Glutathione Content in Various Seedling Plants, Vegetables, and the Processed Foods (각종 유식물체, 채소 및 가공식품 중의 글루타치온 함량)

  • In this study, we investigated the levels of glutathione (GSH) and its oxidized form (GSSG) in more than 40 kinds of plant materials including seedling plants, grains, vegetables, and processed foods. The glutathione contents in the seedling plants were ranged from 0 to $120{\mu}mol/100g$. In addition, the different levels of glutathione were observed within the same family and between species. In the case of marketed grains and vegetables, azuki and kidney beans of leguminosae contained the high levels of glutathione, whereas glutathione was scarcely detected in the processed bean foods (bean paste, soybean sauce, etc.). Overall, a higher GSH content in food may contribute to a higher added value.

Pathogenicity of infectious in vitro transcripts and comparison of RNA3 of Alfalfa mosaic virus Korean isolates

  • Two Korean isolates of Alfalfa mosaic virus (AHV-AZ, AMV-KR) were isolated from azuki bean and potato plants, respectively, and their pathologies were confirmed on some susceptible host plants including pepper, tobacco and red bean plants. Full length cDNAs to RNA1, RNA2 and RNA3 of the two Korean strains were amplified using the long-template reverse transcription (RT)-polymerase chain reaction (PCR) method. RT-PCR products covering entire regions for the three AMV genome RNAs were cloned. RNA transcripts were synthesized in vitro from each clones using T7 RNA polymerase and infectivity test was peformed in 9 reassortment sets of transcripts. All the combinations of reassorted transcripts were found to be infectious when inoculated onto Nicotiana benthamiana plants, and were not distinguishable to those of wild types. The full-length cDNA clones that were confirmed infectious were sequenced their nucleotide sequences. We will discuss sequence analysis of the two Korean isolates of AMV genomic RNA3 and compare reported foreign isolates of AMV.

Development of a Multiplex Reverse Transcription-Polymerase Chain Reaction Assay for the Simultaneous Detection of Three Viruses in Leguminous Plants

  • A multiplex reverse transcription-polymerase chain reaction (mRT-PCR) assay was developed for the detection of Clover yellow vein virus (ClYVV), Peanut mottle virus (PeMoV), and Tomato spotted wilt virus (TSWV), which were recently reported to infect soybean and azuki bean in Korea. Species-specific primer sets were designed for the detection of each virus, and their specificity and sensitivity were tested using mixed primer sets. From among the designed primer sets, two combinations were selected and further evaluated to estimate the detection limits of uniplex, duplex, and multiplex RT-PCR. The multiplex RT-PCR assay could be a useful tool for the field survey of plant viruses and the rapid detection of ClYVV, PeMoV, and TSWV in leguminous plants.

A PCR Method for Rapid Detection of Peanut Ingredients in Food (식품에서 땅콩 성분의 신속검출을 위한 PCR 방법)

  • Peanut (Arachis hypogaea) often causes severe allergic reactions in sensitive people. Agglutinin is known to be one of the allergenic proteins in peanut. A polymerase chain reaction (PCR) method was developed to detect peanut ingredients in food using a primer pair corresponding to the agglutinin gene. This primer pair enabled PCR amplification of specific regions of agglutinin DNA from peanut, but not from 11 other nuts, beans, and cereals (pistachio, almond, sunflower seed, pine nut, walnut, soybean, black bean, kidney bean, azuki bean, rice, and black rice). The proposed PCR method successfully identified all of the 6 processed foods containing peanut whereas 13 other processed foods, which don't declare peanuts as an ingredient, were all negative. The detection limit of this method for purified peanut DNA was 100 pg/reaction. The sensitivity of this method was sufficient to detect peanut DNA in soybean DNA mixture which had been spiked with 0.1% peanut DNA.

Effects Of Phosphate Application Rate on the Growth Characteristics, Yield and Feed Value of Whole Crop Azuki bean in Jeju Island (제주지역에서 인산시비량 차이에 따른 청예팥의 생육반응, 수량 및 사료가치 변화)

  • This study was determined to the growth characteristic, yield and chemical content of Azuki bean(Vigna angularis W.F. $W_{IGHT}$) based on five phosphate rates (0, 40, 50, 120, 160, 200kg/ha) from May, 2002 to August, 2002 in Jeju province. Plant height was 80.7cm at 0kg/ha of phosphate level md, as phosphate rates were increased to 160kg/ha and 200kg/ha, lengthened 88.8cm and 88.9cm, respectively, but between the two phosphat levels were no significance(P>0.05 or 0.0l). Numbs. of branches and leaves per plant, stem diameter, and weight of leaves and stems per plant were the same response with plant height. Fresh forage, dry matter, crude protein, and TDN(total digestible nutrient) yield at the control were 23.8MT/ha, 3MT/ha, 0.5MT/ha and 1.8MT/ha, respectively, at the 200kg/ha plot were 47.3MT/ha, 7.2MT/ha, 1.3MT/ha, and 4.7MT/ha, respectively, as phosphate rate was increased. As phosphate rate increased from 0kg/ha to 200kg/ha, the content of crude protein, crude fat NFE(nitrogen free extract) and TDN increased 15.2%~18.6%, 3.4%~4.5%, 41.4%~45.5% and 58.3%~65.5%, respectively, whereas the content of crude fiber and crude ash were decreased 31.5%~24.8% and 8.1%~6.6%, respectively.

