Effects of Astragali Radix Herbal Acupuncture Extracts on the Collagenase Activity and Procollagen Synthesis in HS68 Human Fibroblasts and Tyrosinase Activity (황기 약침액의 사람 섬유아 세포에서 Collagen 생성과 Tyrosinage 활성에 미치는 영향)

  • 목적 : 본 연구는 황기약침액의 collagen 대사과정과 tyrosinage 활성에 대한 효과를 관찰하여 항노화 약침소재로의 개발 가능성을 알아보고자 계획되었다. 방법 : 황기 약침액이 사람 정상 섬유아세포 HS68에 UVB 조사 후 type I procollagen의 생산량 회복과 collagenage 활성에 미치는 효과를 ELISA법을 이용하여 측정하였다. 황기약침액의 tyrosinage 활성도에 미치는 영향을 측정하였다. 결과 : 황기 약침액은 사람 섬유아세포 HS68 실험에서 UVB 조사로 감소된 type I procollagen생성을 회복시키고, 통계적으로 유의하게 collagenage 활성을 억제하는 것을 관찰하였고 tyrosinage 활성을 통계적으로 유의하게 억제하지만, L-DOPA 산화는 억제하는 경향이 있는 것을 관찰하였다. 결론 : 본 연구 결과에 의하면, 황기약침액은 collagenage 활성을 억제하고, tyrosinage 활성을 억제하므로 주름개선과 미백효과가 있어 미용약침 소재로 개발가능성이 있을 것으로 사료된다.

Flowering Habit, Pollination Patterns and Seed Setting in Astragalus membranaceus Bunge (황기의 개화습성과 수분양식 및 결실 특성)

  • This study was carried out to obtain the basic informations on the flowering habit, pollination patterns and seed setting in Astragalus membranaceus. The first flowering appeared at the 20-22 node on main stem and there is the indeterminate inflorescence that the flowering progress developed toward the tip of the main stem and also toward the tips of branches. According to the flowering time, the number of peduncles were increased in early of Aug. Then, the number of bearing pods and that of seed setting were increased at that time. The length of peduncles with small flower was 6.6-7.3cm, and the number of small flowers were 13-17 per peduncles. The percentage of seed setting was 30-45% per peduncles and seeds were 4.8-5.3 per pod in peduncles. On 30 days after pollination, the pod was developed with the length about 3.8cm and the width about 8mm and the seeds were 7.3per pod. According to pollination treatment, wrapping sotted no seeds and the artificial pollination sotted seeds . about 5% in late of jul. to late of Aug. but increased about 13% of seed set after early of Sep. in A. membranaceus.

  • This study was carried out to find favourable soil conditions to give quality of Astragali Radix. Plants were grown in the soils of different physico-chemical properties, and ash, ethanol extract and free sugars from their roots were analyzed. Fructose and sucrose extracted from the root were determined by HPLC. The contents of Fe were 6.2~7.6 ppm. The ash contents of the root were positively correlated with the available $P_2O_5$ in the soils as well as P and K in the root.

  • Radix Astragali, the root of Astragalus (A.) membranaceus, has been applied in a variety of diseases for a long time in Asian countries such as Korea and China. In addition, the aerial parts such as leaves and stems of A. membranaceus have received a great deal of attention. Recently, the polysaccharide fraction showing a potent immunomoduating activity was isolated from the aerial parts of A. membranaceus. Thus, the aerial parts of A. membranaceus would be worthy enough for a food material and a dietary supplement. However, they should be safe even though valuable. In our previous study, it was estimated that NOAEL for female rats are 5000 mg/kg/day of the crude polysaccharide fraction from A. membranaceus-aboveground parts. As a series of safety evaluation, genotoxicity test for the crude polysaccharide fraction was carried out in this study. In conclusion, the three genotoxicity assays provided strong overall support that the crude polysaccharide fraction lacks mutagenic and/or clastogenic potential under the GLP-based test conditions. This indicates the aerial parts of A. membranaceus would be safe enough for a food material and a dietary supplement.

