  • Fecal malodor is an acute environmental issue to be solved for the intensive animal agriculture in Japan. Among these substances volatile sulfur such as hydrogen sulfide (HS), methanethiol, and dimethyl sulfide, and dimethyl disulfide are the ones most strictly controlled in the Japanese national regulations. In this experiment, we have screened a range of standard strains of chemoheterotrophic bacteria and of the presently isolated soil bacteria for their capacity to decompose HS. We have demonstrated that Comamonas testosteroni $JCM5832^T$ and our isolate Bacillus sp. had a potential to reduce malodor when applied to the pig feces.

  • The genetic diversity of domestic quail and two wild quail species, Japanese (Coturnix coturnix)and Common quail (Coturnix japonica), found in China was studied using microsatellite DNA markers. According to a comparison of the corresponding genetic indices in the three quail populations, such as Polymorphism Information Content (PIC), Mean Heterozygosity ($\bar{H}$) and Fixation Index, wild Common quail possessed rich genetic diversity with 4.67 alleles per site. Its values for PIC and $\bar{H}$ were the highest, 0.5732 and 0.6621, respectively. Domestic quail had the lowest values, 0.5467 and 0.5933, respectively. Wild Japanese quail had little difference in genetic diversity from domestic quail. In addition, from analyses of the fuzzy cluster based on standard genetic distance, the similarity relationship matrix coefficient between wild Japanese quail and domestic quail was 0.937, and that between wild Common quail and domestic quail was 0.783. All of these results showed that the wild Japanese quail were closer to the domestic quail for phylogenetic relationship than wild Common quail. These results at the molecular level provide useful data about quail's genetic background and further supported the hypothesis that the domestic quail originated from the wild Japanese quail.

  • Anaerobic chytridiomycete fungi are now well recognized as one of the major components of rumen microflora. Since the discovery of anaerobic fungi, the knowledge upon their morphology and physiology has been accumulated. It is certain that they gave roles in ruminal fiber digestion, although their quantitative contribution to rumen digestion is still unclear. Their role in fiber digestion is complicated by the dietary factors and the interaction with other microorganisms. We aim at reviewing such information in this article. Considerable attention gas been paid to the polysaccharidase of these fungi. Analysis on the fungal genes encoding these enzymes has been performed in several laboratories. This article also covers the genetical analysis of fungal polysaccharidases.

  • Though a variety of non-conventional feedstuffs have been successfully evaluated in research stations in Asian countries, there had been little adoption of these technologies at the small farm level. The methodology and results of three separate on-farm trials evaluation the possible use of some non-conventional feedstuffs in broiler diets under small farm conditions in Sri Lanka are reported in this paper. In all trials, the commercial mash, that in normally used in the farms, served as the control. In trials 1 and 2, 5 and 10% cassava leaf meal, respectively, was substituted (w/w) for the commercial mash and fed to broilers for six weeks. In trial 3, several non-conventional feedstuffs (cassava leaf meal, rubber seed meal, ipil ipil leaf meal and died poultry manure) were substituted (w/w) for the commercial mash at levels of 15-20%. The results demonstrated that these non-conventional feed resources can be used in broiler diets under small farm conditions, with no adverse effects on performance. The salient features of on-farm animal research are highlighted.

  • The use of Korean native chicken is increasing, and the discovery of new genetic resources is very important from both economic and genetic conservation points of view. In this study, mtDNA D-loop sequences from 272 privately-owned Korean native chickens from a Hyunin farm were investigated. Seventeen nucleotide substitutions were identified from the sequence analysis and they were classified as 6 haplotypes. Previously investigated haplotypes in five Korean native chicken populations have been compared with the Hyunin chicken population. The results indicated that two haplotypes, H10 and H15, in the Hyunin chicken population were not previously identified in other Korean native chicken populations, representing 33.09% (90/272) and 1.1% (3/272) of the Hyunin population, respectively. On the other hand, four other haplotypes were identical to those of a previous study of Korean native chicken populations. This result is indicative of conservation strategies of Hyunin chicken populations for expanding the genetic diversity in the Korean native chicken population.

