibited the proliferation of B16F10 cells and caused a 80% inhibition of B16F10 cells at concentration of $500\;{\mu}g/ml$. B16F10 cells exposed to BBE displayed the DNA fragmentation ladder and nucleus chromatin condensation characteristic for apoptosis. The enzyme activity of caspase-3 and actived caspase-3 protein was markedly increased in B16F10 cells treated with the BBE. The expression of Bcl-2, anti-apoptotic protein, was decreased by treatment of the BBE in a dose-dependent manner. And the expression of pro-apoptotic Bax protein was increased. In conclusion, we can suggest that BBE induce the apoptotic death of B16F10 cells via activation of caspase-3, cleavage of PARP protein and Bcl-2 degradation.

Clinical Significance of SH2B1 Adaptor Protein Expression in Non-small Cell Lung Cancer

  • The SH2B1 adaptor protein is recruited to multiple ligand-activated receptor tyrosine kinases that play important role in the physiologic and pathologic features of many cancers. The purpose of this study was to assess SH2B1 expression and to explore its contribution to the non-small cell lung cancer (NSCLC). Methods: SH2B1 expression in 114 primary NSCLC tissue specimens was analyzed by immunohistochemistry and correlated with clinicopathological parameters and patients' outcome. Additionally, 15 paired NSCLC background tissues, 5 NSCLC cell lines and a normal HBE cell line were evaluated for SH2B1 expression by RT-PCR and immunoblotting, immunofluorescence being applied for the cell lines. Results: SH2B1 was found to be overexpressed in NSCLC tissues and NSCLC cell lines. More importantly, high SH2B1 expression was significantly associated with tumor grade, tumor size, clinical stage, lymph node metastasis, and recurrence respectively. Survival analysis demonstrated that patients with high SH2B1 expression had both poorer disease-free survival and overall survival than other patients. Multivariate Cox regression analysis revealed that SH2B1 overexpression was an independent prognostic factor for patients with NSCLC. Conclusions: Our findings suggest that the SH2B1 protein may contribute to the malignant progression of NSCLC and could offer a novel prognostic indicator for patients with NSCLC.

Expression of Heregulin and ErbB Family Proteins in Gastric Adenocarcinomas: Correlation with Clinopathologic Prognostic Factors (위선암에서 Heregulin과 ErbB Family 단백 발현과 임상.병리학적 예후인자와의 상관관계)

  • Purpose: Heregulin is a natural ligand for erbB3 and erbB4. However, very little is known about their roles in the gastric cancer This retrospective study was performed to evaluate the frequencies of heregulin and erbB family protein expression and to compare their expressions with clinicopathologic parameters. Materials and Methods: Immunohistochemical expressions of heregulin and erbB family proteins were examined with tissue micro-array slides. A total of 251 gastric adenocarcinomas were classified as early cancers and advanced cancers and as having and not having lymph node metastases. Results: The positive rates of the heregulin, erbB1, erbB2, erbB3, and erbB4 protein stainings were 64%, 68%, 6%, 88%, and 76%, respectively. Intestinal type gastric adenocarcinomas showed higher expression of heregulin, erbB2, erbB3, and erbB4 proteins. Heregulin and erbB4 proteins showed lower expressions in advanced gastric carcinomas. However, erbB2 protein showed higher expression in advanced gastric carcinomas. The protein expressions of heregulin and erbB family proteins showed no relationship with survival rate. Co-expression groups of heregulin and erbB3 proteins or heregulin and erbB4 proteins showed higher expressions in intestinal type adenocarcinomas and early gastric carcinomas. Conclusion: Heregulin, erbB3, and erbB4 proteins may play a role in the early stage of adenocarcinomas.

