황기의 알러지 비염 동물실험에 대한 면역조절 효과 (Immuno Modulatory Effect of Astragali Radix on OVA Induced Allergic Mouse Model)

  • Astragali Radix(AR), is a popular tonic herb prescribed for 'insufficient qi' in Korea, Japan and China. The present study examined the effect of AR ethanol extract on ovalubumin induced allergic mouse model. AR administration reduced levels of IFN-gamma, Interleukin(IL)-4, IL-5 and total IgE in the OVA induced allergic inflammation. It also protected the upper airway respiratory epithelium from being damaged by the OVA induced inflammation. Taken together, our results showed that the use of AR alone proved to down-regulate Th1 and Th2 cytokine production and play a protective role in tissue damage in allergic disease.

황기 부탄올 분획물이 생쥐의 체액성 면역기능에 미치는 영향 (Effects of the Butanol Fraction of Astragali Radix on the Humoral Immune Function in Mice)

  • Effect of the butanol fraction of Astragali Radix (BFAR) on the humoral immune response were investigated in ICR mice. Mice were divided into 4 groups and BFAR at doses of 5,25 and 125 mg/kg were administered orally to mice daily for 3 weeks, and the normal animals were given vehicle. The results of this study are summarized as follows; the relative weight of spleen was markedly increased by BFAR treatment, compared with that in normal mice. However, the body weight gain and the relative weight of liver were not affected. Splenic plaque forming cells and hemagglutination titers to sheep red blood cells, and the secondary IgG antibody response to bovine serum albumin were also dose-dependently enhanced by BFAR treatment. In these mice, BFAR did not increase serum alanine aminotransferase total protein, sect albumin and albumin/globulin ratio when compared with those in normal mice. Thus, these findings indicate that BFAR significantly enhances humoral immune response to antigen in concentrations that do not affected liver function.

염화아연의 세포성 면역독성에 미치는 황기 추출물의 효과 (Effects of Astragali Radix extract on the Cell Mediated Immunotoxicity of Zinc Chloride)

  • Effects of Astragali Radix extract (AG) on the cell mediated-and nonlpecific immunotoxic responses of zinc chloride (Zn) were studied usign ICR mice. Mice were divided into 4 groups (10 mice/group), and Zn was given to the mice 1 hr after i.p. injection with 0.5g/kg of AG by i.p. injection daily for 10 days at a dose of 25 mg/kg. Immune responses on the responses on the relative weight of thymus, delayed-type hypersensitivity to SRBC (DTH), phagocytic activity and circulating leukocytes were evaluated. Zn treatment decreased body weight gain, the relative weight of thymus, DTH and circulating leukocytes compared with those in controls. AG treatment increased DTH, phagocytic activity and circulating leukocytes compared with those in controls. Combination of AG and Zn increased DTH and circulating leukocytes compared with those in controls, but decreased body weight gain and the relative weight of thymus. These findings indicated that AG decreased immunotoxicity of Zn on the DTH and circulating leukocytes.

황기의 저밀도지질단백질 (LDL)산화에 미치는 영향 (Effect of Astragali Radix on Low Density Lipoprotein Oxidation)

  • The root of Astragalus membranaceus Bunge (Leguminosae), which has been used for the treatment of hypertension, chronic hepatitis, duodenal ulcers, chronic nephritis and promotion of immunity in folk remedies. Several lines of evidence indicate that oxidative modification of low-density lipoprotein (Ox-LDL) may play an important role in atherogenesis. Hence, the role of antioxidants in the prevention of LDL oxidation needs to be determined. To investigate the antioxidant activity. we determined the MeOH ex. and fractions of Astragali Radix on the inhibition of LDL oxidation. The CH$_2$C1$_2$ and EtOAc orations inhibited the oxidative modification of LDL by a decrease in the lipid peroxide content and the electrophoretic mobility of LDL. Calycosin-7-0-$\beta$-D -glucoside which was isolated from EtOAc fraction inhibits the oxidative modification of LDL.

  • Inhibition of inflammatory response, acceleration of basal cell growth, and balanced synthesis of extracellular matrix (ECM) are important in healing of cutaneous open wounds. In order to evaluate the healing effects of water extracts of Radix Astragali (the root of Astragalus membranaceus (Fisch.)) on open wound at early stage, the experimental open wounds were generated on the dorsal sides of SD rats under anesthesia. The boiled-water extracts of Radix Astragali $(100{\mu}l)$, soaked into an occlusive film dressing were applied once a day for eleven consecutive days. The healing process was assessed by measuring macroscopic appearance and wound areas of the open wounds. The molecular aspects of healing process by Radix Astragali extracts were also investigated by Hematoxylin-Eosin (H-E) double staining and immunohistological staining of collagen type I in the healed skin area, implying cell density and linear alignment of the granulation tissue, and ECM synthesis and its remodeling, respectively. The Astragali radix extracts were found to significantly accelerate the cutaneous wound healing by suppressing the inflammation and stimulating the basal cell growth in wounded area, as compared to epidermal growth factor (EGF).