  • Astragali Radix, known as Huangqi, is the most important tonic in the traditional oriental medicine. It reinforces 'qi' (vital energy), strengthens the superficial resistance and promotes the discharge of pus and the growth of new tissue. It has long been used as an anti-perspirant, anti-diuretic or a tonic. Eleven compounds were isolated from the hexane and EtOAc fractions from the roots of Astragalus membranaceus (Leguminosae) and their structures were identified as four triterpenoids [lupenone (1), friedelin (2), lupeol (3), soyasapogenol E (9)] and seven sterols [${\beta}-sitosterol$ (4), stigmastane-3,6-dione (5), $7{\alpha}-hydroxysitosterol$ (6), $5{\alpha},6{\beta}-dihydroxysitosterol$ (7), $7-oxo-{\beta}-sitosterol$ (8), ${\beta}-sitosterol$ glucoside 6'-O-palmitate (10), ${\beta}-sitosterol$ glucoside (11)]. The chemical structures of these compounds were identified on the basis of spectroscopic methods and comparison with literature values. Among these compounds, lupenone (1), friedelin (2), lupeol (3), stigmastane-3,6-dione (5), $7{\alpha}-hydroxysitosterol$ (6), $5{\alpha},6{\beta}-dihydroxysitosterol$ (7), $7-oxo-{\beta}-sitosterol$ (8), soyasapogenol E (9), and ${\beta}-sitosterol$ glucoside 6'-O-palmitate (10) were isolated from this plant for the first time.

A Literature Review of Experimental Study about Herbal Medicine for Bone Growth (한약제제의 골성장 효과에 대한 국내외 실험 연구 고찰)

  • Objectives The purpose of this study is to investigate the effect of herbal medicine on bone growth and growth factors by collecting and analyzing domestic and foreign experimental research papers to establish a platform for future research. Methods Experimental studies published before March 2021 were searched using 7 Korean databases and 2 foreign databases. Among the numbers of studies searched, 48 experimental studies met the criteria and were selected. The results of those 48 studies upon herbal medicine administration were analyzed by bone growth, growth plate, weight. Results 23 studies mainly used monotherapy of either Astragali Radix and Cervi Parvum Cornu. 28 studies used combination of Poria Sclerotium, Dioscoreae Rhizoma, and Acanthopanacis Cortex. Among all studies selected, weight, bone growth, Insulin-like growth factor-1 (IGF-1) and Growth hormone (GH) were increased, but the increase was significant only in some of the studies. Conclusions The result of this study can be utilized as a background information for further studies and treatment in pediatric growth. Further randomized control studies are needed to underscore the finding.

Moisturizing and Anti-oxidation Effect of Astragalus membranaceus Root Extract (황기추출물의 보습 및 항산화 효과)

  • We investigated the effect of moisturizing and anti-oxidation of Astragalus membranaceus (Astragali Radix) root extract with respective to growing districts and extract methods for the purpose of development of cosmetic ingredients. Astragalii Radix was collected in Jecheon, Jeongseon, Taebaek, and Yeongju in Korea and China as growing districts. Formononetin was determinated by HPLC method as one of the various active agents in Astragalus membranaceus root extract. The 75% ethanol extract demonstrated to be more effective than $H_2O$-extracted one for a scavenging activities to DPPH radicals and reactive oxygens. The 75% ethanol extract showed $IC_{50}$ (50% scavenging concentration) of 2.162 mg/mL and 2.981 mg/mL in case of free radical scavenging activity test and reactive oxygen scavenging activity test, respectively Especially, free radical scavenging activity of isoflavonoids isolated from ethylacetate fraction was similar to scavenging activity of genistein. Astragalus membranaceus root extract of Jecheon district by sonicating extraction method was more effective in skin hydration compared with others.

