Schwann cells play a critical role for myelination in peripheral nerve system. It also plays an important role in nerve protection and regeneration. In peripheral nerve damage, regeneration is induced by the migration and proliferation of Schwann cells which were promoted by suppressing the oxidative stress. In this study, Human placental extract was prepared by homogenization and estimated its efficacy in RSC96 cells. Placental extract exhibited a protective effect against hydrogen peroxide-induced oxidative stress in RSC96 cells, confirmed by MTT assay. Furthermore, placental extract decreased intracellular ROS against oxidative stress, confirmed by DCFH-DA assay. Autophagy was visualized with Cyto-ID staining to confirm the autophagy activity of placental extracts. The activity of autophagy was confirmed by immunoblot analysis of autophagy flux-associated proteins such as LC3 conversion and SQSTM1 degradation. Thus, we confirmed the antioxidant effect of placental extract to protect RSC96 cells from oxidative stress, and observed that it activated autophagy and restored autophagy flux.
This study aims to evaluate the antitumor effect of Adonis multiflora, one of the plants in the Ranunculaceae, on mice to which hepatoma cells were transplanted and to suggest its possibility as a candidate natural substance to replace antitumor drugs. We performed the MTT assay to assess the extract had a decrease in the growth rate of hepatoma cells depending on concentration. In particular, 100 ㎍/㎖ of the extract showed 40% of growth retardation rate. We assessed the autophagy activity to identify the inhibitory autophagy mechanism of tumor cells in the extract. This proved that the activity increases more as the concentration of the extract is higher. We conducted the Western blot test to confirmed the expression of two proteins LC3 and p62. The expression of p62 was in inverse proportion to the concentration of the extract whereas LC3-Ⅱ increased more as the concentration of the extract was higher. This showed that an increase in the autophagy relies on the conentration of the extract. We performed a test to discover the influence of the extracts on hepatoma cells transplanted to mice. The test proved that the extract triggers a significant decrease in the growth rate of tumor cells. Compared to the start of the test, the size of tumor cells with 50, 100 and 200 ㎎/㎏ of the extract respectively increased by 4, 3.7 and 3.5 times whereas in the controlling group by 6.3 times. The size of tumor cells in benign tumor controlling group increased by 3.1 times. This showed a significant decrease in the growth rate of tumor cells compared to the controlling group. We carried out the experiment of influence of the extract on the expression of two proteins LC3 and p62 in the tumor tissue transplanted into mice. The experiment showed that LC3-II increases more as the concentration of the extract is higher. However, there was a rapid decrease in p62 with 200 ㎎/㎏ of the extract compared to the controlling group. In this study, we proved that the autophagy activity of Adonis multiflora extract inhibits the growth of hepatoma cells by in vitro and in vivo experiments. In conclusion, the inhibitory autophagy mechanism of tumor cells in the extract can be used as a new treatment of antitumor.
BACKGROUND/OBJECTIVES: Morusin, a marker component of Morus alba L., possesses anti-cancer activity. The objective of this study was to determine autophagy-inducing effect of morusin in non-small cell lung cancer (NSCLC) cells and investigate the underlying mechanism. SUBJECTS/METHODS: Autophagy induction and the expression of autophagy-related proteins were analyzed by LC3 immunofluorescence and western blot, respectively. The role of autophagy and AMP-activated protein kinase (AMPK) was determined by treating NSCLC cells with bafilomycin A1, an autophagy inhibitor, and compound C, an AMPK inhibitor. Cytotoxicity and apoptosis induction were determined by MTT assay, trypan blue exclusion assay, annexin V-propidium iodide (PI) double staining assay, and cell cycle analysis. RESULTS: Morusin increased the formation of LC3 puncta in the cytoplasm and upregulated the expression of autophagy-related 5 (Atg5), Atg12, beclin-1, and LC3II in NSCLC cells, demonstrating that morusin could induce autophagy. Treatment with bafilomycin A1 markedly reduced cell viability but increased proportions of sub-G1 phase cells and annexin V-positive cells in H460 cells. These results indicate that morusin can trigger autophagy in NSCLC cells as a defense mechanism against morusin-induced apoptosis. Furthermore, we found that AMPK and its downstream acetyl-CoA carboxylase (ACC) were phosphorylated, while mammalian target of rapamycin (mTOR) and its downstream p70S6 kinase (p70S6K) were dephosphorylated by morusin. Morusin-induced apoptosis was significantly increased by treatment with compound C in H460 cells. These results suggest that morusin-induced AMPK activation could protect NSCLC cells from apoptosis probably by inducing autophagy. CONCLUSIONS: Our findings suggest that combination treatment with morusin and autophagy inhibitor or AMPK inhibitor might enhance the clinical efficacy of morusin for NSCLC.
