Objectives : Atractylodes rhizomes, which have been widely used to treat gastrointestinal disorders, consist of numerous chemical compounds. Polyacetylenes are the parts of characteristic compounds of importance required to understand the therapeutic properties of Atractylodes rhizomes. It is necessary to understand the physicochemical and pharmacological properties of polyacetylenes in the Atractylodes rhizomes. Methods : The literatures from 1970 to January 2016 were searched using Korean and international electronic databases. The chemical structures of polyacetylenes were drawn by structure-drawing software. Results : The reported polyacetylenes were classified by their chemical skeletons and original resources, and their physicochemical and pharmacological features were discussed. Polyacetylenes with skeletal moieties were reported, such as diene-diyne types (two double and two triple carbon-bonds), triene-diyne types (three double carbon bonds and two triple carbon bonds), and monoene-diyne types (one double carbon bonds and two double carbon bonds), with various functional groups. Atractylodin was most frequently reported from many Atractylodes species. Atractylodin-related polyacetylenes showed chemical instability in both high and freezing temperatures. Processing of the Atractylodes rhizomes by stir-frying with bran could affect the contents of polyacetylenes and their bioavailability in vivo. Several polyacetylenes showed structure-related anti-inflammatory activities and gastrointestinal activities. Conclusion : Polyacetylene compounds in Atractylodes rhizomes were based on three chemical backbones and showed diverse physicochemical and pharmacological features. The present study provides structural, physicochemical, and pharmacological information of polyacetylene from Atractylodes rhizomes. This information provides fundamental data for further research.
Crop productivity would be determined by relative capacity of source and sink. The study was carried out to measure the effects of shading and flower bud pinching on growth and yield of Atractylodes japonica Koidz. Shading treatment after transplanting was done at 100%, 65, 45 or 25% level of natural light intensity but the pinching treatment was to remove all the inflorescences formed after June 10, July 10, August 10 or none. Growth and yield at early and late stages were periodically measured. At the early stage, shading mainly influenced fresh weight rather than morphological characters. At the late stage, however, severer shading decreased numbers of stems, leaves, roots, fraction and total fresh weight. 65% shading from July 1 to September 30 after skipped the emerging and early growth stages showed nearly same growth and morphological characters but more rhizome yield per unit area than non-shading. In flower bud pinching treatment, earlier pinching increased number of roots, fraction fresh weights per plant and rhizome yield per unit area.

Background: Atractylodes japonica Koidz. and Atractylodes macrocephala Koidz. belong to the family (Asteraceae). Their rhizomes, called white Atractylodes rhizomes, are used in traditional medicine. To address some issues with their cultivation, we recently developed eight hybrid cultivars by interspecific hybridization of A. japonica and A. macrocephala. This study was conducted to screen the hybrid cultivars that have high amounts of active ingredients and yield ability. Methods and Results: Experiments were conducted using the eight hybrid cultivars and A. macrocephala (control cultivar) in the experimental field of the department of Herbal Crop Research located in Eumseong, South Korea. We investigated the growth characteristics of the aerial and underground parts. Among the cultivars, 'Sanwon' had the highest rhizome dry weight (53.8 g/plant), followed by 'Dachul' (50.0 g/plant). In addition, the content of atractylenolide I, II, III and total active ingredients were investigated using high-performance liquid chromatography. Compared with A. macrocephala, most of the inter-specific hybrid cultivars had a higher content of active ingredients and yield ability. Conclusions: Through study, we established the superior quality of Atractylodes inter-specific hybrid cultivars. In particularly, it was found that 'Dachul' may be grown as a superior cultivar, with high amount of active ingredients as well as yield ability.
The rhizome extract of Atractylodes japonica Koidzumi(Compositae) exhibited a particular inhibition on the proliferation of cultured human tumor cell lines, in vitro. Thus, the intensive phytichemical investigation of the MeOH extract of Atractylodes japonica have been conducted by the way of activity-guided purification. The repeated column chromatographic separation of the n-hexane soluble part of extract resulted in the isolation of four sesquiterpenes (1-4) and a polyacetylene component (5). Chemical structures of them were identified as atractylon (1), atractylenolide Ⅰ(2), atractylenolide Ⅲ(3), eudesma-4(15),7(11)-dien-8-one (4) and 1,3-diacetyl-atractylodiol (5) by spectroscopic means. Among the isolates, compound 2-4 were shown to give moderate inhibitory effect in a dose dependent manner on the proliferation of cultured human tumor cell lines such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT 15(colon), respectively.
Antiproliferative activity of the ethanol extract of Atractylodes japonica rhizomes (AJEX) was investigated using methyl thiazolyl tetrazolium (MTT) assays with various cancer cell lines (HL-60, MCF-7, SK-Br-3, MDA-MB-453, HepG2, Hep3B, PC-3, LNCaP, MKN 28, MKN 45, and HT-29 cells). Gastric carcinoma cell lines were the most responsive in terms of cell proliferation. The $IC_{50}$ of MKN 28 and MKN 45 cells were 35.98 and 27.57 ${\mu}g/mL$, respectively. Moreover, gastric carcinoma cells exposed to AJEX underwent apoptosis, as determined by Annexin V binding assay. Compared to respective control level, exposure to the AJEX at each $IC_{50}$ concentration resulted in a remarkable increase in the shift of cell populations. Present results suggest that AJEX possess potential anticancer properties.
This study was aimed at investigating hepatic toxicity and immunomodulating effects of defatted methanol extracts of two kinds of medicinal plants, Rubus coreanus Miq. and Atractylodes japonica Koidz. in mice. Defatted methanol extracts of fruits of Rubus coreanus Miq. and rhizome of Atractylodes japonica Koidz. were added at the level of 0.5% or 5%(w/w) to cholesterol-supplemented AIN-76 diet. Each diet was fed to 8 ICR male mice for 30 days. Weight gain and food efficiency ratio of the mice fed 5.0% extract of Rubus coreanus Miq. were significantly lower than those of the mice fed 0.5% extract Relative liver weight and activity of plasma alanine aminotransfernse were significantly increased only in the mice fed 5% extract of Atractylodes japonica Koidz. compared with the others. Splenocyte proliferation was not significantly different between the groups fed 0.5% or 5.0% extract of Rubus coreanus Miq. However, splenocyte proliferation was significantly decreased in the mice fed 5.0% extract of Atractylodes japonica Koidz. compared with that in the mice fed 0.5% Production of interleukin-2 by splenocytes from the mice fed 0.5% extract of Atractylodes japonica Miq. was significantly higher than the control value and it became lower with 5.0% dietary level. Secretion of $interferon-\gamma$ was not significantly different among groups. In conclusion, the defatted methanol extract of Atractylodes japonica Koidz. was likely to exert immunomodulating effect at the level of 0.5% but it may exert adverse effects on immune and liver functions at the level of 5.0%.