  • In current animal production in Japan, a large surplus of nitrogen and phosphorus is given to animals as their feed which are mostly imported from outside of our own country. Today, an excess of nitrogen and phosphorus from animal manure has been spread out of the area of animal production and the surroundings. These components have become the major reason for eutrophication of ground, surface and inland water. Nutritional studies for the reduction of nitrogen and phosphorus from animal waste has been done by many researchers. The reduction of excess protein in animal feed and the supplementation of deficient essential amino acids to feed have a possibility to increase the biological value of feed and to reduce nitrogen excretion, especially, via urine. The use of phytase activity to degrade phytate and to release utilizable inorganic phosphorus make it possible to cut an excess supply of feed additive inorganic phosphorus and to reduce phosphorus excretion from animal waste.

  • The objective of this study was to investigate single nucleotide polymorphisms (SNPs) in the bovine nephroblastoma overexpressed (NOV) gene and to evaluate whether these polymorphisms affect carcass traits in the Korean cattle population. We resequenced to detect SNPs from 24 unrelated individuals and identified 19 SNPs within the full 8.4-kb gene, including the 1.5-kb promoter region. Of these 19 SNPs, four were selected for genotyping based on linkage disequilibrium (LD). We genotyped 429 steers to assess the associations of these four SNPs with carcass traits. Statistical analysis revealed that g.7801T>C and g.8379A>C polymorphisms in the NOV gene were associated with carcass weight (p = 0.012 and 0.008, respectively), and the g.2005A>G polymorphism was associated with the back fat thickness (BF) trait (p = 0.0001). One haplotype of the four SNPs (GGTA) was significantly associated with BF (p = 0.0005). Our findings suggest that polymorphisms in the NOV gene may be among the important genetic factors affecting carcass yield in beef cattle.

  • Bovine blastocysts were obtained by in-vitro culture of embryos derived from in-vitro fertilization of oocytes matured in-vitro. These blastocysts and blastocysts from inseminated donors were bisected by a simple method (without a holding pipette) using a microblade operated by a micromanipulator. A pair of demi-embryos was transferred nonsurgically into each uterine horn of a recipient cow 6 or 8 days after estrus. Pregnancy resulted from the third transfer. Ultrasound examination done 52 days after estrus (46 days after transfer) confirmed the presence of at least one fetus in the each uterine horn. This is the first report to show the viability of bisected bovine blastocysts obtained from in-vitro culture of embryos derived from in-vitro fertilization of oocytes matured in-vitro. In addition, a simple method to bisect bovine embryos is described.

  • To develop an efficient DNA typing system for Chinese Holstein cattle, 17 microsatellites, which were amplified in four fluorescent multiplex reactions and genotyped by two capillary electrophoresis injections, were evaluated for parentage verification and identity test. These markers were highly polymorphic with a mean of 8.35 alleles per locus and an average expected heterozygosity of 0.711 in 371 individuals. Parentage exclusion probability with only one sampled parent was approximately 0.999. Parentage exclusion probability when another parent' genotype was known was over 0.99999. Overall probability of identity, i.e. the probability that two animals share a common genotype by chance, was $1.52{\times}10^{-16}$. In a test case of parentage assignment, the 17 loci assigned 31 out of 33 cows to the pedigree sires with 95% confidence, while 2 cows were excluded from the paternity relationship with candidate sires. The results demonstrated the high efficacy of the 17 markers in parentage analysis and individual identification for Chinese Holstein cattle.

  • The genetic diversities of 10 Chinese pig populations were analyzed by using microsatellite DNA polymorphisms. The results showed that the mean heterozygosities of the 10 populations were between 0.4561 and 0.6446, the mean polymorphism information contents were 0.4241-0.6184 and the mean effective number of alleles were 2.4295-3.7573. These indicated that the genetic diversity of local Chinese pigs was high. The clustering of the 10 populations was nearly inaccordance with their geographical distributions.