Determination of growth performance of crossbred Korean native chickens for twelve weeks after hatching

  • The study was conducted to evaluate the growth performance, viability, and uniformity of female crossbred Korean native chickens (KNC) for the first twelve weeks after hatching. Six crossbreed lines of chickens (1B, 2B, 3B, 4B, 5B, and 6B) bred from 1 paternal line and 6 maternal lines of KNC were compared in this study. Sixty newly hatched 1-day old chicks from each crossbreed, for a total of 360 chickens, were allocated into 6 replicates (10 birds per replicate) in a complete randomized design. Experimental diets used in this study consisted of 3 phases: 1) week 0 to 5, crude protein 20.3% and metabolizable energy 3,059 kcal/kg; 2) week 5 to 8, crude protein 18.6% and metabolizable energy 3,123 kcal/kg; and 3) week 8 to 12, crude protein 16.7% and metabolizable energy 3,187 kcal/kg. Body weight and feed intake were measured bi-weekly during the experiment. The 4B crossbreed had a higher body weight, average daily gain, and average daily feed intake on week 12 (p < 0.05) than the other groups. The 4B crossbreed had a lower (p < 0.05) uniformity during weeks 10 to 12 than crossbreeds 1B and 5B. However, a lower viability was observed (p > 0.05) in 4B in weeks 2 to 12 than those in the other groups. Results of the current study indicate that the 4B crossbreed had a higher growth performance in week 12 but had a lower viability for weeks 2 to 12 than the other crossbreeds.

Salvianolic Acid B Inhibits Hand-Foot-Mouth Disease Enterovirus 71 Replication through Enhancement of AKT Signaling Pathway

  • Hand, foot, and mouth disease (HFMD) is caused by enterovirus 71 (EV71) in infants and children under six years of age. HFMD is characterized by fever, mouth ulcers, and vesicular rashes on the palms and feet. EV71 also causes severe neurological manifestations, such as brainstem encephalitis and aseptic meningitis. Recently, frequent outbreaks of EV71 have occurred in the Asia-Pacific region, but currently, no effective antiviral drugs have been developed to treat the disease. In this study, we investigated the antiviral effect of salvianolic acid B (SalB) on EV71. SalB is a major component of the Salvia miltiorrhiza root and has been shown to be an effective treatment for subarachnoid hemorrhages and myocardial infarctions. HeLa cells were cultured in 12-well plates and treated with SalB (100 or 10 ㎍/ml) and 106 PFU/ml of EV71. SalB treatment (100 ㎍/ml) significantly decreased the cleavage of the eukaryotic eIF4G1 protein and reduced the expression of the EV71 capsid protein VP1. In addition, SalB treatment showed a dramatic decrease in viral infection, measured by immunofluorescence staining. The Akt signaling pathway, a key component of cell survival and proliferation, was significantly increased in EV71-infected HeLa cells treated with 100 ㎍/ml SalB. RT-PCR results showed that the mRNA for anti-apoptotic protein Bcl-2 and the cell cycle regulator Cyclin-D1 were significantly increased by SalB treatment. These results indicate that SalB activates Akt/PKB signaling and inhibits apoptosis in infected HeLa cells. Taken together, these results suggest that SalB could be used to develop a new therapeutic drug for EV71-induced HFMD.

Simple Sequence Repeat Markers Linked to Quantitative Trait Loci Controlling Seed Weight, Protein and Oil Contents in Soybean (콩에서 종실의 무게와 oil 및 단백질 함량을 조절하는 양적 형질 유전자좌와 연관된 simple sequence repeat marker)

  • Soybean [Glycine max (L.) Merr.] is an important crop, accounting for 48% of the world market in oil crops. Improvement of the quality and quantity of soybean seed constituents is one of the most important objectives in soybean breeding. Protein content and seed size are important properties to determine the quality of tofu and soy sprouts respectively. The objective of this study was to identify quantitative trait loci (QTLs) that control seed weight, protein and oil content in soybean. The 117 $F_{2:10}$ recombinant inbred lines (RlL) developed from a cross of 'Keunolkong' and 'Shinpaldalkong' were used. Narrow-sense heritability estimates based on a plot mean on seed weight, protein and oil content were 0.8, 0.78 and 0.71, respectively. Four independent QTLs for seed weight were identified from linkage group (LG) F, I and K. Five QTL for protein content were located on LG D1b, E, H, I and L. Oil content was related with six QTLs located on LG D1b, E, G, I, J and N. Protein and oil content have three common QTLs on LG D1b, E and I. Thus, we identified major loci improving soybean seed quality.
