Development and Evaluation of Multi-Wavelength Excitation light Source for Fluorescence Imaging to Diagnose Malignancies (악성종양의 형광영상 진단을 위한 다파장 여기광원장치의 개발과 평가)

  • This study aims at designing and evaluating light source devices that can stably generate light with various wavelengths in order to make possible PDD using a photosensitizer and diagnosis using auto-fluorescence. The light source was a Xenon lamp and filter wheel, composed of an optical output control through Iris and filters with several wavelength bands. It also makes the inducement of auto-fluorescence possible because it is designed to generate a wavelength band of 380-420nm, 430-480nm, and 480-560nm. The transmission part of the light source was developed to enhance the efficiency of light transmission. To evaluate this light source, the characteristics of light output and wavelength band were verified. To validate the capability of this device as PDD, the detection of auto-fluorescence using mouse models was performed.

  • Photodynamic diagnosis(PDD) is a method to diagnose the possibility of cancer, both by the principle that if a photosensitizer is injected into an organic tissue, it is accumulated in the tissue of a malignant tumor selectively after a specific period, and by a comparison of the intensity of the fluorescence of normal tissue with abnormal tissue after investigating the excitation light of a tissue with accumulated photosensitizer. Since the selection of the wavelength band of excitation light has an interrelation with fluorescence generation according to the selection of a photosencitizer, it plays an important role in POD.   • Two-photon microscopy (TPM) is minimally-invasive 3D fluorescence microscopy based on nonlinear excitation, and TPM can visualize cellular structures based on auto-fluorescence. Line-scanning TPM is one of high-speed TPM methods without sacrificing the image resolution by using spatial and temporal focusing. In this paper, we developed line-scanning TPM based on spatial and temporal focusing for auto-fluorescence imaging by exciting the tryptophan. Laser source for this system was an optical parametric oscillator (OPO) and it made near 570 nm femtosecond pulse laser. It had 200fs pulse width and 1.72 nm bandwidth, so that the achievable depth resolution was 2.41um and field of view (FOV) is 10.8um. From the characterization, our system has 3.0 um depth resolution and 12.3 um FOV. We visualized fixed leukocyte cell sample and compared with point scanning system.

  • In the study of the fluorescence detection characteristics of the UV light intensity control function for automatic reading sterilization devices, the fluorescent medium of the biological indicator meets the spore bacteria, causing a fluorescence reaction. We try to find certain characteristics by measuring the characteristics of the detected amount of fluorescence while adjusting the intensity. Research to find the appropriate intensity of UV light for automatic reading sterilization devices aims to increase the accuracy of biological indicators and increase the accuracy of reading through appropriate UV light intensity over time.

A Review of SERS for Biomaterials Analysis Using Metal Nanoparticles (바이오 물질 분석을 위한 금속 나노입자를 이용한 SERS 분석 연구동향)

  • Surface enhanced Raman scattering (SERS) was first discovered in 1974 by an unexpected Raman signal increase from Pyridine adsorbed on rough Ag electrode surfaces by the M. Fleishmann group. M. Moskovits group suggested that this phenomenon could be caused by surface plasmon resonance (SPR), which is a collective oscillation of free electrons at the surface of metal nanostructures by an external light source. After about 40 years, the SERS study has attracted great attention as a biomolecule analysis technology, and more than 2500 new papers and 500 review papers related to SERS topic have been published each year in recently. The advantages of biomaterials analysis using SERS are as follows; ① Molecular level analysis is possible based on unique fingerprint information of biomolecule, ② There is no photo-bleaching effect of the Raman reporters, allowing long-term monitoring of biomaterials compared to fluorescence microscopy, ③ SERS peak bandwidth is approximately 10 to 100 times narrower than fluorescence emission from organic phosphor or quantum dot, resulting in higher analysis accuracy, ④ Single excitation wavelength allows analysis of various biomaterials, ⑤ By utilizing near-infrared (NIR) SERS-activated nanostructures and NIR excitation lasers, auto-fluorescence noise in the visible wavelength range can be avoided from in vivo experiment and light damage in living cells can be minimized compared to visible lasers, ⑥ The weak Raman signal of the water molecule makes it easy to analyze biomaterials in aqueous solutions. For this reason, SERS is attracting attention as a next-generation non-invasive medical diagnostic device as well as substance analysis. In this review, the principles of SERS and various biomaterial analysis principles using SERS analysis will be introduced through recent research papers.

NADH Variation and Process Control with NADH Fluorometer in Full Scale Biological Nutrient Removal Process (실규모의 고도처리공정에서 NADH변화 유형과 이를 활용한 공정제어)

  • In this study, we investigated the possibility of auto control and the proper operating factors in the BNR(Biological Nutrient Removal) process using an NADH(Reduced Nicotinamide Adenine Dinucleotide) fluorometer, which characterized the emitted fluorescence when activated by flashes of UV light at 460 nm. In terms of finding adequate operating parameters, results indicted that nitrification efficiency decreased in the controlled DO while denitrification efficiency decreased in the controlled pH. The above results indicated that controlled operating condition after combination with NADH, DO and pH was resonable. Result obtained from the correlation between NADH and pH showed that variation trend of influent loading was similar to those of NADH and pH, and also the variation cycle was repeated on a daily basis. Consequently, this result showed the increase of BOD loading caused the nitrification efficiency to decrease because air-flow, required for nitrification, was reduced, and so the NADH value was increased. From these results, it is possible to use NADH flourimetry to assess the variation of organic load and nitrification efficiency in the case of small change in influent pH such as in sewage and also to handle and operate the load variation in the auto control system using the NADH fluorometer.

  • Cheju island is situated at about 90km south of southern most tip of the Korean peninsula. Its shape is an ellips with size of 80$\times$40$km^2$, and it is characterized by a symmetrical form whose peak is Mt. Halla (1950m). Major chemical composition of the rock samples from Manjang gul cave is determined by XRF using the fundamental parameter method consisting of fully auto quantitative analysis, semi quantitative analysis (order estimation) and group quantitative analysis. Judging from chemical composition ($SiO_2 = 53.07Wt% Fe_2 O_3=11.34Wt%, MgO=6.48Wt%, Na_2 O=3.07Wt%, K_2 O=1.05Wt%$), this rocks may belong to non alkalic basalt. K-Ar ages of two rocks samples from the Manjang gul cave are also determined. The discrepancy of K-Ar ages is found. They are 0.03Ma and 0.42Ma, respectively. This paper describes some problems experienced in dating young volcanic rocks and then discusses chemical composition, X-ray fluorescence analysis and the age of the formation of a lava tunnel such as Manjang gul cave in cheju Island.
