In order to improve the quality of Takju, the effect of combined use of Aspergillus oryzae and Aspergillus kawachii in brewing was investigated. The quality of Takju which was brewed by the combined use of Aspergillus oryzae CF7-koji and Aspergillus kawachii CF1-koji in equal amount was better as cmapared with that brewed by existing method using Aspergillus kawachii-koji only as koji. But the good result did not obtained when the koji was made by mixed culture of Aspergillus oryzae CF7 and Aspergillus kawachii CF1.

This study examined the physicochemical characteristics of mashing prepared using different Nuruks (traditional Nuruk, Nuruk added Aspergillus oryzae, Nuruk added Aspergillus kawachii, and Nuruk added Aspergillus oryzae and Aspergillus kawachii), and to obtain the basic data for Korean Cheongju production. The four different mashes were compared for their brewing characteristics and indicated a normal fermentation pattern. The Nuruk made from A. oryzae or A. kawachii showed higher fermentation ability than the other samples tested. In addition, the analyzed results of fermented mashing showed that the Nuruk made from A. oryzae or A. kawachii had a significantly higher aroma profile, such as higher alcohol and ester than the other samples tested, whereas the Nuruk made from a mixture of A. oryzae and A. kawachii had a much higher organic acid content. In conclusion, the Nuruk prepared using A. oryzae or A. kawachii had a positive effect on the complete fermentation and enhanced aroma compounds for Korean Cheongju production.

Aspergillus kawachii extracellular pectinases were screened in liquid cultures with different carbon sources. The fungus grown on citrus pectin or lemon pomace produced at least one of these inducible pectinases: acidic polygalacturonase, pectin lyase, pectin methylesterase, $\alpha$-L-arabinofuranosidase, $\alpha$-1,5-endoarabinase, $\beta$-D-galactosidase/exogalactanase, and $\beta$-1,4-endogalactanase. The lemon-pomace filtrates also contained significant $\alpha$-L-rhamnosidase and $\beta$-D-fucosidase activities. Most of the screened pectinases were active at pH 2.0-2.5, indicating that the A. kawachii enzymes were acidophilic. Under the culture conditions employed we could not detect enzymatic degradation of soybean rhamnogalacturonan. The A. kawachii pectinase-production-related regulatory phenomena of induction-repression resemble those described for other Aspergillus sp.

The objective of this experiment was to improve the quality of Kochujang and to shorten the aging time of it by using the Kojis of various Aspergillus strains. During the autumn season Kochujangs were mashed with Koji of Aspergillus oryzae, Aspergillus kawachii and Aspergillus shirousamii. Chemical composition of the Kochujang was determined during the aging period. There was no significant difference in quality and component between the Kochujang made with Aspergillus oryzae cultured Koji and Aspergillus kawachii cultured Koji. But the Kochujang with Aspergillus shirousamii cultured Koji was better in flavor, color and amino nitrogen contents than the Kochujang with Aspergillus oryzae cultured Koji, though two Kochuangs were almost the same in general components. The significant difference between the two tested Kochuangs was observed in pH, and the pH of the Kochujong with Aspergillus shirousamii cultured Koji was extremely low during the aging period. Thus it is presumed that Aspergillus shirousamii Koji can be used for the improvement of quality of Kochujang if a suitable method is applied to prevent pH from lowering.