Autophagy targets cytoplasmic cargo to a lytic compartment for degradation. Autophagy-related (Atg) proteins, including the transmembrane protein Atg9, are involved in different steps of autophagy in yeast and mammalian cells. Functional classification of core Atg proteins in plants has not been clearly confirmed, partly because of the limited availability of reliable assays for monitoring autophagic flux. By using proUBQ10-GFP-ATG8a as an autophagic marker, we showed that autophagic flux is reduced but not completely compromised in Arabidopsis thaliana atg9 mutants. In contrast, we confirmed full inhibition of auto-phagic flux in atg7 and that the difference in autophagy was consistent with the differences in mutant phenotypes such as hypersensitivity to nutrient stress and selective autophagy. Autophagic flux is also reduced by an inhibitor of phosphatidylinositol kinase. Our data indicated that atg9 is phenotypically distinct from atg7 and atg2 in Arabidopsis, and we proposed that ATG9 and phosphatidylinositol kinase activity contribute to efficient autophagy in Arabidopsis.
Objectives : The purpose of the study is to examine the cell growth effect and autophagy effect of Scutellariae Radix by ethanol extract in SCC 25 cells. Methods : Cell growth inhibitory effect and autophagy induced by Scutellariae Radix were confirmed by WST-1 assay, monodansylcadaverine(MDC) stain, and flow cytometry by acridine orange(AO) stain. Results : The Scutellariae Radix treatment decreased the cell proliferation in a dose and time dependent manner. Scutellariae Radix has anticancer effects that autophagic vacuoles were apparent by MDC and AO staining in SCC 25 cells. Conclusions : Scutellariae Radix showed anticancer activity against SCC 25 cells via autophagy. The data provided the possibility that Scutellariae Radix may potentially contribute to oral cancer treatment.

Gastric adenocarcinoma is among the top causes of cancer-related death and is one of the most commonly diagnosed carcinomas worldwide. Benzyl isothiocyanate (BITC) has been reported to inhibit the gastric cancer metastasis. In our previous study, BITC induced apoptosis in AGS cells. The purpose of the present study was to investigate the effect of BITC on autophagy mechanism in AGS cells. First, the AGS cells were treated with 5, 10, or 15 μM BITC for 24 h, followed by an analysis of the autophagy mechanism. The expression level of autophagy proteins involved in different steps of autophagy, such as LC3B, p62/SQSTM1, Atg5-Atg12, Beclin1, p-mTOR/mTOR ratio, and class III PI3K was measured in the BITC-treated cells. Lysosomal function was investigated using cathepsin activity and Bafilomycin A1, an autophagy degradation stage inhibitor. Methods including qPCR, western blotting, and immunocytochemistry were employed to detect the protein expression levels. Acridine orange staining and omnicathepsin assay were conducted to analyze the lysosomal function. siRNA transfection was performed to knock down the LC3B gene. BITC reduced the level of autophagy protein such as Beclin 1, class III PI3K, and Atg5-Atg12. BITC also induced lysosomal dysfunction which was shown as reducing cathepsin activity, protein level of cathepsin, and enlargement of acidic vesicle. Overall, the results showed that the BITC-induced AGS cell death mechanism also comprises the inhibition of the cytoprotective autophagy at both initiation and degradation steps.
Objectives: Human rhinoviruses (HRVs) are the major cause of the common cold. Currently there is no registered, clinically effective, antiviral chemotherapeutic agent to treat diseases caused by HRVs. In this study, the antiviral activity of dexamethasone (DEX) against HRV1B was examined. Methods: The anti-HRV1B activity of DEX was assessed by sulforhodamine B assay in HeLa cells, and by RT-PCR in the lungs of HRV1B-infected mice. Histological evaluation of HRV1B-infected lungs was performed and a histological score was given. Anti-HRV1B activity of DEX via the glucocorticoid receptor (GCR)-dependent autophagy activation was assessed by blocking with chloroquine diphosphate salt or bafilomycin A1 treatment. Results: In HRV1B-infected HeLa cells, treatment with DEX in a dose-dependent manner, resulted in a cell viability of > 70% indicating that HRV1B viral replication was reduced by DEX treatment. HRV1B infected mice treated with DEX, had evidence of reduced inflammation and a moderate histological score. DEX treatment showed antiviral activity against HRV1B via GCR-dependent autophagy activation. Conclusion: This study demonstrated that DEX treatment showed anti-HRV1B activity via GCR-dependent autophagy activation in HeLa cells and HRV1B infected mice. Further investigation assessing the development of topical formulations may enable the development of improved DEX effectiveness.

