The Effects of Lactic Acid Bacteria on the Growth and Aflatoxin $B_1$ Production of Aspergillus parasiticus (유산균이 Aspergillus parasiticus의 성장과 Aflatoxin $B_1$ 생성에 미치는 영향)

  • This study was performed to investigate the competitive effect of Leuconostoc mesenteroides and Lactobacillus plantarum on the growth and aflatoxin $B_1$ production of Aspergillus parasiticus. Leu. mesenteroides and Lac. plantarum were grown with A. parasiticus in a modified APT broth at $28^{\circ}C$ for 9 days. It was observed that the growth of A. parasiticus got lower when the two bacteria were inoculated simultaneously than when the mold grew alone. The pH of mixed culture of Leu. mesenteroides and Lac. plantarum showed significantly lower value than the pH of the pure culture of A. parasiticus (p<0.05). The acidity of the mixed culture group significantly increased compared with the control group (A. parasiticus alone group) (p<0.05). The aflatoxin $B_1$ production was significantly decreased in the mixed culture group than in the A. parasiticus alone group (p<0.05). Leu. mesenteroides showed the more efficient effect than Lac. plantarum. These results indicate that the two lactic acid bacteria have inhibitory effect on the growth and aflatoxin $B_1$ production of Aspergillus parasiticus.

Are Current Aspergillus sojae Strains Originated from a Native Aflatoxigenic Aspergillus Species Population Also Present in California?

  • Aspergillus sojae has long been considered a domesticated strain of Aspergillus parasiticus. This study delineated relationships among the two species and an Aspergillus PWE36 isolate. Of 25 examined clustered aflatoxin genes of PWE36, 20 gene sequences were identical to those of A. sojae, but all had variations to those of A. parasiticus. Additionally, PWE36 developmental genes of conidiation and sclerotial formation, overall, shared higher degrees of nucleotide sequence identity with A. sojae genes than with A. parasiticus genes. Examination of defective cyclopiazonic acid gene clusters revealed that the PWE36 deletion pattern was identical only to those of A. sojae. Using A. sojae SMF134 genome sequence as a reference, visualization of locally collinear blocks indicated that PWE36 shared higher genome sequence homologies with A. sojae than with A. parasiticus. Phylogenetic inference based on genome-wide single nucleotide polymorphisms (SNPs) and total SNP counts showed that A. sojae strains formed a monophyletic clade and were clonal. Two (Argentinian and Ugandan) A. parasiticus isolates but not including an Ethiopian isolate formed a monophyletic clade, which showed that A. parasiticus population is genetically diverse and distant to A. sojae. PWE36 and A. sojae shared a most recent common ancestor (MRCA). The estimated divergence time for PWE36 and A. sojae was about 0.4 mya. Unlike Aspergillus oryzae, another koji mold that includes genetically diverse populations, the findings that current A. sojae strains formed a monophyletic group and shared the MRCA with PWE36 allow A. sojae to be continuously treated as a species for food safety reasons.

  • Effect of allylisothiocyanate on the enzyme activites including malate degydrogenase, isocitrate dehydrogenase, NADPH and acetyl CoA which were related to aflatoxin production of Aspergillus parasticus R-716 were invetigated. The activities of malate dehydrogenase (EC.1.1.1.37), isocitrate dehydrogenase (E.C.1.1.1.42) and NADPH oxidase (E.C.1.6.99.1) indicated relatively high in the 50 ppm allylisothiocyanate-added-culture. In contrast, the activity of acetyl CoA in the 50 ppm allylisothiocyanate-added-culture showed rather lower level through the cultivation.

  • The effect of allyisothiocyanate, the mahor compound of radish on the growth of Aspergillus parasiticus R-716 and aflatoxin production, was investigated. An increase in the level of allylisothiocyanate results in a decrease both growth and aflatoxin per myclial weight, and the addition of 125 ppm allylisothocynate completely inhibited the growth of the strain. The addition of allylisothiocyanate to the culture of R-716 strain the production of aflatoxin. The inhibition of aflatoxin was more B-group than G-group and M-group during cultural period. The growth of strain and aflatoxin production were greatly affected by the addition of allylisothiocyanate.

