Damping-off of Edible Aster Caused by Rhizoctonia solani AG-4

  • In August 2021, we surveyed diseases of wild vegetables grown in Taebaek, Gangwon Province, Korea. During the disease survey, we observed severe damping-off symptoms in young edible aster (Aster scaber) plants in a vinyl greenhouse investigated. The incidence of the disease in the plants ranged from 5% to 20%. Diseased plants of edible aster were collected from the vinyl greenhouse, and fungi were isolated from petiole lesions of the diseased plants. Rhizoctonia sp. was consistently isolated from the petiole lesions. We examined morphological characteristics and anastomosis groups of nine Rhizoctonia sp. isolates obtained from the petiole lesions. The examination results revealed that all the isolates corresponded to Rhizoctonia solani AG-4 based on the morphological characteristics and anastomosis test. Three isolates of R. solani AG-4 were tested for their pathogenicity on edible aster plants by artificial inoculation. Inoculation tests showed that the tested isolates caused damping-off symptoms on the inoculated plants. The induced symptoms were similar to those observed in the vinyl greenhouse investigated. Damping-off of edible aster caused by R. solani AG-4 is first reported in this study.

자생 초본류 몇 종의 종자특성과 발아율에 대한 연구 (A Study on the Seed Characteristics and Germination Percent of Several Native Herb Species)

  • 곰취를 비롯한 15종의 자생 초본류의 종자특성인 종자수득율, 천립중, 종자길이와 폭을 측정하였으며, 6종의 자생 초본류의 종자를 가을파종으로 임간 발아시험을 하였다. 참취를 비롯한 13종의 자생 초본류 종자가 7mm미만 의 길이와 3mm미만의 폭을 지닌 세립종자들이었다. 곰취를 비롯한 10종의 자생 초본류의 종자수득율이 5% 미만으로 매우 낮은 편이었다. 참나물을 비롯한 5종의 자생초본 발아율은 생육지간 통계적 유의차가 인정되었다. 평균 발아율은 참당귀가 47.00%로 가장 높았고, 곰취가 38.25%, 참나물이 30.88% 독활이 15.75%, 고려엉겅퀴가 10.00% 였고, 참취가 9.32%로 가장 낮았다.

Improvement of the Quality of Dried Wild Vegetables by Micro Oil Sprayed Thermal Air Technique

  • Wild vegetables, such as Cirsium setidens, and Aster scaber, are commonly distributed as dried materials because the wild vegetables lose their freshness quickly after harvest and distribution. Dried wild vegetables require rehydration to use as a food ingredient, and the quality of rehydrated wild vegetables is affected by pre-drying and drying methods. Here, we introduce the newly developed pre-drying and drying method, termed "micro oil sprayed thermal air (MOTA) technique". The three wild vegetables processed by the MOTA technique showed improved rehydration rate and reduced time to achieve maximum rehydration rate. Color characteristics were also improved in C. setidens. These results indicate that the MOTA technique improves the overall quality of rehydrated wild vegetables. It is expected that our findings could enhance the marketability of dried wild vegetables by improving overall quality and reducing preparation time.

참취(Aster scaber) 추출물의 생리활성 (Biological activities of Aster scaber extracts)

  • 참취로부터 phenolic compounds를 추출 후 항산화 효과, 건강 기능성 식품 및 미용식품 활성을 검정하여 기능성 소재로 활용 가능성을 살펴보았다. 참취에 함유되어 있는 페놀성 물질은 물과 60% ethanol을 용매로 하여 추출하였을 때 각각 11.1 mg/g, 4.18 mg/g 함량을 나타내었다. 추출물의 phenolic 농도를 $50{\sim}200{\mu}g/mL$로 조절하여 DPPH를 측정한 결과, 물 추출물과 ethanol 추출물 $50{\mu}g/mL$ phenolics 농도에서 각각 87%, 91%였고, ABTS radical decolorization도 $50{\mu}g/mL$ phenolics 농도에서 각각 99%, 99%였다. Antioxidant protection factor는 $200{\mu}g/mL$ phenolics 농도에서 물 추출물과 ethanol 추출물에서 각각 1.87 PF와 2.22 PF로 측정되었으며, TBARs값은 $50{\mu}g/mL$ phenolics농도에서 물 추출물이 95%, ethanol 추출물에서 97%를 나타내었다. Angiotensin converting enzyme(ACE) 저해능은 물 추출물에서 50.8%, ethanol 추출물에서 69.4%의 저해율을 나타내었으며, xanthine oxidase 저해능은 물 추출물은 $50{\mu}g/mL$ phenolics농도에서 91%, ethanol 추출물은 $200{\mu}g/mL$ phenolics농도에서 80%의 높은 효능을 보였다. ${\alpha}$-Amylase 저해능은 물 추출물과 ethanol 추출물에서 각각 24%, 89%로 ethanol 추출물에서 높은 효능을 보였다. Tyrosinase 저해능을 측정한 결과 물 추출물과 ethanol 추출물에서 각각 47%, 25%의 미백 효과를 나타내었다. Elastase 저해능 측정 결과 물 추출물과 ethanol 추출물에서 각각 46%, 14%로 물 추출물에서 주름 개선 효과가 상대적으로 높게 나타내었다. 이러한 결과로 보아 참취 추출물은 항산화 고혈압억제, 통풍억제, 당분해억제, 미백효과, 주름개선효과 등의 건강 기능성 식품 및 미용식품 활성 소재로서 활용 가능성이 큰 것으로 판단되었다.