Effect of Seeding Dates on the Growth Characteristics, Yield and Feed Value of Whole Crop Azuki bean in Jeju Island (제주지역에서 파종기에 따른 청예팥의 생육반응.수량 및 사료가치 변화)

  • This study was conducted to determine the influence of seeding date (May 3, 13, 23, June 2, 12) on the growth characteristics yield and feed value Azuki bean(Vigna angularis W.F. Wight) from May 3, 2002 to August 3, 2002 in Jeju province. Plant height was the longest (84cm) when seeded on 13 May and was the shortest (48.4cm) when seeded on 12 June. Numbers of branches and leaves per plant, stem diameter and weight of leaves and stems per plant were the same trend with plant height. Fresh forage, fry matter, crude protein and TDN(total digestible nutrient) yield per ha at were increased to 54.6MT, 7.7MT, 1.4MT and 4.5MT, respectively, whereas decreased as seeding date was delayed and then at seeding on 12 June were decreased to 11.5MT/ha 2.12MT/ha 0.4MT/ha and 1.3MT/ha, respectively The content of crude protein, crude fat, NFE(nitrogen free extract) and TDN were incresed 17.6%∼21.3%, 3.3%∼4.5%, 34.3%∼41.4% and 56%∼64.8%, respectively, as seeding was delayed from 3 May to 13 June. Whereas the content of crude fiber and crede ash were decreased 33.8%∼23.5% and 11%∼9.3%, respectively.

Effects of Aluminium on Growth, Chlorophyll Content, ALAD Activity and Anatomy of Root rind Shoot in Azuki Bean (Vigna angularis) Seedlings (Aluminium이 팥(Vigna angularis) 유식물의 생장, 엽록소함량, ALAD활성 및 뿌리와 경엽부의 형태에 미치는 영향)

  • The toxic effects of aluminium (Al) on growth, chlorophyll content, $\delta-aminolevulinic$ acid dehydratase (ALAD) activity and anatomy of root and shoot were investigated in 7-day-old azuki bean (Vigna angularis) seedlings. Significant depressions in root elongation was observed in the low concentrations of Al (50, 100 $\muM)$ and increasing Al concentrations caused a sharp decline of root and shoot growth. The degree of inhibition was dependent upon Al supply. Exposure to 50 $\muM$ Al or more inhibited root elongation within 1 day. In the 50 $\muM$ Al treatments, a recovery of root growth was seen after 7 days exposure. In contrast, lateral root initials was little affected by Al exposure. Al toxicity symptoms and growth responses were more well developed in the roots than in the shoots. Analysis of Al localization in root cells by hematoxylin stAlning showed that Al entered root apices and accumulated in the epidermal and cortical cells immeadiately below the epidermis. There was a good positive correlation between the level of chlorophyll and ALAD activity. Increasing Al concentrations caused a decrease in total chlorophyll contents, accompanied by proportional changes in ALAD activity, suggesting a cootr-dinated reduction of a photosynthetic machinery. Al exerted specific influence on the morphology of root ann shoot. At higher concentrations of Al the roots induced drastic anatomical changes. The epidermal cells were disorganized or destructed while the cortical cells exhibited distortion of cell shape and/or disintegration. The diameter of root and transectional area of cortical cells decreased considerably with Al treatment. In the shoot Al also enhanced reduction of diameter of shoot and cell size. Gross anatomy of leaves treated with Al did not differ significantly from the controls, except for fewer and smaller chloroplast. Our results indicate that toxic effect of Al appear to be manifested primarily in roots and secondarily on shoots, and changes in root morphology are related to changes in the root growth patterns. Results are further discussed in re181ion to the findings in other plant species, and it is concluded that Al causes morphological, structural and, presumably, functional damage to the roots of the species investigated.

  • This experiment was conducted to verify the responses of major crop species to saline stress.To determine the saline tolerance of crop species, sevral crop species were cultivated under sand-culture system using Hoagland's nutrient solution whit 200mM NaCl. The crop species showing saline tolerance were cotton(Gossypium indicum), maize(Zea mays), barely(Hordeum vulgare), and wheat(Triticum aestivum) but perilla (Perilla frutescens) and leguminous crops, mung bean(Phaseolus radiatus), azuki bean(Phaseolus angularis) and soy bean(Glycin max) showed very por tolerance. One the typical symptom was the darkening of leaf color due to increase of chlorophyll concentration.Among of the plant families, Fabaceae was the most susceptible but crop species belonging to Poaceae were more proper for cultivating on reclaimed tidal land in the course of desalinazation. It was suggrsted that the crop species belonging to Fabaceae, a sensitive family to soil salinity, must be cultivated when the soil salinity decreased below 10ds/m. To know the critical salinity level for crop growth,salinity of saline soil collected from reclaimed tidal land was adjusted from 10.0ds/m tp41.7ds/m with tap water. It was suggested that the ECs of the soil in which the plant height of each crop spicies was reduced to 50% of control plant were 22.6 and 21.7 ds/m in rice, barley, corn, mung bean, and soy bean,respectively.

Desmutagenic Effect of Legumes and Plants Crude Saponins in Salmonella typhimurium TA 98 (콩과 식물에서 추출한 사포닌의 돌연변이원성 억제효과)

  • Crude saponins of soybean and Azuki bean (2.0 mg/plate)were most effective against Trp-p-2, and also all of legume saponins (2.0 mg/plate)were excellent effective against aflatoxin $B_1$. Crude saponins of taro, burdock and ginseng were remarkably effective at ranging from 1.0 mg to 2.0 mg per plate against MeIQ. Especially ginseng saponin was excellent effective and arrow root saponins was remarkably effective against MeIQ, respectively. Plant saponins of taro, lotus burdock, arrow root and ginseng except for dodok were most effective activities against aflatoxin $B_1$.