주류는 미생물에 의한 효소작용으로 원료 성분이 분해됨으로서 생성되는 당분, 아미노산, 유기산등의 맛 성분과 발효과정 동안 생성되는 휘발성 성분들이 조화되어 상당히 다양한 성분들이 향미에 관여하므로 향기의 주성분 규명이나 향기성분을 이용한 품질의 규격화가 매우 어려운 문제로 보여진다. 술덧의 안전한 발효와 잡균오염을 방지하여 품질이 균일한 술을 생산할 목적으로 재래누룩과는 별도로 Aspergillus kawachii등의 균을 이용한 개량곡자(koji)가 술 제조시에 많이 이용되고 있으나 이러한 방법은 탁주의 대량생산과 수율은 증가시켰으나 다양한 품질이나 맛의 향상은 기대하기 어렵다. 본 실험은 재래누룩에서 분리된 곰팡이 10종과 대조균주로 Aspergillus kawachii CF1002 그리고 효모 Saccharomyces cerevisiae를 이용하여 술을 담금하여 그 제품의 headspace gas를 포집하여 GC/MS로 휘발성 향기성분을 분석하였으며 또한 관능평가를 실시하여 검토하였다. 각 균주별로 총 27∼40종의 향기성분이 확인되었으며 전체 시료에서 알콜(peak area% 52.3 9∼70.36%), 알칸류(3.49∼14.17%), 에스테르류(3.23∼7.37%), 유기산류(0∼2.85%), 알데히드류(0∼0.44%)의 순으로 함유량이 높은 것으로 나타났다. GC/MS를 통한 향기성분의 분석 결과 각각의 선발 균주로 제조된 술의 향기성분의 패턴이 서로 상이하였으며 이것은 통계분석에 의해 그 특징이 유의적으로 분석되어 Aspergillus sp. SH-607, Aspergillus sp. SH-613, Aspergillus sp. SH-667, Aspergillus sp. SH-696 그리고 Aspergillus sp. SH-412는 기호도가 높은 것으로 관찰되었다. 수집된 우수균주 10종과 Aspergillus kawachii등으로 담금하여 제조한 술 시료 11종류를 대상으로 하여 쓴맛, 미묘한 맛, 떫은 맛, 신맛, 좋은 맛의 정도를 5단계로 나누어 관능검사를 실시한 다음 자료를 Duncan's multiple range test로 분석한 결과 전체적으로 좋은 맛에 대한 기호도는 Aspergillus sp. SH-607 시료와 Aspergillus sp. SH-412, Rhizopus sp. SH-606, Aspergillus sp. SH-613, Rhizopus sp. SH-654, Aspergillus sp. SH-696 Aspergillus kawachii 시료가 유의성 있게 좋은 맛을 나타내 기호도가 높았으며 맛과 기호도가 가장 좋았던 것은 Aspergillus sp. SH-607 시료로 나타났다.
Aflatoxin accumulation by Aspergillus flavus in rice was inhibited by A. kawachii and A. Shirousamii so that the rate of toxin accumulation and the maximum concentration of accumulated aflatoxins were considerablly reduced, although the initiation of aflatoxin accumulation was not affected. The maximal accumulated aflatoxin $B_1$ in rice by A. flavus at $28^{\circ}C$ and 85% RH was $40{\mu}g/50g$ rice after 35 days. Under the same condition but the additional inoculation of A. kawachii, $25{\mu}g\;of\;aflatoxin\;B_1$ was accumulated maximally in 50g rice after 45 days. When A. shirousamii was inoculated simultaneously with A. flavus on rice, however, only trace levels of aflatoxins were detected throughout 60 days of storage. Aflatoxins added to rice were reduced by 97% with A. kawachii and by 98% with A. shirousamii after 7 days during rife koji preparation. They were also reduced after 48 Hours of incubation by 30-67%, with A. kawachii koji and by 16-75% with A. shirousamii koji.
This study was carried out to investigate the influences of cultural conditions of koji on the production of saccharogenic amylase during rice-koji making by Aspergillus awamori var. kawachii which is now widely used as koji-mold in brewing Tikju and Yakju in Korea. The optimum cultural temperature for the production of saccharogenic amylase by this mold was 36$^{\circ}C$, and at this temperature it needed 40 hours of cultivation for maximum production of this enzyme. It was favorable for high production of both organic acid and saccharogenic amylase to shift the cultural temperature form initial 36$^{\circ}C$ to 32$^{\circ}C$ after 20~25 hours of cultivation. The production of saccharogenic amylase was low when the water content of steamed rice was below 35%, but its production was high at 40~60% of water content. When the quantity of conidial inoculation was too small, the production of saccharogenic amylase was low in initial phase, but it was retrived after 40 hours of cultivation. When koji-thickness was over 3cm, the production of saccharogenic amylase was markedly restricted. The saccharogenic amylase of this koji was stable at pH 2~7, and showed high activity at pH 2~5.
The aflatoxin production by Aspergillus ATCC 15517 decreased in the mixed culture with Aspergillus awamori, Aspergillus kawachii, Aspergillus niger, Aspergillus oryzae, or with Aspergillus shirousamii to 1.3%, 13.8%, 1.3%, 0.7%, or 38.5% of that of monoculture respectively. These koji molds degraded $75%{\sim}100%$ of added aflatoxin $B_1\;(791{\mu}g/50ml\;YES medium)$. A. awamori secreted during growth aflatoxin degrading factor(s) which was heat-labile. The degraded aflatoxin by the factor(s) showed no toxicity against Bacillus megaterium NRRL B-1368.
Six strains of mold which had high saccharifying and acid-productive ability were isolated from wild sources. The strain M-80 among them and Aspergillus kawachii, which was generally used as rice makkulli koji were used for this studies. The results obtained were summarized as follows. 1) $\alpha$-amylase activities of the strain M-80 and Asp, kawachii showed similarly as about 140W. V., while $\beta$-amylase activity of M-80 was 54 A. U. and Asp. kawachii was 40 A. U. 2) Acid protease activity of M-80 was higher then Asp. kawachii but alkaline protease activity was lower Asp. kawachii respectively. 3) The contents of total acid, ethanol and fusel oil in makkulli brewing with M-80 were higher those of Asp. kawachii and methanol contents of the tested two strains were about 33mg/%. 4) Fifteen kinds of free amino acid were detected from makkulli brewing of two strains, and free amino acid contents of M-80 were 10% higher than those of Asp. kawachii.
Biological activities (${\alpha},{\alpha}'$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) free radical scavenging activity, fibrinolytic activity and reducing power) and biochemical properties (protein content and electrophoretical protein patterns) were examined in solid state fermentation with Bacillus subtilis and Aspergillus kawachii using silkworm powder (SP) as substrate. The highest protein contents and free radical scavenging activities were seen in the SP fermented for 12 days with B. subtilis and A. kawachii, and these were in a time-dependent manner. The highest reducing power was seen in the SP fermented for 6 days with B. subtilis and for 12 days with A. kawachii, respectively. The highest fibrinolytic activities were seen in silkworm fermented for 6 days with B. subtilis and A. kawachii, but this activity was higher in the A. kawachii fermented SP than that of B. subtilis. When total protein patterns were analyzed by SDS-polyacrylamide gel electrophoresis (PAGE), the proteins of the SP fermented with B. subtilis for 3 days were completely degraded, while the protein degradation in the SP fermented with A. kawachii occurred after 12 days and this degradation increased proportionally to culture time. As a result, the SP fermented with both B. subtilis and A. kawachii showed higher fibrinolytic activities after 6 days of fermentation and antioxidative activity after 12 days, indicating that physiological activities of the fermented SP using these strains were highly improved compared to the unfermented SP, and that this compound could be a candidate material as a dietary supplement of healthy functional foods.