Extracts of Aster species Inhibit Invasive Phenotype and Motility of H-ras MCF10A Human Breast Epithelial Cells Possibly via Downregulation of MMP-2 and MMP-9

  • Cancer metastasis represents the most important cause of cancer death and antitumor agents that may inhibit this process have been extensively pursued. Invasion and metastasis of malignantly transformed cells involve degradation of the extracellular matrix (ECM) components by matrix metalloproteinases (MMP), especially MMP-2 and -9. We previously showed that H-ras-induced invasive phenotype may involve MMP-2, rather than MMP-9, in MCF10A cells. In the present study, we investigated the chemopreventive effect of Aster, a widely used culinary vegetable in Korea. We screened twelve extracts from three Aster species (Aster scaber, Aster oharai and Aster glehni) for the inhibitory effect on MMP activities of H-ras MCF10A human breast epithelial cells. All of the extracts tested in this study efficiently inhibited the gelatinolytic activities of MMP-2 and MMP-9. A more prominent inhibition was observed in MMP-2 activity compared to MMP-9. Out of twelve extracts, eight extracts showed>90% inhibition of MMP-2 activity in H-ras MCF10A cells while only one extract showed>90% inhibition of MMP-9 activity. We selected three extracts (AO-3, AG-3 and AS-EA) for further studies since they exerted a marked inhibition in the ratio of MMP-2 to MMP-9. Treatment with AO-3, AG-3 and AS-EA in H-ras MCF10A cells caused a significant inhibition of invasive phenotype and migration, proving a chemopreventive potential of these extracts. Taken together, our results demonstrate that extracts of Aster effectively inhibit invasion and migration of highly malignant human breast cells, possibly via downregulation of MMP-2 and MMP-9.

재배환경에 따른 가공식품 소재로서 참취의 품질 비교 (Quality assessment of Aster scaber cultured at different conditions for processed food production)

  • 산채류의 생리활성에 대한 소비자 관심이 증가함에 따라 산채류의 소비가 늘고 있지만 아직 수확시기 외에는 신선한 산채의 섭취가 어려운 실정이다. 특히 바쁜 현대인들의 직접적인 조리는 더욱 어렵기 때문에 가공식품으로의 이용이 증가하고 있다. 본 연구에서는 주요 산채작물인 참취의 재배 방법에 따른 품질비교와 데치기 처리 전후의 물리화학적 특성 변화를 분석하였다. 노지재배 참취는 시설재배에 비해 총 페놀성 화합물 양은 물론 산화방지능도 우수한 것으로 나타났다. 플라본올 중에서는 특히 퀘세틴이 시설재배에 비하여 5배 이상 높은 것으로 나타났다. 데치기 처리 시 경도와 영양성분, 기능성성분의 감소를 보였으나 그 변화 정도에 있어 재배방법에 따른 차이는 뚜렷하지 않았다. 색도의 경우 노지재배 참취가 시설재배에 비하여 확연히 진한 색을 나타내었는데 데치기 전후의 색도변화는 두 시료 모두 크지 않은 것으로 나타났다. 자유당 함량은 노지재배 참취가 두 배 가량 많은 것으로 나타났는데, 특이적으로 엿당은 검출되지 않았다. 결과를 종합하여 보면, 노지재배 참취의 영양적 가치가 시설재배에 비해 월등히 높은 것으로 판단되며 데치기 처리에 따른 성분변화와 색도변화 양상은 재배방법에 따른 차이가 뚜렷하지 않은 것으로 나타났다.