  • The effects of allylisothiocyanate on the biosynthesis of various fungus metabolites such as sterigmatocystin, lipid, protein, citrate RNA and AMP from the culture of Aspergillus Parasiticus R-716 were investigated. The content of sterigmatocystin, the precursor of aflatoxin, was lower in the culture added with 50ppm allylisothicoyanate after 48 hours, however was rather higher after 144 hours compared to that of the control. The addition of allylisothiocyanate resulted in the increase of lipid, protein, RNA in mycelium and the content of citrate in the media, but the amount of AMP was low.

  • The possible effects of garlic (Aliium sativurn L.) extract on growth and aflatoxin production by Aspergillus parasiticus R- 716 were investigated. Various solvent extracts of garlic strongly inhibited growth and sporulation by Aspergillus parasiticus R-716, and effective solvents used for extraction of garlic were chloroform, benzene, and water-chloroform. The growth and aflatoxin production decreased with the increase in extract concentration, and extract equivalent 1.5g of raw garlic weight in 25ml SLS medium completely inhibited, and at a level of 1.25g garlic, total aflatoxin was reduced 64% (472 ${\mu}g$/25ml) of that produced in the control (1, 352 ${\mu}g$/25 ml). During cultivation inhibitory rate of growth was reduced from 89.1% to 40% and aflatoxin $B_2$, $G_1$ production increased with the laps of time. Especially garlic extract appeared to have a stimulatory effect on lipid accumulation on the contrary aflatoxin production.

  • In this study, th effective extraction of 5 vegetables, which have been shown to inhibit the growth of the aflatoxin production of Aspergillus parasiticus R-716, was investigated and their effects were examined. Radish, cabbage, garlic and zinger were effectively with water-chloroform, but crown daisy with n-hexane. Among them, water-chloroform extract of radish was remarkably effective, and garlic extract only inhibited the growth strongly. The growth and the aflatoxin production of the strain were showed 0.758g/25ml, 763ug/25ml with the addition of water-chloroform extract equivalent 15g of raw radish, and an increase in the level of radish extract resulted in a decrease both the growth and the aflatoxin production per mycerial weight.

  • ABSTRACT-This study was designed to observe the effects of temperature cycling on the aflatoxin production by Aspergillus parasiticus R-716 in modified SLS medium. Temperature cycling resulted in total aflatoxin production more than did constant incubation at either $28^{\circ}C$, which was considered to be optimum for aflatoxin production, or $17.5^{\circ}C$, which had the same total thermal input as the temperature cycling. The aflatoxin biosynthesis correlated with the color intensity of media, but was controversal with lipid biosynthesis, and aflatoxin concentration is not related to changes in the fatty acid compositions of used strain.strain.

  • 21 herbal drugs registered in K.P. III were tested for contamination of fungi and isolation of aflatoxin producible strains. Initially contaminated fungi were Aspergillus group (41.28%) and Penicillium (47.26%) and the other fungi were contaminated somewhat. The most frequent isolation of Aspergillus group was Cnidii Rhizoma and that of Penicillium was Piperis Fructus nigri. Cnidii Rhizoma was the most contaminated drug and Cassiae Cortex was the least among them. Aspergillus flavus was isolated from 10 samples and Aspergillus parasiticus was detected in Glycyrrhizae Radix, Phellodendri Cortex. Aspergillus ochraceus was isolated from only Scutelariae Radix, and Fusarium nivale was isolated from Cnidii Rhizoma and Torreya Semen. None of Aspergillus and Penicillium was detected in only Coptidis Rhizoma. No strains of Aspergillus flavus and Aspergillus parasiticus isolated from were produced aflatoxin.

  • Author investigated fluorence antibody reaction for the antigenic relationships between Asp niger group, Asp flavus and Asp parasiticus which was indicated as follows: 1. It was concluded that there are complete differences in the antigenic properties each other because it has not cross reaction, therefore identification of strains will be simpley classified. 2. A complete cross reaction between Asp flavus and Asp parasitic us in the Asp flavus groups existed, accordingly this reaction could not identified the strain and classified between Asp. flavus and Asp. parasiticus. 3. This experiment also followed with the separated each strains from the origin (Meju, Nuruk, ATCC, NRRL), but there no differences. From the above results, this method could be classified between Asp flavus group and Asp niger group in the genus Aspergillus, but classification of Asp. flavus and Asp. parasiticus should hardely conclude with this method.
