Antiradical activity of Azadirachta indica extracts and fractions

  • In present study, total phenolic content of crude extract and antiradical activity of crude extract and different solvent fractions of Azadirachta indica were evaluated. Crude extract and most of the polar fractions showed higher radical scavenging activity. Among the crude extract and nine different fractions, the aqueous/methanol (3:1) fraction showed the highest activity.

  • Potintiodynamic 분광작용과 임피던스 기술을 이용하여 요드화 이온으로 Azadirachta Indica 엑스가 존재하는 0.5 M HCI용액 속의 Al 에 대한 부식 방지 작용을 상승시켰다. 이는 0.5 M HCI 용액에서 AZI 엑스가 Al의 부식을 방지함을 발견하였다. AZI 엑스 농도가 24% v/v이전에는 농도가 커짐에 따라 방지효율도 따라서 커진다. 하지만 AZI 엑스 농도가 더 커지면 Inh. %는 오히려 작아진다. 요드화 이온의 증가는 방지효율을 상당한 양으로 강화시킨다. 요드화 이온이 혼합적으로 존재할 때 Inh. %값은 증가한다. 이는 낮은 AZI 엑스 농도에서 AZI 엑스는 joint adsorption을 통하여 insoluble complex을 형성한다. 하지만 높은 AZI 엑스 농도에서는 요드화 이온과 형성한 complex사이에 competitive adsorption이 나타나 낮은 Inh. %를 초래한다. At all studied요드화 이온의 농도에서 요드화 이온과 AZI 엑스가 동시에 존재할 때의 Inh. %값은 AZI 엑스만 존재할 때보다 감소된다. Synergism parameter Sq는 표면 Coverage 값으로 계산된다. 이 Parameter는 AZI 엑스경우 unity 보다 더 큼을 알 수 있다. 이는 요드화 이온의 증가로 인하여 방지효율이 강화 되였음을 제시한다.

  • It was confirmed that extracts of some plants were used in folklore medicine as hypoglycemic agents. Of these plants is Azadirachta indica ("neem") which grows in tropical regions. The present study deals with biochemical effects of the "neem" leaf water extract given orally to experimental aminals, especially the hypoglycemic characteristics. Normals as well as alloxan diabetic rats have been used in this work. The results showed that the "neem" leaf extract produced some hypoglycemia in normal rats when given in two doses, while in diabetic rats there was a decrease in blood sugar, but it could not alleviate the diabetic in body weight loss and high percentage mortality, especially with a high dose. It was observed increased with concomitant decrease in the liver fat as compared to normal levels. There was also a drop in liver proteins which was dose-related. The results were compared wiht those obtained with an oral hypoglycemic drug (Glibenclamide).glycemic drug (Glibenclamide).

Effect of Azadirachta indica flower extract on basal and experimentally elevated blood glucose in rats

  • Azadirachta indica A. Juss (N.O. Meliaceae), popularly known, as 'Neem' is an indigenous tree widely available in India. Almost every part of the tree has long been used in Unani system of medicine for the treatment of a variety of human ailments. The flowers have been mentioned as a remedy useful in controlling diabetes mellitus. The present study had been designed to investigate the hypoglycemic/anti-hyperglycemic effects of the methanolic extract of the flowers of A. indica (Gule-Neem) and its different fractions on normal, glucose fed hyperglycemic, adrenaline induced hyperglycemic and alloxan induced diabetic rats. The methanolic extract was resolved into water soluble and water insoluble fractions. Water soluble portion of the methanolic extract was found to possess significant blood sugar lowering effect in glucose-fed and adrenaline-induced hyperglycemic rats but it did not show such effect in normal and alloxan induced mild and severe diabetic rats. Water-soluble portion was fractionated by employing the polarity criterion with ethyl acetate and butanol. The ethyl acetate fraction was further fractionated into phenolic and non-phenolic fractions. Hypoglycemic effect of these fractions was also evaluated. The results suggest that the flowers of A. indica contain at least two different constituents, responsible for the said activity. These investigations validate the use of flowers of A. indica in diabetes by Unani physicians.

