Phytochemical Study on the Rhizome of Atractylodes japonica from Korea (한국산 창출의 성분연구)

  • From the rhizome of Atractylodes japonica Koidzumi (Compositae) which is the original plant of oriental medicine 'Cang-Zhu', four essential oil compounds were isolated. Three of them were identified as atractylon, hydroxyatractylon and $5{\alpha}H,\;10{\beta}-selina-4(14),\;7(11)-diene-8-one$, which were already known as the constitutents of Atractylodes Rhizoma. The fourth is a novel polyacetylene type compound, and the structure is postulated as 1, 4-diacetoxytetradeca-6, 12-diene-8, 10-diyne by the spectral data.

Volatile Flavor of Atractylodes japonica koidzumi (삽주의 휘발성 향기성분)

  • An attempt was made in this study to analyze volatile flavor components of A. macrocephala Koidz. and A. lanacea DC. (Atractylodes japonica Koidzumi). Essential oils in A. macrocephala Koidz. and. A. lanacea DC. were isolated by a simultaneous steam distillation and extraction(SDE) method using n-pentane/diethy ether as solvent. A total of 30 and 28 components were identified by GC/MS from the essential oils of A. macrocephala Koidz. (18 hydrocarbons. 2 carbonyls, 5 alcohols, 5 esters) and A. lanacea DC.(14 hydrocarbons, 6 carbonyls, 4 alcohols, 3 esters, 1 acids), respectively. The major volatile flavor components in A. macrocephala Koidz. and A. lanacea DC. were furanodiene(27.9%, 15.7%), $\alpha$-cyperone(8.1%. 22.5% ), alloaromadendrene(2.9%, 4.7% ), (1,1-biphenyl)-4-carbon aldehyde 0%, 8.7% ) were found, respectively. Ten components including limonene, p-cymene, p-hymen-8-ol, (1,1-biphenyl)-4-carbox aldehyde were identified in A. lanacea DC, but not in A. macrocephala Koidz. and eight components including $\alpha$-copanene, isocaryophyllene, $\beta$-himahalene. germacrene B were and identified in A. macrocephala Koidz. but not in A. lanacea DC.

Development of LC-MS/MS analytical methods for metalaxyl in Atractylodes macrocephala Koidzumi and Achyranthes japonica Nakai (LC-MS/MS를 이용한 생약 백출 및 우슬 중 Metalaxyl 잔류분석법 개발)

  • A new rapid and simple method for metalaxyl in Atractylodes macrocephala Koidzumi and Achyranthes japonica Nakai has been developed and validated. This study was conducted to develop a method for analyzing metalaxyl by a method based on QuEChERS using liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). Samples were extracted with acetonitrile and purified using amino-propyl (NH2) Solid Phase Extraction cartridge. The method limit of quantitation (MLOQ) was 0.01 mg/kg. The linearity of matrix-matched calibration curve (r2) was ≥0.99 at the calibration range of 0.001-0.05 mg/kg. For recovery test, Atractylodes macrocephala Koidzumi or Achyranthes japonica Nakai was treated with standard solutions at MLOQ and 10MLOQ levels. Recovery rates were in the range of 88.1-109.1% with <5.5% coefficient of variation. This established analytical method was fully validated. Based on these results, it can contribute to improving the safety of residual pesticides in Atractylodes macrocephala Koidzumi and Achyranthes japonica Nakai.

Safety and Effects on Lipid Parameters of Rubus coreanus and Atractylodes japonica in Ovariectomized Rats

  • Defatted methanol extracts of the medicinal plants, Rubus coreanus Miq. (RC) and Atractylodes japonica Koidzumi (AJ) were added at the levels of 0.1, 0.5, or $2\%$ (w/w) to high cholesterol diets and fed to ovariectomized Sprague-Dawley female rats, weighing 212.6 $\pm$ 1.8 g for four weeks. Weight gains were lower in RC and AJ groups than the control group, but there were no changes in uterus weights. Serum levels of triglyceride decreased by 20-$27\%$ in the experimental groups fed $0.1\%$ of each extract (O.1RC and O.1AJ), compared with that of control (Ovx). Serum cholesterol levels were not changed in the RC groups but increased in the group fed $2\%$ of the AJ extract. Liver levels of cholesterol and triglyceride were reduced in both the RC and AJ groups. Microscopic observation revealed that there were no morphological alterations in liver, lung, heart, spleen and kidney tissues of the experimental groups. Plasma levels of albumin, BUN, creatinine, sodium, potassium and phosphate in the IRC and AJ groups were in normal ranges. Serum GOT and GPT activities were, however, higher in the 2.0AJ than Ovx group. These results suggest that the extracts of the Rubus coreanus Miq. and Atractylodes japonica Koidzumi at dietary levels as low as $0.1\%$ may be utilized as hypotriglyceridemic ingredients for functional foods.

Study on Atractylodes Rhizomes (I) : Pharmacognosy and Chemical Identification of Atractylodes Species (출류(朮類)에 관한 연구( I ) : 한국(韓國)과 일본(日本)의 창출(蒼朮)과 백출(白朮) 의 생약학적(生藥學的) 연구)

  • Traditional Chinese herbal drugs, namely, Atractylodes Rhizome and Atractylodes Rhizome White were identified by means of chemical analysis such as TLC and GC/MS. In Korea, the old-grown and the newly-grown rhizomes of Atractylodes japonica Koidzumi (Compositae) are used as the Atractylodes Rhizome and the Atractylodes Rhizome White, respectively. Both rhizomes contain the atractylon asa major spot on TLC. The content of atractylon in a newly-grown rhizome of A. japonica (Atractylodes Rhizome White called in Korea) appears to contain much larger quantity in comparison with that of an old-growon one (Atractylodes Rhizome). The TLC spot pattern analysis showed that rhizomes of A. japonica and A. macrocephala contain the atractylon as a major indicator spot, but no atractylodin (extreamly small amount). However, rhizomes of A. lancea De Candolle and A. koreana Kitamura contain the atractylodin as a major indicator spot.