This study was carried out to obtain the basic information for effective conservation and cultivation of Atractylodes japonica Koidz. The soil texture was sandy loam with low pH and high organic matter content compared to general cultivative land. Aerial part growth such as plant height, number of leaf, leaf length and leaf width was the highest in Tongyong indigenous species, but fresh rhizome weight was the highest in Pyongchang. Frequency of light penetration rate was high at $60{\sim}80%$ of full sun-light, but growth was better in high light penetration rate. The correlation between growth characteristics and habitat environment were investigated in 59 districts. Correation among growth characteristics in habitat, fresh rhizome weight was significant with plant height, number of leaf and stem diameter.

Two Atractylodes species, A. japonica Koidz. ex Kitam. (AJ) and A. macrocephala Koidz (AM) were used in this study. AJ population had higher amounts of Sesquiterpenoids and stronger tolerance to root rot but less vigor of root growth than AM population. Two populations (AJ and AM) were crossed to make interspecific hybrid population. A total of 98 lines propagated clonally were selected from a cross of AJ and AM, and evaluated for contents of sesquiterpenoids, atractylon (ATLN) and atractylenolide III (AT3) using high performance liquid chromatography (HPLC), and growth characters such as plant height, stem number and root weight. HPLC profiles of the hybrids were compared with those of parent plants, and it demonstrated the production of introgression hybrid by crossing between AJ and AM. Of 98 clonal lines,10 lines were selected by 10% level based on the growth vigor and tolerance to root rot, and AJM2102-51 line showed the heaviest root weight (117.1 g/plant) among them. A total of 98 hybrid lines contained on average $0.16\;{\pm}\;0.10\;mg/g$ of $AT3,\;2.00\;{\pm}\;1.37\;mg/g$ of ATLN, and $2.16\;{\pm}\;1.40\;mg/g$ of total sesquiterpenoids, showing high coefficients of variation (above 65%). Ten lines having high contents of sesquiterpenoids were selected, and AJM2101-15 had the highest amount (9.83 mg/g) of ATLN, and showed 40.8 g/plant of root weight similar to mean value (39.9 g/plant) of hybrid lines. The result showed that the introgression of both characters of vigorous growth from AM and high sesquiterpenoids content from AJ could be possible to make new hybrid lines by crossing between AJ and AM.
Objectives: Type 2 diabetes mellitus is a metabolic disease characterized by insulin resistance and high blood glucose level from progressive insulin secretory defect. The rhizome of Atractylodes japonica Koidz. (AJ) has been used for treatment of retention of water in oriental medicine. The aim of this study is to examine the effects of AJ on type 2 diabetes rats. Methods: Type 2 diabetes was induced by 60% high fat diet and low dose streptozotocin. Rats were divided into 4 groups (n = 6); Nor (normal control group), Con (diabetic group treated with vehicle), Met (diabetic group treated with 200 mg/kg metformin) and AJ (diabetic group treated with 100 mg/kg AJ). The body weights and food intakes were measured during the treatment period. After 4 weeks treatment, blood glucose level, HOMA-IR, and protein expressions of IRS-1, p-IRS-1, PPAR-${\gamma}$, and GLUT4 were measured, and histopathological examination of beta cell was performed. Results: Compared with the control group, blood glucose level and HOMA-IR were reduced in rats treated with AJ. Impaired beta cells in pancreas of rats were recovered and phosphorylation of IRS-1 was increased in rats treated with AJ. And also, protein expressions of PPAR-${\gamma}$ and GLUT4 were increased by treatment of AJ. Conclusions: The results suggest that Atractylodes japonica Koidz. may have anti-diabetic effect on type 2 diabetic rats through regulation of blood glucose level and insulin resistance. Therefore Atractylodes japonica Koidz. may have positive effects on patients with type 2 diabetes.

The new assay method was developed by using young chicks instead of frogs for screening of anti-emetic compounds from natural sources. Comparing with our previous method using leopard and ranid frogs, the advantages of the new method were not only completely parallel results but also decreasing standard errors. Hinesol and eudesmol were isolated from MeOH extract of Atractylodes lancea rhizome as the anti-emetic principles.