  • Natural goods, especially therapeutic plants, are abundant in the World. Because they have the ability to provide all humanity with countless advantages as a source of medicines, medicinal plants are presently receiving more attention than ever. These plants' therapeutic efficacy is based on bioactive phytochemical components that have clear physiological effects on the human body. The drying process is crucial for the preparation of plant materials prior to extraction since freshly harvested plant materials include active enzymes that create active components, intermediates, and metabolic processes. Many of the phytoconstituents may be extracted using the semi-polar solvent methanol. The goal of the current work is to use the GC-MS gas chromatography- mass spectrometry technology to identify the phytochemicals and review their biological activity. In methanol leaf extract, 5 phytocompounds were found in Ricinus communis, 5 phytocompounds in Pongamia pinnata, 12 phytocompounds in Datura metal, 7 phytocompounds in Azadirachta indica, 11 phytocompounds in Acalypha indica.

Effect of oil and aqueous extract of Neem (Azadirachta indica) seeds on growth of Aspergillus species and biosynthesis of aflatoxin

  • Aflatoxin contamination is a major problem in several food crops. Aflatoxin, a mycotoxin, produced by Aspergillus flavus has gained immense concern in the scientific world because of its tremendous harmful effects. The study was focused to see the effect of oil and aqueous extract of neem (Azadirachta indica) seeds on the growth of Aspergillus and production of aflatoxin by the mold. Various amounts of neem oil $(5\;-\;50\;{\mu}l/ml)$ and aqueous extract of neem (5 - 50 mg/ml) were used both in the broth as well as the solid medium. Fungistatic (MIC) and minimal fungicidal concentrations (MFC) were found to be $10\;{\mu}l/ml$ and $50\;{\mu}l/ml$ respectively for neem seed oil. At the concentration of $5\;{\mu}l/ml$ neem oil and 5 mg/ml of aqueous extract, a significant decrease in the aflatoxin content was found in broth medium. Aflatoxin production was totally inhibited at $50\;{\mu}l/ml$ and 50 mg/ml for neem oil and aqueous extract of neem respectively, in both treatments. There was significant inhibition of mycelium dry weight by the neem seed oil. Mycelial growth was totally inhibited at $20\;{\mu}l/ml$ of neem seed oil concentration in broth, whereas it was not affected at all by aqueous extract. It can therefore be inferred that the oil and extract from the neem seed leads to inhibition of aflatoxin production while neem seed oil also significantly inhibits the mycelial growth. Neem seed oil thus can be used as potent, natural and easily available anti-aflatoxigenic agent.

Anti-Termite Activity of Azadirachta excelsa Seed Kernel and Its Isolated Compound against Coptotermes curvignathus

  • Azadirachta excelsa, is a plant belonging to the same genus as Indian neem (Azadirachta indica), and its use as a pesticide is reported by few studies. Despite being a different species, it is expected to have the same biopesticide potential as A. indica. Therefore, this study aims to investigate the anti-termite activity of n-hexane and methanol extracts of A. excelsa seed kernel at various concentrations against Coptotermes curvignathus. The methanol extract demonstrated greater termicidal activity than n-hexane at doses test of 2%, 4%, and 8%. It also showed 100% termite mortality on the third day of administering the 8% dose. According to the gas chromatography with mass spectrometry data, the putative main components of the n-hexane extract were hexadecanoic acid, ethyl ester (18.99%), 9,12-octadecadienoic acid (Z,Z)- (16.31%), and 9-octadecenal (16.23%). In contrast, the principal constituents of methanol extract were patchouli alcohol (28.1%), delta-guaiene (15.15%), and alpha-guaiene (11.93%). Furthermore, limonoids profiling of A. excelsa methanol extract was determined using Ultrahigh-performance liquid chromatography coupled with quadrupole-Orbitrap high-resolution mass spectrometry. The number of limonoids identified tentatively was fifteen, such as 6-deacetylnimbin, nimbolidin C, nimbolide, 6-acetylnimbandiol, 6-deacetyl-nimbinene, salannol, 28-deoxonimbolide, gedunin, nimbandiol, epoxyazadiradione, azadirone, 2',3'-dihydrosalannin, marrangin, nimbocinol, and azadirachtin. They were the same as those reported in the seed and leaves of A. indica, but its largest component in A. excelsa was 6-deacetylnimbin. As a result, the presence of these compounds may be responsible for the anti-termite activity of A. excelsa seed kernel extract. Additionally, column chromatography of methanol extract yielded 6-deacetylnimbin, which was found to be antifeedant and termiticide against C. curvignathus.