Variation analysis of Atractylodes japonica Koidzumi ex Kitamura Based on Quantitative Characters (삽주(Atractylodes japonica Koidzumi ex Kitamura)의 정량적 형질에 의한 변이분석)

  • Sixteen Atractylodes japonica Koidz. accessions were collected in Jeonbuk, Kyeongnam, Kangwon and Kyeonggi province, and their quantitative characteristics were compared with each other. The relationship of variation between Atractylodes japonica Koidz. accessions was also investigated by means of multivariate analysis. Hamyang 1 accession had the most desirable quantitative characteristics among the accessions: plant height was 86.2cm, number of leaves, 117.2 and number of stems, 34.3. Sudo 1 accession had 12 stems, the most among the accessions. The length of leaf margin serras found in all of them was on average 1.6mm. Most accessions had simple or palmately compound leaf with oval or elliptical type of the leaf shape. The accessions could be classified into three groups by average linkage cluster method. The most remote relationship was found between Group 1 containing Jinan 1, Jinan 2 and Group 3 containing Jeongsu, and Geoje.

Effects of Methanol Extracts of Rubus Coreanus Miquel and Atractylodes Japonica Koidzumi on Hepatic Toxicity and Immunomodulating Activity in Mice

  • This study was aimed at investigating hepatic toxicity and immunomodulating effects of defatted methanol extracts of two kinds of medicinal plants, Rubus coreanus Miq. and Atractylodes japonica Koidz. in mice. Defatted methanol extracts of fruits of Rubus coreanus Miq. and rhizome of Atractylodes japonica Koidz. were added at the level of 0.5% or 5%(w/w) to cholesterol-supplemented AIN-76 diet. Each diet was fed to 8 ICR male mice for 30 days. Weight gain and food efficiency ratio of the mice fed 5.0% extract of Rubus coreanus Miq. were significantly lower than those of the mice fed 0.5% extract Relative liver weight and activity of plasma alanine aminotransfernse were significantly increased only in the mice fed 5% extract of Atractylodes japonica Koidz. compared with the others. Splenocyte proliferation was not significantly different between the groups fed 0.5% or 5.0% extract of Rubus coreanus Miq. However, splenocyte proliferation was significantly decreased in the mice fed 5.0% extract of Atractylodes japonica Koidz. compared with that in the mice fed 0.5% Production of interleukin-2 by splenocytes from the mice fed 0.5% extract of Atractylodes japonica Miq. was significantly higher than the control value and it became lower with 5.0% dietary level. Secretion of $interferon-\gamma$ was not significantly different among groups. In conclusion, the defatted methanol extract of Atractylodes japonica Koidz. was likely to exert immunomodulating effect at the level of 0.5% but it may exert adverse effects on immune and liver functions at the level of 5.0%.

Inhibitory Effects of the Rhizome Extract of Atractylodes japonica on the Proliferation of Human Tumor Cell Lines (백출 추출물의 암세포증식 저해 효과)

  • The rhizome extract of Atractylodes japonica Koidzumi(Compositae) exhibited a particular inhibition on the proliferation of cultured human tumor cell lines, in vitro. Thus, the intensive phytichemical investigation of the MeOH extract of Atractylodes japonica have been conducted by the way of activity-guided purification. The repeated column chromatographic separation of the n-hexane soluble part of extract resulted in the isolation of four sesquiterpenes (1-4) and a polyacetylene component (5). Chemical structures of them were identified as atractylon (1), atractylenolide Ⅰ(2), atractylenolide Ⅲ(3), eudesma-4(15),7(11)-dien-8-one (4) and 1,3-diacetyl-atractylodiol (5) by spectroscopic means. Among the isolates, compound 2-4 were shown to give moderate inhibitory effect in a dose dependent manner on the proliferation of cultured human tumor cell lines such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT 15(colon), respectively.

A study on the frequencies of medicinal herb combinations in the prescriptions of "Bangyakhappyeon(方藥合編)" ("방약합편(方藥合編)" 수록(收錄) 처방(處方) 내(內)의 약물(藥物) 조합 빈도(頻度) 연구)

  • Through analyzing the frequencies of medicinal herb combinations in the prescriptions of "Bangyakhappyeon(方藥合編)", we could understand the characteristics of prescription composition and author's medical thoughts. So we conducted this study that analyze combination of 324 medicinal herbs in 467 prescriptions, and found meaningful combinations of 5,045. Among these, the most meaningful combination is the one of 10 herbs, Glycyrrhiza uralensis Fischer, Angelica gigas Nakai, Ziziphus jujube Mille, Atractylodes japonica Koidzumi, Poria cocas Wolf, Zingiber officinale, Rehmanniae radix preparata, Cinnamomum loureirii, Panax ginseng C.A.Meyer, Astragalus membranaceus. This combination is Sibjeondaebo-tang(Shiquandabu-tang) without Cnidium officinale and Paeonia japonica which compose Samul-tang(Siwu-tang). It means this combination is kind of modified Sibjeondaebo-tang(Shiquandabu-tang), which is more effective in tonifying qi than tonifying blood. Prescriptions of "Bangyakhappyeon(方藥合編)" involve herbs mostly used to improve weak and frail health and the herbs which are more effective in tonifying qi than tonifying blood. Through this study, we know the author' s one of medical view points on tonifying qi and blood, attaching importance to tonifying qi relatively to tonifying blood.