  • Background: Pressure injuries are localized areas of damage to the skin and/or underlying tissue, usually over a bony prominence due to pressure. Cerebrovascular disease increases the risk of pressure injuries due to the immobility caused by physical paralysis. The general approach to managing a patient with pressure injuries should include pain relief, the treatment of the infection, optimizing nutritional intake, proper positioning, and contamination prevention. Nonetheless, the duration of treatment for pressure injuries varies from person to person. Case report: An 80-year-old female intracerebral hemorrhage patient developed a pressure injury. To improve the injury faster, a Radix Astragali pharmacoacupuncture solution was applied to the pressure sore. The pressure injury's width, length, and depth was assessed using a ruler, and the exudate amount and tissue types were assessed. The treatment was performed for 35 days. The rate at which the size of the pressure sore lessened increased since the Radix Astragali pharmacoacupuncture solution was applied to the pressure sore. In addition, the tissue type of the pressure injury improved, and the exudates decreased. There was no significant difference in the Pressure Ulcer Scale for Healing Tool 3.0, since the Radix Astragali pharmacoacupuncture solution was applied to the pressure injury. Conclusion: This clinical case study suggests that the Radix Astragali pharmacoacupuncture solution might be effective in speeding up the healing of pressure injuries.

Quantitative Evaluation of Radix Astragali through the Simultaneous Determination of Bioactive Isoflavonoids and Saponins by HPLC/UV and LC-ESI-MS/MS

  • The three major active isoflavonoids (calycosin-7-O-β -glucoside, isomucronulatol 7-O-β-glucoside, formononetin) and two main saponins (astragaloside I, astragaloside IV) in an extract of Radix Astragali were determined using rapid, sensitive, reliable HPLC/UV and LC-ESI-MS/MS methods. The separation conditions employed for HPLC/UV were optimized using a phenyl-hexyl column (4.6 × 150 mm, 5 μm) with the gradient elution of acetonitrile and water as the mobile phase at a flow rate of 1.0 mL/min and a detection wavelength of 230 nm. The specificity of the peaks was determined using a triple quadrupole tandem mass spectrometer equipped with an electrospray ionization (ESI) source that was operated in multiple reaction monitoring (MRM) in the positive mode. These methods were fully validated with respect to the linearity, accuracy, precision, recovery and robustness. The HPLC/UV method was applied successfully to the quantification of three major isoflavonoids in the extract of Radix Astragali. The results indicate that the established HPLC/UV and LC-ESI-MS/MS methods are suitable for the quantitative analysis and quality control of multi-components in Radix Astragali.

보기약(補氣藥)의 부량반응(不良反應)에 관한 문헌적 고찰 (A philological study on adverse effect of restoratives for invigorating qi(補氣藥))

  • Objectives & Method:We investigated adverse symptoms, toxicity, treatment and prevention against adverse effects of restoratives for invigorating qi(補氣藥) in order to use herbal medicines accurately. Result:Ginseng Radix(人參), Codonopsis Pilosulae Radix(黨參), Panacis Quinquefolii Radix(西洋參), Astragali Radix(黃芪), Atractylodis Rhizoma Alba(白朮), Dioscoreae Rhizoma(山藥), Dolichoris Semen(白扁豆), Glycyrrhizae Radix(甘草), Jujubae Fructus(大棗) and Mel(蜂蜜) may give rise to some side effects, allergic reaction or toxic symptoms in restoratives for invigorating qi(補氣藥). The representative methods of poisoning treatment in western medicines are stopping medication, washing out the stomach, promotion of vomiting, causing diarrhea, supplies of grape sugar and symptomatic treatment, etc. The representative methods of poisoning treatment in oriental medicine take advantage of various herbs. And Oriental medical doctor should meet symptoms as patients call for attention. In order to prevent against poisoning of restoratives for invigorating qi(補氣藥), the patients should keep usage, dosage and notes. Conclusion:We should pay attention to clinical using of Ginseng Radix(人參), Codonopsis Pilosulae Radix(黨參), Panacis Quinquefolii Radix(西洋參), Astragali Radix(黃芪), Atractylodis Rhizoma Alba(白朮), Dioscoreae Rhizoma (山藥), Dolichoris Semen(白扁豆), Glycyrrhizae Radix(甘草), Jujubae Fructus(大棗) and Mel(蜂蜜) in restoratives for invigorating qi(補氣藥).

황기의 메탄올 추출물의 용량에 따른 면역생물학적 연구 (Immunobiological Studies on Doses of Methanol Extract of Astragali Radix)

  • Effects of methanol extract of Astragali Radix (AR) on the immune responses were studied using ICR mice. Mice were divided into 4 groups (10mice/group), and methanol extracts of AR at doses of 0.05, 0.25 and 1.25g/kg were orally administered to ICR mice once a day for 2 weeks. Mice were immunized and challenged with sheep red blood cells (SRBC). The results of this study were summarized as follows; (1) Methanol extract of AR at 0.05, 0.25 and 1.25g/kg didn't affect the weight ratios of thymus to body, as compared with those in controls, but significantly increased spleen weight ratio. (2)Methanol extract of AR at 0.05 and 0.25g/kg significantly increased hemagglutination titer and splenic plaque forming cells corresponding to humoral immunity, as compared with those in controls, but their enhancements were somewhat lowered at a high dose (1.25g/kg). (3) Methanol extract of AR at 0.05 and 0.25g/kg siginificantly increased delayed-type hypersensitivity reaction resulted from cell-mediated immunity, as compared with those in controls, but not so significant increases were observed at a high dose (1.25g/kg). (4) Methanol extract of AR at 0.05 and 0.25g/kg significantly increased phagocytic activity and the number of circulating leukocyte compared with those in controls, but their enhancements were lowered at a high dose (1.25g/kg). These results suggest that methanol extract of Astragali Radix increased humoral and cell-mediated immune responses, phagocytic activity and the number of circulating leukocyte, dependent upon dose, but inhibited their enhancement effects were decreased at a high dose (1.25g/kg).