  • The purpose of this study was to investigate the antioxidant and hangover cure effects of compound prescription containing Phyllanthus emblica and Azadirachta Indica leaf extract (CP). In vitro experiments, HepG2 cells were induced oxidative stress by hydrogen peroxide (H2O2) and treated with CP at 50, 100, 200 ㎍/㎖ concentration. Antioxidant enzyme (superoxide dismutase (SOD), catalse (CAT), glutathione peroxidase (GPx), glutathione reductase (GR) activity and glutathione (GSH) content were decreased by hydrogen peroxide-induced oxidative stress, but CP was increased that. In vivo experiments, experiment rats were orally administered alcohol 3 g/kg and, after 30 min administered CP 200 mg/kg. After 1 and 3 h of alcohol administration, blood was collected from the tail vein, while after 5 h, blood was collected from the heart. CP modulates alcohol dehydrogenase (ADH) and acetaldehyde level, thereby decreased alcohol level in serum. Also, CP decreased the levels of aspartate aminotransferase (AST) and alkaline phosphatase (ALP). These results suggest that CP has antioxidant effects and alleviates alcohol hangover symptoms.

님추출물의 경구투여에 따른 랫드의 신장독성 연구 (Study of Kidney Toxicity of Azadirachta Indica Extract for Oral Administration in Rats)

  • Azadirachta indica는 살충작용을 가진 약용 식물 중 하나로 우리나라에서 유기농업자재로 널리 사용되고 있다. 본 연구에서는 님추출물의 인체 안전성을 확인하기 위하여, SD 랫드를 이용하여 님추출물의 급성경구독성시험과 4주 반복투여 경구독성시험을 수행하여 신장의 조직변화 및 혈액생화학적 지표를 관찰하였다. 급성 독성시험 결과 님추출물의 $LD_{50}$은 2.0 g/Kg이상으로 나타났다. 반복투여 경구독성시험으로 님추출물을 각각 0.5, 1.0, 2.0 g/Kg으로 투여한 결과, 체중변화, 사료 및 물 섭취량에서는 유의적인 차이가 없었으며, 시험물질투여군의 상대 신장중량 또한 유의적인 차이를 보이지 않았다. 혈중 CREA는 수컷에서 님추출물 고용량 투여군에서 유의적으로 증가하였으나 BUN은 투여용량이 증가할수록 유의적으로 감소하였다. 신장과 관련된 혈중 지표인 CHO는 암컷에서 시험물질 투여군의 용량이 증가할수록 유의적으로 증가하였다. 그러나, 조직 병리학적 분석결과 모든 시험물질투여군에서 이상이 관찰되지 않았다. 이상의 결과를 종합하여 볼 때, 님추출물은 암컷에서 혈중 콜레스테롤 증가 경향을 보인 것을 제외하고는 모든 용량처리군에서 신장에 독성 영향을 미치지 않는 것으로 생각된다.

The Methanol Extract of Azadirachta indica A. Juss Leaf Protects Mice Against Lethal Endotoxemia and Sepsis

  • In the present study, the inhibitory effect of neem leaf extract (NLE) on lipopolysaccaride (LPS)-induced nitric oxide (NO) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) production was examined both in vitro and in vivo. In vitro study revealed that NLE treatment ($100{\mu}g/ml$) inhibits LPS (100 ng/ml)-induced NO production by 96% and TNF-${\alpha}$ production by 32%. The reduction in NO production is probably conferred by the complete suppression of inducible nitric oxide synthase (iNOS) expression. Interestingly, in vivo NLE significantly improved the survival rate of mice in an experimental sepsis model. Administration of NLE (100 mg/kg) 24 h before LPS treatment (20 mg/kg) improved the survival rate of mice by 60%. The inhibition of plasma NO and TNF-${\alpha}$ production by NLE is likely to account for the improved survival of mice. Our results suggest that NLE may present a promising avenue in the development of therapeutic agents for the treatment of inflammatory diseases.