Blanching 조건과 가염조건에 따른 참취의 품질 특성 변화 (Effects of Blanching Conditions and Salt Concentrations on the Quality Properties of Aster scaber)

  • 참취의 적정 blanching 조건을 설정하고자 온도와 시간을 변수로 한 여러 blanching조건에서 식염의 첨가량 변화에 따른 취나물의 품질특성변화를 조사하였다. 취나물의 자숙 공정에서 식염을 가하지 않은 대조구의 경우에는 blanching 처리 후 비타민 C, 클로로필 및 페놀함량 및 표면경도가 각각 $39.5{\sim}60.3%$, $26.2{\sim}31.5%$, $45.6{\sim}50.1%$, $50.7{\sim}51.1%$로 감소경향이 크게 나타났으나 1%의 식염을 첨가한 경우에는 식염을 첨가하지 않은 대조구에 비하여 각각 $7.1{\sim}30.2%$, $9.2{\sim}12.2%$, $35.9{\sim}52.0%$, $2.7{\sim}6.8%$의 품질손실 억제효과와 더불어 drip도 $3.3{\sim}23.4%$의 억제할 수 있었다. 또한 식염량을 2% 첨가한 경우에는 대조구에 비하여 $22.7{\sim}34.1%$, $10.4{\sim}20.7%$, $55.5{\sim}80.4%$, $13.8{\sim}17.3%$의 품질 손실억제효과와 $23.9{\sim}27.3%$의 drip수 발생량을 억제하는 효과를 나타내어 1%의 식염량 첨가구보다 우수한 품질보존효과가 있었다. 그러나 식염 첨가량에 따른 관능검사 결과 2%의 가염 처리구에서는 짠맛의 강도가 높게 인식되어 종합적인 기호도에서 대조구보다 낮게 평가되고 있었던 반면 1%의 가염처리구에서는 짠맛의 인식도가 보통으로서 대조구보다 종합기호도도 우수한 것으로 평가받았으며, blanching조건별로는 $100^{\circ}C$/6분, 1% 식염첨가구가 가장 우수한 것으로 나타났다.

깻잎, 쑥, 참취의 건분 및 에탄올 추출물이 흰쥐의 지방대사와 항산화능에 미치는 영형 (Effect of Dried Leaf Powders and Ethanol Extracts of Perilla Frutescens, Artemisia Princeps Var. Orientalis and Aster Scaber on Lipid Metabolism and Antioxidative Capacity in Rats)

  • This study was performed to investigate the effects of dried leaf powders and ethanol extracts of Perilla frutescens(perilla leaf), Artemisia princeps var. orientalis(mugwort) and Aster scaber(chamchui) on lipid metabolism and antioxidative capacity in rats. Forty-nine male Sprague-Dawley rats weighing 105.9$\pm$1.7g were blocked into seven groups according to body weigth and raised for four weeks with diets containing 5% dried powders of perilla leaf, mugwort and chamchui, or ethanol extracts from the same amount of each dried leaf powder. Food intake was higher in the control and chamchui powder groups than the other remaining groups. Weight gain was not significantly different among all experimental groups. Food efficiency ratio was highest in the perilla leaf powder group. Plasma total lipid and cholesterol, liver total lipid and triglyceride levels were highest in the perilla leaf powder group. In contrast, dried powders of mugwort and chamchui showed hypolipidemic effects in plasma and liver. Plasma and liver TBARS levels in both dried powder and ethanol extract groups of the three plants were lower than control. There was also no significant difference between corresponding dried powder and ethanol extract groups. Catalas and GSH-Px activities in erythrocyte and liver were not different among all the experimental groups. However, SOD activities were significantly different among the esperimental groups. In erythrocyte. SOD activiteis of all dried powder and ethanol extract groups except the perilla leaf powder group were higher than control, and the chamchui power group showed the highest activity among them. In liver, all the plant groups showed higher SOD activities than control. There was no significant difference between corresponding dried powder and ethanol extract groups. In conclusion, the dried chamchui powder and liver. All the dried plant powder and ethanol extract were effective in decreasing the TBARS levels of liver and particularly plasma. Among the antiocidative enzymes, SOD activity was most responsive to the experimental diets. All the plant groups showed increases in liver SOD activities and there were more increments in dried powder groups. Since the effects of dried powder groups on lipid metabolism were better than those of extract groups, it is plausible that the high dietary fiber level in dried powders was effective. Antioxidative effects were not significantly different between corresponding dried power and extract groups, and it was thought that ethanol extracts from three plants also had similar antioxidative effects as dried powderers.