The Growth Inhibiting Effect of Enterobacteria, Clostridium pefringens KCTC 5014, by the Combination of Natural Products (Clostridium perfringens KCTC 5014의 성장 억제를 위한 항산화 천연산물의 효과)

  • The growth of C. pefringens was inhibited by the combination of natural products which have antioxidative capacity C. perfringens was controlled very effectively by each or the combination of Paeonia japonica, Atractylodis rhizome alba, Epimedii herba, Acori graminei rhizoma, Schizandraefructus, Lyciifructus, Cinnamomi cortex Sophorae flos Phellodendri cortex, Nelumbo nuclfera gaertner, Asparagi tuber, Mori cortex radicis, Astrafali radix and Coptidis rhizome The combination of Phellodendri cortex, Epimedii herba, Astragali radtx and Glycyrrhizae radix inhibited the growth of C. pefringen by 75% and increased the antioxidative activity by 4.4 times. This combination also eliminated more than 80% of hydroxyl radical. These natural products can be utilized as pharmaceuticals, functional foods, antiaging tea, and multifunctional fermentative beverages.

A Review of Clinical Studies for Chinese Medicine Treatment of Idiopathic Thrombocytopenic Purpura Using the CNKI Database (특발성 혈소판 감소성 자반증의 중의치료에 대한 임상연구 동향 - CNKI검색을 중심으로)

  • Objectives: The aim of this study was to analyze the latest clinical studies on Korean medicine treatment of idiopathic thrombocytopenic purpura (ITP) in the Chinese National Knowledge Infrastructure (CNKI) database. Methods: We searched the last 6 years of clinical studies discussing Oriental medicine-based treatments for ITP in the CNKI database. A meta-analysis of 13 RCTs was performed by synthesizing the outcomes, including the measured platelet count and total effective rate. The quality of the studies was assessed using Cochrane's risk of bias (RoB) tool. RevMan 5.4.1 software was used for data analysis. Results: Of the 15 selected studies, 1 was a non-randomized controlled trial (nRCT), 2 were case series, and 12 were randomized controlled trials (RCTs). Treatments in all studies included oral herbal medicine. The most frequently used herbal decoction was the Liangxue Jiedu prescription (凉血解毒方), and the most commonly used herb was Agrimonia pilosa (仙鶴草), Astragali Radix (黃芪), Fossilia Glycyrrhizae Radix et Rhizoma (甘草), and Rehmannia glutinosa Liboschitz ex Steudel (地黃). The meta-analysis showed significantly better improvement in platelet counts and total effective rate for ITP in the treatment group than in the control group. Conclusion: Treatment with herbal medicine was effective in treating ITP. However, the significance of this conclusion is somewhat limited due to the low quality of the available studies. Multifaceted and scientifically designed clinical studies are required to develop treatments for ITP based on Korean medicine. The results of this study could be used as basic data for further ITP studies.

Study on Pharmacological Activity of Sipjeondaebotang by Difference in Component Ratio between Astragali Radix and Cinnamomi Cortex (십전대보탕에서 황기.육계의 함량 변화에 따른 약리활성 비교 연구)

  • Objective : Pamooltang (PM) and Sipjeondaebotang (SC) are used in Korea to treat many diseases such as sterility, menstrual disorder, and general prostration. We made a comparative study of PM and SC which are different in component ratio between Astragalus membranaceus BUNGE (AC) and Cinnamomum cassia PRESL. (CC). Methods : Anti-oxidation was studied by 1.1.-diphenyl-2-picrylhydrazyl (DPPH) assay and anti-inflammation was investigated by prostaglandin $E_2\;(PGE_2)$ and nitric oxide (NO) inhibition assay. For immune response activities, this study used NO synthesis on RAW 264.7 cells and splenocyte proliferation. Results : The results showed that PM and SC components had no significant effect of anti-oxidation or anti-inflammation. However, we observed their effects upon inducible NO synthesis in Raw 264.7 cells. The SC2 stimulated NO synthesis $11.42\pm1.36{\mu}M$ (control; $0.89\pm0.00{\mu}M$). PM and SC components had the effect of immune response which in a dose-dependent manner significantly induced the splenocyte proliferation. The splenocyte proliferation induced by SC2 was higher than others at the concentration of 1, 10, 100, 1000 and 2000 ${\mu}g/ml$. The SC8 was shown to up-regulate IgG, 100 ${\mu}g/ml$ 3.3 times, 1000 ${\mu}g/ml$ 2.6 times as a control. Conclusions : These results may have important implications for our understanding of the ratios of AC and CC in SC.