Fluoxetine is used widely as an antidepressant for the treatment of cancer-related depression, but has been reported to also have anti-cancer activity. In this study, we investigated the cytotoxicity of fluoxetine to human gastric adenocarcinoma cells; as shown by the MTT assay, fluoxetine induced cell death. Subsequently, cells were treated with 10 or 20 µM fluoxetine for 24 h and analyzed. Apoptosis was confirmed by the increased number of early apoptotic cells, shown by Annexin V- propidium iodide staining. Nuclear condensation was visualized by DAPI staining. A significant increase in the expression of cleaved PARP was observed by western blotting. The pan-caspase inhibitor Z-VAD-FMK was used to detect the extent of caspase-dependent cell death. The induction of autophagy was determined by the formation of acidic vesicular organelles (AVOs), which was visualized by acridine orange staining, and the increased expression of autophagy markers, such as LC3B, Beclin 1, and p62/SQSTM 1, observed by western blotting. The expression of upstream proteins, such as p-Akt and p-mTOR, were decreased. Autophagic degradation was evaluated by using bafilomycin, an inhibitor of late-stage autophagy. Bafilomycin did not significantly enhance LC3B expression induced by fluoxetine, which suggested autophagic degradation was impaired. In addition, the co-administration of the autophagy inhibitor 3-methyladenine and fluoxetine significantly increased fluoxetine-induced apoptosis, with decreased p-Akt and markedly increased death receptor 4 and 5 expression. Our results suggested that fluoxetine simultaneously induced both protective autophagy and apoptosis and that the inhibition of autophagy enhanced fluoxetine-induced apoptosis through increased death receptor expression.
Objective : Inulae Flos(IF) has been used to treat arthritis, sever furuncle, fear and palpitation, vomiting, stroke, asthma and cough in Korean Medicine. Although the anticancer activity of IF has been reported, the molecular mechanism is still not well understood. In this study, we investigated the growth inhibitory activity of an ethanol extract of IF in HT-1080 human fibrosarcoma cells and its underlying mechanisms using two-dimensional (2D) and three-dimensional (3D) cell culture system. Methods : HT-1080 cells were cultured with IF for 9 days in 3D cell culture. To check an inhibition of cell prolifelation by IF, MTT assay was performed. DNA contents were measured using flow cytometry. Western blotting was used to evaluate the regulation of cell cycle- and autophagy-related proteins. Acridine orange staining was performed to confirm autophagy, and DCF-DA staining was performed to confirm the occurrence of ROS. Results : IF controlled a spheroid formation and decreased a cell viability in 3D cell culture. IF-induced cell proliferation inhibition was associated with a distinct increase of S and G2/M phase cell distribution in 2D cell cultre. In addition, IF significantly induced autophagy and generated reactive oxygen species(ROS). Interestingly, IF-induced cell cycle arrest and autophagy were recovered after pre-treatment of N-acetyl-L-cysteine, ROS scavenger. Conclusion : Our results indicate that IF induced ROS-mediated cell cycle arrest and autophagy and it may potentially useful for human fibrosarcoma treatment.

Autophagy has been studied as a therapeutic strategy for cardiovascular diseases. However, insufficient studies have been reported concerning the influence of vascular smooth muscle cells (VSMCs) through autophagy regulation. The aim of the present study was to determine the effects of VSMCs on the regulation of autophagy under in vitro conditions similar to vascular status of the equipped micro-tubule target agent-eluting stent and increased release of platelet-derived growth factor-BB (PDGF-BB). Cell viability and proliferation were measured using MTT and cell counting assays. Immunofluorescence using an $anti-{\alpha}-tubulin$ antibody was performed to determine microtubule dynamic formation. Cell apoptosis was measured by cleavage of caspase-3 using western blot analysis, and by nuclear fragmentation using a fluorescence assay. Autophagy activity was assessed by microtubule-associated protein light chain 3-II (LC-II) using western blot analysis. Levels of intracellular reactive oxygen species (ROS) were measured using $H_2DCFDA$. The proliferation and viability of VSMCs were inhibited by microtubule regulation. Additionally, microtubule-regulated and PDGF-BB-stimulated VSMCs increased the cleavage of caspase-3 more than only the microtubule-regulated condition, similar to that of LC3-II, implying autophagy. Inhibitory autophagy of microtubule-regulated and PDGF-BB-stimulated VSMCs resulted in low viability. However, enhancement of autophagy maintained survival through the reduction of ROS. These results suggest that the apoptosis of conditioned VSMCs is decreased by the blocking generation of ROS via the promotion of autophagy, and proliferation is also inhibited. Thus, promoting autophagy as a therapeutic target for vascular restenosis and atherosclerosis may be a good strategy.
